This application is a continuation of US application 11/958556, filed December 18, 2007, having priority to US application 11/616572 of December 27, 2006, the description of which is incorporated herein by reference in full.
The present invention relates to microencapsulated polyphenolic compositions suitable for use in a food product or beverage, and methods for producing such microencapsulated polyphenolic compositions. Microencapsulation significantly reduces the astringent taste and / or bitter taste of polyphenolic compositions and protects the polyphenolic compositions from oxidation, the interaction of ingredients, enzymatic degradation and / or the like, while maintaining the gastrointestinal bioavailability in the digestive system.
Natural polyphenols are obtained from plants or plant material (for example, tea, cocoa beans and the like), they are known for their antioxidant properties, along with providing other potential positive health effects. Therefore, in recent years, many studies have been conducted regarding methods for producing such polyphenols along with methods for their use.
Typically, however, such polyphenolic compositions have a very astringent and / or bitter taste. Therefore, it is difficult to add them to food products or drinks in biologically significant amounts without adversely affecting the taste and / or organoleptic properties of such food products and drinks enriched in polyphenols.
Encapsulation is known to be used to provide delayed isolation and / or protection of sensitive materials. For example, US Pat. No. 6,190,591 (February 20, 2001) describes a continuous and relatively complex method for producing controlled release of discrete solid particles (i.e., matrices) containing an encapsulated and / or coated component, such as a heat-sensitive or readily oxidizable pharmaceutically, biologically or nutritionally active component. To control the rate of release, the active component is introduced into the matrix. US Pat. No. 6,835,397 (December 28, 2004) describes an encapsulated yeast mixture comprising a core containing yeast and a coating containing emulsified lipid.
Этот способ не является успешным, поскольку, как описано, при потреблении во рту все еще чувствуется вяжущий вкус (смотрите Пример 4 ниже).JP 2005-124540A (published May 19, 2005) describes a method for masking or reducing the bitter taste of polyphenolic compositions that include casein, in particular acid casein. In this method, the polyphenol composition is mixed with a solution or suspension of casein in water, preferably adjusted to a neutral pH. Such a casein-containing polyphenolic composition can be used directly in drinks or dried to obtain a powder form for use in food or drinks. Typically, sufficient “masking” of polyphenols is obtained by combining from about 40 to about 350 parts (and preferably from about 60 to 150 parts) of casein with about 100 parts of the polyphenol composition in water. This method is not successful because, as described, an astringent taste is still felt when consumed in the mouth (see Example 4 below).
There is a need to provide other improved methods for reducing the astringent and bitter taste of polyphenolic compositions. The present invention relates to such improved methods for reducing the astringent and bitter taste of polyphenolic compositions. The present invention also relates to polyphenolic compositions with a significantly reduced astringent and bitter taste. The polyphenolic compositions of the present invention can be added to a food product and / or drink without adversely affecting the taste and / or organoleptic properties of such food products or drinks.
SUMMARY OF THE INVENTION
The present invention relates to microencapsulated polyphenolic compositions to significantly reduce the astringent and bitter taste associated with the polyphenolic components. The polyphenolic compositions of the present invention can be added in significant quantities to a food product and / or drink without adversely affecting the taste and / or organoleptic properties of such food products or drinks. Therefore, the present invention relates to polyphenol compositions preserving the original biological activity of polyphenols, but without a specific bitter and astringent taste, normally associated with polyphenols.
DETAILED DESCRIPTION OF THE INVENTION
The present invention relates to encapsulated and microencapsulated polyphenolic compositions to significantly reduce the astringent and bitter taste normally associated with polyphenolic compositions. The polyphenolic compositions of the present invention can be added in significant quantities to a food product and / or drink without adversely affecting the taste and / or organoleptic properties of such food products or drinks.
The present invention relates to polyphenol compositions substantially protected by passage through the mouth, but releasing polyphenolic compounds contained therein later in the digestive system (i.e., essentially in the stomach, small intestine). By preventing or reducing the amount of contact between polyphenolic compounds and taste buds of the oral cavity, the present invention significantly reduces the astringent and bitter taste normally associated with polyphenols. However, polyphenolic compounds are more fully secreted in the digestive system, where they exert their beneficial effects on health. Microencapsulation of polyphenolic compositions significantly reduces the astringent and / or bitter taste of polyphenolic compounds and protects the polyphenolic compositions from oxidation, the interaction of ingredients, enzymatic degradation and the like, while maintaining the gastrointestinal bioavailability in the digestive system.
