Description
Foreliggende oppfinnelse omhandler en fremgangsmåte for fremstilling av derivater av 2-(1-piperazinyl)-4-fenyl-kinazolin.
I norsk patentsøknad nr. 82.3491 er det beskrevet derivater av 4-fenyl-kinazolin svarende til følgende formel:
i hvilken betegner en tertiær aminogruppe som eventuelt er cyklisk og som inneholder en hydroksylgruppe, og R2 representerer et atom klor eller fluor.
Forbindelsene (I) har krampestillende egenskaper og evne til potensialisering av narkose som tillater deres bruk som medikamenter slik som anxiolytika (angststillende midler), hypnotika og antiepileptika.
Det er funnet ut, på en overraskende måte, ved å variere type av substituenten , at de oppnådde produkter ikke lenger utviste de farmakologiske egenskaper til produktene (I), men hadde meget interessante antidepressive egenskaper.
De nye forbindelser svarer til den generelle formel:
i hvilken:
R^ betegner et halogenatom, fortrinnsvis klor eller en nitro-gruppe,
R4 representerer hydrogen eller en lav-alkylgruppe som har fra 1 til 4 karbonatomer,
R,- representerer et halogenatom, fortrinnsvis klor eller fluor.
Forbindelsene (II) gir sammen med mineral- eller orga-niske syrer løselige salter. Disse salter utgjør sammen med de akseptable farmasøytiske syrer en vesentlig del av oppfinnelsen .
Man vil bemerke at i det spanske patent 453 982 beskriv-es en fremgangsmåte for fremstilling av derivater av 2-amino-kinazoliner, substituert blant annet i posisjon 4 med et fe-nylradikal, som eventuelt er substituert. Produktene som oppnås som følge av denne fremgansmåte og er beskrevet i det nevnte spanske patent, er forskjellige fra de produkter som er beskrevet her; på den annen side gir det nevnte spanske patent ingen brukbare opplysninger angående de fremstilte produkters farmakologiske egenskaper.
Forbindelsene (II) kan i henhold til oppfinnelsen fremstilles ut fra et passende substituert 4-fenyl-2-kinazolon (1) ifølge reaksjonsskjemaet:
Ved virkning av et klorert fosforderivat på kinazolonet (1) får man derivatet (2) klorert i posisjon 2. Man bruker oftest fosforoksyklorid. Man kan operere i en inert løsning slik som et aromatisk hydrokarbon (benzen eller toluen), men oftest foretrekker man å bruke et overskudd av oksykloridet som løsningsmiddel. Det henvises forøvrig til patentkravet.
Reaksjonen finner sted ved en temperatur mellom 60 og 120°C og oftest ved kokepunktet til det anvendte løsningsmid-del.
Ut fra klorderivatet (2) ved virkning av et amin
i overskudd i et inert løsningsmiddel som etanol ,
får mån den korresponderende forbindelsen (II). Man utfører det vanligvis ved å varme opp til løsningsmidlets kokepunkt.
Saltene av forbindelsene (II) fåes på den vanlige måten
ved å omdanne basen til salt under oppvarming med en støkio-metrisk mengde syre i et passende løsningsmiddel, valgt slik at det dannede salt utkrystalliserer ved avkjøling.
Utgangskinazolonene (1) er kjente forbindelser. Når
R, = halogen, kan de i særlig grad fremstilles ved virkning
av kaliumcyanat på et passende substituert 2-amino-5-halogen<-benzofenon.
Når R^ representerer nitrogruppen, kan forbindelsene (1)
fåes ved virkning av urea på et passende substituert 2-amino-5-nitro-benzofenon.
De følgende eksempler er gitt som illustrasjon på frem-stillingen av forbindelsene (II) i henhold til den foregående beskrevne metoden.
EKSEMPEL 1
2-(l-metyl-4-piperazinyl)-4-(2-klorfenyl)-6-nitro-kinazolin
(CM 4 04 9 8)
(II) R3 = N02 ; R4 = CH3 ; R5 = Cl
a) 2-klor-4-_(23klorfenYl)_362nitro-kinazolin Man varmer opp under tilbakeløp i 4t en blanding av 40g
4-(2-klor-fenyl)-6-nitro-2-kinazolon og 600 ml fosforoksy-
klorid. Man fordamper under vakuum fosforoksykloridet til resten er tørr, deretter har man resten i en blanding is/vann.
