ST001714, DSM 13784, i løpet av fermentering, fremgangsmåte for deres fremstilling, deres anvendelse som legemidler, legemidler som inneholder caloporosidderivater så vel som mikroorganismen Gloeoporus dichrous Bres. ST001714, DSM 13784.
Caloporosid ble for første gang beskrevet i 1994 som fosfolipase C inhibitor (W. Weber et al. J. Antibiotics, 47, 1188-1194). I samme år ble to ytterligere, lignende sekundærmetabolitter isolert (R. Shan et al. Nat. Prod. Lett., 4,171-178). Forbindelsene ifølge oppfinnelsen med formel I (se nedenfor) skiller seg fra de der beskrevne substanser med hensyn til deres struktur.
Kreft er i de fleste tilfeller en dødelig sykdom hos mennesker og dyr, som forårsakes av den ukontrollerte veksten av kroppsegne celler. Kreft er betegnelsen for dannelse av ondartede svulster (malignomer), av neoplasma (tumorer henholdsvis karsinomer) eller for den maligne fremmedartethet så vel som modningsforstyrrelse hos hvite blodlegemer (leukemi, blodkreft). Kreft- eller tumorceller oppstår ved omdannelse av kroppsegne celler. Ondartetheten av kreftcellene uttrykker seg i autonomien av veksten, dvs. deres evne til å vokse infiltrerende, uhemmet og uten innordning i byggeplanen til organene og ved ødeleggelse av vevet. Et sikkert tegn på ondartethet er dannelse tumorfjerne avleiringer (metastaser) etter hematogener eller lymfogenutbredelse av tumorceller. Kreft hører til de mest vanlige dødsårsaker hos mennesker og derfor er det et behov for fremgangsmåter og midler for å helbrede eller behandle ondartede vekster.
Muligheten for en terapi av ondartede tumorer omfatter ved siden av - hvis mulig radikal-operativ fjerning av tumoren, den radiologiske terapien med røntgenstråler, a-, P", y-stråler, immunterapi og kjemoterapi. Immunterapien anvendes for tiden kun i begrenset omfang. Som kjemoterapi av tumorer forstår man administrasjon av cellegifter (cytostatika) for behandling av tumorer og av gjenstående tumorceller etter lokal kirurgisk behandling eller bestråling. Disse stoffer griper spesifikt inn i visse prosesser av celledelingen, slik at vev med en høy andel av delende celler, som det raskt voksende tumorvevet, reagerer mer ømfintlig. Man anvender alkylerende forbindelser, som for eksempel cyklofosfamid (The Merck Index, 12. ed. s. 463), antimetabolitter som metotrexat (The Merck Index, 12. ed. s. 1025), alkaloider som vinkristin (The Merck Index, 12. ed. s. 1704) og antibiotika som daunomycin (The Merck Index, 12. ed. s. 479) så vel som adriamycin (The Merck Index, 12. ed. s. 581-582). Alle disse agenser har imidlertid, på grunn av massive bivirkninger, store ulemper slik at tidspunktet for inntredelse av døden kun forlenges for den syke personen, men ikke forhindres. I tillegg foreligger resistenser mot de anvendte midler hos degenererte (kreft-) celler. De for tiden anvendte legemidler virker således ikke lenger cytostatisk, men imidlertid toksisk på grunn av deres virkninger. Ytterligere har det vist seg at en kombinert henholdsvis sekvensiell anvendelse av cytostatika overtreffer virkningen av det enkelte cytostatiske (monoterapi) og derfor er det mulig at de betydelige bivirkninger ikke summeres ved polykjemoterapi. På grunn av dette er det nødvendig med nye kjemoterapeutika, som er etterspurt over hele verden.
Cyklinavhengige kinaser (cyklin-avhengige kinase = CDK'er) spiller en sentral rolle ved regulering av cellesyklusen. De katalyserer fosforyleringsreaksjoner og setter
således en reaksjonskaskade i gang som innleder en overgang fra Gl-fase (Vekstfase 1) til S-fase (Syntesefase) i cellesyklusen. Cyklinavhengige kinaser representerer således et godt terapeutisk mål for behandling av kreft og andre sykdommer med en patologisk forstyrrelse av celleproliferasjon. Lavmolekylære inhibitorer som regulerer cellesyklusen og som forhindrer ukontrollert celledeling, ville være nyttige legemidler for behandling av kreftpasienter.
Det er overraskende funnet at mikroorganismestammen Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784 er i stand til å lage høyvirksomme nye cytostatika som hemmer cyklinavhengige kinaser i meget lave konsentrasjoner.
