Abstract
Description
The invention relates to medicine, in particular to a method for producing, based on earthworms, a powder enriched with trace elements. The invention is based on the fact that the resulting vermicomposites are enriched with vitally important elements for humans and animals and, at the same time, potentially dangerous in free form elements (Zn, Cr, Cu), which will be present in them in a non-toxic bound form. The neutralization of heavy metals by worms is due to the fact that their ions are inducers of thionein biosynthesis. Due to the high content of cysteine residues, this protein has a high affinity for divalent metal ions and blocks their toxicity.
[1].Currently, a wide variety of domestic and foreign companies produce a huge number of preparations from earthworms and multivitamin complexes, which include various trace elements. These complexes are obtained simply by mechanical mixing of various vitamins with vital mineral salts in short supply. Similarly, by mechanical stirring of the mixture, numerous pharmaceutical preparations are produced, which are produced on the basis of combinations of earthworms and medicinal herbs. Some of the ingredients of such mixtures are toxic in themselves, for example, many heavy metals, which are simultaneously important trace elements for living organisms. To eliminate these shortcomings, technically simple special technologies are proposed that allow one to obtain vermicelli preparations in which microelements will be in a bound, easily digestible form. There is information that earthworms are highly resistant to heavy metals, they accumulate well in fairly high concentrations and effectively bind [1].
A known method of obtaining the drug "Dilongjing" / patent CN 1114202, A61K 35/64; A61K 35/56; 1996 /, including washing live earthworms, separating by centrifugation, lyophilization to obtain a dry powder and loading into capsules or mixing with honey to obtain oral liquid.
A known method of producing dry powder from earthworms of antidiabetic, antihypertensive and antihypotonic preparations containing dry powder of earthworms as an active ingredient / US patent No. 5024844, A61K 35/56, 1991 /. Live earthworms are placed in fresh water or a 0.1% aqueous solution of an organic acid such as acetic or citric acid for a period of 1 to 40 hours. Thus, the digestive tract of living earthworms will be largely emptied. After this, live earthworms are washed with water to remove dirt from the surface of their body. Then live wet earthworms are kept at a low temperature of −5 ° C. or lower (preferably −10 ° C. to −60 ° C.). In the next step, the worms are dried by freezing or under vacuum for 10 to 60 hours. Thus, a sterile dried earthworm powder is obtained.
[1].The closest analogue is a method for producing pharmaceutical preparations from earthworms. The collected earthworms are cut into pieces of 1-2 cm, washed in water for 1-2 minutes, and the resulting mass is dried in strong sun. Dried worms are placed in a wooden box and stored in a ventilated place [1].
A disadvantage of the known methods is the low content of trace elements in them.
The difference of our proposed method in the manipulations used. The objective of the invention is to provide a method for producing a vermipreparation with a higher content of biologically related trace elements. The problem is achieved in that at the first stage of the worms are placed in physiological saline. Due to the hyperosmoticity of the physiological saline with respect to earthworms, the latter lose about 30% of their mass and, as a result, absorb aqueous solutions of metals more intensively. Then, using the express bioassay methods developed by us (RF patent No. 2215411), by estimating the rate of burrowing of earthworms into the soil or by the length of time they leave the illuminated cage for shading, they experimentally find the most optimal concentration of metal salts in water (they should be close to maximum, but not lead to the death of earthworms, the deterioration of their vital functions, damage to the skin as a result of osmotic reactions).
The method is as follows. Worms taken from the soil are thoroughly washed in several sterile waters. Withstand for 24 hours without food in 0.5 l jars with wet filter paper to free the intestines from soil lumps. After this, earthworms are transferred for 10 minutes in saline. Then:
- пример 1: до 2-х суток выдерживают в водном растворе соли ZnSO4 или до 10-ти суток в почвенном субстрате, пропитанном водным раствором этой соли, при концентрации, не вызывающей подавление физиологической активности червей (таблица 1);- example 1: they are kept in an aqueous solution of ZnSO 4 salt for up to 2 days or up to 10 days in a soil substrate saturated with an aqueous solution of this salt at a concentration that does not suppress the physiological activity of worms (table 1);
- пример 2: до 2-х суток выдерживают в водном растворе соли K2Cr2O7 или до 10-ти суток в почвенном субстрате, пропитанном водным раствором этой соли, при концентрации, не вызывающей подавление физиологической активности червей (таблица 2);- example 2: up to 2 days they are kept in an aqueous solution of K 2 Cr 2 O 7 salt or up to 10 days in a soil substrate saturated with an aqueous solution of this salt at a concentration that does not suppress the physiological activity of worms (table 2);
- пример 3: до 2-х суток выдерживают в водном растворе соли CuSO4 или до 10-ти суток в почвенном субстрате, пропитанном водным раствором этой соли, при концентрации, не вызывающей подавление физиологической активности червей (таблица 3);- example 3: they are kept in an aqueous solution of CuSO 4 salt for up to 2 days or up to 10 days in a soil substrate soaked in an aqueous solution of this salt at a concentration that does not suppress the physiological activity of worms (table 3);
After this, live worms are cut into pieces 2-3 cm long, dried for about a day in a well-ventilated place, then about 10 hours in an oven at 50 ± 5 ° C. The dried worms are crushed in a sterile dry mortar to obtain a powder. The resulting preparation is stored in a refrigerator in tightly closed vials.
Studying the accumulation of zinc in earthworms, using the above methods and keeping the worms in aqueous ZnSO 4 solutions, the following results were obtained:
In soil with a K 2 Cr 2 O 7 content of 100 mg / kg, 10-, 20-, and 30-day exposure of earthworms was carried out. The results of determining the concentration of chromium in the tissues of earthworms:
When the content of CuSO 4 in the soil in the amount of 400 mg / kg received the following:
The proposed method allows to obtain vermipreparations with a high content of trace elements, and therefore with higher physiological properties.
