Abstract
1. Способ конструирования рекомбинантной бактерии, включающийa) получение экзогенного фрагмента ДНК, который имеет в своем составе по крайней мере 2 сайта минимум по 30 п.о. в длину каждый, идентичных сайтам, расположенным в целевом гене на хромосоме бактерии, принадлежащей к роду Panteoa,b) введение указанного линейного фрагмента ДНК в бактерию, принадлежащую к роду Panteoa, причем указанная бактерия модифицирована таким образом, что является устойчивой к продуктам генов gam, bet, and exo из фага лямбда, иc) отбор рекомбинантной бактерии, в которой указанный целевой ген заменен на указанный линейный фрагмент ДНК или линейный фрагмент ДНК встроен в целевой ген.2. Способ по п.1, отличающийся тем, что указанная бактерия является бактерией Pantoea ananatis SC17(0).3. Способ по п.1, отличающийся тем, что указанный целевой ген в указанной рекомбинантной бактерии модифицирован инактивацией или удалением.4. Способ по п.1, отличающийся тем, что указанный целевой ген кодирует белок, который участвует в процессе расщепления целевого вещества или в процессе переноса целевого вещества в клетку.5. Способ конструирования библиотеки фрагментов ДНК, включающийa) введение набора фрагментов ДНК, которые частично идентичны целевому гену на хромосоме бактерии, принадлежащей к роду Pantoea, в бактерию, отличающееся тем, что указанная бактерия модифицирована таким образом, что является устойчивой к продуктам генов gam, bet, и exo фага лямбда, иb) отбор бактерии, в которой указанный целевой ген заменен на указанные фрагменты ДНК.6. Способ по п.5, отличающийся тем, что указанные фрагменты ДНК содержат регуляторный элемент для экспрессии гена, выбранный из группы, состоящей из п�1. The method according to claim 1, characterized in that said target gene encodes a protein that is involved in the process of cleavage of the target substance or in the process of transferring the target substance into the cell. A method of constructing a library of DNA fragments, comprising a) introducing a set of DNA fragments that are partially identical to the target gene on the chromosome of a bacterium belonging to the genus Pantoea, in a bacterium, characterized in that said bacterium is modified in such a way that it is resistant to gam, bet, and exo phage lambda, and b) selection of a bacterium in which said target gene is replaced with said DNA fragments. 6. The method according to claim 5, characterized in that said DNA fragments contain a regulatory element for gene expression selected from the group consisting of