Oppfinnelsesgjenstanden er således de fra stammen Gloeoporus dichrous (Fr.: Fr.) Bres. ST 001714, DSM 13784 dannede virkestoffer (caloporosidderivater) så vel som deres fysiologisk akseptable salter, estere og åpenbare kjemiske ekvivalenter.
Foreliggende oppfinnelse omfatter
Foreliggende oppfinnelsen angår således forbindelser kjennetegnet med den generelle formel I
Ri, R2og R3, uavhengig av hverandre, betyr H eller en acylrest med 2-10 C-atomer, fortrinnsvis 2 til 6 atomer, spesielt foretrukket 2 atomer; og
R4betyr H eller -C(0)(CH2)„ COOH, hvori n er lik 1 til 7, fortrinnsvis 1 til 3, spesielt foretrukket 1 eller 2;
med unntak av at Ri, R2, R3og R4ikke alle betyr H;
samt deres fysiologisk akseptable salter.
Videre angår forbindelsen anvendelse av en forbindelse med formel I, eller et fysiologisk akseptabelt salt derav ifølge ett eller flere av kravene 1 - 8 for fremstilling av et legemiddel for behandling av kreft eller andre sykdommer med en patologisk forstyrrelse av celleproliferasjonen.
Omfattet av oppfinnelsen er også legemiddel, kjennetegnet ved at det inneholder minst en forbindelse med formel I, eller et fysiologisk akseptabelt salt derav ifølge ett eller flere av kravene 1-8.
Videre er fremgangsmåte for fremstilling av legemidler ifølge krav 14 omfattet, kjennetegnet ved at man bringer minst en forbindelse med formel I, eller et fysiologisk akseptabelt salt derav, ifølge ett eller flere av kravene 1 - 8 i en egnet administreringsform ved hjelp av egnede hjelpe- og/eller bærestoffer.
Også mikroorganisme, kjennetegnet ved at den er Gloeoporus dichrous (Fr.:Fr.) Bres. ST001714, DSM 13784 er omfattet av den foreliggende oppfinnelse.
Acylrestene av forbindelsene med formel I kan være rettkjedet eller forgrenet, mettet eller enkelt eller to ganger umettet.
Som acylrest med 2 C-atomer forstås for eksempel en acetylrest.
Eksempler på mettede, uforgrenede acylrester er en eddiksyrerest (C=2), propionsyrerest (C=3), smørsyrerest (C=4), valeriansyrerest (C=5), kapronsyrerest (C=6), ønantsyrerest (C=7), kaprylsyrerest (C=8), perlagonsyrerest (C=9) og kaprinsyrerest (C=10).
Eksempler på enkelt umettede, uforgrenede acylrester er en acylsyrerest (C=3), krotonsyrerest (C=4) eller en vinyleddiksyrerest (C=4).
Et eksempel på to ganger umettede, uforgrenede acylrester er en sorbinsyrerest (C=6).
Kaloporosid er svakt virkende antibiotika som består av en salisylsyre og et disakkarid. Begge strukturer er forbundet via en alkylkjede. Sukkerdelen av forbindelsen med formel I kan være et disakkarid, som består respektivt av en D-pyranose av en aldoheksose (som for eksempel D-glucopyranose eller d-galaktopyranose) og onsyren av en aldoheksose (som for eksempel D-glukonsyre). Fortrinnsvis er sukkerandelen D-mannopyranosyl-D-mannonsyre som er usubstituert eller substituert ved R2, R3og/eller R4som definert over.
Oppfinnelsen angår ytterligere
a) en forbindelse med formen I hvori Ri = acetyl; R2= R3= R4= H (=caloporosid B: sumformel: C3gH620i6, MG 774,9) så vel som deres
fysiologisk akseptable salter;
b) en forbindelse med formel I hvori Ri = R3= acetyl; R2= malonyl (=caloporosid C: sumformel: C43H66O20, MG 902,99) så vel som deres
fysiologisk akseptable salter;
c) en forbindelse med formel I hvori Ri = R2= H; R3= acetyl; R4= malonyl (=caloporosid D: sumformel: C41H64O19, MG 806,96) så vel som deres
fysiologisk akseptable salter;
d) en forbindelse med formel I hvori Ri = R3= H; R2 = acetyl; R4= malonyl (=caloporosid E: sumformel: C41H64O19, MG 806,96) så vel som deres
fysiologisk akseptable salter;
e) en forbindelse med formel I hvori Ri = R2= R4= H; R3= acetyl (=caloporosid F: sumformel: C3gH620i6, MG 774,9) så vel som deres
fysiologisk akseptable salter.
