Abstract
Description
The invention relates to the field of ecology and environmental management and can be used to clean soils in the territories of urbanized and industrial agglomerations, as well as agricultural soils, in order to reduce the content of toxic concentrations of heavy metals received from human activities.
The problem of cleaning and rehabilitation of soils of territories for various purposes, contaminated with heavy metals, is currently extremely urgent in the field of environmental management and restoration. However, today there are no unambiguous approaches to its solution. Among the known methods for cleaning soils from heavy metals, mechanical, physicochemical, chemical and biological techniques are described. The mechanical method of soil cleaning is based on the removal of the contaminated soil layer to a depth of 0.5 m and its replacement with ecologically clean soil selected from unpolluted areas [1, 2]. The disadvantage of this method is the high cost of work, which is estimated at up to 2.5 million euros per 1 ha. Physicochemical methods for cleaning soils from heavy metals are based on the use of sorption, desorption, ion exchange processes and are carried out by adding adsorbents, ameliorants, and zeolites to the soil [3, 4, 5]. Chemical methods are based on the use of lime and phosphorus fertilizers for converting metals into sparingly soluble compounds, leaching of soils, extraction of metal salts with solvents, leaching processes, addition of organic substances and metal binding to complex compounds, etc. [6, 7]. The disadvantages of these methods include their high cost, complexity in execution, the temporary nature of the effect, the occurrence of undesirable side effects, including the negative impact on the physicochemical parameters of soils, the creation of deposition media that determine delayed negative effects, and the risk of secondary environmental pollution. Biological methods for cleaning soils from heavy metals are based on the use of plants, namely, their ability to absorb and accumulate metals in the roots and aboveground parts of plants. The common advantages of biological soil treatment are the relatively low cost of work that does not violate soil compaction, the possibility of in situ cleaning, and the safety of the work for the environment.
Of the known methods of biological cleaning of soils from heavy metals, the most relevant and close to the claimed invention are methods using phytoremediation technology [8-11], which involves the extraction of metals from contaminated soils by the roots of plants planted on them, followed by the transfer and accumulation of metals in the aerial parts of plants, to be disposed of. At the same time, there is no single approach to the choice of plants for biological soil treatment and its methodology
There is a phytoremediation method for cleaning soils of heavy metals, based on the use of plants - hyperaccumulators, which specifically accumulate significant amounts of heavy metals in the aerial parts of these plants without pronounced signs of phytotoxicity, for example, the thrush Thlaspi caerulescens [12], alissum Alyssum sp. [13] or undersized integumentary grass of Arthraxon bristly (Arthraxon hispidus) (Thunb.) Makino [14]. The disadvantage of this method is the low growth rate and low biomass of hyperaccumulative plants, their narrow specialization in hyperaccumulation of only 2-3 heavy metals, in particular, Cd and Zn by yarut plants [12], Ni and Co alissum plants [13] and Zn , Pb, As plants of arthraxon [14], as well as the growth of hyperaccumulators within climatic zones unusual for Russia.
Also known is a phytoremediation method of soil cleaning based on the use of agricultural plants such as corn and wheat [15] or safflower [16]. The disadvantage of this method is the unacceptability of its use for cleaning contaminated soils in a metropolis and climatic restrictions.
Known phytoremediation method of cleaning soil, based on the use of ornamental plants, such as marigolds (French marigold, Tagetes) [17, 18], as well as Balsamine balsamine) and fern nephrolepis (Nephrolepis auriculata) [19] for cleaning soils contaminated with cadmium [17] , 18], as well as copper and copper-containing materials [19]. The disadvantage of this method is that these plants have a small biomass and are designed to extract only one type of metal.
There is a phytoremediation method for cleaning soils based on the use of genetically modified plants or transgenes with increased metal-accumulating ability, for example, transgenic petunia [20] or genetically modified mustard of Sarepta Brassica juncea [21] for the extraction of several metals - Cr, Mo, Cd , U. The disadvantage of this method is the use of the gene of modified plants prohibited for cultivation in the Russian Federation.
