Abstract
Description
The invention relates to an ion-exchange technology for the isolation of lysine from a culture fluid obtained in the process of its microbiological synthesis.
Lysine is an amino acid that is widely used in medicine, food industry and agriculture.
A known method of extracting lysine from the culture fluid, consisting in the fact that the solution of the culture fluid with a pH of 7 is passed through a column of carboxyl ion exchanger KB-4P-2 in the NH 4 form to achieve equilibrium. Every 100 g of ion exchanger sorb 6-8 g of lysine. The ionite is washed with water and lysine is desorbed with a 0.5-5.0% solution of NH 4 OH. In this case, 80-90% of lysine is desorbed and a 1-2% solution of lysine is obtained.
The proposed method allows to obtain lysine solutions of a higher concentration, to reduce the amount of equipment used, the amount of wastewater obtained, to simplify the process by eliminating the operation of washing the ion exchanger.
The method consists in the fact that a solution of lysine in the culture fluid is passed through a carboxylic ion exchanger in a 1: 1-4 mixed hydrogen-sodium form, followed by desorption with an alkaline reagent. Preferably, the desorption of lysine is carried out with a 14-20% sodium hydroxide solution.
The difference of the proposed method consists in carrying out sorption of lysine on a mixed in a ratio of 1: 1-4 hydrogen-sodium form of ion exchanger and desorption of lysine with a 14-20% sodium hydroxide solution.
It was established that a mixed in the ratio 1: 1-4 hydrogen-sodium form of carboxylic ion exchanger has a maximum specific capacity for lysine.
The decrease and increase in the content of Na-form in the ratio leads to a decrease in the specific capacity of the ion exchanger.
When lysine is desorbed with a sodium hydroxide solution, water is formed that flushes the ion exchanger from the culture fluid, thereby eliminating the special operation of washing the ion exchanger with water and reducing the amount of wastewater obtained.
The concentration of sodium hydroxide solutions is due to the production of more concentrated lysine solutions with complete desorption of lysine from ion exchanger.
П р и м е р 1. PRI me R 1. A solution of lysine in a culture fluid with a concentration of 40 g / l with a pH of 3.5 is passed through a column filled with a carboxyl ion exchanger KB-4 in H + , Na + form in a ratio of 1: 2, until equilibrium is reached . The ratio of the height of the ion exchanger layer to the diameter of the column is 50: 1. Every 100 g of ion exchanger sorb 13 g of lysine. Then lysine is desorbed with a 14% sodium hydroxide solution. The resulting filtrate contains culture fluid in a volume corresponding to the free volume of the column, then a zone of lysine in zwitterionic form leaves with a solution concentration of 35%. Lysine is completely desorbed. The impurity content in the resulting lysine solution does not exceed 0.05 g / l.
П р и м е р 2. PRI me R 2. A solution of lysine in a culture fluid with a pH of 3.0 is passed through KB-4 in H + , Na + form in a ratio of 1: 1 until equilibrium is reached. The height of the ion exchanger layer to the diameter of the column is similar to the height in Example 1. Each 100 g of ion exchanger adsorb 12 g of lysine. Desorption is carried out with a 16% NaOH solution. Lysine is completely desorbed; its concentration in the solution is 35%; the content of impurities in this solution is not more than 0.05 g / l.
П р и м е р 3. PRI me R 3. Carried out under conditions analogous to the example, changing only the ratio of the H + , Na + form of the ion exchanger by 1: 4 and the concentration of the stripping solution to 20%. The capacity of the ion exchanger for lysine is 12 g per 100 g of ion exchanger. Lysine is recovered completely from the culture fluid. The content of impurity ions in the resulting solution is not more than 0.05 g / l.
Thus, the proposed method allows you to fully remove lysine from the culture fluid, increase the concentration of the obtained solutions by 15-20 times, reduce the amount of equipment used ≈2 times, and the volume of wastewater obtained by 5-10 times.