Chiralitetssentere i forbindelsene med formel I kan, hvis ikke noe annet er angitt, foreligge i R- eller i S-konfigurasjon. Oppfinnelsen angår både de optisk rene forbindelser og også stereoisomere blandinger som enantiomerblandinger og diastereomerblandinger.
Ifølge oppfinnelsen oppnås forbindelsene med formel I ved fermentering av Gloeoporus dichrous (Fr.:Fr.) Bres. ST001714, DSM 13784, eller en av dets varianter eller mutanter under egnede betingelser i et kulturmedium, inntil ett eller flere av caloporosidderivatene med formel I anrikes i kulturmedium. Ved etterfølgende isolering av forbindelsene og eventuelt omdannelse til kjemiske ekvivalenter, så vel som deres fysiologisk akseptable salter, utvinnes caloporosidderivatene.
Oppfinnelsen angår ytterligere en fremgangsmåte for fremstilling av en forbindelse med formel I, kjennetegnet ved at mikroorganismen Gloeoporus dichrous (Fr.:Fr.) Bres. ST001714, DSM 13784 eller en av dens varianter eller mutanter fermenteres under egnede betingelser i et kulturmedium, inntil det anrikes en eller flere av målforbindelsene i kulturmediet og at man deretter isolerer de fra kulturmediet og omdanner eventuelt til kjemiske ekvivalenter og/eller fysiologisk akseptable salter.
Fortrinnsvis fermenteres stammen ST001714, DSM 13784, dens mutanter og/eller varianter i en næringsløsning eller et fast medium (også betegnet kulturmedium) med karbon- og nitrogenkilder så vel som de tradisjonelle uorganiske salter inntil forbindelsene ifølge oppfinnelsen anrikes i kulturmedium og deretter isoleres forbindelsene fra kulturmediet og separeres eventuelt til de enkelte aktive bestanddeler.
Fremgangsmåten ifølge oppfinnelsen kan anvendes for fermentering i laboratoriemålestokk (milliliter- til literområdet) eller for industriell målestokk (kubikkmetermålestokk).
Stammen Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714 ble fermert i en forkultur. Et isolat ble sendt til Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH, Mascheroder Weg IB, 3300 Braunschweig, Tyskland, ifølge reglene i Budapestavtalen av 14. desember 1999 med følgende nummer: DSM 13784.
Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784 besitter et hvitt mycel og purpurfargede sporer. Det foreligger fortrinnsvis på Betula, men kan også angripe andre verter som for eksempel Alnus, Salix, Populus, Ulmus, Prunus.
I stedet for stammen Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784 er det også mulig å anvende dens mutanter og varianter som syntetiserer en eller flere av forbindelsene ifølge oppfinnelsen. Slike mutanter kan fremstilles på i og for seg kjent måte ved fysikalske midler, for eksempel bestråling, som med ultrafiolett- eller røntgenstråler, eller kjemiske mutagener, som for eksempel etylmetansulfonat (EMS), 2-hydroksy-4-metoksy-benzofenon (MOB) eller N-metyl-N'-nitro-N-nitrosoguanidin
(MNNG).
Screeningen etter mutanter og varianter som syntetiserer en eller flere av forbindelsene ifølge oppfinnelsen, skjer ifølge følgende skjema: - lyofilisering av platekulturer; - ekstrahering av lyofilisatet med et organisk oppløsningsmiddel; - ekstrahering av forbindelsen fra kulturfiltratet med festfaser; - analyse ved hjelp av HPLC, DC eller ved undersøkelse av den biologiske virksomheten.
De i det følgende beskrevne fermenteringsbetingelser gjelder for Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, det deponerte isolatet DSM 13784, så vel som mutanter og varianter av den samme.
I en næringsløsning som inneholder en karbonkilde og en nitrogenkilde så vel som de vanlige uorganiske salter, produserer Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784, caloporosidderivatene.
Som foretrukne karbonkilder for fermentering er assimilerbare karbohydrater egnet samt sukkeralkoholer, som glukose, laktose, sakkarose eller D-mannitt, så vel som karbohydratholdige naturprodukter, som for eksempel maltekstrakt. Som nitrogenholdige næringsstoffer anvendes: aminosyrer, peptider og proteiner, så vel som deres nedbrytingsprodukter, som kasein, peptoner eller tryptoner, ytterligere kjøttekstrakter, gjærekstrakter, malte frø, for eksempel fra mais, hvete, bønner, soya eller bomullsplante, destilleirngsrester fra alkoholfremstilling, kjøttmei eller gjærekstrakter, men også ammoniumsalter og nitrater, spesielt også syntetisk henholdsvis biosyntetiske utvunnede peptider. Av uorganiske salter kan næringsløsningen inneholde for eksempel klorider, karbonater, sulfater eller fosfater fra alkali- eller jordalkalimetallene, jern, sink, kobolt og mangan.