There is also a method of phytoremediation cleaning of soils contaminated with heavy metals, using some wild plants exhibiting the ability to accumulate metals, in particular, dandelion medicinal Taraxacum officinale Wigg. [22]. The disadvantage of this method is the low efficiency of soil cleaning from heavy metals and significant labor costs.
Of the described phytoremediation methods of soil cleaning, the closest in technical essence and technical result achieved to the claimed invention is a method based on seeding on soils contaminated with heavy metals, seeds of several species of wild plants from the families of Asteraceae, legumes and cereals, followed by repeated mowing them at the stage the growing season, drying and removal from the surface of the soil [23], selected as a prototype.
The disadvantage of this method, selected as a prototype, is the relatively low accumulative ability of the aerial parts of the sown plants with respect to their accumulation of heavy metals such as zinc, copper, manganese and chromium, as well as the limited use thereof for cleaning urban areas due to the presence in the recommended herbal mixture malicious weed of an artisan field, giving a large number of seeds, the spread of which can lead to rapid clogging of adjacent territories.
The claimed invention is free from the above disadvantages.
The technical result of the claimed invention is to increase the amount of heavy metals extracted from contaminated soils, as well as their composition.
The specified technical result is achieved by the fact that when seeding wild plant species typical of the local area, typical and dominant for a given local region, they determine the metal-accumulating ability in terms of heavy metal concentrations and their transfer coefficients in the system “soil-roots-aerial parts of plants »At levels of contamination of specific soils, as well as their indicator identification for the purpose of their further use as phytoremeditors.
The basis of the claimed invention is the concept of providing biological cleaning of soils and soils from heavy metals due to a complex of biogeochemical processes taking place in the system “rhizosphere - roots - aerial parts of plants” and including 2 successive stages - 1) absorption of heavy metals from the soil and their binding to the root system and 2) the transfer of metals from the roots to the shoot and their accumulation in the aerial parts of plants. At the same time, the rhizosphere refers to the part of the soil adjacent to the roots of the plant and directly affected by root secretions and soil microorganisms, and contributing to the migration activity of heavy metals. Evaluation, along with the metal-accumulating ability of the aerial parts of plants, of the metal-accumulating ability of their roots and the establishment of metal transfer coefficients in the “rhizosphere – roots – aerial parts of plants” system will allow for the selection of indicator species for phytoremediation to take into account the contribution of the plant root system to the biosorption of heavy metals, extracted from soil and soil. At the same time, in the rhizosphere during seasonal decay of roots remaining in the soil after mowing of the aerial mass, metals immobilized by the roots transform into more bioavailable forms, which is a factor contributing to their more active absorption and transfer to the aerial part of plants upon repeated sowing of plants for phytoremediation purposes.
The essence of the invention is based on genetically determined molecular mechanisms of absorption and upward transport of ionic and chelated forms of heavy metals in plants that function in different plant species with different efficiencies, as a result of which pollutant metals are absorbed and bound by roots and transferred to the aerial part with different intensities different types of plants
Ni, Cr.The use of such a mechanism can significantly increase both the amount of absorbed heavy metals and their composition. This result can be achieved due to the spread of naturally growing indicator species of metal-accumulating plants characteristic of this local region, as well as more effective exposure of indicator vegetation to soils and the removal of a wide range of polluting heavy metals from them, including Zn, Cu, Pb, Mn, Co, Ni, Cr.
In addition, the above technical result is achieved by the fact that in the claimed phytoremediation method for cleaning soils contaminated with heavy metals, contaminated soils are sown with plant species native to the area and having the maximum rates of extraction and removal of polluting metals characteristic of this local region to the aboveground biomass.
