Abstract
Description
The invention relates to a method for obtaining and processing images of optical interferometry (optical coherence tomography) in order to isolate and visualize image areas containing micromotion. The invention can be used for intravital imaging and quantification of the bloodstream of biological tissues, including human.
For the purpose of diagnosing a number of diseases, including oncological, as well as evaluating, including early, the effectiveness of certain types of therapy, information on the degree of development of the bloodstream in the target area plays an important role. Using the information of fluorescence imaging, or X-ray angiography, or speckle-contrast registration allows visualization of a two-dimensional map of microvessels, however, the construction of a three-dimensional model of microcirculation by these methods seems difficult. The technology of optical interferometry (optical coherence tomography, OCT) makes it possible to obtain intravital three-dimensional images of the structure of biological tissue, which makes it possible to extract three-dimensional information about microcirculation in biological tissue.
There is a method of extracting and visualizing information about micromotion (patent US 8180134, B2 IPC7 G06K 9/00, publ. 05/15/2012), in which information about the movement is highlighted by using the effect of degeneration of images of optical interferometry by the sign of the delay that occurs under complex registration conditions the amplitudes of the probe wave in the presence of a Doppler frequency shift during scattering by moving scatterers.
The disadvantage of this method is a twofold decrease in the effective depth of the image and the overall computational complexity of the processing algorithms.
Joey Enfield, Enock Jonathan and Martin Leahy. «In vivo imaging of the microcirculation of the volar forearm using correlation mapping optical coherence tomography (cmOCT)» Joey Enfield, Enock Jonathan and Martin Leahy (Biomedical optics express, V. 2, I. 5, pp. 1184-1193)»).The closest analogue of the developed method is a method for visualizing areas of an object containing micromotion, including registering a set of interferograms, in which scanning with a probe beam is carried out sequentially in the main and perpendicular to the main directions, recalculating interferograms into a set of two-dimensional images, calculating the correlation angiographic image, performing a binarization procedure with assignment image element value "1", if the value of this element exceeds the value set as the noise level, and the value “0” if this condition is not met, and the visualization of the correlation angiographic image obtained from the binary data array in the form of color or brightness amplitudes (see Joey Enfield, Enock Jonathan and Martin Leahy. “In vivo imaging of the microcirculation of the volar forearm using correlation mapping optical coherence tomography (cmOCT) »Joey Enfield, Enock Jonathan and Martin Leahy (Biomedical optics express, V. 2, I. 5, pp. 1184-1193) ").
In the known method, at least two sets of optical spectra of the sum of interfering waves corresponding to one transverse cut are recorded. In this case, when registering each set of interferograms corresponding to one transverse cut, the probe beam is displaced along the main direction, between the registration of two consecutive sets of interferograms, the probe beam is shifted in the direction perpendicular to the main one by no more than 0.2 probing beam diameters. Interferograms are converted to a set of two-dimensional images using the Fourier transform. A correlation analysis of at least two sequentially obtained two-dimensional images is carried out. It is assumed that the presence of decorrelation (non-zero elements of the correlation image) is due to dynamic processes occurring in the object. Binarization of a two-dimensional image is carried out: the image element is assigned the value “1” if the value of this element exceeds the value set as the noise level, and the value “0” if this condition is not met. The binarized two-dimensional image and the correlation image are multiplied. The resulting correlation angiographic image is visualized in the form of color or brightness amplitudes.
However, the images obtained by implementing this approach turn out to be sensitive to the presence of macroscopic movements of both the object of study and the body of the probe scanner, which is especially important when implementing contactless image registration (in medical applications, the mutual micro movements of the optical elements of the probe scanner and the object under study, physiologically determined movements of individual structures in the studied object, not related to the presence of blood flow). The sensitivity of the approach to the presence of macroscopic motions increases with increasing time between registration of successively obtained two-dimensional images, which ultimately leads to the necessity of realizing a high scanning speed in the main direction, otherwise the time interval between registration of two successively obtained sets of interferograms can be large enough to occur significant decorrelation of two-dimensional images due to external causes.
The technical effect to which the invention is directed is the development of a method for visualizing areas of an object containing micromotion, which improves the accuracy and objectivity of the data obtained.
The specified technical result is achieved by the fact that in the known method of visualizing areas of an object containing micromotion, including registering a set of interferograms, while scanning with a probe beam is carried out sequentially in the main and perpendicular to the main directions, the interferograms are converted into a set of two-dimensional images, calculation of the correlation angiographic image, binarization procedures with assigning a value of “1” to an image element if the value of this element exceeds the value set as the noise level, and the value "0" if this condition is not met; visualization of the correlation angiographic image obtained from the binary data array in the form of a color or brightness amplitude, scanning along the main direction is carried out with a step dx corresponding to the probe beam diameter D as dx <0.25 D, the calculation of the correlation angiographic image is carried out as follows: two-dimensional structural image of the OCT subjected to spectral transformation along the x coordinate, the resulting distribution is multiplied by the window function of

The present invention allows, when used, to obtain the following technical effect - improving the accuracy and objectivity of the obtained data when visualizing areas of the object containing micromotion.