The encapsulated polyphenol compositions of the present invention can be prepared using traditional encapsulation and food encapsulation or coating procedures, provided that the encapsulation allows the polyphenol materials, in particular when they are added to food products, to pass through the oral cavity without significant release of polyphenols, allowing polyphenols stand out when materials pass through the digestive system after the oral cavity and preserve bioavailability and highlighting. For the purposes of the present invention, the term “without significant release” means that the release of polyphenols in the oral cavity, such that the astringent and / or bitter taste normally associated with polyphenols, is effectively eliminated or reduced to an acceptable level for a particular product to which the polyphenols are added .
Although synthetic polyphenols can be used, it is preferred in the present invention to use polyphenols of natural origin derived from plants or plant materials (e.g. berries, tea, cocoa beans, coffee, vegetables, fruits and the like, along with combinations thereof) that are known for their antioxidant properties, along with providing other potential health benefits. Such polyphenolic compounds normally include catechin, epicatechin, gallocatechin, catechingallate, epicatechin gallate, gallocatechin gallate, epigallocatechin gallate, epigalocatechin, tannic acid, gallotannin, ellagitannic acid, gallogentic acid, gallofentic acid, gidogrofenoic acid, gidric acid, gallofentic acid, gidric acid, gentric acid, rosmarinic acid, rutin, quercetin, quercetagin, quercetagetin, gossipetin, anthocyanin, leukoanthocyanin, proanthocyanidin, enocyanin and the like, along with their derivatives, polymers and stereoisomers. Polyphenols can be extracted from these plants using conventional techniques (for example, by extraction with one or more solvents selected from water, ethyl acetate, methanol, ethanol, isopropanol and the like, or mixtures thereof).
Typically, the encapsulated polyphenols used in the present invention are in powder form. Although physical properties may vary depending on the encapsulation method used and the product into which the polyphenols are added, the encapsulated polyphenols in most embodiments of the present invention are preferably particles of a substantially spherical shape with an average size of from about 50 to about 1700 microns, preferably from about 50 to about 500 microns and more preferably from about 70 to about 120. If necessary, the particle size can be reduced before or after the encapsulated addition polyphenols in food. Of course, encapsulated polyphenols of other forms and / or of a different size can be used, provided that a predetermined decrease in the astringent and bitter taste is obtained while maintaining the desired organoleptic properties of the food product.
For some products (for example, having a very smooth texture, such as chocolate), smaller sizes or even sizes less than the limits given here are preferred. Therefore, for example, the encapsulated polyphenols used in the preparation of chocolate preferably have from about 15 to about 100 microns in the finished product d90 (i.e., 90 percent of the particles have a particle size equal to or less than that characteristic of d90) and even more preferably from about 20 to about 30 microns in the finished product, such a particle size can be achieved by reducing the particle size of the encapsulated polyphenols before adding them to the product or by reducing the particle size (e.g. grinding) of the encapsulated poly enols during or after the process of obtaining the product into which the encapsulated polyphenols are added.
The encapsulating material or coating material must be food grade and must provide controlled release when consumed in the human body. For the purposes of the present invention, the term “controlled excretion” does not mean excretion or significantly reduced excretion during normal chewing, but subsequent increased excretion (essentially complete excretion) when passing through the rest of the digestive system (i.e., the stomach and / or small intestine), where bioabsorption. When not excreted and with significantly reduced excretion in the mouth during a normal meal containing such polyphenolic compositions, the astringent and bitter taste normally associated with polyphenols does not occur, thus allowing the encapsulated polyphenolic compositions to be added to a wider range of food products and / or add large quantities, without negatively affecting the taste and / or organoleptic properties of such foods or drinks into which they are added. For the purposes of the present invention, the term “normal chewing process” includes normal chewing actions in the mouth when consuming a food product while swallowing chewed food. For the purposes of the present invention, the term “normal digestion process” includes the normal digestion process that occurs after ingestion of chewed food; usually, this process involves mixing and digesting the food product in the stomach, along with passing the mixed and digested product through the small intestine.
The encapsulating or coating composition must remain intact (thus preventing and / or substantially reducing the release of polyphenols) for at least about 30 seconds and preferably at least about 60 seconds in the oral cavity (essentially for a period of time, sufficient to chew and swallow a food product containing encapsulated polyphenols), but then disintegrating in order to secrete polyphenols in the stomach and / or small intestine. For the purposes of the present invention, the actual mechanism by which the encapsulated polyphenols remain essentially not secreted, or in which the secretion is delayed in the oral cavity, but then the secretion occurs in the rest of the digestive system, is not critical.
