Abstract
Fremgangsmåte for fremstilling av det B-laktamase-inhiberende antibiotikum MM 17880 og alkalimetallsalter derav.
Description
Oppfinnelsen vedrører en fremgangsmåte for fremstill-
ing av (3-laktamase-inhiberende antibiotikum MM 17880 samt alkalimetallsalter derav, og fremgangsmåten er karakterisert ved at en mikroorganisme av slekten Streptomyces dyrkes, hvoretter forbindelsen MM 17880 eller et salt derav, isoleres ved kjente me-, toder.
Fremgangsmåten omfatter kromatografisk separering av
en løsning som inneholder MM 17880 eller et salt derav sammen med ytterligere antibakterielle materialer til fraksjoner som består i alt vesentlig av en løsning av MM 17880 eller et salt derav, og utvinning av det i alt vesentlig rene MM 17880 eller et salt derav fra løsningen.
En slik fremgangsmåte innebærer normalt søylekromato-grafi under anvendelse av en inert bærer, f.eks. cellulose, og et alkanolisk løsningsmiddelsystem som her beskrevet.
Britisk patent nr. 1 363 075 åpenbarer at stammer av Streptomyces olivaceus er i stand til å produsere en nyttig P-laktamase-inhiberende substans. Norske patentsøknader nr.
751074 og 751075 viser at materialet i henhold til britisk pa-
tent nr. 1 363 075 var svært urent og inneholdt bare små mengder av antibakterielt aktive midler. • Disse antibakterielt aktive midler ble karakterisert i de nevnte norske patentsøknader og ble betegnet MM 4550 og MM 13902. Søkeren har nå funnet at i tillegg til MM 4 550 og MM 13902 produserer visse stammer av Streptomyces olivaceus og beslektede organismer en ytterligere antibakterielt aktiv 3-laktamase-inhiberende forbindelse. Dette nye materiale betegnes her som MM 17880.
Ved fremgangsmåten ifølge foreliggende oppfinnelse tilveiebringes MM 17880 og salter derav, idet denne substans MM 17880 er et di-syré-faststoff som i form av et i alt vesentlig rent dinatriumsalt har følgende egenskaper: (a) Det har et karakteristisk infrarødt spektrum som, når det undersøkes i en KBr-tablett er i alt vesentlig som vist i figur 1. (b) Det har et karakteristisk kjernemagnetisk resonans-spektrum som, undersøkt i deuteriumoksyd, er i alt vesentlig som vist i figur 2.
(c) Det har et karakteristisk ultrafiolett spektrum som i vann
har et absorpsjonsmaksimum ved ca. 297 nm i alt vesentlig
som vist i figur 3.
MM 17880 har følgende strukturformel:
Når MM 17880 er i alt vesentlig ren, har MM 17880 som et di-na triumsalt de R^-verdier i tynnsjiktkromatografisystemene som er angitt i tabell 1 nedenunder:
Det fremgår av formelen at MM 17880 inneholder svovel og nitrogen i tillegg til karbon, oksygen og hydrogen,
Når den er i alt vesentlig ren, har MM 17880 som et dinatriumsalt et godt nivå av antibakteriell aktivitet ved bestemmelse med mikrotitermetoden mot visse Gram-positive og Gram-negative organismer f.eks. mot stammer av Bacillus subtilis, Enterobacter cloacae, Escherichia coli, Klebsiella aerogenes, Proteus mirabilis, Salmonella typhimurium, Serratia marcescens og Staphylococcus aureus.
Når den er i alt vesentlig ren, er MM 17880 som et dinatriumsalt i stand til å synergisere den antibakterielle aktivitet hos 13-laktam-antibiotika så som ampicillin, og amoxycillin og lignende mot visse bakterier inklusive visse 13-
laktamaseproduserende stammer av Staphylococcus aureus og Klebsiella aerogenes.