Dannelsen av forbindelsene ifølge oppfinnelsen skjer spesielt godt i en næringsløsning som inneholder ca. 0,05 til 5%, fortrinnsvis 1 til 2% maltekstrakt, 0,05 til 3%, foretrukket 0,05 til 1% gjærekstrakt og 0,2 til 5%, fortrinnsvis 0,5 til 2% glukose, 0,5 til 3%, fortrinnsvis 0,5 til 3% cellulosepulver og sporer av ammoniumsulfat. Angivelsene i prosent er henholdsvis på basis av vekten av den totale næringsløsningen.
I denne næringsløsningen produserer Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784 en blanding av caloporosidderivater. Avhengig av sammensetningen av næringsløsningen kan man variere den mengdemessige andelen av ett eller flere av caloporosidderivatene ifølge oppfinnelsen. Ytterligere er det mulig å styre syntesen av enkelte caloporosidderivater ved næringsmediesammensetning slik at et caloporosid-derivat ikke fremstilles i det hele tatt, henholdsvis i en mengde som ligger under påvisningsgrensen av mikroorganismen.
Kultiveringen av mikroorganismen skjer aerobt, altså for eksempel nedsunket ved risting eller røring i ristekolbe eller fermenter, eventuelt ved å tilføye luft eller oksygen eller på fast medium. Dette kan gjennomføres i et temperaturområde på fra ca. 18 til 35°C, fortrinnsvis ved ca. 20 til 30°C, spesielt ved 25 til 30°C. pH-området bør ligge mellom 5 og 8, fortrinnsvis mellom 5,5 og 6,5. Man kultiverer mikroorganismen under disse betingelser generelt over et tidsrom på fra 24 til 720 timer, fortrinnsvis 288 til 576 timer.
Det er fordelaktig å kultivere i flere trinn, dvs. man fremstiller først en eller flere forkulturer i et flytende næringsmiddel som deretter overføres til det egentlige produksjonsmediet, hovedkulturen, for eksempel i volumsforholdet 1:10. Forkulturen oppnår man for eksempel ved at man overfører et mycel i en næringsløsning og at man lar det samme vokse i ca. 36 til 120 timer, fortrinnsvis 48 til 72 timer. Mycelet kan for eksempel oppnås ved at man lar stammen vokse ca. 3 til 40 dager, fortrinnsvis 10 til 30 dager, på et fast eller flytende næringsmedium, for eksempel malt-gjær-agar eller potet-dekstrose-agar.
Fermenteringsforløpet kan overvåkes ved hjelp av pH-verdien til kulturen og mycelvolumet så vel som ved kromatografiske metoder, som for eksempel høy prestasjonsvæskekromatografi (HPLC) eller utprøving av den biologiske aktiviteten.
Den i det følgende beskrevne isoleringsfremgangsmåte tjener til rensing av caloporosidderivatene ifølge oppfinnelsen.
Isoleringen henholdsvis rensingen av caloporosidderivatene ifølge oppfinnelsen fra kulturmediet skjer ifølge kjente fremgangsmåter, ved å ta hensyn til de kjemiske, fysikalske og biologiske egenskapene til naturstoffene. For testing av konsentrasjonen av de respektive caloporosidderivatene i kulturmediet eller i de enkelte isoleringstrinn, kan man anvende HPLC, hvorved mengden av den dannede substansen kan sammenlignes hensiktsmessig med en justeringsoppløsning.
For isolering av forbindelsene ifølge oppfinnelsen lyofiliserer man kultur- samt fastmediet og deretter ekstraheres caloporosidderivatene fra lyofilisatet med et egnet, organisk oppløsningsmiddel blandbart med vann. Den organiske oppløsningsmiddelfasen inneholder naturstoffene ifølge oppfinnelsen og eventuelt konsentreres disse i vakuum og renses videre.
