State of the art
Receptor tyrosine kinases (PTKs) are key regulators of intercellular communication that control cell growth, proliferation, differentiation, survival, and metabolism. About 20 different PTK families with the same structure have been identified, namely, the extracellular ligand binding site, the transmembrane region, and the intracellular tyrosine kinase domain [1]. Extracellular binding of ligands causes dimerization of the receptor or stabilizes it, which leads to an increase in the kinase activity of PTK. The intracellular catalytic domain exhibits the highest degree of conservatism among PTK and includes an ATP-binding site that catalyzes receptor autophosphorylation at the tyrosine cytoplasmic residues, which serve as a landing site for proteins containing a homology domain with Src 2 (SH2) and a phosphotyrosine (PTB) binding domain, like Grb2, She, Src, Cbl, or Cγ phospholipase. These proteins subsequently attract additional effectors containing SH2, SH3, and PTB domains to the activated receptor, as well as a homology domain with plectrin (PH), which leads to the assembly of signal complexes on the membrane and activation of the cascade of intracellular biochemical signals. The most important cascades of downstream signals activated by PTK include the Ras pathway of the extracellular signal-regulating kinase (ERK) - mitogen-activated (MAP) kinase, the phosphoinositide 3-kinase (
PTK activity and their mediated cellular signaling are well coordinated and strictly controlled in normal cells. Imbalance of the PTK signaling system, both during stimulation with a growth factor and with genetic changes, leads to a violation of the regulation of tyrosine kinase activity. Such disturbances usually lead to the fact that the kinase activity, and thereby the signaling ability of PTK, becomes constitutive (constant) or increases significantly, which leads to malignant degeneration of the cell. Therefore, they are often associated with human cancer, as well as with other hyperproliferative diseases, such as psoriasis [2]. The most important mechanisms causing constitutive PTK signaling include overexpression and / or amplification of PTK genes, such genetic changes as deletions and mutations in the extracellular domain, as well as changes in the catalytic center, or autocrine-paracrine stimulation through abnormal systems of growth factors.
[8].For example, in many types of cancer in humans, gene amplification and / or overexpression of PTK occurs, which can increase the susceptibility of cancer cells to normal levels of growth factors. In addition, overexpression of specific PTK on the cell surface increases the frequency of receptor dimerization even in the absence of an activating ligand. In many cases, this leads to constitutive activation of PTK, which leads to abnormal and uncontrolled cell proliferation and the formation of tumors. An important example of this scenario is HER2, also known as ErbB2, which belongs to the PTK family of epidermal growth factor receptor (EGFR). HER2 overexpression was detected in various types of human cancer, especially in carcinomas of the breast and ovaries [3]. It is very important that abnormally high levels of HER2 correlate with more aggressive disease progression and a shorter life span of patients [4]. EGFR, the first receptor tyrosine kinase that has been cloned at the molecular level [5], also plays a significant role in oncogenesis. EGFR is often overexpressed in non-small cell carcinoma of the lung, bladder, cervix, ovary, kidney, and pancreas, and in squamous cell carcinoma of the head and neck [6]. The predominant mechanism leading to superexpression of EGFR is the amplification of EGFR genes, with up to 60 copies per cell in certain tumors [7]. Typically, increased levels of EGFR expression are associated with a high frequency of metastasis and increased tumor proliferation [8].
[9].Since tyrosine kinases have been implicated in various cancer symptoms, PTK and the signaling cascades they activate are promising areas for developing cancer-selective drugs that are selective for these targets. One approach to inhibiting abnormal PTK signaling is the development of low molecular weight drugs that selectively suppress tyrosine kinase activity inherent in PTK and, thereby, block receptor autophosphorylation and activation of downstream signal transducers [9].
п.In order to identify PTK-specific inhibitors, several methods for screening compound libraries have been developed, most of which use biochemical methods [10]. One of the important considerations when choosing effective tyrosine kinase inhibitors is that these compounds must penetrate cell membranes and function in the intracellular environment for the required time. In addition, in order to become a potential prototype of a drug, kinase inhibitors should not exhibit cytotoxic effects. Therefore, it is desirable to have a cell system for primary screening of compounds capable of inhibiting PTK activity. The requirements for such in vivo methods are the ability to study a specific cellular process triggered by a specific target, and in a tool for easily measuring its result in a high-throughput screening system (HTS). The presence of an increasing number of biotechnology tools for the genetic modification of cells and microorganisms has allowed the development of simple digital methods for the analysis of cellular processes that can be easily applied to automated systems with HTS [11-14]. Cellular screening should ideally be carried out on cells from humans, which, obviously, are the physiologically most reasonable model system. However, it may turn out to be difficult to control the effects of excess processes on the measured result and distinguish them from those effects that should be specific to a given target; and genetic manipulation on mammalian cells is usually problematic and time consuming. Moreover, culturing human cells is expensive and can be difficult to grow in automated systems for HTS. Microorganisms such as yeast provide a convenient alternative for measuring the activity of certain human proteins, albeit in a heterologous, but cellular (eukaryotic) environment. In yeast cells, the functioning of human proteins can often be restored and some aspects of human physiological processes can be reproduced due to the high degree of conservatism of the basic molecular and cellular mechanisms between yeast and human cells [14-17]. The fact that many human proteins function in yeast means that the conformation required for them, stability, protein-protein interaction, etc., takes place in this eukaryotic organism.
