Abstract
Description
The invention relates to medicine, namely to plastic reconstruction of damaged bone tissue.
Calcium phosphate cements are prepared on the basis of a reaction-hardening mixture of powders of a mixture (RPS) from two or more calcium phosphates and a solidifying liquid (GC). The starting powder is a mixture of acidic and basic calcium phosphates. When GIs are added to the mixture, the components begin to interact with each other through the liquid phase by the dissolution-precipitation mechanism with the formation of neutral (pH ~ 7) phosphates. As the initial mixture (V.V. Samuskevich, N.Kh. Belous, L.N. Samuskevich, A.A. Dobryshevskaya. “Water mixing based on hydroxyapatite and heat-treated calcium dihydrogen phosphate.” Inorganic Materials, 2000, vol. 36 , No. 9, p.1148-1152) used a mixture of powders of HA and calcium dihydrogen phosphate, water was used as a coolant. Upon the addition of a shutting fluid between the components of the RPS, an interaction occurs with the formation of an amorphous phase, which in the process of setting passes into crystalline HA.
The proposed materials can be used as cement pastes for filling bone maxillofacial and dental defects. The disadvantage of these materials is low strength.
The closest in technical solution is the material for the replacement of defects in bone tissue (patent RU 228112 C1 from 08/10/06, sections: A61K 6/033, A61L 27/00), consisting of a mixture of powders of calcium hydroxide and monosubstituted potassium phosphate, as a fatty acid - a solution of magnesium phosphates in phosphoric acid. When a mixture of powders is mixed with a coolant, a dough-like mass is formed, which over time sets to form a strong cement stone. A significant drawback of this material is its low crack resistance (0.1 MPa · m 0.5 ) and strength (30 MPa), which does not allow its use to eliminate defects in complex areas of bone tissue.
The technical result of the invention is to increase the strength of calcium phosphate cement material. To achieve a technical result, it is proposed to use as a RPS a mixture of powders of HA, TKF, tetracalcium phosphate (TTKF) with the addition of titanium powder, as a ZH solution of magnesium, potassium and sodium phosphates in phosphoric acid, which can significantly increase the crack resistance and strength of a HA-based cementitious material .
Cement consisting of a powder mixture: GA, TTKF and TKF (RPS) and additives - titanium powder and ZH on the basis of magnesium, sodium and potassium phosphates - is not known. The content in the RPS GA is 0-80 wt.%, TTKF 0-100 wt.%, TKF 0-100 wt.%. The content of the additive (titanium powder) 5-60 wt.% In a mixture of RPS-titanium.
The ratio of the amount of introduced coolant liquid ZH (ml) to the amount of the mixture of RPS-titanium (g) should be in the range 0.45-0.75 (ZZ (ml) / RPS - titanium (g) = 0.45-0.75). The setting time ranged from 2 to 40 minutes depending on the amount and composition of the coolant, the ratio of calcium phosphates in the RPS and the amount of additive (titanium powder). After the addition of the coolant to the RPS-titanium mixture, the liquid phase reacts with the RPS, while the RPS is partially dissolved, followed by precipitation in the form of a strengthening amorphous or crystalline phase. In the process of setting, a structure is formed consisting of crystals of calcium phosphates and titanium particles, which are coated with layers of a hardening cementing phase, which ensures strong adhesion of the particles to each other. The introduction in amounts of more than 80 wt.% HA and less than 20 wt.% TTKF or TKF in RPS, as well as the use of more than 60% of the addition of titanium powder in a mixture of RPS-titanium, leads to a quick setting of the hardening mixture and its poor formability due to high viscosity , which makes it difficult to use this material to close bone defects. In addition, the content of more than 60 wt.% Titanium in the mixture of RPS-titanium leads to softening of the material to values of strength less than without titanium-containing cement. When using less than 5 wt.% Titanium powder, an increase in crack resistance of cement samples is not achieved. In the case of using a shutting fluid in an amount less than the lower limit (GJ (ml) / solid (g) <0.45) or the use of highly concentrated solutions of GJ with a magnesium phosphate content of more than 75 wt.% And a total content of potassium and sodium phosphate of more than 25% , the resulting mixture has a high viscosity, which leads to the formation of numerous cracks during molding of the product of the desired configuration. When using a coolant in an amount above the upper limit (coolant (ml) / CPS (g)> 0.75) and dilute solutions with a high water content of more than 45 wt.%, Magnesium phosphate content of less than 15 wt.%, The mixture is too liquid that does not allow to form products due to spreading of the mixture. In addition, the setting time is significantly increased, which leads to a decrease in strength, especially in the first minutes of hardening. With the introduction of a total content of potassium phosphate and sodium of less than 3.5 wt.%, The porosity of the samples increases, which leads to a sharp decrease in strength.
0,5.An example of obtaining sample No. 1. An RPS powder containing 15 g of HA, 10 TCF and 15 g of TTFC is mixed with an additive of 10 g of titanium powder in a planetary mill with corundum balls for 10 minutes. The resulting mixture of RPS - titanium in an amount of 0.4 g is mixed with 0.25 ml of ZH (65 wt.% Magnesium phosphate and 10 wt.% Potassium phosphate, 10 wt.% Sodium phosphate, phosphoric acid 8 wt.%, 7 wt. % water). Mixing is carried out for 1-2 minutes with a metal spatula on the glass to a sour cream-like state, after which the mixture is placed in a cylindrical mold with a diameter of 8 mm After several minutes, the molded sample is taken out and placed in a thermostat at a temperature of 37 ° C in a solution of SBF (Simulated Body Fluid), corresponding to human blood plasma. After 24 hours, the cured sample has a compressive strength of 120 MPa and a crack resistance of 0.8 MPa · m 0.5 .
