Abstract

DNA sekvenser, avledet fra Lactococcus lactis subsp. lactis er nyttige som promotere og promoter/sekresjonsaktiverende signaler for heterolog eller homolog uttrykking i gram- positive bakterier. Ved et ytterligere aspekt vedrører oppfinnelsen vektorer, som plasmider, omfattende sekvensene i samsvar med den foreliggende oppfinnelse, og vertceller transformert med slike vektorer. Ytterligere aspekter av den foreliggende oppfinnelse vedrører metoder for fremstilling av ønskede heterologe eller homologe peptider eller proteiner . under anvendelse av sekvensene, vektorene eller transformerte celler i samsvar med oppfinnelsen. Ved hjelp av oppfinnelsen kan det oppnås sterkt forbedret heterolog og homolog uttrykking og sekresjon i E. coli og i gram-positive bakterier som B. subtilis, Lactococci og Lactobacillus.DNA sequences derived from Lactococcus lactis subsp. lactis are useful as promoters and promoter / secretion activating signals for heterologous or homologous expression in gram-positive bacteria. In a further aspect, the invention relates to vectors, such as plasmids, comprising the sequences of the present invention, and host cells transformed with such vectors. Further aspects of the present invention relate to methods for preparing desired heterologous or homologous peptides or proteins. using the sequences, vectors or transformed cells according to the invention. By means of the invention, greatly improved heterologous and homologous expression and secretion in E. coli and in gram-positive bacteria such as B. subtilis, Lactococci and Lactobacillus can be obtained.