Betegnelsen "i alt vesentlig ren" skal her bety minst 75% ren og fortrinnsvis minst 85% ren, f.eks. 90-100% ren.
Generelt er saltene av MM 17880 meget mer stabile enn den ikke-saltdannede syre som har tendens til å bli spaltet i løsning.
De dibasiske salter av MM 17880 som tilveiebringes ved foreliggende oppfinnelse, er fortrinnsvis farmasøytisk akseptable salter, f.eks. av natrium, kalium, kalsium, magnesium, aluminium eller de av konvensjonelle ammonium- eller substitu-erte ammoniumsalter.
Ved fremgangsmåten i henhold til oppfinnelsen fremstilles fortrinnsvis farmasøytisk akseptable alkalimetallsalter av MM 17880, f.eks. et dinatrium- eller dikaliumsalt av MM 17880.
Nyttige farmasøytiske preparater inneholder MM 17880
eller et farmasøytisk akseptabelt salt derav.
Slike preparater er egnet for bekjempelse av bakteri-ell infeksjon eller for profylakse hos pattedyr inklusive mennes-ket. Eksempelvis kan preparatene anvendes ved behandling av sykdommer i åndedrettsorganene, urinveiene, de bløte vev, huden og lignende. For anvendelse ved slike behandlinger kan preparatene tilveiebringes i konvensjonell form for oral, topisk, intra-mammal, injiserbar eller infuserbar administrering.
Et foretrukket farmasøytisk preparat som inneholder MM 17880 eller et farmasøytisk akseptabelt salt derav, er tilpas-set for oral eller parenteral administrering til mennesker.
Slike foretrukne preparater vil generelt inneholde fra 10 mg til 5 g av MM 17880 eller ét salt derav og vil mer vanligvis inneholde 50 mg til 1,25 g av et salt av MM 17880, f.eks. 150 mg til 1,0 g.
Preparatene kan inneholde MM 17880 eller et salt derav som eneste terapeutiske middel eller de kan inneholde MM 17880 eller et salt derav sammen med andre terapeutiske midler, f.eks. et penicillin eller cefalosporin, f.eks. ampicillin, amoxycillin, karbenicillin, benzylpenicillin, in vivo hydrolyser-bare estere av hvilke som helst av de foregående penicilliner,
fenoksymetylpenicillin, cefaloridin, cefalotin, cefaloglycin, cefamandol,
cefazolin, aceton- eller formaldehydaddukter av hvilket som helst av de forannevnte penicilliner eller cefalosporiner som inneholder en a-aminogruppe i acylaminosidekjeden, cloxacillin, dicloxacillin, flucloxacillin eller andre konvensjonelle p-lak-tam-antibiotika.
Preparatene kan inneholde bærere på konvensjonell
måte, slik det vil forstås for fagmannen på området hva angår fremstilling av antibakterielle preparater.
Dyrkning av Streptomyces olivaceus kan utføres som beskrevet i britisk patentskrift nr. 1 363 075 eller norske pa-tentsøknader nr. 751074 og 751075, og vil generelt innebære aerob fermentering i nærvær av assimilerbare kilder av karbon, nitrogen, svovel og mineralsalter. De nødvendige næringsstoffer kan tilveiebringes av komplekse organiske kilder eller ved kjemisk defi-nerte media på konvensjonell måte.
Betegnelsen "Streptomyces olivaceus" skal her menes å være definert i henhold til klassifiseringen av R. Hutter (i Systematic der Streptomyceten, S. Korger, Basle, s.8-32). Det bemerkes at ved denne definisjon kan Streptomyces fulvovoridis, Streptomyces flavus og Streptomyces flavovirens betraktes som synonyme med Streptomyces olivaceus.
Sporemorfologien og karbon-forbrukskarakteristikaene
til "Streptomyces olivaceus" er beskrevet i norske patentsøknader nr. 751074 og 751075.