Den ytterligere rensingen av en eller flere av forbindelsene ifølge oppfinnelsen, skjer ved hjelp av kromatografi på egnede materialer, fortrinnsvis for eksempel en molekylsikt, på kiselgel, aluminiumoksid, på ionebyttere eller på adsorbérharpikser henholdsvis med omvendt fase (reversed phase, RP). Ved hjelp av kromatografien separeres caloporosidderivatene. Kromatografien av caloporosidderivatene skjer med bufrede vandige oppløsninger eller blandinger av vandige og organiske oppløsninger.
Under blanding av vandige eller organiske oppløsninger forstår man alle organiske løsningsmidler blandbare med vann, fortrinnsvis metanol, propanol og acetonitril, i en konsentrasjon på fra 5 til 80% løsningsmiddel, fortrinnsvis 20 til 50% løsningsmiddel eller også alle bufrede vandige oppløsninger som kan blandes med organiske løsningsmidler. Bufferne som skal anvendes er de samme som beskrevet over.
Separasjon av caloporosidderivatene på grunn av deres forskjellige polaritet skjer ved hjelp av den omvendte fasekromatografien, for eksempel på MCI® (adsorberharpiks fra Mitsubishi, Japan) eller Amberlite XAD <D (TOSOHAAS), på ytterligere hydrofobe materialer, som for eksempel på RP-8- eller RP-18-faser. Dessuten kan separasjon også skje ved hjelp av normalfasekromatografi, for eksempel på kiselgel, aluminiumoksid eller lignende.
Kromatografien av caloporosidderivater skjer med bufrede eller surgjorte vandige oppløsninger eller blandinger av vandige oppløsninger med alkohol eller andre, ved vann blandbare organiske løsningsmidler. Som organiske løsningsmidler anvendes fortrinnsvis propanol og acetonitril.
Som bufrede eller surgjorte vandige oppløsninger forstår man for eksempel vann, fosfatbuffer, ammoniumacetat, citratbuffer i en konsentrasjon på fra 0 til 0,5 M så vel som maursyre, eddiksyre, trifluoreddiksyre eller alle i handelen vanlige, for fagmannen kjente syrer, fortrinnsvis i en konsentrasjon på fra 0 til 1%. Med bufrede vandige oppløsninger foretrekkes spesielt 0,1% ammoniumacetat.
Man kromatograferer med en gradient som begynner med 100% vann og slutter med 100% oppløsningsmiddel, fortrinnsvis kjørt med en lineær gradient på fra 20 til 100% propanol eller acetonitril.
Alternativt kan det også utføres en gelkromatografi eller kromatografi på hydrofobe faser.
Gelkromatografien gjennomføres på polyakrylamid- eller blandingspolymergeler, som for eksempel Biogel-P 2® (Fa. Biorad) eller Fractogel TSK HW 40® (Fa. Merck, Tyskland eller Toso Haas, USA).
Rekkefølgen av de tidligere nevnte kromatografier kan snus.
Forbindelsene ifølge oppfinnelsen er stabile i fast tilstand og i oppløsninger i pH-området mellom 3 og 8, spesielt 5 og 7, og lar seg dermed innarbeide i vanlige galeniske tilberedninger.
En eller flere forbindelser av forbindelsene ifølge oppfinnelsen egner seg på grunn av deres verdifulle farmakologiske egenskaper for anvendelse i human- eller dyremedisin som legemidler.
Den foreliggende oppfinnelse angår således anvendelse av forbindelsen med formel I eller et fysiologisk akseptabelt salt av den samme for fremstilling av et cytostatikum for behandling av tumorsykdommer.
Foreliggende oppfinnelse angår ytterligere alle åpenbare kjemiske ekvivalenter av forbindelsene med formel I ifølge oppfinnelsen. Slike ekvivalenter er forbindelser som oppviser en liten kjemisk forskjell, men som har den samme virkningen eller som omdannes under milde betingelser til forbindelsene ifølge oppfinnelsen. Til de nevnte ekvivalenter hører for eksempel også salter, reduksjonsprodukter, estere, etere, acetaler eller amider av forbindelsene ifølge oppfinnelsen, samt ekvivalenter som fagmannen kan fremstille ved hjelp av standardfremgangsmåter i tillegg til alle optiske antipoder, diastereomerer og alle stereomere former.
Som fysiologisk akseptable salter av forbindelsene med formel I forstår man både deres organiske og uorganiske salter som er beskrevet i Remington's Pharmaceutical Sciences (17. opplag, s. 1418 (1985)). På grunn av den fysikalske og kjemiske stabiliteten og oppløseligheten foretrekkes for sure grupper blant annet natrium-, kalium-, kalsium- og ammoniumsalter; for basiske grupper foretrekkes blant annet salter av saltsyre, svovelsyre, fosforsyre eller av karbonsyre eller sulfonsyre, som for eksempel eddiksyre, sitronsyre, benzosyre, maleinsyre, fumarsyre, vinsyre og p-toluolsulfonsyre.