Examples of encapsulating or coating materials for use in the present invention include lipids, gelatin, shellac, guar gum, waxes, polymers of a mixture of proteins and carbohydrates, and the like, along with combinations thereof. The amount of encapsulating or coating material compared to polyphenols is usually an effective amount to reduce the astringent and / or bitter taste normally associated with polyphenols, while maintaining organoleptic properties within acceptable limits. Typically, the resulting encapsulated polyphenols contain from about 60 to about 90 percent of the polyphenols and from about 5 to about 40 percent of the encapsulating material, and preferably from about 70 to about 90 percent of the polyphenols and from about 10 to about 30 percent of the encapsulating material.
B.Most preferred lipid coating compounds for use in fluidized bed encapsulation systems include hydrogenated palm oil, acetylated monoglycerides, hydrogenated cottonseed oil, hydrogenated soybean oil, hydrogenated coconut oil, cocoa butter and the like. Preferably, the gelatin coating composition contains from about 2 to about 20 percent gelatin, from about 1 to about 5 percent glycerol, and from about 75 to about 97 percent water. Type A or B gelatin can be used.
The most preferred protein and carbohydrate mixtures for use in spray drying encapsulation systems are proteins such as (e.g., milk protein isolate, sodium caseinate, all milk proteins, whey protein, and the like) and soy proteins (e.g., soy protein isolate and the like like) and carbohydrates such as maltodextrin, trehalose, corn syrup solids and the like. More preferably, the protein is milk protein in the form of a milk protein isolate, and sodium caseinate is most preferred, and maltodextrin or trehalose with maltodextrin are most preferred. Spray drying typically uses a water-based carrier system containing about 30 to about 70 percent protein and about 30 to about 70 percent carbohydrates, and more preferably about 35 to about 45 percent protein and about 55 to about 65 percent carbohydrates. Typically, the spray-dried encapsulated polyphenols contain from about 20 to about 60 percent of the polyphenols and from about 40 to about 80 percent of the encapsulating material, and preferably from about 30 to about 50 percent of the polyphenols and from about 50 to about 70 percent of the encapsulating material. Not wanting to be limited by any theory, the authors of the present invention believe that in the spray drying system, encapsulation can be achieved by physical absorption and / or adsorption of polyphenols together with individual protein chains. In addition, during this physical absorption and / or adsorption, polyphenols can also essentially be contained in a matrix formed by the carrier material. Regardless of the mechanism or mechanisms involved, spray drying compositions provide an effective system to reduce the effects of polyphenols (e.g., a reduced astringent and / or bitter taste).
The thickness and nature of the composition covering the polyphenolic particles should be effective to prevent and / or delay release in the oral cavity and effective to subsequently ensure release in the rest of the digestive system and, thus, to reduce the astringent and bitter taste in the oral cavity. Typically, the coating composition forms an encapsulating layer, coating or matrix protecting polyphenolic particles, a thickness of from about 1 to about 100 microns (from about 0.001 to about 0.1 mm) and preferably a thickness of from about 10 to about 40 microns (from about 0, 01 to about 0.04 mm). The encapsulating layer, coating or matrix protecting the polyphenolic particles may be uniform or not uniform in thickness. Typically, the encapsulated polyphenolic compositions contain from about 60 to about 95 percent polyphenes and from about 5 to about 40 percent of the encapsulating material, preferably from about 70 to about 90 percent of the polyphenols and from about 10 to about 30 percent of the encapsulating material. Of course, the relative amount of polyphenols and the encapsulating material and the thickness of the encapsulating layer, coating or matrix can vary, provided that the astringent and bitter taste of the polyphenols is effectively reduced in the oral cavity. In some cases (for example, a matrix formed by spray drying encapsulation technology, this can be achieved by grinding the encapsulated polyphenol compositions to obtain a predetermined particle size distribution. Such additional grinding can be carried out on encapsulated polyphenols before or after adding to a given food product. Usually, the size particle size is preferably adjusted to reduce or avoid sandiness while maintaining a predetermined binder reduction and a bitter taste in the finished polyphenol-containing food product.