Den foretrukne organisme for anvendelse i denne fremgangsmåte er Streptomyces olivaceus ATCC 31126.
MM 17880 oppnås normalt hovedsakelig fra dyrkningsfiltratet slik at det foretrukne innledningstrinn i isolasjons-prosessen er fjerning av fast materiale fra fermenteringen, for eksempel ved filtrering.
Generelt bør alle isolerings- og rensningsprosesser finne sted ved ikke-forhøyede temperaturer, f.eks. under 20°C
og mer passende ikke over 12°C.
Det urene MM 17880 eller salt kan oppnås fra det klarnede dyrkningsfiltrat ved absorbering av MM 17880 eller saltet derav inn i et materiale så som aktivt kull, og deretter eluering med vandig aceton og fordampning av løsningsmidlene under redusert trykk. Råproduktet fra denne prosess kan renses videre ved oppløsning i vann og ekstrahering inn i en organisk fase under anvendelse av et lipofilt kvaternært ammoniumsalt for dannelse av et organisk løsningsmiddelløselig salt, fulgt av tilbakeekstraksjon i vann eller fortynnet natriumjodid.
Alternativt kan dyrkningsfiltratet ekstraheres under anvendelse av et' lipofilt kvaternært ammoniumsalt og et organisk løsningsmiddel fulgt av tilbakeekstraksjon i vandig natriumjodid-løsning. Dette er ofte mer effektivt enn den fremgangsmåte som anvender intermediær konsentrering av filtratet.
Det menes at den kromatografiske rensning av MM 17880 best utføres under anvendelse av et salt av MM 17 880, f.eks. nat-riumsaltet. Salter av MM 17880 er normalt mer løselige i vandige og vandig/alkohol-løsningsmiddelsystemer enn i sterkt lipofile løsningsmidler, og følgelig foretrekkes det å anvende vandige og vandig/alkohol-løsningsmiddelsystemer i de kromatografiske rens-ninger som anvendes ved fremgangsmåten i henhold til oppfinnelsen.
Ved forsøk har det vist seg at vandige løsninger av elektrolytter som er pufret til tilnærmet nøytralitet, har vist seg egnet for anvendelse i tilknytning til polare bærermaterialer så som basiske ionebytteharpikser for rensning av MM 17880. Således kan en vandig løsning av natriumklorid som er pufret til ca. pH 7 med en konvensjonell puffer, f.eks. fosfatpuffer, anvendes i tilknytning til bærermaterialer som inneholder kvaternære ammoniumgrupper. Man har funnet at basiske ionebyttecelluloser og basiske ionebytte-tverrbundne dekstraner er egnede bærermaterialer og at QAE "Sephadex" A25 spesielt er et meget godt bærer-materiale.
Separering av MM 17880 fra uorganiske salter spesielt, men også fra andre kontaminerende substanser, kan utføres ved absorbering av antibiotikumet på en lipofil harpiks som uorganiske salter ikke adsorberes til. I forbindelse med oppfinnelsen har det vist seg at en polystyren/divinylbenzen-kopolymer, f.eks. "Amberlite" XAD-4 er spesielt godt egnet, idet det ønske-de antibiotikum kan fjernes fra søylen ved eluering (eluering med vann eller vandig alkanol) og den resulterende løsning konsentre-res ved inndampning og frysetørkes slik at man får et materiale med forbedret renhet. Separering av MM 17880 fra uorganiske salter kan også gjerne utføres ved kromatografi på en søyle som består av et gelfiltreringsmiddel, f.eks. tverrbundne dekstran-geler f.eks. "Sephadex" G15 og polyakrylamidgeler, f.eks. "Biogel" P2.