Estere, etere og acetater kan fremstilles ved hjelp av fremgangsmåter beskrevet i litteraturen, for eksempel i Advanced Organic Synthesis, 4. utgave, J. March, John Wiley & Sons, 1992 eller Protective Groups in Organic Synthesis 3. utgave, T.W. Greene & P.G.M. Wuts, John Wiley & Sons, 1999.
Karboksylgruppene kan for eksempel reduseres med LiAlFLttil alkohol.
Fra forbindelser med formel I kan først og fremst glykosiddelen avspaltes ved hjelp av alkalisk hydrolyse (W. Weber et al. J. Antibiotics, 47,1188-1194). Deretter kan ved hjelp av glykosylering (for eksempel Konigs-Knorr-reaksjonen) innføres hvilke som helst sukkerrester. Tilsvarende fremgangsmåter er beskrevet i litteraturen, for eksempel i Carbohydrate Chemistry, J.F. Kennedy, Oxford University Press, 1988.
Virkningsmekanismen av caloporosidderivatene er ukjent, imidlertid kunne man påvise en betydelig virkning.
For påvisning av inhibitorene av CDK'er bruker man en undersøkelse ved hvilken fosforyleringshastigheten til et spesifikt peptidsubstrat måles ved hjelp av cyklinavhengige kinaser. De cyklin-avhengige kinaser aktiveres ved binding til det respektive cyklin. [y-P]-fosfat overføres fra [y-P]-ATP ved enzymet på peptidsubstratet. Undersøkelsen gjennomføres i 96-brønn mikrotiterplater: Radioaktiviteten av fosfatet av [y-P]-fosfatet som ble overført på substratet måles.
IC50-verdiene for caloporosidderivatene vises i Tabell 1; det er konsentrasjonen som inaktiverer CDK-4 med 50%.
Ytterligere angår den foreliggende oppfinnelsen legemidler med et innhold på minst en forbindelse ifølge oppfinnelsen.
En eller flere forbindelser av caloporosidderivatene ifølge oppfinnelsen kan prinsipielt administreres som sådan i substansen. Man foretrekker anvendelsen av blandingen med egnede hjelpestoffer eller bærematerialer. Som bæremateriale kan man anvende ved legemidler de vanlige og farmakologisk akseptable bærematerialer og/eller hjelpestoffer.
Legemiddelet ifølge oppfinnelsen administreres generelt oralt eller parenteralt, men det er også prinsipielt mulig med en rektal anvendelse. Egnede faste eller flytende galeniske tilberedningsformer er for eksempel granulater, pulvere, tabletter, drageer, (mikro-)kapsler, stikkpiller, siruper, emulsjoner, suspensjoner, aerosoler, dråper eller injiserbare oppløsninger i pulverform så vel som preparater med protraherte virkestoff-frigivning, ved hvis fremstilling man anvender vanligvis bærestoff og tilsetninger og/eller hjelpemidler som spreng-, binde-, overtrekks-, svelle-, glide- eller smøremidler, smaksstoffer, søtningsmidler eller oppløsningsformidlere. Som ofte anvendte bære- og hjelpestoffer nevnes for eksempel magnesiumkarbonat, titandioksid, laktose, mannitt og andre sukkere, talkum, melkeprotein, gelatin, stivelse, vitaminer, cellulose og dens derivater, oljer fra plante og dyr, polyetylenglykoler og oppløsningsmidler, som for eksempel sterilt vann, alkoholer, glycerin og flerverdige alkoholer.
Eventuelt kan doseringsenhetene for den orale administrasjonen mikroinnkapsles for å forsinke frigivningen eller for å utvide den over et lengre tidsrom som for eksempel ved overtrekking eller innlegging av virkestoffet i partikkelform i egnede polymerer, vokser eller lignende.
Fortrinnsvis fremstilles og administreres de farmasøytiske preparater i doseringsenheter hvorved hver enhet inneholder som aktiv bestanddel en bestemt dose av en eller flere forbindelser av caloporosidderivatene ifølge oppfinnelsen. Ved faste doseringsenheter som tabletter, kapsler og stikkpiller kan denne dosen utgjøre opp til ca. 500 mg, fortrinnsvis imidlertid ca. 0,1 til 200 mg og ved injeksjonsløsninger i ampulleform opp til ca. 200 mg, fortrinnsvis ca. 0,5 til 100 mg, pr. dag.