Of course, the appropriate particle size can be determined on a case-by-case basis and will likely depend on the type of food product given. For example, for foods that normally have a crispy texture, larger particles can be used, while for a product with a smooth texture, smaller particles are likely to be required. For products with a very smooth texture (for example, high-quality chocolate), smaller particles are usually preferred. Therefore, for example, the encapsulated polyphenols used in the preparation of chocolate preferably have a d90 of about 15 to about 100 microns (i.e., 90 percent of the particles have a particle size equal to or less than a certain indicator) and even more preferably from about 20 to about 30 microns per finished product; this particle size can be achieved by reducing the particle size of the encapsulated polyphenols, both before adding them to the product, and grinding the enriched product during or after the production process.
Encapsulation can be carried out using any conventional technology. Examples of such technologies include fluidized bed encapsulation, extrusion, spray drying, granulation with centrifugal disk spray dryers, and the like. Preferred for encapsulation are fluidized bed systems or spray drying.
The encapsulated polyphenol compositions of the present invention are essentially intended to add a significant amount of polyphenols to foods for human or animal consumption without the astringent and bitter taste normally associated with polyphenols. Typically, the encapsulated polyphenol of the present invention is added to a given food product in an amount of from about 1 to about 20 percent, preferably from about 1 to about 10 percent, using suitable technology. Therefore, for example, encapsulated polyphenolic compositions essentially obtained by spray drying of polyphenols using mixtures of proteins and carbohydrates can be added to dark or milk chocolate to significantly increase the amount of polyphenols without adversely affecting the taste and / or organoleptic properties of chocolate. For polyphenol-containing chocolate, it has been found that spray-dried encapsulated polyphenolic material can be added at various stages of the chocolate production process. Such a spray-dried encapsulated polyphenolic material is preferably added to the chocolate at or shortly before the end of the conching treatment in the conventional chocolate process. The conched material containing the encapsulated polyphenolic material is then preferably ground using a ball mill and further processed using conventional chocolate technology. The grinding stage in a ball mill makes it possible to reduce the particle size of the encapsulated polyphenolic material to sizes effective to reduce or avoid the feeling of sandiness while maintaining a predetermined level of reduced astringent and bitter taste. Of course, other methods can be used to obtain a given particle size of the encapsulated polyphenols in the finished product. For example, spray-dried encapsulated polyphenolic material may be ground to a predetermined particle size before being added to chocolate. Dark chocolate with a content of about 500 mg or more polyphenols per 60 g of chocolate and milk chocolate with a content of about 200 mg or more polyphenols per 60 g of chocolate with good organoleptic properties can be obtained. This is a significantly higher polyphenol content compared to traditional chocolate, along with currently available enriched chocolate polyphenol (for example, about 200 mg of polyphenol per 60 g of dark chocolate or about 100 mg of polyphenol per 60 g of milk chocolate). The currently available polyphenol-enriched chocolate is usually prepared either by (1) selecting a raw material with a relatively high polyphenol content and / or adjusting the production conditions to maintain the polyphenol content in the raw material or (2) adding an increased amount of polyphenols. When using the first technology, a significantly higher polyphenol content (that is, as high as obtained in the present invention), usually cannot be achieved without a significant astringent and bitter taste; and when using the second technology, although a high polyphenol content can be achieved, the astringent and bitter taste associated with the polyphenols becomes very noticeable. However, the present invention allows to obtain a significantly higher content of polyphenols without an astringent and bitter taste, normally associated with polyphenols.

Examples 1-4 illustrate encapsulated polyphenolic compositions obtained in a fluidized bed system. Examples 5-8 illustrate encapsulated polyphenolic compositions obtained in a spray drying system; then the spray dried polyphenol composition is added to chocolate.
This Example illustrates microencapsulation of a polyphenolic composition with lipids (hydrogenated palm oil from Humko Oil Products, Cordova, TN). As the polyphenolic composition, VitaBerry ™ (Van Drunen Farms, Momence, IL) was used; until use, it was stored frozen. VitaBerry ™ is a powdered mixture of concentrated fruit extracts and powdered whole fruits / whole fruit powders that contain natural antioxidants with a high free radical absorption capacity (ORAC) and phytochemicals; the polyphenol content is about 30 percent. Polyphenolic compositions were sieved to obtain particles passing into sieve cells with sizes from 0.089 to 0.122 mm (mesh size about 140-100).
