Abstract
Analogifremgangsmåte for fremstilling av terapeutisk aktive aryloksyalkyl-diketoner og -keto-estere.
Description
Foreliggende oppfinnelse angår en analogifremgangsmåte for fremstilling av terapeutisk aktive aryloksyalkyl-diketoner og -ketoestere med formel
der:
Alk er rettkjedet alkylen med 4-7 karbonatomer;
R er alkyl med 1-5 karbonatomer;
R<1> er alkyl med 1-5 karbonatomer eller alkoksy
med 1-5 karbonatomer;
R^, R2 og R^ uavhengig av hverandre er hydrogen, alkyl med 1-4 karbonatomer, alkoksy med 1-4 karbonatomer, halogen, trifluormetyl, hydroksy, benzyloksy, lavere alkylbenzoyloksy, karboksy, alkoksykarbonyl med 3-4 karbonatomer, metoksybenzoyloksy, dialkylamino der alkyl har fra 1-4 karbonatomer, dialkylaminoalkoksy der alkyl har fra 1-4 karbonatomer og alkoksy har fra 2-4 karbonatomer, eller to av R^, R~ og R^ sammen danner 3,4-metylendioksy, med antiviral virkning.
US-PS 3.541.155 beskriver forbindelser med formelen
der n er et helt tall fra og med 0 til og med 5. Forbindelsene er angitt å være brukbare som "biological toxicants", spesifikt som agrikulturelle baktericider, fungicider og herbicider.
Forbindelsene ifølge formel I, fremstilt ved fremgangsmåten ifølge oppfinnelsen, skiller seg fra de som er beskrevet i dette US-PS ved nærværet av en alkylenbro inneholdende minst 4 karbonatomer mellom oksygenatomet og det ter-tiære karbonatom. Videre har de to grupper av forbindelser forskjellige egenskaper, fordi forbindelsene ifølge dette US-PS ikke er vist å ha antiviral virkning.
I den ovenfor angitte generelle formel I står Alk for en mettet alifatisk hydrokarbonbro inneholdende 4-7 karbonatomer.
Karbonkjedene i R og R' kan være rette eller for-grenede, selv om primære eller sekundære alkyldeler er foretrukket.
Substituentene R-i , R2 og R 7 kan være
like eller forskjellige.
Ifølge oppfinnelsen fremstilles en forbindelse med formel I ved at man i et inert oppløsningsmiddel og under vannfrie betingelser kondenserer enten
a.) en halogenalkylfenyleter med formelen:
med et alkalimetallsalt eller tungmetallchelat av et diketon med formelen:
eller
b) en forbindelse med formelen:
med en forbindelse med formelen:
hvori Alk, R og R' har den ovenfor angitte betydning og R^' , R2' og R^' uavhengig av hverandre er hydrogen, alkyl med fra 1-4 karbonatomer, alkoksy med fra 2-4 karbonatomer, halogen, trif luormetyl, benzyloksy eller dialkylamino eller to av R-^' , R2<1> og R3' sammen danner 3,4-metylendioksy, og X er brom eller jod; og M er et alkalimetall; og, hvis ønskelig,
i) underkaster en oppnådd forbindelser der R^', R2' og R^' er 1-3 benzyloksygrupper, en katalytisk hydrogenolyse for å fremstille den tilsvarende forbindelse der R^, R2 og R^ er 1-3 hydroksygrupper;
ii) underkaster en oppnådd forbindelse der R-^, R^ eller R^ er hydroksy en forestring for å fremstille den tilsvarende forbindelse der R^, R2 og R-j er benzoyloksy, alkylbenzoyloksy, metoksybenzoyloksy ved behandling av hydroksyforbind-elsen med det egnede benzoylhalogenid eller substituerte benzoylhalogenid; eller iii) fremstiller en forbindelse hvori R^, R2 eller R-^ er karboksy ved hydrogenolyse av den tilsvarende forbindelse hvori R-^, R2 eller R^ er alkoksykarbonyl.
