Description
Foreliggende oppfinnelse vedrører nye forbindelser som hemmer dopamin-p-hydroksylase.
Under biosyntesen av katekolamin blir tyrosin omdannet i tre trinn til norepinfrin (NE). Mellomprodukter er dihydroksyfenyla-lanin (DOPA) og dopamin (DA). Dopamin blir hydroksylert til norepinfrin ved hjelp av dopamin-p-hydroksylase (DBH) i nærvær av okskygen og askorbinsyre.
En hemming av katekolaminaktiviteten vil nedsette blodtrykket. Weinshilboum , Mayo Clin. Proe. 55, 39 (1980) har gitt en oversikt over de forbindelser som hemmer katekolaminaktiviteten ved å virke på de adrenergiske reseptorer. Alternativt kan katekolaminbiosynteseveien undertrykkes på ethvert av de nevnte trinn, noe som resulterer i reduserte NE-nivåer. I tillegg til å gi en antihypertensiv effekt, vil inhibitorer av NE-syntesen også virke som diuretika, natriuretika, kardiotoniske midler foruten som vasodilatorer. En hemming av DBH-aktiviteten kan videre ha den fordel at man får øket DA-nivå, noe som er angitt av Ehrreich e_t a_l. , "New Antihypertensive Drugs," Spectrum Publishing, 1976, s. 409-432, og som har selektiv vasodilaterende aktivitet ved visse konsentrasjoner.
En har også kunnet påvise at DBH-inhibitorer reduserer eller hindrer dannelsen av mavesår hos rotter, Hidaka e_t aJL. , "Catecho-lamine and Sress," redigert av Usdin e_t al. , Permagon Press, Oxford, 1976, s. 159-165 og Osumi et_ al. , Japan J. Pharmacol. 23, 904 (1973) .
Det er kjent en rekke DBH-inhibitorer. Disse kan generelt deles i to grupper, nemlig metallgelaterende midler som binder seg til kobberet i enzymet, og fenetylaminanaloger. Rosenberg et al. "Essays in Neurochemistry and Neuropharmacology," Vol. 4, redigert av Youdin et al., John Wiley & Sons, 1980, s. 179-192 og Goldstein, Pharmacol. Rev. 18J1), 77 (1966), gir en oversikt over DBH-inhibotrer. Førstnevnte forfatter angir at mange virksomme DBH-inhibitorer har en hydrofob sidekjede som størrelsesmessig kan sammenlignes med den aromatiske ringen i DA, noe som har gjort at nevnte forfattere antyder at inkorporeringen av en terminal hydroksylgruppe i en 4- til 6-leddet karbonsidekjede på en fenetylaminanalog, kan gi sterke inhibitorer.

Alle de ovennevnte forbindelser med unntak av benzyloksyamin og benzylhydrazin har en hemmende effekt p.g.a. metallgelaterende egenskaper. Alkylderivater av imidazol-2-tiol er mer virksomme, noe som antagelig skyldes en ikke-spesifikk reaksjon mellom alkylsubstituenten og enzymet. Benzyloksyamin og benzylhydrazin er fenetylaminanaloger som åpenbart virker som konkurrerende inhibitorer.
I tillegg til de ovennevnte forbindelser angir Runti et_ aJL. , Il Farmaco Ed. Sei. 36, 260 (1980) at andre fusarinsyrederivater og analoger hemmer DBH. Disse forbindelser innbefatter fenylpik-olinsyre som har to ganger sterkere hemmende aktivitet enn fusarinsyre, og 5-(4-klorbutyl)-pikolinsyre og andre såsom substituerte amider av fusarinsyre, foruten syrer og amider av 5-butyroylpikolinsyre, 5-aminopikolinsyre og 5-hydrazinopikolinsyre og derivater derav.
Hidaka et al., Moleclar Pharmacology, 9, 172-177 (1972) angir at 5-(3,4-dibrombutyl)pikolinsyre og 5-(dimetylditiokarba-moyl)metylpikolinsyre er DBH-inhibitorer.
Bupikomid, 5-(n-butyl)pikolinamin er angitt av Ehrreich et al., "New Antihypertenisve Drugs", Spectrum Publications, 1976, s. 409-432, å være en DBH-inhibitor som har antihypertensiv aktivitet
I europeisk patentsøknad nr. 125.033 (publisert 14. november 1984) er det beskrevet en serie 1-fenyl og 1-fenylalkylimidazol-forbindelser med en merkapto eller alkyltiogruppe i 2-stillingen. Disse forbindelser er angitt å ha en DBH-hemmende aktivitet.