To achieve the specified technical result in accordance with the claimed invention, in the area to be cleaned of heavy metals, a site with a size of at least 100 sq. M is selected, the concentration of heavy metals is determined in the soil of this site, naturally growing and dominant plant species are distinguished on it, after which they the roots and aboveground parts determine the accumulating ability by the concentrations of heavy metals and their transfer coefficients in the system "soil-roots-aboveground part of plants", select indicator f plant species based on the accumulating capacity obtained by the highest indicators of heavy metal accumulation and the maximum values of their transfer coefficients to the aerial parts of plants, after which they are selected as phytoremeditors by these indicators

As shown in FIG. 1, metals entering plants from contaminated soil are distributed unevenly in them and accumulate mainly in the roots. It has also been shown that selected plants accumulate polluting metals with different intensities.
As shown in FIG. 2, the efficiency of metal extraction and transfer, measured in the soil-root system, differs in selected plant species and in clover more than in mother and stepmother, and is minimal in dandelion.
As can be seen from FIG. 3, the transfer coefficients of metals from the soil to the aerial part of plants vary within 0.3-0.8 and are highest for clover for Mn, Zn, Cu, Pb and Ni, and for mother and stepmother for Cr and Co.
As shown in FIG. 4, the metal transfer coefficients in the root – aerial part of the plant system vary widely from 0.04 to 1.47 and for some metals (Mn, Zn, Cu, Ni) are much higher for dandelion, and for others (Pb, Cr and Co) - in dandelion and mother and stepmother.

Table 1 illustrates the results obtained by the claimed method of evaluating the content of heavy metals in the soils of the studied territories, mg / kg

The essence of the claimed phytoremediation method for cleaning soils contaminated with heavy metals is that cleaning takes place directly on site and is carried out using plant species native to the area and selected as indicators for their intensive distribution on the entire surface of the cleaned soils of the local region. In the process of plant growth and vegetation, polluting heavy metals are absorbed by the root system in amounts that depend on their concentration in the soil and on the species characteristics of plants, are bound by roots and transferred to the aerial part of plants with transfer coefficients inherent in the selected species of indicator plants. Seasonal decay of the roots remaining in the soil transforms the metals bound by them into more bioavailable forms, which is a factor contributing to a more efficient assimilation and transfer of metals to the aboveground parts of plants during re-sowing of plants.
In the territory to be cleaned of heavy metals, the sown plants are brought to the stage of the growing season and mowing and collecting green mass. This procedure is repeated periodically as the aerial parts of plants grow. The higher the concentration of polluting metals in the soil in concentration, the greater the number of mowing green masses that need to be carried out, which, in turn, increases the efficiency of soil cleaning from pollutants. As a result, the present invention reduces labor costs and reduces the cost of activities carried out in contaminated areas, especially in a large area.
The claimed method was tested on the urbanized territory of the Leningrad region on soils contaminated by Zn, Cu, Pb, Cr, Mn, Ni, Co. The claimed method is carried out in 5 stages.
At the first stage, selection and preparation for analysis of soil samples in the territory to be cleaned from pollution with heavy metals is carried out.
In the sampling procedure, the main parameter is the representativeness of the sample, i.e. its correspondence to the structure and composition of the studied object. When assessing total pollution, test sites with a size of at least 100 m 2 are marked on the grid, indicating their numbers and coordinates. Five spot samples are taken from each site by type of envelope. The selection is carried out at a depth of 0-20 cm. The combined sample, consisting of a mixture of samples, should have a mass of at least 1 kg. Samples are taken with a shovel and placed in bags made of polyethylene or fabric. Samples are numbered and recorded in the journal, indicating the location, depth of sampling, type of soil or soil, type of contamination and date of sampling. Then the samples freed from stones and plant roots are scattered evenly on a flat surface and dried in the dark at room temperature to an air-dry state, after which they are sieved through a sieve with a hole diameter of 1 mm and packaged in thick paper bags. The collected samples are stored in a chilled or frozen state (to -20 ° C). To determine the gross content of heavy metals in the selected soil samples by chemical analytical analysis (for example, atomic emission spectroscopy) they are transferred to a chemical analytical laboratory.
