Description
The present invention relates to the field of stimulating the growth of cells of living organisms, and more specifically to devices for electrical control of the growth of cells and tissues by direct application to tissues and cells of an electric field or current. The present invention relates to devices for controlling cell and tissue growth in vitro.
Известны устройства электрической стимуляции (ЭС) роста клеток и тканей in vitro [1, 2, 3].Known devices for electrical stimulation (ES) of cell and tissue growth in vitro [1, 2, 3].
A known device ES [1], containing a set of electrodes placed on an electrode board that makes electrical connection with the electrodes, and a layer of protective coating separating the electrode board from the cell culture medium. A feature of the device is the local supply of ES to cells and tissues located in a nutrient medium, which reduces the power consumption of the device, and allows more efficient to supply voltage or current to cultured cells. However, this design requires the use of special tablets, and cannot be used in traditional culture tablets (tablets for the growth of cell cultures), which limits its scope in the study of various bases for cell culture.
A device ES [2], which contains a set of two electrodes of different geometries located on opposite sides of the wells of the culture plate. Moreover, the device is characterized in that an electric field for ES cells is formed on the electrodes, which reduces the efficiency of exposure to one cell, and also leads to an uneven distribution of electric field lines at the bottom of the well of the culture plate.
However, even the use of arbitrary-shaped electrodes does not allow ES to be carried out on cell culture surfaces in standard culture plates with potential applied directly to each cell.
The closest in technical essence to the claimed invention is an ES device [3] containing a culture plate, a set of chemically and biologically inert electrodes for placement above the culture plate, located on opposite sides of each well, each of which is flat and extended along the well. Compared with the designs of ES devices described above [1, 2], this device can be used in standard multi-well culture plates, and the ES potential can be introduced simultaneously into each well of the tablet. However, like the ES device described above [1,2], it does not have sufficient localization of the electric field near the cells and requires the application of high voltages to deliver the ES potential through the culture medium to the cells, which increases its energy consumption. This is primarily due to the large distance between the electrodes.
The objective of the invention is to increase the locality of the transmitted potential to the cell and reduce power consumption, by organizing a system of electrodes that directly transmit the potential of ES cells through an electrically conductive base. The technical result of the claimed invention is to increase the efficiency of the stimulating effect of the electric field on one cell and the uniform distribution of electric field lines in the bottom zone of the hole.
To achieve this objective, a set of electrodes placed on the cover of the tablet, which are immersed in a nutrient medium before contact with the base at the bottom of the hole, is introduced into the ES device containing a multi-well culture plate and a breadboard with external terminals attached to the cover of the well two electrodes are located on one line passing through the center of the tablet and located at least half the radius from the center and characterized in that each electrode is cylindrical them, the diameter of which is the minimum necessary to ensure the supply of potential to a conductive base located at the bottom of the hole. The electrodes are connected to the breadboard for supplying external electrical impulses in this case, from each pair of electrodes, one is connected to one electric bus, the second to the second electric bus on the breadboard located on the cover of a standard culture tablet, ensuring the supply of the corresponding potential difference, while the contact electrodes with a bus on the breadboard are through a hole in the lid of the tablet with a diameter not exceeding one and a half diameters of the electrode, which ensures freedom of movement ektroda and protection against the penetration of external factors during cell growth.
Due to the free passage of the electrodes, an electrically conductive base with lateral dimensions can be placed on the bottom of the well, not less than the distance between the electrodes, to which an electrical signal can be supplied directly during cell cultivation on the conductive base.
The maximum positive effect is achieved if the base and electrodes form an ohmic contact upon contact, that is, a contact whose resistance does not exceed 100 Ohms. Then the main voltage drop occurs on the electrically conductive base, and not in the medium for cultivation, which minimizes the applied voltage for ES cells located on the electrically conductive base.

The ES device according to the invention contains (FIG. 1) a culture plate containing at least one well, two
The diameter of the electrode is calculated from two conditions: the diameter of the electrodes should be sufficient to ensure elastic contact of one end of the electrode with the conductive base in the hole when the electrode deflection is not more than 10% of the length of the electrode section in the hole, on the other hand, the contact surface area of the electrode with the environment should be minimal to ensure minimal leakage current through the culture medium.
It is known that the stiffness of the electrode in the form of a rod K is determined by its mechanical properties and geometric dimensions according to the expression:
где Е - модуль Юнга, l - длина стержня, r - радиус стержня.where E is Young's modulus, l is the length of the rod, r is the radius of the rod.
