The duration of the experiment was 10 hours when steady state for drug penetration had been achieved for at least several hours. Figure 11 shows penetration profiles for hydrocortisone from percutaneous absorption enhanced by various cyclic ketones through hairless mouse skin. The order of the potency of the improved absorption property for various cyclic ketones is as follows: cyclopentadecanone > cyclotridecanone > cyclododecanone > cyclododecanone > cyclononanone > cycloundecanone > cyclodecanone (in decreasing order). The slope coefficients for the penetration profiles, which represent the steady-state permeation rate for drugs, were calculated and are shown in Table 5. The enhancement factor for various cyclic ketones was calculated based on the control group as 100. There was a slight decrease in the permeation rate for hydrocortisone through hairless mouse skin when cyclodecanone and cycloundecanone were used as respective enhancers. In other words, both cyclodecanone and cycloundecanone weakly inhibit the percutaneous absorption of hydrocortisone through hairless mouse skin. There was a slight effect on the percutaneous absorption of hydrocortisone through hairless mouse skin when cyclononanone was used. There was a 230% increase in the permeation rate of hydrocortisone through hairless mouse skin when cyclododecanone was used in the study. However, there was an increase of 524& and an increase of 590$ in percutaneous absorption of hydrocortisone through hairless mouse skin when, respectively. cyclotridecanone and cyclopentadecanone were used as improvers. Blood samples were again taken after 0, 10, 20, 30, 45, 60, 90, 120 and 180 minutes. Both blood glucose determined using a YSI glucose analyzer and serum insulin determined by radioimmunoassay were examined. Both procedures were commonly practiced in the laboratory.

Thereafter, the blood glucose levels remained approximately constant for approx. 80 minutes. Finally, when serum insulin decreased after 120 minutes to 7.9 pU/ml at 180 minutes, blood glucose levels rose from 45.8 mg/dl to 72.7 mg/dl during the same time period.

The time course of the changes in blood glucose and serum insulin levels is shown in Figure 15. From this investigation it can be determined that the effect of 3-methylcyclopentadecanone on the nasal absorption of insulin in dogs was similar to the for cyclopentadecanolide used in the nasal spray preparation.

Vaginal absorpsjon av fluorogestonacetat for synkronisering av eggløsning i sauer. Vaginal absorption of fluorogestone acetate for synchronization of ovulation in sheep.
Formålet med denne undersøkelsen var å demonstrere at vaginal absorpsjon av terapeutiske midler kan oppnås til ønskede terapeutiske nivåer ved addisjon av permeeringsforbedringsmidler, såsom cyklopentadekanolid. Polymer-svamper femstilt av polyuretan e.l. impregneres med 80% fuorogestonacetat og 20% cyklopentadekanolid. Svampen innføres i vagina på søyer i opptil 12 dager. Blood samples were taken, and the levels of fluorogestone acetate were determined by radioimmunoassay. Table 9 shows the blood levels of fluorogestone acetate in ewes during the course of treatment. The last phase of the treatment is the decisive indicator for synchronization of ovulation in ewes. The results clearly indicated that at the final phase of treatment (on days 6, 9 and 12) the blood levels in the ewes that had received sponges containing permeation enhancers, such as cyclopentadecanolide, were higher than those without permeation enhancers.
