Abstract
Description
The invention relates to the field of modification of natural polymers, and in particular to methods for producing sulfated derivatives of chitosan. The invention can be used to obtain biologically active compounds with anticoagulant, antisclerotic and antiviral effects, as well as an increased ability to specifically bind low-density lipoproteins from the blood.
In known methods for producing sulfated chitosan [1-3] having anticoagulant and antisclerotic effects, sulfating agents, such as a mixture of sulfur dioxide and trioxide, chlorosulfonic acid, fuming sulfuric acid (oleum), as well as sulfuric anhydride or chlorosulfonic complexes, are used, as a rule. acids with pyridine or dimethylformamide, etc. In this case, chitosan is usually activated before sulfation by reprecipitation and washing with various anhydrous organic solvents (most often, pyridino m or dimethylformamide), then in the form of a suspension of the swollen polymer in these solvents is contacted with a sulfating agent. Moreover, the reaction in the initial period proceeds, as a rule, under heterogeneous conditions, then the modification product can go into solution and the sulfation process in this case will continue in a homogeneous environment.
The disadvantages of the described methods are:
1. The need for pre-activation of chitosan, which is associated with large volumes of precipitant (NaOH), multiple solvent changes with extraction and filtering, as a result of which the process is much more complicated.
2. Essentially, the heterogeneous nature of the sulfation reaction can lead to a product with an uneven distribution of substituted groups, and therefore with irreproducible properties.
It is also known sulphation of chitosan under homogeneous conditions consisting in that the chitosan is previously converted into its salt as the hydrochloride, formate, acetate, perchlorate, then the selected salt of chitosan was dissolved in formamide at a temperature of not lower than 50 ° C and subjected to sulphation of chlorosulfonic acid in pyridine or formamide. For low molecular weight desired product preformed polymer degradation salt is an aqueous solution of hydrogen peroxide at 70 ° C for 2 hours and then sulfating the described method is carried out either partial reduction of the degree of polymerization of the final product by hydrolysis sulfated with concentrated hydrochloric acid or 30% aqueous hydrogen peroxide [4]
The disadvantages of the method are:
1. The need for preliminary obtaining formamide-soluble salts of chitosan, which requires large volumes and consumption of precipitant (for example, acetone), additional washing, filtering and drying steps during the isolation of this polymer salt.
2. The need to use an elevated temperature to dissolve chitosan salt in formamide, which can cause decomposition of formamide (emission of CO 2 ), as well as a change in the physicochemical properties of the polymer salt itself.
3. The difficulty of controlling the degree of degradation during the hydrolysis of the sulfated product with hydrogen peroxide or HCl at elevated temperatures, which may be accompanied by the elimination of sulfate groups of the polymer, therefore, a decrease in the degree of substitution of the sulfated product.
The closest to the proposed method for producing a sulfated chitosan with simultaneous depolymerization [5] In this method, the chitosan is treated with a mixture of sulfuric acid and 95% chlorosulfonic acid at a certain ratio of them, a temperature of from -20 to -40 ° C for a time from 5 minutes to 2 hours. The target product is isolated by known methods, for example, the reaction mixture is mixed with an apolar or aprotic solvent in which the target product is insoluble. Then neutralized and purified by dialysis. The molecular weight of the resulting product depends on the reaction time.
The complexity of the method for practical use is due to the lack of solvent and the need for low temperatures.
The objective of the invention is to simplify the method.
This problem is solved by a method for producing sulfated chitosan, including treating chitosan with a sulfating agent, neutralizing the reaction mixture and isolating the target product, in which a complex of chlorosulfonic acid or oleum with amide is used as a sulfating agent, and the treatment is carried out in an amide solvent or dimethyl sulfoxide in the presence of sodium nitrite or potassium nitrite in a molar ratio of chitosan and equal to 1: 0.01-1, from
The presence of the sodium or potassium salt of nitrous acid promotes, firstly, the dissolution of chitosan in these solvents at room temperature without first converting it to any form or activated state, and secondly, it causes partial degradation of the glycosidic bonds of the polymer and, therefore, its molecular the mass or viscosity of the solution, the degree of fall of which depends on the amount of salt added. The latter circumstance is important for regulating the molecular weight of the target product, since the molecular weight of sulfated products to a certain extent determine their scope.
For the first time in the proposed method, the degradation of chitosan by sodium or potassium nitrite is carried out in an anhydrous medium, and the process can be controlled by changing the molar ratio of sodium or potassium nitrite and chitosan. Varying the ratio in the range of (1: 1) - (1: 0.001) is explained by the fact that the lower boundary corresponds to the minimum amount of salt needed to dissolve chitosan. The upper limit corresponds to the equimolar ratio at which all glycosidic bonds break and, therefore, an increase in salt concentration is not advisable. In addition, a noticeable decrease in the viscosity of chitosan solutions in the presence of even a small amount of salt (1 / 100-200 part by weight of the polymer) makes it possible to obtain more concentrated chitosan solutions for sulfation in this way and thereby increase the yield of the final product in one load, i.e. improve process performance.

К 30 мл диметилформамида (ДМФА) или формамида (ФА) при перемешивании и охлаждении (0-5оС) прикапывают 4,5 мл хлорсульфоновой кислоты (ХСК) или 7 мл 30% -ного олеума, затем охлаждение прекращают и реакционную смесь продолжают перемешивать для повышения температуры смеси до окружающей среды (25оС).To 30 ml of dimethylformamide (DMF) or the formamide (FA) with stirring and cooling (0-5 ° C) was added dropwise 4.5 ml of chlorosulfonic acid (HSC) and 7 ml of 30% strength oleum and then cooling was stopped and the reaction mixture was continued to stir to increase the temperature of the mixture to ambient (25 about C).
Preparation of chitosan solution.
To 10 ml of FA or dimethyl sulfoxide (DMSO) add the calculated amount of chitosan to obtain a solution of the appropriate concentration and a predetermined amount of sodium or potassium nitrite in the claimed range of the molar ratio of sodium or potassium nitride and chitosan (basic mol. Unit) (1: 1) - ( 1: 0.001). For example, to prepare a 10% solution of chitosan with a degree of deacetylation (DM) of 90%, take 1 g of polymer (mol. M unit 161 x 0.9 203 x 0.1 165.2) and 0.021 g of sodium nitrite, which corresponds to a molar ratio of 0.05. After making the calculated amounts of the components, the resulting suspension is stirred until the polymer is completely dissolved.

By sulphating complex prepared by the method described, is added with stirring 10 ml of chitosan solution prepared as described above and reacted for 10-60 min at room temperature or at 50 ° C. After reaction, the reaction mixture (solution) was neutralized with 4 n with an aqueous solution of sodium or potassium hydroxide to