Ytterligere rensning av MM 17880 fra materialer som fremstilles under anvendelse av en eller flere av de fremgangs-måter som er beskrevet ovenfor, kan utføres ved søylekromatogra-
fi på en inert, fast fase, f.eks. silikagel eller' cellulose,
under anvendelse av vandig alkoholiske løsningsmiddelsystemer. Egnede løsningsmiddelsystemer vil inneholde vann og minst én. lavere alkanol, f.eks. vann/isopropanol, vann/n-propanol, vann/- metanol/isopropanol, vann/butanol, vann/etanol/butanol eller lignende systemer. Hos søkeren har det vist seg at en 4/1-blanding av n-propanol og vann-har vært et spesielt.godt egnet løsningsmiddelsystem for anvendelse i tilknytning til en cellu-losebærer.
De fraksjoner som skal oppsamles og tilbakeholdes,
er de som viser et UV-spektrum som er karakteristisk for MM 17880 (se f.eks. figur 3) som har et UV-absorpsjonsmaksimum ved ca.
297 nm. Frysetørkingen av den resulterende løsning gir ét i alt vesentlig rent salt av MM 17880.
Følgende eksempler illustrerer oppfinnelsen.
Eksempel 1
Fremstilling av i alt vesentlig rent MM 17880-natriumsalt.
a. Fermentering
Streptomyces olivaceus ATCC 31126 ble dyrket i 7 dager ved 28°C på en fast agar-skråkultur i en Roux-kolbe,.. Agarsubstratet hadde følgende sammensetning:
(Gjærekstrakten var "Yeatex" fra Bovril Food Ingredi-ents, P.O. Box 18, Wellington Road, Burton-on-Trent, og agaren ble levert av Oxoid Limited, Southwark Bridge Road, London, S.E.I.).
Substratet ble justert til pH 6,8 før sterilisering.
50 ml sterilt, avionisert vann som inneholdt 0,02% "Tween" 30, ble tilsatt til en Roux-kolbekultur, og sporene ble suspendert ved risting. Denne sporesuspensjon ble så tilsatt som podestoff til 75 1 av sterilisert kimtrinnssubstrat i en 100 1 fermentator av rustfritt stål. Sammensetningen av kimtrinnsubstratet var som følger:
(Soyabønnemelet var "Arkasoy" 50 fra British Arkady Co. Ltd., Old Trafford, Manchester).
For regulering av skummingen ble 50 ml 10 volum% "Pluronic" L81 i soyabønneolje tilsatt til fermenteringssubstratet før sterilisering. ("Pluronic" L81 ble levert av Jacobsen van den Berg U.K. Ltd., 231 The Vale, London, W.3.). Substratet ble dampsterilisert i fermentatoren i 20 minutter ved 120 oC. Kim-trinnkulturen ble omrørt ved 140 opm med en skiveagitator med vinger med diameter 19,05 cm og forsynt med 75 l/min. steril luft gjennom en spreder som var åpen i enden. Dyrkningskaret var utstyrt med ledeplater. Temperaturen ble regulert til 28 oC, og etter inkubering under disse betingelser i 75 timer bile 7,5 1 av denne kimkultur tilsatt som podestoff til 150 1 sterilt fer-menteringssubstrat i en 300 1 fermentator av rustfritt stål. Fermenteringssubstratet hadde følgende sammensetning:
300 ml 10% "Pluronic" L81 i soyabønneolje ble tilsatt for å forhindre skumming. Fermenteringen ble innhøstet etter 48 timer og klarnet ved sentrifugering. Det klarnede brygg ble vilkårlig gitt en aktivitet på 340 enheter/ml ved undersøkelse ved hjelp av den konvensjonelle hull-i-plate-metode på agar som var kimsatt med Klebsiella aerogenes.