4.The coating of the polyphenol composition is carried out using a high temperature fluidized bed system (Uni-Glatt GmbH, Ramsey, NJ). The fluidized bed system is started in advance for about one hour to achieve operating temperature. Hydrogenated palm fat (99 g; melting point about 57 ° C) was melted using a heating plate. Thirty minutes before coating, the polyphenol composition was taken out of the freezer and thawed to room temperature. Then the polyphenol composition (200 g) was added to the fluidized bed system and the fluidized bed was started. After 1-2 minutes (in order for the polyphenol composition to reach an operating temperature of about 70 ° C), a flow of molten lipids was applied and a speed of about 4 to about 5 ml / min was maintained. A fluidized-bed chamber with the melt in it is tapped with a rubber mallet and vibration function is used approximately every two to three minutes to maintain fluidized bed uniformity. After all hydrogenated palm fat has been supplied, the fluidized bed is stopped and the encapsulated polyphenol composition is removed. The encapsulated polyphenolic composition is laid out on parchment paper and cooled for about 30 minutes. The ratio of polyphenol composition to lipids is about 70:30. The encapsulated polyphenolic composition is sieved to obtain particles passing into sieve cells with a size of 0.250 to 1.7 mm and stored in a glass container sealed with aluminum foil in a freezer. The evaluation of the polyphenol composition coated with lipids is given in Example 4.
Example 2. This Example illustrates the microencapsulation of a polyphenolic composition (ie, VitaBerry ™ used in Example 1) with gelatin (100 Bloom Type A; Great Lakes Gelatin, Grayslake, IL) using essentially the same device and procedure (except as indicated), as in Example 1. A gelatin coating of the composition was obtained by heating water (about 150 g) to a temperature of about 100 ° C. Then, gelatin (about 10 g) was slowly added with stirring. Once all the gelatin has dissolved, glycerol (about 2 g; (Dow Chemical, Pevely, MO)) is added and stirred continuously for 5 minutes to give a uniform mixture. The coating solution was kept at a temperature of about 70 ° C and sealed until use. High-temperature system the fluidized bed was modified so that the inlet pipe for the coating composition could be heated and could maintain the coating composition at a temperature of about 85 ° C when entering the fluidized bed chamber.
4.After the polyphenol composition (about 50 g) has reached operating temperature, a coating composition stream (about 170 g) is fed into the fluidized bed chamber and a speed of about 4 to about 5 ml / min is maintained. A fluidized-bed chamber with the melt in it is tapped with a rubber mallet and the filter function with a sludge blower is used approximately every five to ten minutes to keep the filter clean and reduce excessive powder loss. After the entire gelatin solution has been supplied to the polyphenol composition, the encapsulated polyphenol composition in the form of a dry powder is removed. After cooling, sieved to obtain particles passing into the sieve mesh with a size of 0.250 to 1.7 mm, and stored in a glass container sealed with aluminum foil in the freezer. The ratio of the polyphenol composition to the gelatin coating is about 70:30. Evaluation of the polyphenol composition coated with gelatin is shown in Example 4.
4.Comparative Example 3. This Example illustrates for comparison polyphenolic compositions obtained by the methods described in JP 2005-124540A. The same starting polyphenol composition was used as in Example 1 and 2. The ratings of the three comparative samples described here are also given in Example 4.
Sample 1. Acid caseinate (7.5 g; Dairygold Co-Operative Society Limited, Ireland) was mixed with 7.5 ml of 0.1 N sodium hydroxide in 80 ml of deionized water for one hour at room temperature. A polyphenol composition (12 g) was then added and mixed continuously for an additional hour at room temperature. The solution was then poured into a small bread pan and sealed with aluminum foil and stored in the freezer overnight. Then, the frozen solution was placed in a freeze dryer (Virtis Genesis 25XL) and freeze-dried for about 3.5 days. After freeze-drying, a casein-containing polyphenol composition is obtained in powder form and then stored in a sealed glass container in a freezer.
Sample 2. The second comparative sample was obtained in the same way as the above Sample 1, except that the initial casein solution contained 7.5 g of acid casein, 7.5 ml of 0.1 N sodium hydroxide and 0.475 g of sodium tripolyphosphate in 80 ml of deionized water. A sample of the polyphenol composition (12 g) was added and processed according to Example 1. After freeze-drying, the casein-containing polyphenol composition was obtained in powder form and then stored in a sealed glass container in a freezer.
Sample 3. Acid casein (49.4 g; the same as used in the previous samples) and sodium carbonate (3.3 g) were added to deionized water (1769.3 g) and mixed until dissolved. A sample of the polyphenolic composition (79 g), as used in the previous samples, was slowly added and the entire solution was mixed. The resulting solution was then spray dried using an APV Anhydro Laboratory Spray Dryer Type PSD 52 at a flow rate of about 10 ml / min, an inlet temperature of 170 ° C, an outlet temperature of about 72 ° C (initial) to about 87 ° C (final) to obtain a casein-containing polyphenolic mixture, which is stored in a freezer.

