Abstract
Description
The invention relates to compositions based on siloxane rubber, having a wide range of applications and intended primarily for the manufacture of medical devices in contact with blood and biosubstrates. In addition, the antimicrobial composition can be used for the manufacture of products (elements, parts) intended for installation in devices for storing and / or pumping liquid media in which it is necessary to prevent the propagation of unwanted microflora for a long time.
1975].A known composition based on silicone rubber, including organic peroxides as a vulcanizing agent [Schetz M. Silicone rubber. - Leningrad: Chemistry. 1975].
The specified composition is widely used to obtain a variety of medical devices with an implantation period of up to 1 month due to insufficient biocompatibility with body tissues.
Compositions based on siloxane vulcanized rubbers using silica hydrides with crosslinking activators (Pt compounds) as higher crosslinking agents have higher medical and technical parameters. The vulcanization process proceeds without the release of toxic products, unlike peroxides, and the resulting vulcanizates have a high level of medical and technical indicators.
The use of such compositions allows you to produce products intended for long-term implantation - for a period of more than six months.
A significant drawback of compositions containing catalysts based on platinum compounds is their high activity in the crosslinking process even at room temperature. This dramatically reduces the shelf life of such mixtures, worsens their rheological properties, which creates difficulties in the processing of rubber compounds.
Resistance to premature vulcanization of such compositions is achieved, as a rule, by introducing special compounds (inhibitors) into the composition of the mixtures, which slow down crosslinking at the processing temperature of the rubber mixture, but have a minimal effect on the speed of the process at the vulcanization temperature.
Т. Method of preparing a platinum catalyst inhibitor: пат. [Preparation of stable mixtures of organosilicon compositions in the presence of a nitrogen-containing ligand: пат. [Preparation of stable copolymerizable organosilicon compositions containing a platinum catalyst and a phosphorus ligand: пат. [Garden W.D. Catalyst compositions and their use in the preparation of release coatings: пат. - 1981].So known compositions containing acetylene alcohol as inhibitors [Bergstrom F.A., Lee C.L., Maxson M.T. Method of preparing a platinum catalyst inhibitor: US Pat. 3933880 United States. - 1976], nitrogen-containing compounds [Preparation of stable mixtures of organosilicon compositions in the presence of a nitrogen-containing ligand: US Pat. 3,188,299 United States. - 1965], organophosphorus compounds [Preparation of stable copolymerizable organosilicon compositions containing a platinum catalyst and a phosphorus ligand: US Pat. 3,188,300 US. - 1965.], amines [Garden W.D. Catalyst compositions and their use in the preparation of release coatings: US Pat. 4,281,093 - 1981].
Although it is possible to significantly slow down the process of cross-linking the compositions at room temperature, the development of infection is a frequent complication associated with the use of various medical devices made from these compositions — catheters, endotracheal tubes, and various implants.
Given that catheter-associated infections are based on the formation of a biofilm on the surface of a foreign body - a catheter, giving it antimicrobial properties can be a reliable way to protect against infection. The presence of antimicrobial agents on the surface of the catheter for a long time creates a zone of antimicrobial action in the surrounding tissues and acts as a system of a local source that continuously releases antimicrobial agents. In this case, microorganisms entering the catheter, even if they do not die, then lose the ability to reproduce and form a film.
[Suh K.I. Mixture of silicon oxide, aluminum oxide and silver or silver oxide added to silicone or latex: пат. - 1995].A known composition based on siloxane rubber containing, as an antimicrobial agent, a mixture of oxides in an amount of 0.2-1.0 mass parts. the following composition: 40-79% SiO 2 , 20-50% Al 2 O 3 , 1-10% silver or its oxide [Suh KI Mixture of silicon oxide, aluminum oxide and silver or silver oxide added to silicone or latex: US Pat. 5476881 United States. - 1995].
- 1995].Also known is a composition based on siloxane rubber, comprising as an antibacterial agent a zeolite containing from 0.1 to 15% silver ion [Inoue Y., Susumu S., Kurihara Y. Antibacterial, antifungal silicone rubber composition: US Pat. 5466726 United States. - 1995].
A significant drawback of these technical solutions is that the resulting compositions do not provide the required degree of antimicrobial efficacy, since these compositions are characterized by a slight diffusion of silver ions from rubber.