b. Isolering av i alt vesentlig rent MM 17880 som natriumsalt
Klarnet brygg som var fremstilt i alt vesentlig
som angitt under (a) (1050 1; 340 enheter/ml) ved 10°C og pH
6,8, ble ekstrahert med 310 1 diklormetan ved 10°c, inneholdende 1200 g cetyldimetylbenzylammoniumklorid ved pumping av de to væsker i for et fastsatte strømningshastigheter gjennom en "in-line"-mikser. Fatene ble separert i en kontinuerlig Sharples-sentrifuge etter at de var blandet i ca. 2 minutter. Diklormetan-fasen (300 1) ble tilbakeekstrahert med vandig natriumjodid. Tilbakeekstraksjonen ble utført i 4 satser under anvendelse av ialt 7 1 vann som inneholdt 210 g natriumjodid. Fasene ble separert ved hjelp av gravitasjon. Vannfasen ble justert fra pH 7,7 til pH 7,0 med saltsyre og filtrert. Natriumjodidekstrakten (7 1) inneholdt 21.900 enheter/ml.
En ionebyttekolonne ble fremstilt ved pakking av
QAE "Sephadex" A25 (fra Pharmacia Ltd.) i fosfatpuffer pH 7
(0,05 m) som inneholdt 0,3 m natriumklorid, i en glasskolonne med 10 cm diameter, til en høyde av 40 cm. Natriumjodidekstrakten (7 1) ved 5°C ble perkolert gjennom QAE "Sephadex" med 50 ml/min. Kolonnen ble eluert med 0,7 m Nacl i 0,05 m fosfatpuffer med
pH 7 også ved 5°C ved en strømningshastighet på 25 ml/min. 2 1 eluat ble forkastet, og 90 fraksjoner (100 ml) ble oppsamlet. Fraksjonene ble kjørt i et UV-spektrofotometer, og de som viste et absorbsjonsmaksimum ved ca. 285 nm, som ble funnet å inneholde en blanding av MM 17880 og en urenhet som var ansvarlig for
absorpsjonen ved 285 nm, ble samlet og justert til pH 7 (frak-sjonstall 25-35, samlet volum 1230 ml ved 8450 enheter/ml) .

samlede fraksjon som deretter ble perkolert ved 5 C gjennom en 6,3 cm diameters kolonne pakket med "Amberlite" XAD 4 harpiks (fra Rohm & Haas Ltd.) til en høyde av 30 cm ved en strømnings-hastighet på 20 ml/min. Antibiotikumet ble adsorbert til har-piksen under disse betingelser, mens de uorganiske urenheter ikke ble det. Antibiotikumet ble eluert ved romtemperatur ved 200 ml destillert vann fulgt av 50% vandig metanol. Eluatet (1 1) ble inndampet under 30°C under redusert trykk til 70 ml, justert til pH 7 og frysetørket til et brunt, fast stoff (2,18 g) ved 5.000 enheter/mg. En annen sats av materialet som var fremstilt ved ovennevnte fremgangsmåte, var noe renere, med en aktivitet på. 5.700 enheter/mg. Dette faste stoff (0,55 g) ble kromatografert på en cellulosekolonne (4 x 29 cm) (whatman CC 31) likevektsatt med n-propanol/vann (4/1). Kolonnen ble eluert med n-propanol/- vann (-4/1) , de første 135 ml av eluatet ble forkastet, og deretter ble 15 ml fraksjoner oppsamlet. De fraksjoner som hadde et UV-absorpsjonsspektrum som var karakteristisk for MM 17880, ble kombinert (45 ml), inndampet under redusert trykk for fjerning av n-propanol og deretter frysetørket slik at man fikk 33 mg av et gult, fast stoff. Det faste stoff ble undersøkt og hadde en aktivitet på 16.000 enheter/mg.