At the second stage, selection and preparation for analysis of samples of plant species that naturally grow and dominate the territory to be cleaned is carried out.
Sampling of plants is carried out at the same test sites as the selection of soils and soils. Samples of plant species growing at all test sites are selected, which allows us to consider these species as predominant and representative for this contaminated region. When taking plant samples, samples of roots and aerial parts of plants are taken separately. Of the point samples, a combined sample weighing 1 -1.5 kg is made up. Root samples are freed from soil particles, all samples are placed in plastic bags and delivered to the laboratory.
In the laboratory, raw plant samples are removed from the bags and washed thoroughly with water. In the presence of adhering particles on the surface of plants, they are washed first with tap water and then with distilled water. Uncontaminated plants are washed only with distilled water. Plant samples are slightly blotted with filter paper (without squeezing so as not to squeeze out the contents of the cells) and placed in a dry room on sheets of filter paper for drying. At a temperature of 20 ° C and low humidity, the samples dry in 2-3 days. With high humidity and low room temperature, additional dry air sampling is recommended. If there are special drying cabinets in the laboratory, the samples are placed in the cabinet, placing them in cuvettes (not metal) and left at a temperature of + 60 ° С for 12-24 hours until complete drying. The dried plant material is ground to a powder state using special electric grinders or ground manually in agate or porcelain mortars.
The next step in the preparation of plant samples for analysis is to take samples and transfer the samples to solution, which is necessary to determine the metal content using chemical analytical methods. This is achieved by ashing plant material, i.e. releasing it from the organic matrix. Modern methods for analyzing the content of chemical elements make it possible to use either dry plant ash or solutions obtained either by dissolving the ash residue in acids or by wet ashing of plant material using concentrated acids for analysis.
At the third stage, the determination of the concentrations of heavy metals in the selected plant samples is carried out, and the coefficients of their transfer in the system "soil-roots-aerial parts of plants"
At this stage of the action using the certified method of atomic absorption spectrophotometry, the concentration of heavy metals in the roots and in the aerial parts of plants is established for each of the plant species selected on the studied area. Next, an assessment is made of the coefficients of metal accumulation in the roots and aerial parts of selected plant species relative to the concentrations of the corresponding metals in the studied soils and the coefficients of their transfer from the roots to the aerial parts of the plants are determined.
At the fourth stage, indicator species of plants are selected for their further distribution with the aim of phytoremediation cleaning of the contaminated area.
Based on the results of determining the accumulative capacity of dominant plant species by the highest indicators of heavy metal accumulation in them and the maximum values of their transfer coefficients to the aerial parts of plants, indicator species of plants are established and selected as phytoremeditors for intensive distribution in the contaminated area.
At the fifth stage, the seeds of indicator species of plants selected as phytoremediators are sown, brought to the stage of the growing season, mowing and collecting green mass.
The sowing of seeds of indicator plants for the purpose of phytoremediation of soils contaminated with heavy metals is carried out in the spring. This period is convenient in that the moisture reserve in the soil provides an optimal water regime for the entry of heavy metals from the soil into the roots of plants and transfer to the aerial parts of plants. Next, the plants are brought to the stage of the growing season and mowing and collecting green mass. This procedure is repeated periodically as the aerial parts of plants grow.
The results of testing the inventive method are implemented as specific examples.
The following are examples of specific testing of the claimed phytoremediation method for cleaning soils contaminated with heavy metals.