Anti-microbial silicone rubber composition and method for making same: пат. - 2004].To increase the rate of release of silver ions from rubber, it is proposed to introduce dispersed in an organic liquid (aliphatic or aromatic hydrocarbon, mineral oil, petrolatum, glycerin, fatty alcohol, propylene glycol, polypropylene glycol, animal and / or vegetable oil, or fat or silicone into a composition based on siloxane rubber oil) nanosilver particles (size from 1 to 50 nm) [Hanke B., Bast T. Anti-microbial silicone rubber composition and method for making same: US Pat. 6822034 United States. - 2004].
- 1982].The introduction of monocarboxylic acids into silicone compositions can increase the duration of antimicrobial activity from several days to several weeks [Mustacich R.V., Lucas D.S., Stone R.L. Antimicrobial polymer compositions: US Pat. 4,343,788 to the United States. - 1982].
However, like the previous technical solutions, this composition does not have a pronounced prolonged antibacterial effect both on the entire surface of the resulting products and in its volume.
[Sankar G.G. et al. Polydimethyl siloxane based nanocomposites with antibiofilm properties for biomedical applications //Journal of Biomedical Materials Research Part B: Applied Biomaterials. - 2016].Of the other organometallic antimicrobial agents introduced into siloxane rubbers, copper oxide modified with cetyltrimethylammonium bromide is known [Sankar G.G. et al. Polydimethyl siloxane based nanocomposites with antibiofilm properties for biomedical applications // Journal of Biomedical Materials Research Part B: Applied Biomaterials. - 2016].
The disadvantages of all of these compositions include the insufficient duration of the antimicrobial effect, which limits their use in medical devices with a long implantation period.
[Nelson J.N. Antimicrobial elastomer composition and method for making: пат. - 2008].Closest to the proposed technical solution is a rubber mixture based on polydimethylmethylvinylsiloxane rubber, including aerosil, anti-structuring additive, silica, platinum catalyst and silver oxide dispersion as an antimicrobial agent [Nelson J.N. Antimicrobial elastomer composition and method for making: US Pat. 7393547 United States. - 2008].
Silicone rubber exhibiting effective antimicrobial activity: заяв. - 2008].A significant drawback of such compositions is the low diffusion rate of silver ions from rubber due to their adsorption on the surface of the filler, which does not provide the necessary degree of protection of products from infection [Zhu A., Wang Y.F. Silicone rubber exhibiting effective antimicrobial activity: application. US Pat. 12/072, 486 United States. - 2008].
The problem solved by the invention is to eliminate the above disadvantages of analogues, including the closest analogue, namely improving the antimicrobial effect and technological properties.
The technical result, which is achieved by the claimed invention, is the creation of a biologically active composition based on silicone rubber with a wide range of applications, with bio- and hemocompatibility, optimal technological and physico-mechanical properties and a long-term antimicrobial effect.
To date, a number of methods are known for imparting antimicrobial properties to medical devices made from siloxane rubber using rifampicin.
- 2013].So known is the method of introducing rifampicin into a liquid non-vulcanized silicone compound with its subsequent application (spraying) on the finished product with further vulcanization [Berg E.P., Stella R. Coating with antimicrobial agents: US Pat. 8419793 United States. - 2013].
The main disadvantage of this method is the impossibility of its use for medical devices, where the key factor is the accuracy of geometric shapes and sizes, as well as the formation of a uniform film thickness on the surface of the product.
Известен также способ введения рифампицина в готовые изделия на основе силоксанового каучука путем их обработки раствором рифампицина в неполярных растворителях [Darouiche R.О., Raad I. Antimicrobial impregnated catheters and other medical implants: пат.5902283 США. - 1999]. There is also a method of introducing rifampicin into finished products based on siloxane rubber by treating them with a solution of rifampicin in non-polar solvents [Darouiche R. O., Raad I. Antimicrobial impregnated catheters and other medical implants: US Pat. No. 5,952,283. - 1999]. After solvent removal, rifampicin remains in the finished product.
A significant drawback of this technical solution is that it does not provide the desired degree of effectiveness, since it does not allow you to control the uniform concentration of antimicrobial agent throughout the thickness of the finished product.
In addition, the above methods cannot be used to obtain products of complex geometric shape and provide a long antimicrobial effect.
In connection with the foregoing, the claimed invention is directed to the development and creation of compositions based on siloxane rubber, intended for the manufacture of implantable medical devices (implantation period of more than six months) and having a long (prolonged) antibacterial effect.