Eksempel 2
Fermenterings- og isoleringsmetoden ble fulgt i alt vesentlig som beskrevet i eksempel 1, opp til og inklusive elu-eringen fra "Amberlite" XAD-4. Eluatet fra "Amberlite" XAD-4-kolonnen ble konsentrert under redusert trykk til tilnærmet 20 ml volum. Denne løsning ble kjørt på en QAE "Sephadex" A25-kolonne (3,8-x 30 cm) fremstilt i 0,18 m NaCl. Kolonnen ble eluert inn-ledningsvis med en eksponensiell gradient av natriumklorid fra 0,18 til 0,28 m over et totalt volum på 2 liter fulgt av eluering ved en konstant natriumkloridsammensetning på 0,28 m. Kolonnen ble eluert ved 4°c ved en hastighet av 3 ml/min., og 25 ml fraksjoner ble oppsamlet. Fraksjoner med et UV-absorpsjonsspektrum karakteristisk for MM 17880 (90-105) ble oppsamlet (400 ml).
De kombinerte fraksjoner ble inndampet under redusert trykk til tilnærmet 10 ml og belagt på en 3,8 x 28 cm kolonne av "Biogel" P2 (200-400 mesh) (Bio Rad Laboratories) ekvilibrert i 1% butanol. Kolonnen ble eluert med 1% butanol ved en hastighet av 2 ml/min., og 5 ml fraksjoner ble oppsamlet. Reaksjoner med et UV-absorpsjonsspektrum som var karakteristisk for MM 17880 og som ga en negativ sølvnitratreaksjon for klorid, ble kombinert. De kombinerte fraksjoner ble inndampet i vakuum for fjerning av butanol og frysetørket slik at man fikk 73 mg av et amorft, fast stoff.
Det faste stoff (70 mg) ble oppløst i et minimum av
•n-propanol/vann (4/1) og kjørt på en cellulosekolonne (1,5 x 15 cm)
(Whatman CC 31) ekvilibrert i den samme løsningsmiddelblanding. Kolonnen ble eluert med n-propanol/vann (4/1) ved 1 ml/min., og 6 ml fraksjoner ble oppsamlet. Fraksjonene ble undersøkt med hensyn på sine u.V.-absorpsjonsspektra, og de som hadde et spektrum som var karakteristisk for MM 17880 (13-16) , ble kombinert (24 ml), inndampet under redusert trykk for fjerning av propanol og frysetørket slik at man fikk et lysebrunt, amorft faststoff (17 mg).

Det materiale som ble fremstilt i alt vesentlig som beskrevet i eksempel 2, hadde de I.R.-, N.M.R.- og U.V.-spektra som er vist i henholdsvis figurer 1, 2 og 3.
Den antibakterielle aktivitet for det materiale som
er fremstilt som angitt ovenfor, ble bestemt ved mikrotitermetoden under anvendelse av podestoffet som hadde henstått natten over fra testorganismen i 0,2% konsentrasjon. Resultatene er vist i tabell 2. Den synergistiske aktivitet for dinatriumsaltet av MM 17880 i kombinasjon med ampicillin og med cefaloridin mot Klebsiella aerogenes og Staphylococcus aureus Russell er vist i tabell 3.
Dinatriumsaltet av MM 17880 som bar produsert på denne måte, hadde en I5Q-verdi på • 250 x 10 —12 g/ml mot I5-laktamasen fra E.coli JT4 ved bestemmelse i henhold til den metode som er beskrevet i belgisk patentskrift nr. 827.331.
Elektroforese antyder at det materiale som er produsert, er et dinatriumsalt slik at MM 17880 er en disyre.
En elementæranalyse som ble utført på en prøve av dinatriumsaltet av MM 17880 produsert som angitt ovenfor, ga følgende resultater: C:31,9%; H:4,2%; N:5,4%; S:13,2%; Na:ll,6%
Man skal huske på at det materiale som ble utsatt for analyse, var fremstilt ved frysetørking og således kan inneholde vann eller andre forurensninger. Dinatriumsaltet av MM 17880 inneholder (i tillegg til karbon, hydrogen og oksygen) elementene svovel, nitrogen og natrium i forholdet 2:2:2.

- 2. Fremgangsmåte som angitt i krav 1, karakterisert ved at det som mikroorganisme anvendes Streptomyces olivaceus ATCC 31126.