Pharma (East Sussex, UK). J. Enol. Vitic, 16:144-158 (1965)). Dissolution. To evaluate the processing methods and their effect on the astringent and bitter taste associated with polyphenolic compositions, various samples were tested in various solutions that mimic (1) saliva in the mouth, (2) gastric juice in the stomach, and (3) intestinal juice in the small intestine. Imitation of saliva was obtained from A.S. Pharma (East Sussex, UK). Imitation of gastric juice and intestinal juice was obtained according to the Pharmacopoeia of the United States of America (Vol. 29, p. 3171). To simulate gastric and intestinal digestion, 25 mg of the sample was weighed into 15 ml propylene centrifuge tubes, 10 ml of the solution (heated to 37 ° C) was added to the tube and sealed. The tubes were rotated by inverting at a speed of 25 rpm at 37 ° C for 1 hour, then they were immediately filtered through a glass microfiber filter (VWR degree / class / gradient 691), on which insoluble material remained. A sample of the filtrate was collected and analyzed for all polyphenols using a Folin-Ciocalteu assay (see, Singleton et al., Am. J. Enol. Vitic, 16: 144-158 (1965)). A similar procedure was used to simulate contact with saliva, except that the tubes were rotated only 30 seconds before being filtered.
J. Enol. Powdered polyphenolic compositions (usually about 25 mg) were incubated in various simulating solutions (usually about 10 ml) at 37 ° C for about 30 seconds to simulate a saliva solution and for 1 hour to simulate gastric juice and intestinal juice. After each incubation, the amount of polyphenols released in the respective solutions of the test samples was determined using the Folin-Ciocalteu assay (see, Singleton et al., Am. J. Enol. Vitic, 16: 144-158 (1965)); then the percentage of polyphenol release was calculated. The following results were obtained.

As can be seen from these data, the samples of the present invention showed a significantly lower release of polyphenols in the simulated saliva solution compared to comparative samples. Therefore, when the samples of the present invention are consumed, a significantly lower amount of polyphenols is released in the mouth, thereby significantly reducing the astringent and bitter taste. Data simulating gastric and intestinal juice showed that the samples of the present invention secrete polyphenols during digestion. Therefore, compared with comparative samples, the samples of the present invention provide polyphenols during the digestion process (i.e., in the stomach and small intestine), and not in the mouth.
The destruction of polyphenols by oxidation. Various examples were also evaluated by determining the ability of polyphenols to avoid oxidation degradation. A set of experimental models of the destruction of polyphenols on models of food matrixes due to the presence of an enzyme that oxidizes polyphenol is presented. To determine the degree of protection against the polyphenol oxidizing enzyme, Comparative samples (about 7 mg; processed casein) or samples of the present invention (10 mg; encapsulated) of Comparative Example 3 (Samples 1 and 2 only) and Examples 1-2, respectively, were placed in 15 ml propylene centrifuge tubes. Additionally, an unencapsulated polyphenolic sample (about 7 mg) was treated in the same manner. Test samples (5 ml) 80:20 glycerol / water with or without polyphenol oxidation products (2 mg / ml; Sigma-Aldrich, St. Louis, MO) were added and sealed with a tube. The tubes were rotated and turned over for one hour at a temperature of 22 ° C and then sparged with air for 30 minutes. Then, any enzymatic reactions were stopped by heating the tubes in boiling water for 10 minutes; then the samples were cooled on ice. The resulting solutions were centrifuged for 20 minutes at 10,000 G. The supernatant was collected and then evaluated for total polyphenols using a Folin-Ciocaltheu assay as described above. The following results were obtained:
Example 5. This example illustrates the traditional spray drying procedures used to encapsulate polyphenol compositions, along with methods for assessing the effect of encapsulated polyphenols on chocolate.
All powdered ingredients, including polyphenols, were mixed in a Hobart mixer (Hobart -50, 5 quart, equipped with paddles, maximum speed 1725 rpm): dry powders were mixed at low speed. Then, to prevent clumping, water was slowly added to the Hobart mixer at low speed, whipping and / or increasing viscosity; if necessary, sieving is carried out. The resulting suspensions typically had a solids content of about 45 percent.