Et alkalimetallsalt av en fenol (IV) der M er alkalimetall og fortrinnsvis natrium eller kalium, omsettes med et alkylendihalogenid, X-Alk-X, der X er brom eller jod. Reaksjonen skjer under moderat oppvarming, mellom ca. 50 og 100°C, i et inert oppløsningsmiddel ved bruk av ekvimolare mengder av reaktanter, eller fortrinnsvis et støkio-metrisk overskudd av dihalogenidet for å få minimal dannel-
se av di-eteren. Denne di-eteren som måtte dannes, kan lett skilles fra den ønskede mono-eter (II), fordi den førstnevnte er et relativt høytsmeltende faststoff som lett lar seg skille fra organiske oppløsningsmidler hvori mono-eteren er oppløselig.
Det er foretrukket å utføre den første foretringen med et dibromid (X-Alk-X, hvor X er Br), fordi man vanligvis lettere har bedre tilgang på dibromider enn dijodider. Det resulterende aryloksyalkylbromid n , hvor X er Br), kan reageres direkte med alkalimetallenolatsaltet (IIA) eller, hvis ønskelig, omdannes til det tilsvarende jodid
II , hvor X er I), som reagerer noe lettere med enolat-saltet enn bromidet. Omdannelsen av II (X = Br) til II
(X = I) utføres ved å oppvarme den førstnevnte forbindelse
med natrium-eller kaliumjodid i et inert oppløsningsmiddel, f.eks. aceton.
Det er også mulig å bruke klorbromalkaner som di- . halogenidreaktanten, nemlig Cl-Alk-Br. Bruken av slike bland-ede dihalogenider har den fordel at man totalt eliminerer eller nedsetter til et minimum di-eterdannelsen, ettersom reaksjonen nesten utelukkende skjer i nabostilling til brom-atomet, spesielt hvis man bruker støkiometriske mengder av fenolen og dihalogenidet.
Den resulterende kloralkoksyaryleter, II,x=Cl , må deretter omdannes til den tilsvarende bromalkoksyaryleter eller jodalkoksy-aryleter før den kan reageres med alkalimetallenolatsaltet av et diketon eller ketoester. Klorbromalkanutgangsforbind-elsene kan fremstilles ved reduksjon, f.eks. ved hjelp av litiumaluminiumhydrid av en klor-ester, Cl-Alk'-COOCH^, hvorved man får en kloralkanol, Cl-Alk-OH, hvoretter man erstatter
hydroksygruppen med brom, f.eks. ved hjelp av fosfortribromid.
I det avsluttende trinn blir så mono-eteren,
(II) behandlet med et alkalimetallenolatsalt av et di-
keton eller en keto-ester med formelen (IIA), hvor R og R' har samme betydning som angitt ovenfor, og M<+> er et alkalimetallkation, fortrinnsvis litium. Reaksjonen skjer i et inert oppløsningsmiddel under vannfrie betingelser ved romtemperatur eller noe høyere (25-70°C).
Alternativt kan alkalimetallenolatsaltet i det avsluttende trinn erstattes med et tungmetallchelat av diketon-
et eller keto-esteren. Passende tungmetallchelater innbefat-
ter kopper- nikkel-og koboltchelater .
En passende syntesevei for forbindelser ifølge foreliggende oppfinnelse er angitt ved hjelp av følgende reaksjonsskjerna:
I denne alternative syntesevei blir et alkalimetallenolatsalt eller et tungmetallchelat av et diketon eller en keto-ester ii reagert med et alkylendihalogenid, X-Alk-X. Reaksjonen skjer i et inert oppløsningsmiddel under vannfrie betingelser ved romtemperatur eller noe høyere (20-70°C), idet man bruker ekvimolare mengder av reaktantene eller et støkiometrisk overskudd av dihalogenidet. Det resulterende halogenalkyldiketon eller keto-ester med formel III, blir så reagert med et alkalimetallsalt av en fenol (HOAr) med formel IV , og denne reaksjon skjer ved moderat oppvarming, dvs. mellom 50 og 100°C, i et inert oppløsningsmiddel under vannfrie betingelser.
Hvis det er ønskelig å fremstille forbindelser med formel I hvor R-| , R 2 °9 R3 er fra 1-3 hydroksygrupper, er det nødvendig å utføre reaksjonen ved hjelp av de tilsvarende forbindelser hvor R-| / <K>2 og R3 er fra 2-3 benzyloksygrupper. Benzyloksygruppene eller benzyloksygruppen kan så spaltes ved hjelp av katalytisk hydrogenolyse.
