Abstract
Description
The invention relates to biotechnology and can be used in biology, pharmacology, cosmetic industry, veterinary medicine and food industry.
Preparations based on liposomes or vesicles can be used in biology, pharmacology, and the food industry to create systems for the targeted transport of biologically active substances into cells, in particular, to increase the therapeutic activity of drugs.
Preparations based on phospholipid vesicles (FLV) appeared relatively recently and in a short time became widespread, first in medicine and then in cosmetics. The effectiveness of exposure to FLV is due to the possibility of their deep penetration into tissues. In this way, in-depth delivery of not only lipids useful to the body, of which the membrane of microcapsules usually consists, but also other substances that are in their internal volume, can be ensured.
Depending on the production method, phospholipid vesicles (FLV) can have different layers (one or more lipid bilayers). In cases where it is necessary to enclose a substance with a high molecular weight in a liposome (for example, when solving genetic engineering problems when incorporating DNA molecules), large liposomes, including multilayer ones, are used. In other cases, small single-layer vesicles are usually preferable.
A 61 K 9/127, 1991). From previously developed methods for producing liposomes, a method for producing a liposome composition is known (see RF patent N 2014070, class A 61 K 9/127, 1991).
The disadvantage of this method is that it does not provide liposomes having small sizes. In addition, the known method is characterized by considerable complexity.
There is also known a method for producing a liposomal composition (see European patent N 0130577, class A 61 K 9/50, 1989), comprising mixing the components of the liposome membrane with a water-soluble non-volatile physiologically acceptable organic solvent and dispersing the resulting mixture in an aqueous medium using a conventional mixer.
The liposomes obtained by this method have relatively large sizes ≈ 2500 nm, which is the main disadvantage of the known method.
The closest in technical essence to the present invention is a method for producing a liposomal composition, comprising forming a mixture containing at least one component of the liposomal membrane, water and an additive, dispersing the mixture, reduces to the fact that either submicro powder of a substance insoluble in water and in an organic solvent, or water-swellable polymers, or a liquid not miscible with water and other components of the mixture (RF patent N 2104691, MKI 7 A 61 K 9/127, 9/50, 9/66, BI N 5, 1998 year )
а) для стабильной воспроизводимости метода необходимо специальное оборудование (перемешивающие устройства с мощными двигателями или рамочными мешалками, проточный режим перемешивания;
б) строгий отбор всех липидных компонентов, чтобы избежать их агрегации в кислой среде;
The disadvantage of this method is that with industrial scaling there are a number of difficulties:
a) for stable reproducibility of the method, special equipment is needed (mixing devices with powerful motors or frame mixers, flow-through mixing mode;
b) strict selection of all lipid components in order to avoid their aggregation in an acidic environment;
c) the use of distilled water only, since the system is very sensitive to the presence of salts, even which are present in micro-quantities in water-alcohol extracts.
In addition, in some cases, the additive must be removed where aerosil or perfluorodecane is used. The second variant of the method, where a swollen sacap polymer is used, the vesicles are initially in an acidic medium, when the medium is neutralized, the polymer unfolds, which provides a gel-like substance. This is convenient for certain tasks, but does not provide versatility.
The objective of the invention is to obtain phospholipid vesicles with certain parameters when using industrial dispersing devices. Siliconorganolipid single-layer microcapsules containing biologically active substances (BAS) and possessing aggregative stability in media with a pH range from 5.0 to 9.0 and a salt content in the aqueous phase of up to 0.1% (mainly Ca ++ and Mg ++ ) are highly effective and universal for the creation of medical, cosmetic preparations.
As a result of using the present invention, it becomes possible to obtain a more perfect basis for the preparation of medical, cosmetic, pharmacological, veterinary, dental and other drugs.
The technical result is achieved by the fact that before mixing the lipophilic and hydrophilic components, organosilicon polymer compounds are introduced into the lipid fraction in acceptable quantities, and the dispersion is carried out in a rotary cavitation unit, which has high shear forces and powerful sonar and ultrasonic sonar phenomena that provide fine particle redistribution at the time of their passage through the installation.
Ethylene diamine tetraacetic acid (EDTA) products are added to the aqueous medium in micro amounts. Organosilicon compounds, for example, silicoctamethylcyclotetrasiloxane, or polyethylsiloxane, or alkylmethylsiloxane, are used as a stabilizer of the lipid phase, which makes it possible to achieve lecithin homogeneity, decrease its viscosity and facilitate its subsequent mixing with other lipids. The organosilicon compounds are added to the lipid phase in an acceptable amount before mixing. As a dispersing device, a rotary-cavitation installation (RKU) is used, which provides high-speed dispersion of a mixture of the aqueous and lipid phases with simultaneous low-frequency ultrasonic treatment. As a result, small single-layer vesicles are formed with an average size of 20-60 nm.
In the proposed method for producing silicon organolipid microcapsules for the creation of medical and cosmetic preparations, including the formation of a mixture containing the lipid phase, water and biologically active substances (BAS) in an aqueous medium, EDTA is added as a stabilizer in trace amounts. EDTA - Ethylenediaminetetraacetic acid is used in the form of disodium salt dihydrate (Trilon B) as a chelating agent, blood anticoagulant and food stabilizer, for the production of liquid soaps and shampoos, plant protection products, as a water softener / Chemical Encyclopedia. Scientific publication of the Great Russian Encyclopedia. M., 1999.V.5. S.988). The use of Trilon B as a softener of tap water and using the rotary-cavitacin dispersion method can produce products for medicine, cosmetics and veterinary medicine with lower energy costs (the distillate is very expensive) without losing quality. Organosilicon compounds are added in acceptable amounts as a stabilizer of the lipid phase, homogeneity of lecithin and the ease of its subsequent mixing with other lipids, and a rotary-cavitation unit, which provides high-speed dispersion of a mixture of the aqueous and lipid phase with simultaneous low-frequency ultrasonic treatment, is added as a dispersing device.
Silicon octamethylcyclotetrasiloxane, or polyethylsiloxane, or dimethicone are used as organosilicon compounds. Silicones are widely used to improve the organoleptic and aesthetic characteristics of cosmetics, in particular sunscreens. There is an assumption that, in addition to improving chemical characteristics, organosilicon compounds enhance the sun-protection properties of drugs with the active substance remaining unchanged. The use of a lecithin / silicone mixture in various ratios (from 1: 1 to 1: 0.25) makes it possible to obtain a uniform thin lipid film, which is very important in the formation of membrane structures.
The proposed method allows the use of lipids without organic solvents, due to the introduction of organosilicon compounds.
The essence of the invention is reduced to the use of a rotary-cavitation installation (ECU) to obtain small single-layer vesicles. The use of CGS allows you to combine two well-known methods for obtaining intensive dispersion MOB and a method for processing ultrasonic vibrations. RKU is a simple and reliable design, which combines a powerful generator of hydroacoustic vibrations and an efficient mixer, which allows to obtain fine emulsions of water-fuel mixtures.
The use of the rotor-cavitation treatment method to obtain silicon organolipid microcapsules made it possible to obtain large volumes of suspensions of vesicles of various concentrations by lipid and with different composition of biologically active substances in a very short period of time, 5-10 min of the unit operation provides 10 - 50 l of liposomal suspension. The installation allows metered through special holes to introduce both hydrophilic and lipophilic compounds, stop the process if necessary, change the MOB parameters by adjusting the time and temperature conditions.
RKU allows you to use the aqueous phase with various indicators of hardness (from distilled to tap).
The power of the installation allows the use of low purification water. To stabilize the lipid ingredients, a small amount of organosilicon compounds (up to 2%) is introduced into the fat mixture.