Dagsdosen som skal administreres avhenger av kroppsvekt, alder, kjønn og tilstand hos pattedyret. Under visse omstendigheter kan imidlertid også høyere eller lavere dagsdoser anvendes. Administrasjon av dagsdosen kan både skje ved engangsadministrering i form av en enkel doseringsenhet eller i flere små doseringsenheter og også ved flere gangers administrering av avdelte doser i bestemte intervaller.
Legemidlene ifølge oppfinnelsen fremstilles ved at man bringer en eller flere av forbindelsene ifølge oppfinnelsen med i henholdsvis en egnet administreringsform med de vanlige bære- henholdsvis eventuelt tilsetnings- og/eller hjelpestoffer.
I de følgende eksempler beskrives oppfinnelsen nærmere. Prosentangivelser baserer seg på vekt. Blandingsforhold ved væske baserer seg på volum, hvis ikke noe annet er angitt.
Eksempler
Eksempel 1: Fremstilling av en glycerinkultur av Gloeoporus dichrous (Fr.:Fr.)
Bres. ST 001714, DSM 13784
100 ml næringsløsning (maltekstrakt 2,0%, gjærekstrakt 0,2%, glukose 1,0%
(NH4)2HP040,05%, pH 6,0) i en steril 300 ml Erlenmeyerkolbe ble inkubert med stammen Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784 og holdt i 7 dager ved 25°C og 140 omdreininger pr. minutt på en roterende ristemaskin. 1,5 ml av denne kulturen ble deretter fortynnet med 2,5 ml glycerin med en konsentrasjon på 80% og lagret ved-135°C.
Eksempel 2: Fremstilling av en forkultur av Gloeoporus dichrous (Fr.:Fr.) Bres.
ST 001714, DSM 13784 i en Erlenmeyerkolbe
30 ml næringsløsning (maltekstrakt 2,0%, gjærekstrakt 0,2%, glukose 1,0% (NH4)2HPO40,05%, pH 6,0) i en steril 100 ml Erlenmeyerkolbe ble inokulert med stammen Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784 og inkubert i 4 dager ved 25°C og 140 omdreininger pr. minutt på en roterende ristemaskin. Med respektivt 2 ml av denne forkulturen ble deretter platene inokulert ved fremstilling av hovedkulturene.
Eksempel 3: Fremstilling av en hovedkultur av Gloeoporus dichrous (Fr.:Fr.)
Bres. ST 001714, DSM 13784 på fastmediumplater
1 sterile 25 x 25 cm plater (Fa. Nunc) ble det helt 200 ml av den følgende næringsløsningen: 20 g/l maltekstrakt, 2 g/l gjærekstrakt, 10 g/l glukose og 0,5 g/l (NFLO2HPO4pH 6,0. Disse platene ble inokulert med respektive 2 ml av en forkultur. Den maksimale produksjonen av en eller flere av forbindelsene ifølge oppfinnelsen ble oppnådd etter ca. 480 timer.
Eksempel 4: Fremstilling av caloporosidderivater
50 stykk 25 x 25 cm plater ble fremstilt og inokulert med forkultur:
Næringsmedium:
20 g/l maltekstrakt
2 g/l gjærekstrakt
10 g/l glukose
0,5 g/l (NH4)2HPO4
pH 6 (før sterilisering)
Inkubasjonstid: 480 timer
Inkubasjonstemperatur: 25°C.
Eksempel 5: Isolering av caloporosidblandingen fra platekulturer av Gloeoporus
dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784
Etter avslutningen av fermenteringen av Gloeoporus dichrous (Fr.:Fr.) Bres. ST 001714, DSM 13784 ble platekulturene, utvunnet ifølge Eksempel 3, lyofilisert og lyofilisatet ble ekstrahert med 5 liter metanol. Den virkestoffholdige, metanolske oppløsningen ble behandlet ved filtrering for å fjerne resten og konsentrert i vakuum. Konsentratet ble fortynnet med vann og påført på en forberedt, 1,0 liter MCI GEL, CHP20P-søyle. Man eluerte med en gradient av vann etter 100% acetonitril. Søylegjennomstrømningen (25 ml pr. minutt) ble oppfanget fraksjonert (respektive 25 ml) og caloporosidderivatene som inneholdes i disse fraksjoner (fra 40% til 100% acetonitril) ble sammenfattet. Konsentrering i vakuum og etterfølgende lyofilisering gir 8,5 g av et gulbrunt pulver.