Det er foretrukket å fremstille forbindelser med formel I hvor R1, R9 og R3 er karboksyl
(COOH) ved en hydrolyse fra de tilsvarende forbindelser med formel I hvor R-| , Rp og R3 er alkoksykarbonyl. ■
Videre er det foretrukket å fremstille forbindelser med formel I hvor R-| , R2 og R3 er dialkylaminoalkoksy ved en foretring av de tilsvarende forbindelser med formel I hvor R-), R2 og R3 er hydroksy,
noe som utføres ved at man reagerer et alkalimetallsalt av sistnevnte forbindelse med et dialkylaminoalkylhalogenid.
En biologisk undersøkelse av forbindelser ifølge foreliggende oppfinnelse har vist at de har aktivitet overfor virus. Man har funnet at de er effektive in vitro mot en rekke forskjellige typer virus, heri innbefattet rhino-2, heste-rhino, menneske-rhino, para-influensa, herpes og respi-ratorisk syncitial virus med en minimumsveksthemmende konsentrasjon på fra 0,5-50 ug pr. ml. Nevnte verdier ble bestemt ved hjelp av standard seriefortynningsprøver.
Strukturen på forbindelser ifølge foreliggende oppfinnelse ble fastslått ved hjelp av syntesemåtene, elemen-tæranalyse og ved infrarøde og kjernemagnetiske resonans-spektralbestemmeIser.
Preparater mot virus kan opparbeides ved å fremstille en fortynnet oppløsning eller suspensjon i et organisk eller vandig organisk medium. Denne oppløsning eller suspensjon kan påføres det sted som skal desinfiseres på vanlig måte. Forbindelsene kan også opparbeides som salver eller kremer eller som aerosolforstøvningspreparater eller som skumpreparater.

De etterfølgende eksempler skal illustrere oppfinnelsen nærmere.
Eksempel 1
a) 4-( 3, 4- metylendioksyfenoksy) butylbromid
3,4-metylendioksyfenol (sesamol) (13,8 g, 0,1 mol)
ble oppløst i 100 ml IN natriumhydroksyd, og oppløsningen ble deretter filtrert og konsentrert i vakuum for å fjerne vannet. Residuet ble opparbeidet med eter og tørket i vakuum ved 40°C i 5 timer. En blanding av 10 g (0,062 mol) av det resulter- . ende natriumsaltet av 3,4-metylendioksyfenol "og 21 g (0,1 mol) av 1,4-dibrombutan i 75 ml dimetylformamid ble rørt ved 60°C
i ca. 16 timer under nitrogen. Reaksjonsblandingen ble konsentrert i vakuum for å fjerne oppløsningsmidlet, og residuet ekstrahert med 150 ml metylendiklorid. Det faste natri-umbromid ble fjernet ved filtrering, og filtratet ble vasket
med en fortynnet natriumbikarbonatoppløsning og så med vann, tørket over vannfritt magnesiumsulfat og konsentrert for å fjerne oppløsningsmidlet. Den gjenværende olje ble utkrystallisert fra 50 ml 95% etanol, hvorved man fikk 9,5 g 4-(3,4-metylendioksyfenoksy)butylbromid, sm.p. 52-54°C.
b) 4- [ 4-( 3, 4- metylendioksyfenoksy) butyl]- 3, 5- heptandion
(I; R1 = R2 = 3,4-metylendioksy, R3 = H, Alk = CH2CH2CH2CH2,
R = R' = CH3CH2 ) .
En blanding av 21 g (0,15 mol) av litiumsaltet av 3,5-heptandion (fremstilt fra 22,8 g 3,5-heptandion i 300
ml eter og 108 ml 1,6N butyllitium i heksan dråpevis tilsatt i løpet av 30 minutter ved -15°C) og 26,1 g 4-(3,4-metylen-dioksyf enoksy) butylbromid i 200 ml dimetylformamid ble rørt og oppvarmet ved 52-53°C i ett døgn under nitrogen. Reaksjonsblandingen ble konsentrert i vakuum for å fjerne oppløs-ningsmidlet, og residuet ble delt mellom eter og vann. Eterlaget ble. vasket suksessivt med 5% vandig svovelsyre, vann,
5% vandig natriumbikarbonat og vann, og deretter konsentrert for å fjerne oppløsningsmidlet. Residuet som veide 25 g ble kromatografert på en kolonne av 400 g silisiumdioksydgel idet man påsatte det hele i en pentan:benzen 90:10 oppløsning. Kolonnen ble eluert med pentan-benzen-kloroformoppløsnings-middelserier, og de forbindelser (14,0 g) som kom ut med fraksjonen benzen:kloroform 8:2, ble kromatografert på nytt på 240 g silisiumdioksydgel. Sistnevnte gel ble eluert med samme oppløsningsmiddelserier, og pentan:benzen 80:20 til 50:50 bragte ut 8,0 g av det forønskede produkt, 4-[4-(3,4-metylendioksyfenoksy)butyl]-3,5-heptandion.