ч.:The above technical result is achieved due to the fact that in the composition based on siloxane rubber, including aerosil, anti-structuring additive, silica hydride, platinum catalyst and antimicrobial additive, the latter contains rifampicin in the following ratio of components, parts by weight:
Rifampicin is a versatile drug, effective against a wide range of bacteria, with high antibacterial activity in very low doses. Moreover, the given combination of ingredients in the above amounts allows, along with antibacterial activity, to provide good technological properties of the composition, namely the optimal physical and mechanical properties necessary for the manufacture of products from the claimed composition.
The invention is illustrated by the following examples.
1.Example 1
1On the rollers, a rubber mixture is prepared, the composition of which is given in table. one
The rubber used was SKTB polydimethylmethylvinylsiloxane rubber.
150.Aerosil grades 300 and 150 were used as filler.
%.As the silica used polydimethylhydride siloxane with an active hydrogen content of 1.19 mol. %, or polyethylsiloxane with an active hydrogen content of 1.09 mol. %
В качестве антиструктурирующей добавки использовали α,ω-дигидроксиолигодиметилсилоксан - продукт НД-8 и дифенилсиландиол.Α, ω-dihydroxyoligodimethylsiloxane, a product of ND-8 and diphenylsilanediol, were used as an anti-structuring additive.
To activate the crosslinking process, H 2 PtCl 6 ⋅ 6HCl was introduced into the rubber mixture in the form of 0.02 M p-in tetrohydrofuran.
The degree of dispersion of rifampicin was evaluated visually by the sheen of the slice of the mixture and the degree of roughness of its surface, as well as quantitatively after vulcanization according to physical and mechanical tests.
Samples for determining the physicomechanical properties were prepared by vulcanizing the composition in a press at a temperature of 150 ° C for 10 minutes. Evaluation of the properties of vulcanizates was carried out on a tensile testing machine "Monsanto" in accordance with GOST.
The study of the rheological properties of the composition was carried out on a rotorless rheometer MDR-2000 by determining the torque corresponding to the viscosity of the mixture.
В качестве тестовых штаммов были выбраны St. aureus (золотистый стафилококк), St. Epidermidis (эпидермальный стафилококк).To study the antibacterial activity of the samples, the modified Kirby-Bauer method was used. As test strains, St. aureus (Staphylococcus aureus), St. Epidermidis (epidermal staphylococcus).
Evaluation of antibacterial activity was carried out in Petri dishes on a dense nutrient medium Agar Mueller-Hinton (pH 7.4 ± 0.2). The inoculum dose of bacteria was 106 CFU / ml (colony forming units). 3-4 samples from each test product group were placed on the surface of the seeded plates. All samples were incubated at 37 ° C for 24 hours, after which the diameters of the zones of growth inhibition of bacteria around the product samples were measured.
To determine the duration of the antibacterial activity of the product, samples around which there were growth inhibition zones (even minimal ones) were transferred daily to freshly seeded plates until there were no zones of inhibition of microorganism growth.
As can be seen from the table. 2 data, the uniform distribution of rifampicin in the volume of rubber allows you to get a reliable prolonged antibacterial effect and at the same time maintain high medical and technical properties of the resulting rubber.
The declared amount of rifampicin was determined by the following parameters: technological properties of rubber compounds, physical and mechanical properties of rubbers, and antibacterial effect.
It turned out that with a rifampicin content of less than 1 part by weight the duration of antimicrobial action is reduced and the technological properties of rubber compounds are deteriorated.
With a rifampicin content of more than 2 parts by weight the color of the composition acquires a brown hue, which does not satisfy the aesthetic requirements for medical devices. At the same time, the main operational indicators of the obtained vulcanizates practically do not change.
High biocompatibility and functionality of the developed compounds was confirmed in animal experiments.
Tube rubber samples were implanted into white rats subcutaneously and into the lumen of the digestive tract.
The dynamics of body weight was studied, the behavior of animals in the postoperative period for 2.5 months.
All animals after implantation of samples and further throughout the experiment were healthy. There were no differences in the mass of animals and the masses of the internal organs of the control and experimental groups. Macroscopic examinations of internal organs and histological studies of sections of tissues of internal organs at all periods did not reveal negative changes.
Thus, the proposed composition has a pronounced antibacterial effect while maintaining high physical and mechanical properties. This extends the service life of products made from it, including medical supplies, and provides a long antibacterial effect.