U.S. patent nr. 4.487.761 beskriver flere metylpyridinderi-vater som er isolert fra fermenteringsmediet for en rase av Streptoverticillium. Disse forbindelser hemmer DBH-aktivitet.
U.S. patent nr. 4.532.331 beskriver forskjellige l-benzyl-2-aminometylimidazolderivater som hemmer DBH-aktivitet foruten farmasøytiske preparater som inneholder disse derivater, samt fremgangsmåter når man bruker disse derivater for å hemme DBH-aktivitet .
Friedman et al., Psychosomatic Med. 40, 107 (1978) angir at pasienter som ble behandlet med alfa-metyl-DOPA, guanetidin og reserpin, men ikke propranolol og diuretika, hadde nedsatt DBH-nivå, skjønt betydningen av denne observasjonen er usikker.
Ikke-spesifikke ofte toksiske effekter av kjente DBH-inhibitorer har gjort at man ikke klinisk har brukt disse forbindelser. Således er f.eks. fusarinsyre hepatotoksiskk. Se f.eks. Teresawa et a_l. , Japan. Cir. J. 35, 339 (1971) og de der angitte referanser. Antagelig virker pikolinsyrestrukturen sammen med en rekke metallproteiner og enzymer på en ikke-spesifikk måte, slik at man får de observerte sideeffekter.
I U.K. patent nr. 1.155.580 er det beskrevet forbindelser med følgende formel:
hvor R<2>og R<3>kan være H, og R<1>kan være substituert fenyl. Disse forbindelser er angitt å ha smertestillende, anti-inflamma-
toriske og antipyretiske egenskaper. Gerbert et al., U..S. patent 3.915.980 beskriver slike forbindelser hvor R<1>kan være fenyl eller fen(Ci - 3)alkyl, som mellomprodukter for fremstilling av imidazolyl-2-tioalkanoinsyreestere.
Iverson, Acta Chem. Scand. 21, 279 (1967) angir forbindelser med følgende formel:
hvor R kan være -CO2Heller -CH2NHC6Hg , men angir ikke farmasøy-tisk anvendelse av forbindelsene.
SAMMENDRAG AV OPPFINNELSEN
Man har nå oppdaget at DBH hemmes av substituerte-1-(4'-aminoaralkyl)-imidazol-2-tiol- og substituerte-1-(4'-aminoaral-kyl)-2-alkyltioimidazolforbindelser. Disse forbindelsene er virksomme og gir en forlenget DBH-hemming.
Foretrukne forbindelser ifølge foreliggende oppfinnelse innbefatter følgende: 1-(4'-amino-3',5'-difluorbenzyl)imidazol-2-tiol;
og
1-(4'-amino-3',5<1->diklorbenzyl)imidazol-2-tiol.
Videre angår oppfinnelsen nye mellomprodukter som kan brukes for fremstilling av substituerte-1-(4'-aminoaralkyl)imidazol-2-tiol og substituerte-1-(4'-aminoaralkyl)-2-alkyltioimidazolfor-bindelser. Hvert av mellomproduktene er et substituert N-acetylanailin som også er substituert i 4-stillingen.
Foreliggende oppfinnelse angår også en fremgangsmåte for å hemme DBH-aktivitet i pattedyr, og heri inngår mennesker, som innbefatter at man internt tilfører pasienten en effektiv mengde av en substituert-1-(4'-aminoaralkyl)imidazol-2-tiol- eller en substituert-1-(4'-aminoaralkyl)-2-alkyltioimidazolforbinde1se.
Foreliggende oppfinnelse angår også farmasøytiske preparater som inneholder forbindelser som kan brukes ifølge foreliggende fremgangsmåte, samt et farmasøytisk bæremiddel eller fortynnings-middel.
DETALJERT BESKRIVELSE AV OPPFINNELSEN
Foreliggende oppfinnelse angår forbindelser som hemmer DBH og med følgende formel:
hvor:
X er hydrogen, brom, klor, fluor, jod eller enhver kombinasjon av disse i et antall på opptil fire substituenter;
n er 0-5; og
R er hydrogen eller Ci-4-alkyl; eller
et farmasøytisk akseptabelt salt eller hydrat av en slik forbindelse.