4-[4-(3,4-metylendioksyfenoksy)butyl]-3,5-heptandion ble funnet å ha antivirusaktivitet in vitro overfor heste-rhinovirus ved en minimumseffektiv konsentrasjon på 12 ug pr. ml.
Eksempel 2
a) 5-( 3, 4- metylendioksyfenoksy) pentylbromid
En blanding av 27,6 g (0,2 mol) sesamol, 56 g
(0,4 mol) kaliumkarbonat, 92 g (0,4 mol) av 1,5-dibrompentan i 400 ml aceton ble kokt under tilbakeløp og under røring og nitrogen i 3 døgn. En del av oppløsningsmidlet (200 ml)
ble avdestillert, hvoretter 400 ml benzen og 200 ml vann ble tilsatt. Det vandige lag ble utskilt og ekstrahert med benzen. De samlede organiske lag ble vasket med en 5% vandig natriumbikarbonatoppløsning og så med vann, tørket over vannfritt magnesiumsulfat og konsentrert i vakuum. Residuet som var en rødaktig olje,, ble opparbeidet med 500 ml absolutt eter, og blandingen ble filtrert for å fjerne 5,0 g fast bis-eter, 1,5-bis(3,4-metylendioksyfenoksy)pentan, sm.p. 129-130°C. Filtratet ble fordampet og residuet destillert, kokepunkt 130-135°C (0,03-0,005 mm), hvorved man fikk 30 g 5-(3,4-metylendioksyfenoksy)pentylbromid, sm.p. 54-57°C. Eksempel 3
a) 7-( 3, 4- metylendioksyfenoksy) heptylbromid ble fremstilt fra 27,6 g sesamol og 100 g 1,7-dibromheptan i nærvær av
53,8 g kaliumkarbonat i aceton ved hjelp av den fremgangsmåte som er beskrevet i eksempel 2a. Man fikk ialt 43,5 g 7-(3,4-metylendioksyfenoksy)heptylbromid, sm.p. 45-47°C.
b) 7-( 3, 4- metylendioksyfenoksy) heptyljodid
En blanding av 43,5 g 7-(3,4-metylendioksyfenoksy)-heptylbromid, 20,7 g natriumjodid og 300 ml aceton ble oppvarmet ved koking under tilbakeløp i 2 timer. Reaksjonsblandingen ble filtrert, filtratet fordampet i vakuum, hvoretter residuet ble delt mellom vann og metylendiklorid. Metylendikloridlaget ble utskilt, vasket med vann, tørket
over vannfritt magnesiumsulfat og så konsentrert. Residuet som besto av 7-(3,4-metylendioksyfenoksy)heptyljodib, ble brukt uten ytterligere rensing i den følgende reaksjon.
c) 4-[ 7-( 3, 4- metylendioksyfenoksy) heptyl]- 3, 5- heptandion
(I; R1 = R2 = 3,4-metylendioksy, R., = H, Alk = (CH2>7,
R = R' = CH3CH2CO) ble fremstilt fra 38,5 g 7-(3,4-metylen-dioksyf enoksy) heptyl jodid og 22 g av litiumsaltet av 3,5-heptandion slik det er beskrevet i eksempel lb. Produktet ble kromatografert på 1000 g aktivert magnesiumsilikat og eluert med pentan-benzen-kloroform-oppløsningsmiddelserier. Kloroformelueringsmidlene frembragte 23,5 g 4-[7-(3,4-metylen-dioksyf enoksy) heptyl ] -3 , 5-heptandion som et fargeløst faststoff .

4-[7-(3,4-metylendioksyfenoksy)heptyl]-3,5-heptandion viste seg å ha aktivitet overfor virus såsom heste-rhinovirus ved en minimumhemmende konsentrasjon på 6 yg pr-ml.