Although methods for isolating receptor tyrosine kinase inhibitors already exist, there is a need for a reliable cellular method for identifying and / or testing such receptor tyrosine kinase inhibitors that penetrate cell membranes and are not cytotoxic.

The implementation of the invention
The present invention provides a cell system for identifying and / or testing specific receptor tyrosine kinase (PTK) inhibitors, preferably human PTK. In this system, recipient cells, preferably yeast cells expressing specific PTK fragments under certain conditions, can grow only when protein kinase activity is inhibited or inactivated. This positive feature allows the selection of specific kinase inhibitors, which should also be soluble, stable and not cytotoxic.
Анализ экстрактов клеток методом Western-блот-гибридизации с помощью антител к фосфотирозину показывает, что эффект ингибирования роста PTK, такими как, например, c-Met и PDGFRβ, коррелирует с их способностью катализировать фосфорилирование тирозина (фиг.2b).In the cell system of the present invention, the tyrosine kinase domain of PTK or its functional fragments, preferably the cytoplasmic domain of PTK or its functional fragments, are expressed in the cytoplasm of the recipient cells either “as is” or as fusion peptides having known protein sequences that form strong dimers. The expression of these proteins in cells, preferably yeast cells, is preferably controlled by inducible promoters such as the GAL1 gene promoter. Expression of dimerizable PTK derivatives significantly reduces the growth of yeast cells under selective conditions (FIGS. 1a and 1b). Such growth inhibition is caused by dimerization-dependent activation of tyrosine kinase activity, since the introduction of a point mutation into the active center, which eliminates the tyrosine kinase function of PTK, removes the effect of growth inhibition (Figs. 1a and 1b). Analysis of cell extracts by Western blot hybridization using antibodies to phosphotyrosine shows that the effect of inhibiting the growth of PTK, such as, for example, c-Met and PDGFRβ, correlates with their ability to catalyze tyrosine phosphorylation (Fig.2b).
Встраивание активных цитоплазматических доменов PTK в цитозольную часть клеточных мембран, по-видимому, не существенно для фосфорилирования тирозина и эффектов ингибирования роста, так как не наблюдается различия между экспрессией простых цитоплазматических доменов PTK и тех же последовательностей, несущих сигнал миристоилирования для заякоривания в мембране (фиг.1a, 1b и 2a).Expression of PTK peptides, including the transmembrane domain or its functional fragment, in particular the transmembrane domain of PTK, and the signal peptide for localizing the N-terminal portion of the protein in the lumen of the endoplasmic reticulum (ER) and extracellular space, completely eliminates the effect of growth inhibition (figa and 1b ), while it does not reduce the ability of transmembrane PTK to catalyze tyrosine phosphorylation (Fig.2b). The incorporation of active cytoplasmic PTK domains into the cytosolic part of cell membranes is apparently not essential for tyrosine phosphorylation and growth inhibition effects, since there is no difference between the expression of simple cytoplasmic PTK domains and the same sequences carrying a myristoylation signal for anchoring in the membrane (Fig. .1a, 1b and 2a).
In the prior art, it has been shown that expression of the full-length cytoplasmic tyrosine kinase c-src in S. pombe results in cell death [18], and pp60v-src expression in S. cerevisiae leads to cessation of growth [19]. The pp60v-src mutants lacking a functional N-terminal myristoylation signal cause only partial suppression of growth arrest [19], i.e. complete cessation of growth caused by expression of pp60v-src in S. cerevisiae can be observed only when the pp60v-src kinase enters the cell compartment natural for it. The authors of the present invention, in contrast to the prior art, found that expression of full-sized PTK in yeast cells does not lead to growth arrest, i.e. the presence of PTK in the natural cell compartment (transmembrane localization) in yeast cells does not stop growth.
Ингибирование роста под действием другой PTK (напр., RET), на которую не влияет специфический ингибитор киназ Gleevec, не устранялось в присутствии этого соединения (фиг.3).In a typical embodiment of the present invention, the addition of a specific tyrosine kinase inhibitor imatinib mesylate (Gleevec ®, Novartis) a modified yeast cells expressing dimerized fragment PTK PDGFRβ, leads to blocking dimerization-dependent kinase activity of the PTK and to restore growth under selective conditions (Figure 3 ) Growth inhibition by another PTK (eg, RET), which is not affected by the specific Gleevec kinase inhibitor, was not eliminated in the presence of this compound (FIG. 3).