Formel I innbefatter tautomeren av forbindelsene hvor R er hydrogen, dvs. forbindelser med ovennevnte formel hvor imidazol-gruppen har en av de to nedenfor angitte formler:
Forbindelsene med formel I fremstilles fra de tilsvarende substituerte-4-cyanoaniliner ved hjelp av kjente fremgangsmåter, f.eks. slik det er vist på reaksjonsskjerna I som er angitt i det etterfølgende. Utgangsforbindelsene, dvs. substituerte-4- cyanoaniliner er kjente og beskrevet i flere referanser og kan lett fremstilles. Eventuelt kan de nevnte substituerte-4-cyanoaniliner fremstilles fra de analoge substituerte aniliner. De substituerte aniliner behandles med N-bromsuccinimid i et egnet dipolart, aprotisk oppløsningsmiddel, f.eks. dimetylformamid, ved hjelp av den fremgangsmåte som er beskrevet av Mitchell, et al., J. Org. Chem., 44, 4733 (1979), hvorved man får fremstilt substituerte-4-bromaniliner. Deretter blir 4-brom-forbindelsene behandlet med kobbercyanid i et egnet dipolart, aprotisk oppløs-ningsmiddel, såsom dimetylformamid, ved hjelp av fremgangsmåten til Friedman og Schechter, J. Org. Chem., 26, 2522 (1961) , hvorved man får de forønskede substituerte-4-cyanoanilinforbind-elser.
Skjema I illustrerer reaksjonen mellom substituerte-4-cyanoaniliner (A) med X substituenter som er de samme som X i formel I, med et acyleringsmiddel såsom trifluoreddiksyreanhy-drid, noe som gir det tilsvarende substituerte 4-acetamidocyano-benzen (B) fulgt av en reduksjon, f.eks. ved behandling med en egnet hydrogeneringskatalysator såsom Raney-nikkel og en organisk syre såsom maursyre, hvorved man får fremstilt substituerte 4-acetamidobenzaldehyder (C). Ved reaksjon med et aminoacetalde-hydacetal, f.eks. aminoacetaldehyd-dimetylacetal, fulgt av reduksjon, f.eks. ved behandling med et sterkt reduseringsmiddel sAsom NaBH4og en uorganisk syre såsom saltsyre, såvil 4-acetami-dobenzaldehydene (C) gi substituerte 4-acetamidobenzylaminoacet-aldehyd-acetalhydroklorider (E). Deretter kan de substituerte-1-(4'-aminoaralkyl)imidazol-2-tioler (F) fremstilles ved at man reagerer nevnte 4-acetamidobenzylaminoacetaldehyd-acetalhydroklo-rider (E) med kaliumtiocyanat i nærvær av en sterk uorganisk syre, f.eks. saltsyre.
Som vist i Reaksjonsskjerna I er n 1, men n kan imidlertid ha en verdi fra 0 til 5. Forbindelsene hvor n er 2, 3, 4 eller 5 blir fortrinnsvis fremstilt som beskrevet i eksempel 4. I syntesen av disse forbindelser blir fremgangsmåten fra Reaksjons-skjema I brukt, bortsett fra at de substituerte-4-cyanoaniliner blir erstattet med substituerte-4-aminofenylalkylnitriler, f.eks. substituerte-4-aminobenzylnitriler, substituerte-4-aminofenetyl-nitriler, substituerte-4-aminofenylpropylnitriler og substitu-er te-4-aminofenylbutylnitriler.
1-(4'-aminosubstituert fenyl)imidazol-2-tioler (n er 0) blir fortrinnsvis fremstilt ved å reagere et passende substituert, eventuelt beskyttet, 4-aminofenylisotiocyanat med et aminoacetal-dehydacetal fulgt av en sterk syrekatalysert cyklisering, f.eks. slik det er vist i eksempel 3.
Forbindelser hvor R er en metylgruppe, kan fremstilles ved å alkylere de tilsvarende 1-(4'-aminoaralkyl)-imidazol-2-tioler med metyljodid i metanol ved hjelp av kjente fremgangsmåter. Andre alkylhalogenider såsom metylbromid eller metylklorid kan brukes istedenfor metyljodid i et passende oppløsningsmiddel. Forbindelser hvor R er en alkylgruppe som er forskjellig fra metyl, kan videre fremstilles ved at man reagerer det tilsvarende substituerte-1- (41-aminoaralkyl)-imidazol-2-tiol med et alkylhalogenid, såsom butyljodid, hvorved man får den forønskede substituerte-1-(4'-aminoaralkyl)-2-alkykltioimidazolforbindelsen ifølge foreliggende oppfinnelse.