Man alkaliserer med en 10% natriumhydroksyd-løsning. Man
tørker bunnfallet, og man vasker med acetonitril. Man renser produktet ved kromatografi på en silikakolonne. Ved å
eluere med en blanding kloroform/metanol (95-5 volum/volum), isolerer man det forventede produkt. Vekt: 27 g; Smeltepunkt:
228°C (isopropanol).
b) CM_40498
Man varmer under avtrekk i 3t en blanding av 3,2g av det'
klorderivatet man fikk foran, og 3g N-metyl-piperazin i 120ml absolutt etanol. Man fordamper løsningen under vakuum til resten er tørr, deretter har man resten i etylacetat. Man vasker løsningen med vandig løsning av natriumkarbonat, deretter med vann. Man tørker løsningen over natriumsulfat og fordamper så til resten er tørr. Resten rekrystalliseres i en blanding metanol/diklormetan. Man får krystaller. Vekt: 3,1 g; Smeltepunkt: 204°C.
EKSEMPLER 2 til 6
a) Idet man går fram som i eksempel la) men varierer kinazolonet, får man på samme måte: -2-klor-4-(2-fluorfenyl)-6-nitrokinazolin;
smp.: høyere enn 260°C (isopropanol) -2,6-diklor-4-(2-klorfenyl)-4-kinazolin smp.: 175-176°C (etanol) -2,6-diklor-4-(2-fluorfenyl)-kinazolin;
smp.: 208-210°C (acetonitril)
b) Ut fra de forskjellige derivater klorert i posisjon 2 nevnt foran og idet man varierer det anvendte amin
får man som påpekt i Eksempel lb) de forskjellige
forbindelsene (II) som er samlet x tabell I bak.
Produktene i henhold til oppfinnelsen har vært underlagt farmakologiske forsøk med sikte på å bestemme deres virkning på sentralnervesystemet.
Under angis de forskjellige prøver som produktene har vært underlagt. I alle disse tilfeller ble produktene som skulle studeres, administrert per os.
A. PORSOLTS TEST
Denne test ble gjennomført på hunmus, CDI (Charles Rivers, Frankrike) som veide fra 18 til 23 g, i henhold til metoden beskrevet av Porsolt (Archives Internationales de Pharmacologie, 1977, 229, 327-336).
Prinsippet i denne testen er følgende: Når en mus "blir plassert i et trangt kar fylt med vann, slår den om seg, så,
i løpet av 2 til 4 min., blir den ubevegelig og flyter på magen med rund rygg og med baklabbene ført inn under kroppen; den gjør bare noen få bevegelser som er nødvendige for å holde hodet over vannet. Dette er fortvilelsesreaksjonen (despair reaction).
Bestemte psykofarmaka, særlig antidepressive, forlenger tiden under hvilken musen slår om seg.
Følgende skjema ble valgt:
Produktene som skulle studeres, ble administrert gjennom munnen til grupper på 10 mus: 1 t senere blir dyrene plassert i et trangt kar (10 x 10 x 10 cm) fylt med vann til en høyde av 6 cm, vanntemperaturen er 24°C. Man lar dyrene være 6 min. i vannet og måler tiden når dyret blir ubevegelig mellom det andre og det sjette minuttet. Jo kortere tiden er, desto mer aktivt er stoffet.
Resultatene er uttrykt som forkortelsen av immobiliser-ingstiden i forhold til kontrolldyrene.
B. ANTAGONISME TIL PTOSIS INDUSERT VED RESERPIN
De fleste antidepressiva virker antagonistisk til ptosis indusert ved reserpin. Denne test, beskrevet ved Gouret [Journal de Pharmacologie (Paris), 1973, 4 (1), 105-128], ble gjennomført på hunmus CDI (Charles Rivers, Frankrike) som veide fra 18 til 23 g. Reserpin medfører en ptosis 1 t etter intravenøs administrering; bestemte antidepressiva, særlig imipraminene, motvirker denne ptosis.
F ølgende skjema ble valgt:
Stoffene som skulle studeres, ble administrert per os til grupper på 10 mus. Samtidig ble reserpin administrert intravenøst i en dose på 2 mg/kg. 1 t etter administrasjonen av reserpin legger man merke til antall dyr som ikke har
ptosis.
C. ANTAGONISME TIL HYPOTERMI INDUSERT VED RESERPIN
De fleste antidepressiva virker antagonistisk til hypotermi indusert ved reserpin. Denne test ble gjennomført i henhold til metoden beskrevet av HINO og Coll [Chem Pharm Bull 28 (9), 2618-2622, 1980] på hunmus CDI (Charles.Rivers, Frankrike) som veide fra 18 til 23 g.