Ved hjelp av foregående fremgangsmåter idet man gikk ut fra passende substituerte fenoler og alkylendibromider som i eksempel 2a, ble følgende forbindelser fremstilt fra de respektive mellomliggende aryloksybromider i henhold til fremgangsmåten i eksempel lb.
Eksempel 4

En oppløsning av 4,5 g 4-[6-(p-benzyloksyfenoksy)-heksyl]-3,5-heptandion (eksempel 8) i 100 ml absolutt etanol ble hydrogenert i nærvær av 0,4 g av en palladium på karbon-katalysator ved et trykk på 3,2 kg/cm 2. Hydrogeneringen var_ fullstendig i løpet av 45 minutter. Reaksjonen ble gjentatt med en ytterligere mengde på 4,5 g benzyleter, og blandingene ble slått sammen, filtrert og fordampet. Residuet ble utkrystallisert fra 30 ml pentan og 60 ml eter, og man fikk 5,0 g 4-[6-(p-hydroksyfenoksy)heksyl]-3,5-heptandion, sm.p. 65-66°C.

(I; Rx = 4-HO, R2 = R3 = H, Alk = (CH^, R = R' = CH3CH2 ), sm.p. 46-48°C, ble fremstilt ved hydrogenolyse av 4-[7-(p-benzyloksyfenoksy)heptyl]-3,5-heptandion (eksempel 11).

4-[ 7- ( p- karboksyfenoksy) heptyl]- 3, 5- heptandion
(I; R1 = 4-HOOC, R2 = R3 = H, Alk = (CH^, R = R' = CH3CH2 ).
En oppløsning av 10,7 g 4-[7-(p-etoksykarbonylfenoksy)heptyl]-3,5-heptandion (eksempel 12) og 3 ml konsentrert saltsyre i 150 ml dioksan ble oppvarmet ved koking under tilbakeløp i 48 timer. Oppløsningen ble behandlet med aktivert trekull, filtrert og fordampet til tørrhet. Residuet ble oppløst i eter og ekstrahert med 10% kaliumkarbonat-oppløsning. De basiske ekstrakter ble gjort sure, og denne blanding ble så ekstrahert med eter. Eteroppløsningen ble tørket og fordampet, og residuet utkrystallisert fra eter, hvorved man fikk 5,5 g 4-[7-(p-karboksyfenoksy)heptyl]-3,5-heptandion, sm.p. 105-107°C.

4-{ 6-[ p-( 2- dietylaminoetoksy) fenoksy] heksyl}- 3, 5- heptandion (I; Rx = 4-(C2H5)2NCH2CH20, R2 = R3 = H' Alk = ^ CE2) 6' R =
R' = CH3CH2 ).
En blanding av 11 g (0,034 mol) 4-[6-(p-hydroksyfenoksy)heksyl]-3,5-heptandion (eksempel 48), 4,97 g (0,034 mol) av 2-dietylaminoetylklorid og 9,66 g (0,07 mol) av kaliumkarbonat i 200 ml dimetylformamid ble rørt ved 25-30°C i 3 døgn. Reaksjonsblandingen ble konsentrert for å fjerne oppløsningsmidlet, og residuet ble delt mellom vann og eter. Eterlaget ble vasket med vann, tørket og konsentrert. Residuet ble kromatografert på 280 g aktivert magnesiumsilikat og eluert med pentan-benzen-metanolserier. Benzen som inneholdt 1% metanol, inneholdt 5,5 g 4-{6-[p-(2-dietylaminoetoksy)fenoksy]heksyl1-3,5-heptandion. Sistnevnte forbindelse ble oppløst i 30 ml absolutt eter og denne ble dråpevis tilsatt 3,3% eteroppløsning av saltsyre under røring inntil pH ble sur. Det faste stoff ble oppsamlet ved filtrering og tørket i vakuum ved romtemperatur. Materialet ble omkrystallisert ved at det ble oppløst i isopropylalkohol og tilsatt eter og pentan inntil oppløsningen ble turbid. Man fikk på denne måten ialt 2,0 g 4-{6-[p-(2-dietylaminoetoksy)fenoksy]-heksyl}-3,5-heptandion i form av sitt hydrokloridsalt, sm.p. 90-92°C.