Ved fremstillingen av de nevnte substituerte-1-(4'-amino-aralkyl )-imidazol-2-tioler, så syntetiserte man også nye forbindelser med følgende formel: X er H, Br, Cl, F, I eller enhver kombinasjon av disse i et antall på opptil fire substituenter;
n er 1-5;
bortsett fra forbindelser hvor:
X er fire H'er; og
Y er CN eller CHO.
Farmasøytisk akseptable syreaddisjonssalter av forbindelser ifølge foreliggende oppfinnelse kan fremstilles ved hjelp av sterke eller moderat sterke organiske eller uorganiske syrer ved hjelp av kjente fremgangsmåter. F.eks. kan basene reageres med en uorganisk eller organisk syre i et vandig blandbart oppløs-ningsmiddel såsom etanol, hvoretter man isolerer saltet ved å fjerne oppløsnignsmidlet, eller i et vandig ublandbart oppløs-ningsmiddel hvor syren er oppløselig, f.eks. etyleter eller kloroform, hvoretter det forønskede saltet skiller seg ut direkte eller kan isoleres ved å fjerne oppløsningsmidlet. Eksempler på salter som inngår i foreliggende oppfinnelse er maleat, fumarat, laktat, oksalat, metansulfonat, etansulfonat, benzensulfonat, tartrat, citrat, hydroklorid, hydrobromid, sulfat, fosfat og nitratsalter.
Etter som forbindelsene med Formel I hemmer DBH-aktivitet, så har de terapeutisk verdi som diuretika, natriuretika, kardiotoniske midler, antihypertensive midler og som vasodilaterende forbindelser, foruten at de kan brukes ved behandling av mavesår og Parkinsons sykdom. Tabell I angir forbindelser ifølge foreliggende oppfinnelse som er prøvet for iri vitro DBH-hemming ved en standardprøve for å måle omdannelsen av tyramin til oktopamin i nærvær av DBH. J. J. Oktopamin ble målt ved å følge natriumperjodatoksydasjon til p-hydroksybenzaldehyd ved å måle den spektrofotometriske absorpsjon ved 330 nm. I Tabell I er inhiberingen eller hemmingen gitt i molar konsentrasjon av forbindelsen som ga en halvert DBH-aktivitet (ICso). Smeltepunk-tene (mp) er angitt i °C. Man fant at fusarinsyre i denne prøven hadde en ICso-verdi på 8x10"7 M.
En av forbindelsene ifølge foreliggende oppfinnelse ble prøvet for sin effekt in vivo på perifere dopamin (DA) og norepinfrin (NE) nivå i alt vesentlig ved hjelp av den fremgangsmåte som er beskrevet av DaPrada og Zurcher, Life Sciences, 19, 1161, (1976). Grupper på fem spontane hypertensive rotter ble dosert oralt to ganger, og den andre dosen var ca. 18 timer etter den første, hvoretter rottene ble obdusert ca. 2 timer etter andre dose. Midlere resultater uttrykt i mikrogram DA og NE pr. gram vev er gitt i Tabell II.
Videre ble spontant hypertensive rotter dosert med en suspensjon eller oppløsning av 1-(4'-amino-3',5'-difluorbenzyl)- imidazol-2-tiol i en dose på 50 mg/kg intraperitonealt, hvoretter man målte det midlere arterieblodtrykket i 260 min., idet man brukte en ilagt kanyle som var plassert i halearteriene. Man kunne observere ca. 40% reduksjon av blodtrykket 15 min. etter at man hadde tilført denne forbindelsen. 260 min. etter tilførselen av forbindelsen forble blodtrykket redusert med ca. 10% sammen-lignet med kontrollprøvene.