In the approximation that all the voltage drops on the contact region of the electrode surface and the culture medium, the expression for the resistance R takes the following form:
где ρ - удельное сопротивление электрода, l - длина стержня, r - радиус стержня.where ρ is the resistivity of the electrode, l is the length of the rod, r is the radius of the rod.
According to the invention, the resistance of the contact area with the medium should be maximum, to ensure the passage of current directly through the conductive base to the cells, while the stiffness should be high enough to ensure ohmic contact of the end of the electrode with the conductive base. According to figure 2, the resistance will be maximum with decreasing radius, while the rigidity increases with increasing radius of the electrode. Thus, the optimum radius value for a particular electrode material should be sought. Given the fact that the dependence of the change in resistance on the radius decreases faster than the stiffness increases, the radius should be chosen in the region of low values. In the claimed invention, the radius of the cylindrical electrode may be at least 0.1 mm and not more than 5 mm.
Electric potentials in the form of a constant voltage, alternating, pulsed, or signal of any other form are applied to two electrodes located in the well. In this case, the electrodes are made or coated of chemically and biologically inert material having high electrical conductivity. In this case, the diameter of the electrodes is selected from the condition of ensuring their rigidity to form contact with the base: that is, upon contact with the base, the electrodes should only experience elastic deformation. The electrode material may be platinum, gold, stainless steel, or other biocompatible alloys coated with gold or platinum.
The material of the conductive base can be made of any biologically inert and chemically stable conductive material, for example, gold, platinum, tin alloyed titanium oxide, a film of nanotubes, graphenes.
The cylindrical electrode can be mounted on a breadboard under the shoulder 90 °, providing free vertical movement of the electrode at a distance of at least 10% of the electrode height, providing spring contact to the conductive base at the bottom of the well of the tablet.
The maximum efficiency of the device is achieved when all the voltage applied to the electrodes linearly drops on the conductive base, while the maximum localization of the electric field is achieved if the base is made in the form of a percolated grid with a cell pitch of no more than the size of one cultured cell.
Concrete example
As an example of a specific embodiment of the invention, a standard 6-well plate was selected with a diameter of each well of 3.5 cm and a depth of 1 cm (Figure 3). Two holes above each well were formed in the lid of the plate. The hole diameter was 0.5 mm. On the lid there was a breadboard with a formed system of adjustable tires. Each busbar above each well was soldered with one electrode with a diameter of 0.3 mm from a 40KKHNM-VI biocompatible alloy with a formed gold coating with a thickness of at least 1 μm. The electrode with a total length of not more than 5 cm was bent at an angle of not more than 100 ° and not less than 80 ° to the base, so that the working part of the electrode was placed through the hole in the lid perpendicular to the base of the hole, touching it with its end. The remainder was positioned horizontally to the breadboard and soldered with the other end to the supply bus. In this case, the length of the electrode section between the hole into which the electrode enters and the place of solder is at least 1 cm in order to ensure free vertical movement of the electrode in a range no more than 1 mm.
At the bottom of the well was placed an electrically conductive substrate made of a base in the form of a coverslip with a size of 24 × 24 mm 2 , a system of gold contact pads along the edges of the coverslip with a length of 24 mm and a width of at least 10 mm. A thin film of carbon nanotubes was formed between the gold electrodes. The resistance between the electrode and the gold pad was no more than 100 ohms.
Human embryo fibroblast cells (PEF) were cultured on carbon nanotube-modified glass coverslips with electrodes placed in 6-well plates in Eagle's medium supplemented with 10% fetal calf serum. Cultivation and ES cells were carried out in a CO 2 thermostat at 37 ° C, 5% CO 2 for 72 hours without antibiotics.
A potential difference of 10 mV to 5 V was applied to the conductive base through the electrodes. After 24 hours of incubation without an applied voltage, a signal of 5 pulses with a pulse length of 5 ms, a pulse spacing of 5 ms and a period between pulse trains of 1 second for 48 hours was applied. .
The result of using the invention in the form of a specific embodiment is shown in FIG. 4 and FIG. 5. The results demonstrate that, thanks to the proposed design of the invention, cell proliferation begins to change already - at voltages of 50 mV, at a voltage of 200 MB and above, the spatial orientation of the cell monolayer on a conductive base is observed. Moreover, the maximum increase in proliferation is observed at a potential difference of 100 mV, which corresponds to a power consumption of ~ 1 μW. Thus, the proposed contraction leads to the locality of the transmitted potential directly to each cell and a decrease in power consumption during ES cell growth