The proposed method is as follows.
An optical interferometry image is recorded, for which the string image elements of the optical interferometry are sequentially recorded and the position of the probe beam on the surface of the object of study is measured along at least one of the two scanning directions - the main and perpendicular to it. To register string image elements, probing optical radiation is directed to the object of study; the optical radiation scattered in the opposite direction on the structural elements of the object of study is recorded interferometrically; the received information is transmitted for storage and processing on a computer. Two- and three-dimensional images of optical interferometry are obtained by a combination of string image elements in accordance with the scanning algorithm.
Scanning along the main direction is performed with a step dx corresponding to the diameter of the probe beam D as dx <0.25 D, i.e. scanning is carried out with a step much smaller than the diameter of the probe beam, which ensures the non-isotropic speckle structure constituting the image (Fig. 1) - the elements become elongated along the main direction. Moreover, due to the fact that individual lines are recorded at different points in time, the presence of dynamics in separate areas of the object (indicated by a dotted line in Fig. 1) leads to an observed change in the transverse scale of individual image elements.
2,б).The spatial spectrum in the main direction of the stationary regions of the image shown in FIG. 1 is limited due to the smallness of the scanning step and the associated elongation of image elements. In the region of the image in which dynamics is present, the spatial spectrum in the main direction experiences broadening (Fig. 2a), which is proportional in the general case to the speed of the scatterers in the object in the direction of the axis of the probe beam. At high speeds v (at which, in the interval between sequential recording of two interferograms t, the condition ) or transverse to its direction, the spatial spectrum in the main direction becomes unlimited under the registration conditions (Fig. 2, b).
The broadening of the spatial spectrum in the main direction in the areas of the image containing motion allows the calculation of the correlation angiographic image. To do this, spectral conversion of a two-dimensional image is carried out along the main direction, after which a two-dimensional image is filtered by multiplying the obtained spectral distribution and the window function of the notch filter, the width of which is determined in the general case by the width of the spectral distribution obtained for the stationary part of the studied object (Fig. 3). Since the broadening value of the elements of the speckle image structure is determined by the scan step size and the properties of the probe radiation source used, the procedure for determining the width of the notch filter can be performed once on a model object.
4).The obtained two-dimensional spectral distribution is subjected to the inverse Fourier transform procedure in the main direction, as a result of which only regions in which phase displacements due to the presence of motion led to a broadening of the spatial spectrum in the main direction are preserved in the image (Fig. 4).
5).Visualization of the angiographic correlation image can be carried out both separately (as shown in Fig. 4), and on a two-dimensional image by coloring or highlighting the area of the two-dimensional image that coincides with the area of the presence of movement (as shown in Fig. 5).
An example of a specific implementation of the method of visualizing areas of an object containing micromotion
BalbC.For a specific implementation of the method of visualizing areas of an object containing micromotion, an object was chosen - the ear of the mouse of the BalbC line.
Image of optical interferometry was recorded, for which probing radiation was directed to the object of study, sequential registration of string image elements of optical interferometry was performed when the position of the probe beam on the surface of the object of study along the main and perpendicular to it was changed. Scanning along the main direction was carried out with a step dx much smaller than the probe beam diameter D (dx <0.15 D). The obtained set of interferograms was subjected to Fourier transform in the direction of the axis of the probe beam, as a result of which two-dimensional images of the object under study were obtained. The resulting images were subjected to Fourier transform in the main direction. The obtained images were subjected to a filtration procedure by multiplying the obtained spectral distribution and the window function of the notch filter. The obtained two-dimensional spectral distributions were subjected to the inverse Fourier transform in the main direction, as a result of which only the areas in which the presence of micromotions were noted on the obtained angiographic image were preserved (Fig. 6.c). The resulting areas correspond to the position of the vessels in the test volume. After binarization of the obtained angiographic image and color coding of the structural image elements, the mutual arrangement of the vessels and the structure of the object under study was compared with its direct photographic image (Fig. 6.a) and the image obtained using fluorescence microscopy (Axiom Zoom.V16 steriomroscope (Zeiss, Germany) with a FITC fluorophore conjugated to 150 kD dextran (Sigma, USA). A comparison showed that the structural elements shown in the image obtained using Imaging method for an object area containing micromotion correspond grid blood vessels in the object under study.