The slurry is fed into a spray dryer Niro Mobile Minor TM (L / W / H 1800/925/2200 mm) using a peristaltic pump (Cole Parmer Masterflex L / S Easy -Load). The product was sprayed as an aerosol using an impeller rotating at a high speed (typically about 27,000 rpm). Hot air circulated in the chamber around the wheel, drying the aerosol to obtain a powder, which was then separated from the air in the cyclone. Air under pressure (4-5 bar) was used to actuate the atomizer and dryer cover (pneumatically lifted). During the operation, the inlet air temperature is about 155-170 ° C and the outlet temperature is about 95-105 ° C. The spray dryer and atomizer were disassembled and washed with water once or twice a day to prevent excessive powder deposition, along with significant cross-contamination of the samples. Spray dried polyphenol powder was collected for evaluation.
Then, the resulting encapsulated polyphenols were added to a mass of commercial liquid dark chocolate taken from a working production line (i.e., after the conching step). This commercial chocolate mass contains about 300 mg of polyphenols per 100 g (several months' average of standard batches of products). The spray dried polyphenolic compositions were then manually added and mixed with the chocolate mass and then manually tempered and molded to form chocolate bars. The added amount of the encapsulated polyphenol composition was adjusted to achieve a total polyphenol content of at least about 500 mg per 60 g (or about 830 mg polyphenol per 100 g). The polyphenol content of the polyphenol extracts used to prepare the spray dried compositions was determined using HPLC; this method allows the measurement of intact procyanidin molecules (e.g., epicatechin and catechin) along with dimers, trimers, tetramers and similar forms. The chocolate bars were then stored at a temperature of about 16 ° C for about three weeks so that the cocoa fat or butter crystallized before a sensory evaluation with the participation of a qualified tasting panel.
Example 6. This example demonstrates previous experiments using spray drying to encapsulate grape seed polyphenol extracts obtained from Planteextrakt. Typically, the same spray drying and evaluation procedures were used as described in Example 5. The compositions used for spray drying consist of aqueous suspensions containing the extract and test vehicles in a 30/70 ratio. Encapsulated compositions were added in an amount of about 5.3 percent to a sample of liquid chocolate taken from an industrial production line. The following control samples were also obtained: Control 1 — chocolate (no additives); Control 2 - grape seed extract (1.6 percent) mixed with chocolate (without carrier or spray drying); and Controls 3, 5, and 7 — mixing the powdered grape seed extract and carrier in the same proportions (without spray drying) with chocolate. Sensory evaluation of chocolate samples was performed after about 3 weeks.
Received and evaluated the following samples. Except for Control 1, all samples contained 1.6 percent grape seed extract; no control sample included encapsulation; Samples 4, 6, 8, and 10 contained encapsulated polyphenols obtained with various carriers (30/70 ratio of grape seed extract to carrier). With the exception of Control 1, all samples contained grape seed extract.

Example 7. Using the procedures of Example 5 and guided by the results of Example 6, the following components were used to obtain encapsulated polyphenols. Many of these encapsulated polyphenols were then added to chocolate for evaluation, as described in Example 5. Spray-dried formulations are presented below:
Cargill.
Natraceutical Group.
Mafco.a partially hydrolyzed sodium caseinate; Alanate 385 is a calcium caseinate; TMP 1104 is a common milk protein (i.e. milk protein isolate), all obtained from Fonterra (New Zealand). Pro-Fam 873 is an isolated soy protein ADM.
** Carbohydrates obtained from Cargill.
+ CocoanOX 45 and CocoanOX 70 are natural cocoa extracts, as stated, contain from 45 to 70 percent polyphenols, respectively, from the Natraceutical Group.
ǂ MM-100 is a masking agent based on monoammonium glycyrrhizinate from Mafco.
Chocolate bars were prepared containing spray dried compositions as shown in the table above using the procedure as described above in Example 5. Additionally, standard chocolate samples containing only CocoanOX 45 and CocoanOX 70 (i.e., without carriers or spray drying) were also obtained. ; Controls 1 and 2, respectively). The amounts of added spray dried polyphenol compositions and CocoanOX standards were adjusted to achieve a polyphenol content of about 850 mg per 100 g chocolate. Sensory results are given below.