Eksempel 52
Ved valg av egnede forbindelser ble det også fremstilt 4-{6-[p-(p-metoksybenzoyloksy)fenoksy]heksyl}-3,5-heptandion (I; R1 = 4-(4-CH3OC6H4COO), R2 = R3 = H, Alk =
(CH2)6, R = R' = CH3CH2 ), sm.p. 68-70°C.
E ksempel 53
Litiumsaltet av 3,5-heptandion (0,1 mol) og 0,15
mol 1,6-dibromheksan i 200 ml dimetylformamid ble holdt på 50°C i ett døgn under nitrogen. Reaksjonsblandingen ble oppvarmet som beskrevet i eksempel 1 del (a). Det resulterende produkt 4-(6-bromheksyl)-3,5-heptåndion ble blandet med 0,1 mol av natriumsaltet av 2-klor-4-metoksyfenol i 100 ml dimetyl-
formamid, hvoretter blandingen ble rørt ved 60°C i ca. 16 timer under nitrogen. Reaksjonsblandingen ble opparbeidet som beskrevet i eksempel 1 del (b) hvorved man fikk 4-[6-(2-klor-4-metoksyfenoksy)heksyl]-3,5-heptandion identisk med den forbindelse som ble fremstilt som beskrevet i eksempel 9.
Claims (2)
1. Analogifremgangsmåte for fremstilling av terapeutisk aktive aryloksyalkyl-diketoner og -ketoestere med formel:
der: Alk er rettkjedet alkylen med 4-7 karbonatomer; R er alkyl med 1-5 karbonatomer; R' er alkyl med 1-5 karbonatomer eller alkoksy
med 1-5 karbonatomer; Rl' R2 0<^ R3 uavhengig av hverandre er hydrogen, alkyl med 1-4 karbonatomer, alkoksy med 1-4 karbonatomer, halogen, trifluormetyl, hydroksy, benzyloksy, lavere alkylbenzoyloksy, karboksy, alkoksykarbonyl med 3-4 karbonatomer, metoksybenzoyloksy, dialkylamino der alkyl har fra 1-4 karbonatomer, dialkylaminoalkoksy der alkyl har fra 1-4 karbonatomer og alkoksy har fra 2-4 karbonatomer, eller to av Rl' R2°^R3 sammen danner 3,4-metylendioksy, karakterisert ved at man i et inert oppløs-ningsmiddel under vannfrie betingelser kondenserer enten a) en halogenalkylfenyleter med formelen:
med et alkalimetallsalt eller tungmetallchelat av et diketon med formelen;
eller b) en forbindelse med formelen:
med en forbindelse med formelen:
hvori Alk, R og R<1> har den ovenfor angitte betydning og R-^1 , R2' og R^' uavhengig av hverandre er hydrogen, alkyl med fra 1-4 karbonatomer, alkoksy med fra 2-4 karbonatomer, halogen, trifluormetyl, benzyloksy eller dialkylamino eller to av R^<1>, R,,1 og R3<1> sammen danner 3,4-metylendioksy, og X er brom'eller jod; og M er et alkalimetall; og,hvis ønskelig, i) underkaster en oppnådd forbindelse der R^', R2 1 og R^' er 1-3 benzyloksygrupper, en katalytisk hydrogenolyse for å fremstille den tilsvarende forbindelser der R^, R2 og R^ er 1-3 hydroksygrupper; ii) underkaster en oppnådd forbindelse der R^, R2 eller R^
er hydroksy en forestring for å fremstille den tilsvarende forbindelse der R^ , R ? og R^ er benzoyloksy, alkylbenzoyloksy, metoksybenzoyloksy ved behandling av hydroksyforbind-elsen med det egnede benzoylhalogenid eller substituerte benzoylhalogenid; iii) fremstiller en forbindelse hvori R^, R^ eller R^ er karboksy ved hydrogenolyse av den tilsvarende forbindelse hvori R^, R2 eller R^ er alkoksykarbonyl; eller iv) underkaster en oppnådd forbindelse der minst en av R^, R^ og R^ er hydroksy, en foretring for å oppnå en tilsvarende forbindelse der nevnte minst ene av R^, R^ og R^ er dialkylaminoalkoksy.
2. Fremgangsmåte ifølge krav 1 for fremstilling av 4-[6-(2-klor-4-metoksyfenoksy)heksyl]-3,5-heptandion, karakterisert ved at 6-(2-klor-4-metoksyfenoksy)-heksylbromid omsettes med litiumsaltet av 3,5-heptandion.