1.Example 1
С этой целью полученные вытяжки фильтровали и проводили определение тяжелых металлов в фильтратах методом атомно-эмиссионного анализа с индуктивно-связанной плазмой на ICPE-9000 (Shimadzu, Япония).The purpose of the experiment: to establish the concentration of heavy metals (Mn, Zn, Cu, Cr, Pb, Co, Ni) in soils on the urbanized territory of the Leningrad region contaminated with heavy metals, and to establish the accumulation indicators of these metals in the roots and aboveground parts of plants growing on this territory. To do this, in the study area, a plot of 100 m 2 was selected, on which 3 experimental reference plots covered with grassy vegetation with an area of 1 m 2 each were marked. At each experimental site, soil samples were taken by an envelope method to a depth of 20 cm. Averaged soil samples weighing 50 g were dried in an oven for 5 hours at 105 ° C. Then 10 g of air-dry soil, crushed and passed through a sieve with 2 mm holes, was subjected to chemical decomposition by heating using 50 cm 3 HNO 3 (1: 1) to determine the gross content of heavy metals in soil samples. To this end, the obtained extracts were filtered and heavy metals in the filtrates were determined by the method of atomic emission analysis with inductively coupled plasma on ICPE-9000 (Shimadzu, Japan).
Содержание тяжелых металлов в пробах, полученных в результате озоления, определяли методом атомно-эмиссионного анализа с индуктивно-связанной плазмой на ICPE-9000 (Shimadzu, Япония).During the experiment, 3 species of naturally growing and dominant plants were isolated in experimental plots - mother and stepmother Tussilago fdrfara, medicinal dandelion Taraxacum officinale and clover meadow Trifolium pratense. Samples of the roots and aerial parts of these plants were taken in conjunction with soil samples in the same metering plots of 1 m each for chemical analysis. Plants were completely extracted from the soil, they were divided into the aerial part and the roots, which were carefully washed from the soil with tap water and then with distilled water. Samples of the roots and aerial parts of plants of each species weighing up to 300-500 g of wet weight were recorded in a thermostat at 105 ° C for 1 hour and then dried at 70 ° C for 24 hours. The dried samples were ground to a powder state, samples of 0.5 g mass were taken from them, which were wet-ashed with a mixture of concentrated acids HNO 3 : HClO 4 in a volume ratio of 4: 1 at 160 ° C. The content of heavy metals in the samples obtained as a result of ashing was determined by the method of atomic emission analysis with inductively coupled plasma on an ICPE-9000 (Shimadzu, Japan).
Based on the analysis of soil samples taken from the experimental plots, the concentrations of heavy metals were established, the average values of which are shown in Table 1. From Table 1 it follows that the soil in the experimental plots of the urbanized area selected for the experiment is contaminated for a number of heavy metals. Moreover, the excess in it of gross concentrations of metals such as Pb, Zn and Cu relative to the regulated MPC values was 7.87, respectively; 3.67 and 1.72 times.
The results of the analysis of the content of heavy metals in plant samples taken in experimental plots are presented in FIG. 1, where it is shown that metals entering plants from contaminated soil are distributed unevenly in them and accumulate mainly in the roots. It has also been shown that selected plants accumulate polluting metals with different intensities. The metal content in the root biomass generally corresponded to the general order: Mn> Zn> Cu> Cr> Pb> Ni> Co. At the same time, the concentrations of Mn, Zn, Cr, Pb, Co, and Ni in the roots of clover were noticeably higher than in the roots of other plant species, while the accumulation of Cu was equally high in the roots of clover and mother and stepmother. In the aboveground biomass, the highest concentrations of Zn, Cu, and Pb were found in clover leaves (184; 71 and 37 mg / kg dry biomass, respectively), and the maximum concentration of Cr (135 mg / kg) was found in the leaves of mother and stepmother. Differences in the levels of accumulation of individual metals in the roots and in the aboveground biomass were more pronounced in clover and less in dandelion.
2.Example 2
4.The purpose of the experiment is to establish the conformity of naturally occurring and dominant in the selected area of the contaminated territory plant species to the requirements for plants used for phytoremediation. As an evaluation criterion for this correspondence, coefficients were calculated that characterize the efficiency of the movement and accumulation of metals in the integrated system "soil - roots - aerial parts of plants". The coefficients were calculated based on the results of determining the concentrations of heavy metals in the soil (mg / kg) and in the roots and aboveground parts of plants (mg / kg of dry biomass), which were used to plot the graphs shown in FIG. 2 - FIG. four.