Det var relativt uventet å finne at substituerte-1-(4'-aminoaralkyl)-imidazol-2-tiolforbindelsene hadde effekt som DBH-inhibitorer, p.g.a. at nærstående forbindelser hadde liten eller ingen effekt. Denne prøvingen ble utført ved å bruke forbindelser fremstilt ved å bruke forskjellige fenoliske isoestere istedenfor den fenoliske hydroksygruppen i en annen DBH-inhibitor, 1-(3',5'-difluor-4'-hydroksybenzyl)imidazol-2-tiol (U.S. patentsøknad 590.665, innsendt 19. mars 1984). Forbindelser hvor den fenoliske hydroksygruppen ble erstattet med -NHCHO, -NHCOCF3 , -NHSO2NH2og -CH2 SO2 CH3 ble prøvet in vitro, og man fant at ikke hadde noen DBH-hemmende aktivitet. I motsetning til dette slik det fremgår av Tabell I, så lar ICa0-verdien for l-(4'-amino-3',5<1->difluorbenzyl)imidazol-2-tiol seg meget gunstig sammenligne med den tilsvarende verdien for 1-(3',5'-difluor-4'-hydroksyben-zyl )imidazol-2-tiol som er 8 x 10~<8>M.
Forbindelser med formel I kan lett opparbeides i hensiktis-messige doseringsformer såsom kapsler, tabletter eller injiser-bare preparater. Man kan bruke faste eller flytende farmasøy-tiske bærere. Faste bærere innbefatter stivelse, laktose, kalsiumsulfat, dihydrat, terra alba, sakkarose, talkum, gelatin, agar, pektin, akasie, magnesiumstearat og stearinsyre. Flytende bærere innbefatter sirup, jordnøttolje, olivenolje, saltoppløsing og vann. På lignende måte kan bærestoffet eller fortynningsmidlet innbefatte ethvert materiale med forlenget frigjøring, såsom glyseryl-monostearat eller glyseryl-distearat alene eller sammen med en voks. Mengden av fast bærestoff varierer meget sterkt, men vil fortrinnsvis være fra ca. 25 mg til ca. 1 g pr. doseringsenhet. Når man bruker en flytende bærer så vil preparatet være i form av en sirup, en saft, en emulsjon, en myk gelatinkapsel, en steril injiserbar væske, f.eks. i form av en ampulle, eller som en vandig eller ikke-vandig flytende suspensjon.
De farmasøytiske preparater fremstilles ved å bruke vanlige kjente teknikker i den farmasøytiske industri, ved at man blander, granulerer og presser når dette er nødvendig for fremstilling av tabletter, eller blander, fyller og oppløser ingrediensene alt etter behov, hvorved man får de forønskede orale eller parenterale produkter.
Doser av de foreliggende forbindelser i en farmasøytisk doseringsenhet vil være en effektiv, ikke-toksisk mengde valgt fra området fra 0,1-100 mg/kg kroppsvekt av den aktive forbindelsen, fortrinnsvis 0,1-50 mg/kg. Den valgte dose tilføres en pasient som har behov for DBH-hemming, fra 1-6 ganger daglig, enten oralt, rektalt, ved injeksjon, eller kontinuerlig ved infusjon. Doseringsenheter for oral tilførsel til mennesker inneholder fortrinnsvis fra 1 til 500 mg av den aktive forbindelsen. Det er foretrukket å bruke parenteral tilførsel hvor man bruker lavere doser. Oral tilførsel ved høyere doseringer kan imidlertid brukes når dette er trygt og hensiktsmessig for pasienten.
De følgende eksempler illustrerer fremstillingen av forbindelser med Formel I. Eksemplene begrenser ikke oppfinnelsen slik denne er definert ovenfor og angitt i de etterfølgende krav.
EKSEMPEL 1
i) Fremstilling av N- trifluoracetyl- 4- cyano- 2, 6-
dikloranilin
25 g (0,1337 mol) 4-cyano-2,6-dikloranilin ble oppløst i 250 ml metylenklorid, hvoretter man langsomt under røring tilsatte 30,9 g (20,8 ml, 0,1471 mol) av trifluoreddiksyreanhyd-rid. Oppløsningen ble hensatt over natten og ble så nøytralisert ved å tilsette 5% vandig natriumkarbonat, og det utfelte krystal-linske hvite faste stoffet ble frafiltrert og tørket til 28,0 g (74% utbytte) av N-trifluoracetyl-4-cyano-2,6-dikloranailin, smp.: 134-136°C.
