По известным литературным источникам - эндотелиальная дисфункция лежит в основе патогенеза гестоза. Одним из путей коррекции эндотелиальной дисфункции является увеличение концентрации оксида азота (NO), который относится к основным фактором вазодилатации и индукторам ангиогенеза (Марков, X.М. Молекулярные механизмы дисфункции сосудистого эндотелия / X.М. Марков // Кардиология. - 2005. - Т.45, №12).According to well-known literature, endothelial dysfunction underlies the pathogenesis of gestosis. N-нитро-L-аргинин метиловый эфир (L-NAME) вводится внутрибрюшинно в дозе 25 мг/кг/сут в течение 7 дней. Неселективный блокатор NO-синтазы N-нитро-L-аргинин-метиловый эфир (L-NAME) вводился внутрибрюшинно в дозе 25 мг/кг/сут однократно в течение 7 дней. ПРИМЕР КОНКРЕТНОГО ВЫПОЛНЕНИЯEXAMPLE OF SPECIFIC PERFORMANCE
Блокада NO-синтазы, вызванная 7-дневным введением L-NAME беременным крысам, приводила к нарушению взаимоотношений вазодилати-рующих и вазоконстрикторных механизмов регуляции сосудистого тонуса, о чем свидетельствуют результаты сосудистых проб на эндотелийзависимое (ацетилхолин) и эндотелийнезависимое (нитропруссид натрия) расслабление сосудов и увеличение КЭД с 1,28±0,23 у интактных беременных животных до 3,06±0,32 (р<0,05). Длительное, в течение 7 суток, ежедневное внутрижелудочное (через зонд) введение азитромицина (30 мг/кг) на фоне ADMA-подобной модели гестоза у беременных крыс приводило к достоверному снижению КЭД до 1,82±0,09, что достоверно меньше, чем в группе животных с введением L-NAME (р<0,05). Артериальное давление у интактных животных составляло: систолическое (САД) - 125,0±6,3 мм. рт.ст., диастолическое (ДАД) - 82,0±5,8 мм. рт. ст. При введении L-NAME происходило увеличение артериального давления до 183,1±9,4 мм. рт. ст. и 136,7±7,4 мм. рт.ст. соответственно.