Since bacterial and yeast cells do not have endogenous tyrosine kinases of the same type as mammals, this cell system offers a zero background advantage for the expression of PTK and screening for specific inhibitors of these membrane kinases, which creates a privileged situation that cannot be achieved on mammalian cells.
Sambrook et al., Molecular Cloning: A Laboratory Manual. New York: Cold Spring Harbour Laboratory, 2001.The peptide of the present invention can be expressed from an extrachromosomal gene construct, for example, from an episomal vector that allows expression of a fusion protein in recipient cells. A nucleic acid construct encoding a fusion peptide can be integrated into the genome of recipient cells. The nucleic acid can be introduced into the cell by any transfection method, resulting in the capture of the nucleic acid sequence by the cell. Such methods are known to those skilled in the art and are described, for example, in Sambrook et al., Molecular Cloning: A Laboratory Manual. New York: Cold Spring Harbor Laboratory, 2001.

These conditions depend on the genetic constructs and recipient cells used.
There are at least three different categories of compounds that can be screened by the screening method of the present invention: chemical libraries, natural product libraries, and combinatorial libraries. Chemical libraries consist of structural analogues of known compounds. Natural product libraries are collections of microorganisms, animals, plants or marine organisms that are used to form screening mixtures, for example, by fermenting and extracting culture fluid from soil, plant or marine microorganisms, or by extracting plants or marine organisms. Combinatorial libraries consist of a large number of peptides, oligonucleotides or organic compounds in a mixture. They can be relatively easily obtained, for example, by traditional methods of synthesis, PCR or cloning.
The invention will now be described in more detail using examples.
В отличие от изолированных цитоплазматических доменов этих PTK, включение соответствующих трансмембранных доменов этих белков, вместе с сигнальным пептидом для локализации в ER и секреции, вызывало устранение эффекта ингибирования роста клеток (фиг.1a и 1b, дорожки 2 и 5).The results of this experiment are presented in FIG. 1 and FIGS. 2a and 2b. Cell growth and colony formation on glucose plates that repress PTK genes are shown in the left row. Cell growth and colony formation on galactose plates, on which the induction of PTK genes occurs, are presented in the right row. Expression of the active cytoplasmic domains of PTK c-Met and PDGFRβ causes kinase-dependent inhibition of yeast growth (FIGS. 1a and 1b,
Suc2 (SS).For constitutive activation of c-Met and PDGFRp, the heterologous dimerization domain was fused with the cytoplasmic domains of c-Met and PDGFRβ or with longer derivatives of c-Met and PDGFRβ, including the natural transmembrane domain. In one reading frame, the c-Jun leucine fastener or the c-fos leucine fastener or to the N-ends of the cytoplasmic c-Met domains (amino acid residues, amino acid residues 932-1366) and PDGFRR (ai 524-1067) were sewn, or to the N-ends of the constructs for c-Met (a.o. 905-1366) and PDGFRβ (a.o. 496-1067) bearing transmembrane domains. To analyze the expression of constructs in yeast cells, an HA or Myc epitope was inserted between the fusion site of the dimerization domain and the kinase domain. The cytoplasmic constructs were imparted with membrane localization using a myristoylation signal (black column anchored in the membrane, FIG. 1). In order to ensure proper secretion of fusion proteins containing transmembrane domains, the yeast signal sequence Suc2 (SS) was sutured at the N-ends of the dimerization domains.
Как и ожидалось, мутанты с инактивированной киназой не проявляли фосфорилирования тирозина (фиг.2b, дорожки 3 и 6).The expression of these fusion proteins was investigated by SDS-PAGE followed by Western blot hybridization using anti-Myc antibodies (Fig. 2a). All of these hybrid proteins carrying the Myc epitope for detection, the c-Fos leucine zipper for heterodimer formation, and the Myroylation signal (Myr) for anchoring in the membrane were expressed in the same way as similar constructs carrying the HA epitope and the other c-Jun leucine zipper instead of functionally equivalent sequences of marked proteins. Protein phosphorylation (PTK autophosphorylation and transphosphorylation of unknown substrate proteins) was detected by Western blot hybridization using anti-pTyr antibodies (Figure 2b). Despite the fact that c-Met and PDGFRβ derivatives bearing transmembrane domains (TM) were expressed and possessed activity at a level comparable to the isolated cytoplasmic domains of these PTKs (cf.
3.The results of this experiment are presented in figure 3.
RET.Selection for specific inhibition of receptor tyrosine kinase activity was carried out on yeast. The expression of human PDGFRβ and RET receptor tyrosine kinases in yeast causes a strong slowdown in cell growth, judging by measurements of light scattering at 600 nm in both cell cultures. The addition of the kinase inhibitor imatinib mesylate (Gleevec ® , Novartis), which inhibits PDGFRp, but not RET, at a concentration of 50 μM in yeast cultures led to the resumption of growth of PDGFRβ-expressing yeast cells, but not the growth of cells expressing RET.