ii) Fremstilling av 4- trifluoracetamido- 3, 5-
diklorbenzaldehyd
27 g (0,0954 mol) av ovennevnte N-trifluoracetylcyanoanilin ble oppløst i 270 ml 88% maursyre, hvoretter man tilsatte 27 g Raney-nikkel. Blandingen ble rørt og oppvarmet under tilbakeløp i 1,5 timer.. Reaksjonsblandingen ble så avkjølt, filtrert og
filtratet fortynnet med vann, hvoretter blandingen tre ganger ble ekstrahert med metylenklorid. De samlede ekstrakter ble tilbakeekstrahert med 5% vandig natriumbikarbonat, hvoretter metylenklo-ridoppløsningen ble konsentrert til en gul olje. Denne ble
oppløst i eter og man tilsatte heksan (1:1 til 1:2) inntil man fikk en uklar oppløsning. Avkjøling av blandingen ga et krystallinsk bunnfall som ble filtrert og tørket til 18,85 g (69% utbytte) av 4-trifluoracetamid-3,5-diklorbenzaldehyd, smp.: 88-90° C.
iii) Fremstilling av 1-( 4'- amino- 3', 5'- diklorbenzyl)-imidazol- 2- tiol 5 g (0,175 mol) av det ovennevnte benaldehyd ble oppløst i 50 ml metanol og man tilsatte 1,84 g (0,0175 mol) aminoacetalde-hyddimetylacetal. Denne oppløsningen ble kokt under tilbakeløp i én time, avkjølt og man tilsatte 0,68 g (0,0175 mol) natriumborhydrid i små porsjoner under røring. Blandingen ble kokt under tilbakeløp et par minutter, avkjølt og fortynnet med vann, og så ekstrahert med tre ganger med eter. De samlede eterekstraktene ble konsentrert i vakuum til en gul olje. Denne oljen ble omrørt med 25 ml vann, 12 ml etanol og 6 ml 12N (vandig) saltsyre, og man tilsatte 2,04 g (0,0210 mol) kaliumtiocyanat. Blandingen ble rørt og kokt under tilbakeløp 30 min., så avkjølt og fortynnet med vann som ga et bunnfall. Dette ble frafiltrert og tørket til 6,7 g faststoff. Dette faststoff ble behandlet med vann og så med metanol og ga 5,7 g av et fast produkt etter tørking. Dette ble oppløst i 25 ml 2,5N (vandig) natriumhydroksyd, og oppløs-ningen ble kokt under tilbakeløp i én time, avkjølt og nøytrali-sert til ph 7 med fortynnet saltsyre. Man fikk et gult bunnfall som ble frafiltrert og tørket til 3,63 g, smp.: 223-230°C ved dekomponering. Produktet ble omkrystallisert fra dimetylformamid-acetonitril, til 1,18 g (24% utbytte) av 1-(4'-amino-3',5'-diklorbenzyl)imidazol-2-tiol, smp.: 233-236° dek..
EKSEMPEL 2
i) Fremstilling av 1-( 4'- amino- 3', 5'- difluorbenzyll)-imidazol- 2- tiol- 4- brom- 2, 6- difluoranilin 50 g (0,39 mol) 2,6-difluoranilin ble behandlet med 71,0 g (0,39 mol) N-bromsuccinimid i 250 ml dimetylformamid ved hjelp av fremgangsmåten til Mitchell, Lai og Williams, J. Org. Chem., 44, 4733 (1979), noe som ga et totalt utbytte på 50,2 g (62%) av 4-brom-2,6-difluoranailin, smp.: 64-66°C.
ii) Fremstilling av 4- cyano- 2, 6- difluoranailin
22,6 g (0,11 mol) av ovennevnte bromforbindelse ble behandlet med 11,7 g (0,13 mol) kobbercyanid i 17 ml dimetylformamid ved hjelp av fremgangsmåten til Friedman og Schechter, J. Org. Chem., 26, 2522 (1961), idet man brukte etylendiamin-natriumcya-nidkompleks-dekomponeringsmetoden i denne referansen [25 ml etylendiamin og 20 ml 10% vandig natriumcyanid], noe som ga 4,3 g (26%) 4-cyano-2,6-difluoranilin, som ble utkrystallisert fra eter-heksan, smp.: 110-111°C (mykner ved 107°C).
iii) Fremstilling av N- trifluoracetyl- 4- cyano- 2, 6-
difluoranilin
4,07 g (0,026 mol) av ovennevnte cyanoanilin ble oppløst i 50 1 metylenklorid, hvoretter man tilsatte 10 ml trifluoreddiksy-reanhydrid under røring. Oppløsningen varmet seg selv opp til koking under tilbakeløp og dette ble fortsastt i et par min. på et dampbad. Oppløsningen ble avkjølt og behandlet med 5%
(vandig) natriumkarbonat, hvorved man fikk utfelt et rødhvitt faststoff. Dette ble frafiltrert og vasket med heksan og vann. Den vandige delen av filtratet ble nøytralisert til pH 7 med 3N (vandig) saltsyre. Et hvitt bunnfall ble frafiltrert og tørket, og var N-trifluoracetyl-4-cyano-2,6-difluoranilin, smp.: 133,5-135°C, totalt utbytte 6,15 g (93%).