In FIG. Figure 2 shows that the efficiency of metal extraction and transfer, measured in the soil – root system, varies among selected plant species and decreases in the order clover – mother and step-mother – dandelion.
As the calculation results show, the transfer coefficients of most metals in the soil-root system in clover and mother and stepmother plants are significantly higher than 1.0, reaching 3.6 and 3.5 for clover in clover and mother and stepmother 3.5 and 2.3, respectively, which indicates the high ability of these plants, naturally growing on contaminated soils, to extract and accumulate heavy metals at the root level. The transfer coefficients of other metals from the soil to the roots are also higher for clover and in relation to Mn, Zn, Co, Ni are from 1.5 to 2.5. The lowest indices of plant root extractability are shown for Pb, which is consistent with the well-known concept of low bioavailability of this metal.
In FIG. Figure 3 shows that the majority of heavy metals polluting soils in the studied urbanized area accumulate in the aerial parts of naturally growing and dominating plant species with different intensities, and their transport coefficients in the soil – aboveground system vary within 0.3 -0.8. The efficiency of moving a number of metals (Zn, Cu, Pb, Ni) from the soil to the aerial parts of plants is higher for clover, with respect to Cr and Co the highest coefficients of their transfer from the soil to the aerial parts of plants are shown for mother and stepmother. The transfer coefficients of Pb and Ni from the soil to the aerial part of the plants turned out to be lower than other metals.
In FIG. Figure 4 shows that the transfer coefficients of metals in the root – aerial part of the plant system vary widely from 0.04 to 1.47 and, in contrast to the soil – root transfer coefficients for one metal (Mn, Zn, Cu, Ni ) they are much higher in dandelion, and in others (Pb, Cr and Co) - in dandelion and mother and stepmother.
3.In general, based on the assessment of metal transport coefficients in the soil – root – aboveground part of plants system, we can conclude that the soil – root and soil – above ground part transport coefficients are of great indicator significance, and for the type of pollution we studied soil heavy metals clover plants have the greatest potential for phytoremediation method of purification. Example 3
79% [23].The purpose of the experiment is to establish the potential metal-accumulating ability of indicator plant species naturally growing in contaminated areas. The experiment consisted in calculating the accumulation of heavy metals in the aerial parts of plants, expressed in kg per 1 sq km of contaminated territory, using the studied territory as an example, based on experimentally established metal contents in the aerial parts of indicator plant species and taking into account the minimum and maximum yields available in the literature green biomass of herbaceous plants, namely, from 600 to 1000 centners per ha with a water content of 79% [23].
Mn, Zn, Cu, Cr.As the results of testing showed, at a specified level of soil contamination with heavy metals, clover plants with a maximum yield of green biomass of 1000 kg / ha can extract and accumulate in the aboveground part in the amount of up to 1509.4 kg of metals, coltsfoot plants - up to 1461.6 kg, and dandelion plants - up to 1291.5 kg of metals from a contaminated area of 1 sq km, which is shown in Table 2, which shows that with the studied soil pollution levels the highest potential bioaccumulation of metals in the aboveground parts indicator plant species have been identified for Mn, Zn, Cu, Cr.
As numerous examples of testing have shown, the experimental results confirm the indicated technical result of increasing both the amount of heavy metals extracted from contaminated soils and their composition when seeding wild plant species typical of a given local region inherent in a given area. Thus, plants naturally growing and dominating in soils contaminated with heavy metals have a high potential for their use for phytoremediation cleaning of these soils from polluting metals. The choice of plants for further sowing should be determined on the basis of establishing indicators of the accumulation of heavy metals in the aerial parts of plants and their transfer coefficients in the system “soil-roots-aerial parts of plants” according to the maximum values of these indicators.