Følgende skjema ble valgt:
Stoffene som skal studeres, blir administrert per os til grupper på 10 mus, idet forsøksdyrene mottar bare løsningen; samtidig blir reserpin administrert i.p. (intraperitonealt)
i en dose på 5 mg/kg. Temperaturen til hvert dyr blir tatt rett før administrasjonen av produktene som skal studeres, og 4t etterpå. For hvert dyr blir differansen mellom temperaturen før og etter behandling regnet ut. Resultatene er uttrykt i prosent av antagonismen til hypotermi observert hos forsøksdyrene.
D. POTENSIALISERING AV TOKSISITETEN TIL YOHIMBIN
De fleste antidepresssiva virker potensialiserende til toksisiteten av yohimbin. Denne test ble gjennomført i henhold til metoden beskrevet ved MALICK (i Antidepressants: Neurochemical Behavioral and Clinical Perspectives, eds
S.J. ENNA, J.B. MALICK, E. RICHELSON, Raven Press, New York,
s. 141-155) på hunmus CDI (Charles Rivers, Frankrike).
Stoffene som skal studeres, blir administrert per os til grupper på' 10 mus. Yohimbin blir administrert i.p. It senere i en dose på 30mg/kg. Dødeligheten blir konstatert 18t senere.
E. ANTAGONISME TIL SKJELVINGER VED OXOTREMORIN
De cholinergiske virkninger av imipramin betraktes som ansvarlige for bestemte sjenerende sekundærvirkninger dette stoffet har. Disse virkningene blir klarlagt gjennom antagonisme til skjelvinger ved oxotremorin. Denne test ble gjennomført på hunmus CDI (Charles Rivers, Frankrike) som
veide fra 18 til 23 g.
Følgende skjema ble valgt:
Produktene som skal studeres, blir administrert gjennom munnen til grupper på 10 mus. Ved tid 60 min. blir oxotremorin administrert per os i en dose på 1 mg/kg. Man legger merke til antall mus som ikke skjelver 30 min. etter administrasjonen av oxotremorin.
Samtlige farmakologiske resultater fått med forskjellige forbindelser i henhold til oppfinnelsen er samlet i tabell II bak. I denne tabell opptrer de oppnådde resultater likt, på
den ene side med imipramin, et stoff hvis antidepressive egenskaper hovedsakelig brukes terapeutisk, og på den annen side med forbindelsen CM 40 331 eller hydrokloridet til 6-klor-4-(2-klorfenyl)-2-(4-hydroksy-l-piperidinyl)-kinazolin,
et stoff som er beskrevet i fransk patentsøknad nr. 81/19767.
Resultatene i tabell II viser klart at de her
beskrevne produkter har kraftig antidepressiv virkning. Denne virkningen er i de fleste tilfeller kraftigere enn virkningen av imipramin og ledsages av en toksisitet og cholinergiske sekundærvirkninger som er klart svakere enn disse hos imipramin.
På den annen side kan man legge merke til at produktet
CM 40 331 som ble studert, i sammenligning er praktisk talt inaktivt ved de praktiske testene.
Følgelig kan de her beskrevne produkter brukes
i human terapi til behandling av nevropsykiske problemer slike som endogene, reaksjonelle og nevrotiske depressive tilstander slik som involusjonsdepresjon hos eldre.
Produktene kan tilbys i galeniske former som tilsvarer
oral bruk (tabletter, geler etc.) og parenteral bruk (injiser-bare ampuller).
Doseringen som varierer avhengig av lidelsen som skal behandles og adminis.trasjonsveien, vil være tiltagende og befinne seg mellom 50 og 300 mg pr. dag for voksne.
Claims (1)
- Analogifremgangsmåte for fremstilling av terapeutisk aktive derivater av 2(1-piperazinyl)-4-fenyl-kinazolin med generell formel:i hvilken R, betegner et halogenatom, fortrinnsvis klor eller en nitro-gruppe,R4 representerer H eller en alkylgruppe som har fra 1 til 4 karbonatomer,Rtj representerer et halogenatom, fortrinnsvis klor eller fluor, og farmasøytisk akseptable salter av de nevnte derivater, karakterisert ved at man som utgangsforbindelse anvender et kinazolon med formelen:i hvilken R^ og R,, er betegnet som ovenfor, at man bringer et klorert fosforderivat til å reagere med dette kinazolon, fortrinnsvis fosforoksyklorid, og at man med det slik oppnådde klorerte derivat bringer et amin med formelen( til å reagere, <p>g om ønsket omdannes den oppnådde forbindelsen med formel II til et farmasøytisk akseptabelt salt ved omsetning med en passende syre.