iv) Fremstilling av 3, 5- difluor- 4- trifluoracetamido-benzaldehyd
5,78 g (0,023 mol) av ovennevnte trifluoracetylforbindelse ble oppløst i 60 ml 88% maursyre, hvoretter man tilsatte 6,0 g Raney-nikkel. Blandingen ble kokt under tilbakeløp i 1,5 timer og ble avkjølt og filtrert. Filtratet ble fortynnet med vann og ekstrahert tre ganger med metylenklorid. De samlede ekstrakter ble tilbakeekstrahert med 5% (vandig) natriumbikarbonat, og ble så konsentrert til et faststoff som ble omkrystallisert fra eter-
heksan, noe som ga 4,45 g (76% utbytte) av 3,5-difluor-4-tri-fluoracetamidobenzaldehyd, smp.: 122-125°C.
v) Fremstilling av N-( 3, 5- difluor- 4- trifluoracetamido-bezyl) aminoacetaldeyd- dimetylacetal- hydroklorid
4.3 g (0,017 mol) av ovennevnte benzaldehyd ble oppløst i
22 ml 95% etanol, hvoretter man tilsatte 1,79 g (0,017 mol) aminoacetaldehyd-dimetyl-acetal. Oppløsningen ble kokt under tilbakeløp i én time, avkjølt, hvoretter man i flere små porsjoner tilsatte 0,64 g (0,017 mol) natriumborhydrid. Reaksjonsblandingen ble kokt under tilbakeløp et par minutter, avkjølt og fortynnet med vann. pH-et ble justert til 7 og blandingen ble så ekstrahert tre ganger med eter. De samlede eterekstraktene ble konsentrert, noe som ga en olje som så ble oppløst i eter, hvoretter oppløsningen forsiktig ble behandlet med hydrogenklo-ridholdig eter, noe som ga et hvitt krystallinsk faststoff,
4,4 g. pH-et på den vandige fasen ble justert til 8,2, hvoretter blandingen ble gjenekstrahert tre ganger med eter. Konsentrasjon av de samlede ekstrakter ga en olje som ble omdannet til det faste hydrokloridsaltet som beskrevet ovenfor, 0,45 g. Totalt utbytte av N-(3,5-difluor-4-trifluor-acetamidobenzyl)-aminoacetaldehyd-dimetyl-acetal-hydroklorid var 4,85 g (83%), smp.: dekomponerer over 120°C.
vi) Fremstilling av 1-( 4'- amino- 3', 5'- difluorbenzyl)-imidazol- 2- tiol
4.4 g (0,012 mol) av ovennevnte N-benzylaminoacetal-hydroklorid ble oppløst i 22 ml vann, hvoretter man tilsatte 1,13 g (0,012 mol) kaliumtiocyanat og derpå 5,5 ml 12N (vandig) saltsyre. Oppløsningen ble rørt og oppvarmet til 90°C, hvorved man fikk utskilt en olje. Blandingen ble rørt og kokt under tilbake-løp i 30 min. uten ytterligere forandring, hvoretter den ble avkjølt og hensatt. Oljen stivnet og ble frafiltrert. Produktet ble oppløst i 25 ml 2N (vandig) natriumhydroksyd. Blandingen ble så kokt under tilbakeløp i én time, og oppløsningen ble så avkjølt og nøytralisert til pH 7, noe som ga et gult bunnfall, 2,2 g etter tørking. Produktet ble oppløst i 10-20 ml metanol, og det faste stoff ble gjenutfelt ved å tilsette 20-50 ml eter.
Produktet ble så omkrystallisert fra etylacetat, noe som ga 1,0 g (36% utbytte) av 1-(4'-amino-3',5'-difluorbenzyl)-imidazol-2-tiol, smp.: 181-184°C.
Behandling av nevnte 1-{4'-amino-3',5'-difluorbenzyl)-imidazol-2-tiol i etanolisk oppløsning med en oppløsning av hydrogenklorid i dietyleter, gir 1-(4'-amino-3',5'-difluorben-zyl ) imidazol-2-tiol-dihydroklorid.
