Description
A device for express analysis of the biochemical oxygen consumption index belongs to the field of environmental environmental monitoring and can be used to assess the state of surface waters and control wastewater treatment processes.
Since the duration of the standard method for determining BOD is 5-10 days [PNDF 14. 1: 2: 3: 4. 123-97. Quantitative chemical analysis of water. The methodology for measuring biochemical oxygen demand after n-days of incubation (BODful) in surface fresh, underground (ground), drinking, waste and treated wastewater. - M .: 1997. - 25 p.] Express analyzers, in particular biosensors, are being intensively developed.
A known model of the device for the rapid determination of biochemical oxygen consumption, which is an oxygen electrode with immobilized microorganisms [Arlyapov V.A., Kamanin S.S., Alferov S.V., Alferov V.A. Device for rapid determination of biochemical oxygen consumption. RF patent No. 106898 of 07/27/2011]. The device consists of a measuring cell, a magnetic stirrer and an oxygen electrode with immobilized D. hansenii yeast. A characteristic feature of the device in question, which coincides with the essential features of the claimed technical solution, is the amperometric method for measuring biochemical oxygen consumption using microorganism cells. Significant disadvantages are the use of an oxygen electrode with a high working potential (-0.7 V), which can cause adverse reactions involving electroactive impurities. Other disadvantages include the fact that the electrode reaction is pH dependent.
The closest in its features, taken as a prototype, is a device for express monitoring of the index of biochemical oxygen consumption [Zaitseva A.S., Yudina N.Yu., Arlyapov V.A., Ponomareva O.N., Alferov V.A. Device for express monitoring of the biochemical oxygen consumption index. RF patent No. 164144 of August 3, 2016]. The device contains a measuring cuvette with a magnetic stirrer, a biosensor system consisting of two electrodes: a silver chloride reference electrode and a working carbon paste modified with a ferrocene mediator, an electrode with immobilized yeast cells Debaryomyces hansenii, an additional solution of a thionine mediator is introduced into the measuring cuvette. The device allows you to determine the index of biochemical oxygen consumption (the lower limit of the determined values of BOD 5 is 4.6 mg / DM).
- М., 2000].Signs of the prototype, which coincides with the essential features of the claimed device, is the replacement of the oxygen electrode with a working carbon paste electrode with immobilized cells and the use of mediators - compounds that transfer electrons from the biomaterial to the electrode. The working potential at which the biochemical oxygen consumption index is analyzed is determined by the redox properties of the mediators used. The disadvantage of this device is the high lower limit of the determined values of biochemical oxygen consumption of 4.6 mg / dm 3 , which does not allow the study of water, the value of BOD 5 of which does not exceed the MPC (2-4 mg / dm 3 ), which belong to the clean category [SanPiN 2.1 .5.980-00. Drainage of populated areas, sanitary protection of water bodies. Hygienic requirements for surface water protection. Approved The chief state sanitary doctor of the Russian Federation on June 22, 2000 - M., 2000].
The task to which the claimed technical solution is directed is to expand technological capabilities by improving the technical characteristics of the BOD sensor, in particular, reducing the lower boundary of the determined values of the biochemical oxygen consumption.
This problem is solved due to the fact that the claimed device for express analysis of biochemical oxygen consumption contains a measuring cell with a mediator solution, a magnetic stirrer, chloride — a silver reference electrode and a working carbon-paste electrode with immobilized biomaterial, and E cells were taken as immobilized biomaterial coli, and potassium ferricyanide was taken as a mediator.