Based on sensory evaluation, compositions containing CocoanOX 45 were rated higher compared to similar compositions containing CocoanOX 70, even though the amounts were adjusted so that all compositions contained a similar total amount of polyphenols. Of the proteins tested, sodium caseinate, milk protein isolate, and soy protein isolate are preferred. Of the carbohydrates tested, maltodextrin, trehalose and dry solids of corn syrup are preferred (25DE; data not shown), maltodextrin is most preferred; although data on high maltose corn syrup, modified starch and fibers (e.g., oligofructose (<10DE) and inulin) were not presented here, they were tested, but were not satisfactory. Carrier systems containing masking agents (e.g. MM-100 or cocoa butter) were generally rated worse than similar systems without masking agents, a system with calcium caseinate (sample 15) was rated as the mildest among them and had a very weak astringent taste; but she also had very faint notes of cocoa and chocolate.
Of course, it will be understood by those skilled in the art that systems that effectively reduce the astringent taste but which reduce or otherwise negatively affect the desired quality characteristics of the chocolate flavor can be used and may even be preferred in other food products with a different taste profile. The overall assessment is important because the encapsulated polyphenols must have, in addition to reducing the astringent taste, a slight negative effect on the organoleptic properties of the food product in which it is used.
5.Example 8. This example illustrates the addition of spray-dried encapsulated polyphenols to chocolate at various stages in chocolate industrial production lines. With the exception of the timing of adding the encapsulated polyphenols to the chocolate and chocolate production lines, the procedures of Example 5 are essentially used.
The first spray dried polyphenolic composition obtained included 47.5 percent trehalose, 22.5 percent sodium caseinate and 30 percent CocoanOX 70. The first spray dried polyphenolic composition had a d90 of about 82 microns; the amount added to the chocolate was adjusted for the final yield of the finished product containing about 500 mg of polyphenols / 60 g of chocolate. Then, the first spray dried polyphenol composition was added to individual batches of products of the pilot production plant in the following stages:
Образец 1 - высушенную распылительной сушкой композицию добавляли с сухими ингредиентами, которые затем обрабатывали на рафинировочных вальцах с получением хлопьев;Sample 1 — a spray-dried composition was added with dry ingredients, which were then processed on refining rollers to give flakes;
Sample 2 — a spray-dried composition was added at the beginning of the conching process (along with the ingredients normally added at this stage, for example, cocoa liquor); and
Sample 3 — a spray-dried composition was added at the end of the conching process (along with the ingredients normally added at this stage, for example, lecithin, flavor, cocoa butter).
A control sample was also prepared by mixing the appropriate amounts of trehalose and sodium caseinate (without any polyphenols and without spray drying) with the finished chocolate sample from a batch of standard products.
Evaluation of chocolate samples was carried out after three weeks, during which crystallization of fat or cocoa butter took place, and the following results were obtained:
Although Sample 3 was the best polyphenol-containing sample, it was very sandy.
3.Sample 2 was similar to Sample 3.
3.Sample 1 had a good texture, but was more astringent compared to Sample 3.
Then, the second spray dried polyphenol composition was added to separate batches of the pilot plant in the following steps:
Образец 1 - высушенную распылительной сушкой композицию добавляли в сухими ингредиентами, которые затем обрабатывали на рафинировочных вальцах с получением хлопьев;Sample 1 — a spray-dried composition was added to the dry ingredients, which were then processed on refining rollers to give flakes;
Sample 2 — a spray-dried composition was added at the end of the conching process (along with the ingredients normally added at this stage, for example, lecithin, flavor, cocoa butter); then, the chocolate containing the encapsulated polyphenols was ground using a ball mill to reduce particle size. The finished chocolate encapsulated polyphenol had a d90 of about 20 microns.
Готовый продукт имел аналогично d90.Sample 3 — a composition comprising a spray-dried composition and coarse-ground cocoa liquor was ground in a ball mill to produce finely ground cocoa liquor containing enough encapsulated polyphenols to deliver substantially the same amount of polyphenols as in Examples 1 and 2 to finished products . Grated finely ground cocoa containing encapsulated polyphenols had a d90 of about 26 microns and was added at the beginning of the conching process. The finished product had the same d90.
Evaluation of chocolate samples was carried out after three weeks of storage at a temperature of about 16 ° C, during which crystallization of fat or cocoa butter took place, and the following results were obtained:
Sample 2 was the best spray dried polyphenol-containing sample from this series; it was perceived as having the best texture and taste, with a stronger fruity note and a slightly weaker astringent taste compared to other samples and with the best balance in overall taste. Therefore, it is preferable to add the encapsulated polyphenols at the end of the conching process and then subject the resulting mixture to an additional grinding step before completing the batch of products to produce a finished chocolate product.