8. The installation comprises an electric motor 1, a container for introducing a lipid fraction 2, a container for introducing hydrophilic substances and solutions 3 with valves for regulating the feed rate of the lipid phase 10 and solutions of biologically active substances 9, a preliminary distribution chamber 4, a sounding chamber 5, a rotary cavitation mill 6, a formation zone cavitation phenomena 7, apparatus for the preparation of finished forms 8.
According to the first option, the problem is solved in that the method of obtaining a liposomal composition using RKU is carried out on distilled water with a certain amount of lipids, a strictly balanced composition. In this way, microcapsules are formed for therapeutic and prophylactic preparations using in dentistry.
Before mixing, organosilicon compounds are added to the lipid fraction in the container 2. Control valves 9 and 10 allow the lipids to be dosed into the pre-film redistribution chamber in the aqueous phase 4. Water-soluble ingredients are introduced simultaneously into the device 4 and enter the sounding chamber at a speed of 3000 rpm 5 The rotation of the rotor provides mechanical dispersion of the lipid mixture in the aqueous phase 5 and the formation of hydroacoustic and ultrasonic vibrations.
ECC provides two methods simultaneously: dispersion and ultrasound, shear phenomena provide a fine redistribution of particles at the time of their passage through the installation.

The wide possibilities of CGS can be used to increase the efficiency of existing serial production lines and newly created ones.
Depending on the specific features of the technological process, ECU allows you to choose the optimal values of performance, power and frequency of acoustic vibrations.

The proposed preparation technique allows one to form silicon organolipid microcapsules in a flow mode, to exclude the use of inappropriate additives, to minimize the use of organic solvents (only pure ethyl alcohol at a concentration of 0.5% or water-alcohol extracts allowed for use are used).
In certain cases, it is possible to use microscopic particles of water-insoluble chemical elements (for example, Ag, Zno, S) or other targeted additives.
The presence in the volume of a certain number of dispersing microparticles sharply increases the surface area on which the process of formation of liposomes can proceed.
As a result of the influence of hydroacoustic vibrations with mechanical dispersion, the process of liposome formation occurs. Qualitatively new results are achieved using ECG without the use of other special additives, and small single-layer vesicles were obtained, the size and ply of which were controlled by electron microscopy.