Т., Abdelkareem М.A. //Old Yeasts-New Questions. InTech, 2017.]. 2018. V. 15. I. 3. 5.The lowering of the lower boundary is associated with the replacement of D. hansenii yeast with E. coli bacteria. E. coli cells are prokaryotes and, in contrast to D. hansenii eukaryotes, are characterized by a simpler structure, which determines the availability of enzymatic systems for the electron transport mediator, which, in turn, increases the efficiency of electron transfer [Sayed E.T., Abdelkareem M.A. . // Old Yeasts-New Questions. InTech, 2017.]. Therefore, when using E. coli cells, only one mediator is required, which is introduced into the measuring cell, in contrast to the adopted prototype, in which the effective operation of the bioelectrode based on D. hansenii yeast requires additional modification of the working electrode by the mediator ferrocene, which greatly simplifies the design of the claimed device. In addition, these microorganisms are present in natural and wastewater, and also are part of the inoculum in a standard analysis method [Galler N. et al. // International journal of environmental research and public health. 2018. V. 15. I. 3. PP. 479.], which increases the sensitivity in determining BOD 5 .
(5).In FIG. shows a diagram of a device for determining the index of biochemical oxygen consumption. The inventive device consists of the following elements: carbon paste electrode (1) with cells of the strain Escherichia coli immobilized on the surface (2). The working electrode and chloride silver reference electrode (3) are placed in a 4 ml measuring cell (4). Mixing the solution with a mediator potassium ferricyanide is carried out using a magnetic stirrer (5).
The technical result provided by the given set of features is to improve the characteristics of the device, in particular, lowering the lower boundary, which is associated with the use of prokaryotes of E. coli as biomaterial. In addition, the design of the working electrode is simplified, since E. coli cells are effective biocatalysts and do not require the use of a second mediator to participate in mediator bioelectrocatalysis.
When registering the signal of the biosensor system, an electrochemical transducer-potentiostat is used, which records the dependence of current strength on time - IPC Micro, to which the electrodes are connected. Measurements are carried out at a constant potential of 288 mV.
A working carbon paste electrode is prepared on the basis of graphite powder and mineral oil (based on 100 mg of powder per 40 μl of oil). The resulting paste is filled into a plastic tube. Cells of E. coli microorganisms are applied to the surface of the working electrode; a dialysis membrane is used to hold the biomaterial, which is fixed with a plastic ring. The principle of operation of the device for express analysis of biochemical oxygen consumption is as follows: the measurements are carried out in a two-electrode system immersed in a 4 ml measuring cell containing potassium sodium phosphate buffer (pH = 7.2). The working electrode is a carbon paste with immobilized bacteria cells of Escherichia coli and a reference electrode - a saturated silver chloride electrode. The measurements are carried out with continuous stirring of the solution with a mediator of potassium ferricyanide located in the measuring cell using a magnetic stirrer (300 rpm) at room temperature.
- М.: 1997. - 25 с.].After establishing a stable current level, the required amount of the analyte solution is introduced into the cell with a micropipette. After each measurement, the cell is washed with potassium-sodium phosphate buffer solution. The biodegradation rate of organic substrates is calculated by the amplitude of the current (sensor response, ΔI, μA) and the value of the biochemical oxygen consumption is determined using a calibration curve previously constructed using a model glucose-glutamate solution. In accordance with PNDF 14. 1: 2: 3: 4. 123-97, the value of the biochemical oxygen consumption of a model solution containing a total of 300 mg / dm 3 glucose and glutamic acid (1: 1) is 205 mgO 2 / dm 3 [PNDF 14. 1: 2: 3: 4. 123-97. Quantitative chemical analysis of water. The methodology for measuring biochemical oxygen demand after n-days of incubation (BODful) in surface fresh, underground (ground), drinking, waste and treated wastewater. - M .: 1997. - 25 p.].
3.Thus, the claimed device for express analysis of biochemical oxygen consumption does not require the involvement of highly qualified personnel and allows you to determine the index of biochemical oxygen consumption in less than 4 minutes, while the lower limit of the determined values is 0.7 mg O 2 / dm 3 .
Техническим результатом заявляемого устройства заключается в улучшении технических характеристик, за счет снижения нижней границы 4,6 мг/дм3 (прототип) до 0,7 мг/дм3 (заявляемое техническое решение).The technical result of the claimed device is to improve technical characteristics by reducing the lower limit of 4.6 mg / dm 3 (prototype) to 0.7 mg / dm 3 (the claimed technical solution).
