Abstract
Description
The invention relates to the field of biotechnology and can be used in the production of highly immunogenic, storage-stable vaccines for the prevention of rabies in cattle and small cattle.
Вакцина антирабическая сухая для крупного и мелкого рогатого скота, включающая антирабический антиген, среду культивирования, защитную среду высушивания на основе пептона, сахарозы и желатина, а также стимулятор иммунитета сапонин, отличающаяся тем, что в качестве антирабического антигена и депонирующего компонента сухая вакцина содержит гидрат окиси алюминия (ГОА) с адсорбированным на нем инактивированным вирусом бешенства при следующем соотношении компонентов (мас.%):Dry anti-rabies vaccine for cattle and small cattle, including anti-rabies antigen, cultivation medium, protective drying medium based on peptone, sucrose and gelatin, as well as saponin immunity stimulator, characterized in that the dry vaccine contains oxide hydrate as an anti-rabies antigen and depositing component aluminum (GOA) with the inactivated rabies virus adsorbed on it in the following ratio of components (wt.%):
причем масса сухой вакцины составляет в среднем 10% от массы жидкой вакцины, получаемой при восстановлении сухой вакцины до первоначального объема стерильной дистиллированной водой.moreover, the weight of the dry vaccine is on average 10% of the mass of the liquid vaccine obtained by reconstituting the dry vaccine to the original volume with sterile distilled water.
Гидрат окиси алюминия в составе заявляемой сухой вакцины создает депо антирабического антигена и этим пролонгирует его специфическое возбуждающее воздействие на иммунную систему организма крупного рогатого скота, что приводит к более длительной циркуляции в крови животных антител с активностью, близкой признанному ВОЗ лимиту надежности - 0,5 МЕ/мл (табл.2).The alumina hydrate in the composition of the inventive dry vaccine creates a depot of the rabies antigen and thereby prolongs its specific stimulating effect on the immune system of the cattle, which leads to longer circulation of antibodies in the blood of animals with activity close to the WHO recognized reliability limit of 0.5 IU / ml (table 2).
Она способна защищать животных от вируса бешенства, введенного непосредственно в мишень поражения (мозг), в дозе, многократно превышающей летальную (табл.3).The inventive vaccine manifests itself as a highly immunogenic drug also in experiments on sheep and goats. It is able to protect animals from rabies virus, introduced directly into the target of the lesion (brain), in a dose many times higher than the lethal one (Table 3).
Включенный в состав заявляемой вакцины адсорбированный на ГОА вирус бешенства при сублимационном высушивании практически полностью сохраняет первоначальную иммуногенную потенцию (93,7-96,4%), тогда как вакцина, высушенная без ГОА (аналог), теряет 16,7-31,3% иммуногенности (табл.1).Included in the composition of the claimed vaccine adsorbed on GOA rabies virus during freeze-drying almost completely retains the original immunogenic potency (93.7-96.4%), while the vaccine dried without GOA (analogue) loses 16.7-31.3% immunogenicity (table 1).
Предлагаемая вакцина обладает большей иммуногенностью и, следовательно, будет с большей эффективностью защищать целевых животных от бешенства (3 табл.).The proposed vaccine has greater immunogenicity and, therefore, will more effectively protect target animals from rabies (3 tab.).
The invention relates to the field of biotechnology and can be used in the production of highly immunogenic storage-stable vaccines for the prevention of rabies in cattle and small cattle.
11.04.80).A known method for producing an anti-rabies vaccine, according to which the finished dry preparation contains the rabies virus Schelkovo-51 inactivated by B-propiolactone, reproduced in the culture of transplantable cells of BHK-21, components of the culture medium and protective drying medium based on peptone, sucrose, gelatin ( RF patent 965049, C12N 7/08 // C12R 1/91, claimed 11.04.80).
This vaccine is produced on an industrial scale and is used in Russia and the CIS countries for the prophylactic immunization of dogs, cats, horses, pigs and other animals against rabies. Its composition has remained unchanged for 15 years, although the technological methods for obtaining a seed viral pool, reproduction of BHK-21 cells and reproduction of the rabies virus of the Schelkovo-51 strain in them are changing and improving (RF patents 2191600, 2002; 2287343, 2006. )
Its disadvantage is that due to the lack of immunogenicity for cattle and small cattle, its use for these animals is possible only in the presence of a special adjuvant solvent.
The closest analogue of the claimed vaccine is a vaccine for the prevention of rabies in cattle, including the rabies antigen in the form of an inactivated rabies virus in the culture medium, a protective drying environment based on peptone, sucrose, gelatin, as well as an immunity stimulator saponin (RU 955577, A61K 39/205 , registered April 22, 1993). For the manufacture of the vaccine, β-propiolactone inactivated rabies virus Schelkovo-51 strain reproduced in the cell culture of BHK-21 is used. Before use, the dry vaccine is restored to its original volume with distilled water. Animals are vaccinated subcutaneously in a dose of 5.0 ml twice.
The disadvantage of the analogue is that during the initial single vaccination of cattle, post-vaccination anti-rabies antibodies almost completely disappear from the bloodstream of animals 11-12 months after the introduction of the vaccine (Table 2). In addition, when freeze-drying, the analogue loses a significant part (16.7-31.3%) of the immunogenic potency (Table 1). Its use for immunization of small cattle is not provided.
The objective of the invention is to improve the dry vaccine for the prevention of rabies in cattle and small cattle by eliminating these disadvantages.