Eksempel 6: Forseparasjon av caloporosidderivatene ved hjelp av RP18-kromatografi
0,5 g av det ifølge Eksempel 4 utvunnede produktet ble påført på en Nucleosil® 100-7 Cl8 HD-søyle (størrelse: 40 mm x 250 mm). Man eluerer med en gradient av 20% acetonitril (+ vann med 0,1% ammoniumacetat-tilsats) etter 100% acetonitril med en strømningshastighet på 35 ml pr. minutt. Søyleutstrømningen ble fraksjonert (35 ml) oppfanget. Hovedsakelig befinner caloporosidderivatene seg i fraksjonene 39 til 68. De sammenfattes, fjernes fra løsningsmiddelet i vakuum og deretter lyofiliseres disse. Således oppnår man Caloporosid B (22,4 mg) og F (10,5 mg) allerede > 95% rent. Caloporosid C (fraksjon 41; 43,4 mg), D (fraksjon 43-45; 76,2 mg) og E (fraksjon 43-45; 76,2 mg) ble oppnådd med en renhet på ca. 70% og derfor ytterligere renset ved hjelp av kromatografi.
Eksempel 7: Rensing av Caloporosid C, D og E
20 mg av det ifølge Eksempel 5 isolerte og anrikede caloporosid C ble påført på en LUNA ® 5 (i C18(2)-søyle (størrelse: 10 mm x 250 mm) og kromatografert med en gradient på fra 25 til 35% acetonitril i 0,1% ammoniumacetat/vann. Gjennomstrømningen av elueringsmiddelet var 6,5 ml pr. minutt, fraksjonsstørrelsen 6,5 ml. I fraksjonene 35 til 42 befinner caloporosid C seg. Lyofiliseringen av de nevnte fraksjoner gir > 95% rent caloporosid C (7,5 mg). 25 mg av den ifølge Eksempel 5 isolerte og anrikede blandingen av caloporosid D og E ble påført på en LUNA® 5 u C18(2) søyle (størrelse: 10 mm x 250 mm) og kromatografert med en gradient på fra 30 til 40% acetonitril i 0,1% ammoniumacetat/vann. Gjennomstrømning av elueringsmiddelet er 6,5 ml pr. minutt, fraksjonsstørrelsen er 6,5 ml. I fraksjonene 18 og 19 befinner caloporosid D seg, i fraksjonene 20 til 21 caloporosid E. Lyofiliseringen av de nevnte fraksjoner gir > 95% rent caloporosid D (7,0 mg) og caloporosid E (6,0 mg).
De fysikalsk-kjemiske så vel som spektroskopiske egenskaper av substansene ifølge oppfinnelsen lar seg sammenfatte som følger:

Eksempel 8: Bioanalyse på CDK-4 inhibitorer
For bestemmelse av ICso-verdien fremstilles stamoppløsninger i en konsentrasjon på 10 mM fra naturstoffene ifølge oppfinnelsen. 384-brønn flash-plater ble belagt med 50 (il (50 mg/brønn) bioinylert peptidsutbrat ved romtemperatur i 2 timer og deretter vasket med 3 x PBS buffer. For reaksjonen ble 30 ul av en med buffer fortynnet oppløsning av caloporosidderivatene og 20 ul av en forblandet ATP/cyklinDl/CDK4 oppløsning (sluttkonsentrasjon: 1 uCi 33P-y-ATP, 2 uM ATP og 1 ug enzymblanding) pipettert på plater. Etter 2 timers reaksjonstid ved 37°C vaskes platene tre ganger med respektive 80 ul fosforsyre 3% og deretter måles disse i en MicroBeta Counter i 30 sekunder. Bestemmelsen av den prosentmessige inhiberingen skjer ved hjelp av matematiske ligninger. For bestemmelsen av ICso-verdiene ble det undersøkt 10 konsentrasjoner av en frisk fortynnet DMSO-oppløsning av substansene ifølge oppfinnelsen.

hvor: Ri, R2, R3, uavhengig av hverandre betyr H eller acylrester med 1 til 10 C-atomer; og R4betyr H eller -C(0)(CH2)„COOH, hvori n er lik 1 til 7; med unntak av at Ri, R2, R3og R4ikke alle betyr H; så vel som deres fysiologisk akseptable salter.

Fremgangsmåte ifølge krav 9,karakterisert vedat fermenteringen gjennomføres under aerobe betingelser ved en temperatur på mellom 18 og 35°C og ved en pH på mellom 5 og 8.