EKSEMPEL 3
En oppløsning av 4-amino-3,5-difluorfenyl-isotiocyanat i triklormetan ble behandlet med en ekvimolar mengde av aminoacetaldehyd-dimetyl-acetal.. Oppløsningsmidlet ble fordampet, og resten ble omkrystallisert fra etanol, noe som ga N-(4-amino-3,5-dif luorf enyl) -N ' - ((3 , (3-dimetoksyetyl) tiourea . En suspensjon av denne tioureaforbindelsen i konsentrert svovelsyre og vann (1:4) ble kokt under tilbakeløp i tre timer. Blandingen ble så avkjølt, og det faste stoff ble frafiltrert, vasket med vann og tørket. Omkrystallisering fra etanol gir 1-(4'-amino-3<1>,5'-difluorfenyl)-imidazol-2-tiol.
EKSEMPEL 4
Man brukte her fremgangsmåten fra eksempel 1 og 4-amino-3,5-diklorfenylpropylnitril istedenfor 4-cyano-2,6-dikloranilin noe som gir 1-(4'-amino-3',5'-diklorfenylpropyl)imidazol-2-tiol.
EKSEMPEL 5
Man anvendte fremgangsmåten fra eksempel 2 og 4-cyanoanilin istedenfor 4-amino-2,6-dikloranailin, hvorved man får fremstilt 1-(4'-aminobenzyl)imidazol-2-tiol.
EKSEMPEL 6
En reaksjon mellom 1-(4'-amino-3'-brombenzyl)imidazol-2-tiol fremstilt som beskrevet i eksempel 1, idet man brukte 2-bromani-lin istedenfor 2,6-difluoranailin, med metyljodid og natrium-metoksyd i metanol ved hjelp av kjente teknikker, gir l-(4'-amino-3 *-brombenzyl)-2-metyltioimidazol. ;EKSEMPEL 7 ; ;
Man brukte her fremgangsmåten fra eksempel 2 og 2,3,5-trifluoranilin istedenfor 2,6-difluoranilin, hvorved man fikk fremstilt 1-(4'-amino-2',3",6'-trifluorbenzyl)imidazol-2-tiol. ;EKSEMPEL 8; ;
Man anvendte fremgangsmåten fra eksempel 2 og 2-klor-6-fluoranilin istedenfor 2,6-difluoranilin, hvorved man fikk fremstilt 1-(4'-amino-3'-klor-5'-fluorbenzyl)imidazol-2-tiol. ;EKSEMPEL 9;En oral doseringsform for tilførsel av de foreliggende forbindelser, kan fremstilles ved at man sikter, blander og fyller i en hard gelatinkapsel de ingredienser som er gitt i den nedenforstående tabell III. ;
EKSEMPEL IO;Sakkaraosen, kalsiumsulfatdihydratet og den substituerte-1-(4'-aminoaralkyl)imidazol-2-tiol som er vist i tabell IV nedenfor, ble blandet og granulert med en 10% gelatinoppløsning. De våte granulatene ble siktet, tørket og blandet med stivelse, talkum og stearinsyre, deretter siktet og så presset til en ;tablett.; ;
EKSEMPEL 11 ;1-(4 *-amino-31-brombenzyl)imidazol-2-tiol, 75 mg ble dispergert i 25 ml normal saltoppløsning, hvorved man fikk fremstilt et injiserbart preparat.
Skjønt oppfinnelsen er blitt illustrert ved foretrukne utførelser som angitt ovenfor, så er det underforstått at oppfinnelsen ikke er begrenset til disse eksempler, og at man lett kan utføre modifikasjoner og variasjoner uten at man derved forlater oppfinnelsens intensjon slik denne fremgår av de etterfølgende krav.

1. Fremgangsmåte for fremstilling av en forbindelse med formelen (X):
hvor:
X er hydrogen, brom, klor, fluor, jod eller enhver kombinasjon av disse i et antall på opptil fire substituenter;
n er 0-5; og
R er hydrogen eller Ci - 4-alkylgruppe;
eller et farmasøytisk akseptabelt salt eller hydrat av en slik forbindelse,
karakterisert ved at man
omsetter et tiocyanatsalt i nærvær av en sterk uorganisk syre, med en forbindelse med formelen:
hvor X og n er som angitt ovenfor.