The technical result of the invention is to maintain the immunogenic potency of the rabies vaccine during freeze-drying (Table 1), to lengthen the circulation of post-vaccination antibodies in cattle at a level close to the reliability limit (Table 2), and also to obtain evidence the high antibody-inducing activity of the claimed vaccine and its ability to effectively protect small cattle (sheep and goats) from rabies virus at the level of the central nervous system (table 3). These results were obtained due to the inclusion in the composition of the proposed dry vaccine, before drying, alumina hydrate (GOA) with rabies virus adsorbed on it.
Вакцина антирабическая сухая для крупного и мелкого рогатого скота, включающая антирабический антиген, среду культивирования, защитную среду высушивания на основе пептона, сахарозы и желатина, а также стимулятор иммунитета сапонин, отличающаяся тем, что в качестве антирабического антигена и депонирующего компонента сухая вакцина содержит гидрат окиси алюминия (ГОА) с адсорбированным на нем инактивированным вирусом бешенства при следующем соотношении компонентов (мас.%):SUMMARY OF THE INVENTION Dry anti-rabies vaccine for cattle and small cattle, including anti-rabies antigen, cultivation medium, protective drying medium based on peptone, sucrose and gelatin, as well as saponin immunity stimulator, characterized in that the dry vaccine contains oxide hydrate as an anti-rabies antigen and depositing component aluminum (GOA) with the inactivated rabies virus adsorbed on it in the following ratio of components (wt.%):
причем масса сухой вакцины составляет в среднем 10% от массы жидкой вакцины, получаемой при восстановлении сухой вакцины до первоначального объема стерильной дистиллированной водой.moreover, the weight of the dry vaccine is on average 10% of the mass of the liquid vaccine obtained by reconstituting the dry vaccine to the original volume with sterile distilled water.
Alumina hydrate, included in the composition of the inventive dry rabies vaccine, in combination with the inactivated rabies virus adsorbed on it, increases the immunogenic potency of the drug and lengthens the circulation period of post-vaccination antibodies in the bloodstream of the cattle (Tables 1, 2) with an activity of 0.5 IU / ml, which is considered sufficient to protect animals and humans from rabies (8th WHO expert report on rabies, Geneva, 1994). The inventive vaccine protects sheep and goats from death even when they are directly infected with rabies virus in the brain, at doses many times higher than the lethal dose (Table 3). Therefore, its use will more effectively protect cattle and small cattle from rabies.
The invention is illustrated by the following examples. The cultivation of rabies virus strain of Schelkovo-51 strain VNK-21 is carried out by roller and pseudosuspension (on microcarriers Cytodex 2) methods. The collection of virus-containing culture fluid is carried out after 96 and 120 hours of cultivation. The result is three series of virus-containing material, part of which is used for the manufacture of vaccines of the claimed (3 series) and known (3 series) composition.
Preparation of the claimed vaccine. To 4 liters of virus-containing culture fluid cooled to 4 ° C, 1 ml of β-propiolactone is added with constant stirring, and 2 hours after this, 600 ml of 6% alumina hydrate are added. After thorough mixing, the mixture is placed in a refrigerator at 8-10 ° C for 24 hours. Then 600 ml of the supernatant are removed from the carrier, and 2 liters of sterile drying medium, which is an aqueous solution of peptone (9-10%), sucrose (11-12%) and gelatin (2.7-3.0%, are added to the remainder of the mixture. ) To 6 liters of the prepared mixture, 61 ml of a sterile 10% aqueous solution of saponin is added, the adjuvant activity of which in the composition of rabies vaccines in cattle and small cattle appears in the range of 1.5-3.0 mg / dose. The result is a series of liquid vaccines containing 1.0 mg / ml saponin and 10% 6% alumina hydrate with the rabies virus Schelkovo-51 adsorbed on it. In two other series, the concentration of 6% GOA was adjusted to 15%. For the manufacture of an analogue, the same virus and components are used as for the manufacture of the claimed vaccine, only without the inclusion of GOA.
The prepared series of liquid vaccines (of the claimed and the analogue) are poured into 10 ml into vials of 20 cm 3 and subjected to freeze-drying. Dry vaccines, as well as three series of liquid vaccine analogues, taken before drying, are examined by the method of the US National Institutes of Health - NIH recommended by WHO in order to assess their immunogenic potency in international units (ME), as well as to determine the persistence of vaccine immunogenicity during the drying process.
As can be seen from table 1, the vaccine of the proposed composition has a greater immunogenic potency, since it almost does not lose its potential during the drying process, unlike the analogue vaccine, which loses 16.7-31.3% of its immunogenicity when dried.
Вируснейтрализующую активность антирабических антител выражают в международных единицах (МЕ/мл).In experiments on cattle using the inventive vaccine with an immunogenic potency of 3.4 IU / ml, a vaccine analogue with activity of 3.5 IU / ml. Dry vaccines are dissolved before use with sterile distilled water (10 ml per vial) and administered to calves weighing 230-260 kg subcutaneously in the neck at a dose of 2.0 ml. Before vaccination, 45 and 350 days after it, blood serum is obtained from animals and tested in the neutralization reaction against the standard rabies virus strain CVS relative to the international serum standard. The neutralizing activity of anti-rabies antibodies is expressed in international units (IU / ml).
The data presented in table 2 indicate that the claimed vaccine provides a longer circulation of post-vaccination anti-rabies antibodies in the body of cattle with activity close to 0.5 IU / ml, recognized by the WHO as sufficient to protect animals and humans from rabies (8th expert report WHO on rabies, Geneva. 1994). Therefore, it will more reliably protect these animals from rabies.
The inventive vaccine proved to be a highly immune preparation, also in experiments on sheep and goats. She was able to protect these animals from death even with the direct introduction of rabies virus into the brain at doses many times higher than lethal (Table 3). Its use will more effectively protect not only cattle, but also small cattle from rabies virus.
