Description
Oppfinnelsen angår en fremgangsmåte for fremstilling
av etanol med høyt utbytte ut fra celluloseholdig materiale.
Hittil er etanol på dette grunnlag fremstilt ved å
la en cellulase innvirke på cellulose for enzymatisk forsukring av cellulosen til glukose med følgende omsetning av glukosen med en alkohol-produserende mikroorganisme for fremstilling av etanol. Omsetningsgraden for omsetning av cellulose til glukose i henhold til denne kjente fremgangs-
måte er lav og følgelig får man store mengder uomsatte cellu-loserester. Hvis derfor produktet fra en slik cellulase-behandling av celluloseholdige stoffer benyttes som utgangsstoff for alkoholgjæring, er det nødvendig å skille glukosen fra produktet f.eks. ved filtrering. I tillegg til den ulempe at glukosekonsentrasjonen i den forsukrede væsken er lav på grunn av lav omsetningsgrad for cellulose til glukose,
er det en mulighet for at man mister glukose ved denne separasjonsprosess. Det oppnås følgelig lavt utbytte av alkohol ved fermentering av den forsukrede væske.
Man har nå funnet at større utbytter av etanol kan oppnås ut fra celluloseholdig utgangsstoff når man samtidig under anaerobe forhold omsetter det celluloseholdige materiale, en cellulase og en alkoholproduserende mikroorganisme.
Selv om man ikke ønsker å være bundet av noen spesiell teori, antar man for tiden at ved vanlig forsukring av cellulose til glukose med cellulase, vil glukoseutbyttet være lavt på grunn av at reaksjonen hemmes av den dannede glukose, samt av cellobiose som fås som biprodukt. Det antas derfor at samtidig nærvær av en alkoholproduserende mikroorganisme sammen med cellulasen i reaksjonsblandingen i henhold til foreliggende oppfinnelse resulterer i omdannelse av glukose til etanol og gjør det mulig at den enzymatiske omdannelse av cellulose til glukose går lengre enn hvis glukosen ikke ble omdannet på denne måten. Uavhengig av teoretiske be-traktninger forløper imidlertid forsukringsreaksjonen i henhold til foreliggende oppfinnelse glatt og med betraktelig øket totalutbytte av alkohol fra cellulose.
Ifølge foreliggende oppfinnelse er det således til-veiebragt en fremgangsmåte for fremstilling av etanol fra celluloseholdige materialer, hvorved cellulosen enzymatisk forsukres til glukose med en cellulase, og glukosen for-
gjæres til etanol under anaerobe forhold med en mikroorganisme av arten Saccharomyces cerevisiae, og denne fremgangsmåte er kjennetegnet ved at man samtidig omsetter cellulosen, cellulasen og mikroorganismen.
De cellulosematerialer som er egnet som utgangs-stoffer omfatter rensede cellulosetyper, celluloseholdige jordbruksprodukter som'bomull, tømmer, strå av ris og hvete, maiskolbekjerner og andre stoffer som overveiende inneholder cellulose som f.eks. avispapir, bølgepapp, magasinpapir og avfallspapir. For at nevnte stoffer skal kunne brukes effektivt som utgangsmaterialer til forsukringsreaksjoner i nærvær av cellulase, er det en fordel om materialene ned-brytes eller oppmales. For hydrolyse av nevnte celluloseholdige stoffer vil vanlig markedsført cellulase være til-strekkelig. Et enzympreparat som f.eks. "Cellulase Onotsuga" kan brukes. En væske som inneholder cellulase, nemlig et dyrkningsmedium fra en cellulaseproduserende mikroorganisme som f.eks. Trichoderma viride, kan også brukes.
Man lager med fordel en vandig suspensjon inneholdende 1-30 vekt-% cellulose eller et celluloseholdig materiale, og varmesteriliserer materialet slik at det danner et egnet utgangsstoff, en cellulase (eller cellulaseholdig væske) tilsettes utgangsstoffet og samtidig tilsettes en alkohol-produserende mikroorganisme, dyrket på forhånd slik at reaksjonen forløper anaerobt ved temperaturer mellom 25 og 35°C.
Når omsetningen er foretatt som ovenfor beskrevet og
i henhold til nedenstående foretrukne utførelse, vil det produseres etanol med et utbytte omtrent fire ganger så
stort som etter kjente metoder. Produksjon av etanol fra cellulose etter denne fremgangsmåten har den ytterligere fordel at den gjennomføres ved en enkel ett-trinns opera-
sjon. I motsetning til de kjente metoder som omfatter to trinn med forsukring av et cellulosemateriale med cellu-
lase og separasjon av det forsukrede produkt ved filtrering med påfølgende alkohol-fermentering av det forsukrede væske-filtrat, gjennomføres i henhold til foreliggende oppfinnelse
forsukringsreaksjonen av cellulose-råmaterialet med cellu-
lase og alkohol-fermenteringen av den dannede glukose sam-
tidig i ett trinn. Metoden må altså sies å være meget effektiv.
Det fremstilles etanol i henhold til oppfinnelsen
ut fra cellulose og jord- og skogbruksproduserte materialer eller avfallsstoffer, overveiende inneholdende cellulose og oppfinnelsen er således ikke bare fordelaktig for fremstil-
ling av alkohol, men utgjør også en effektiv metode til ut-nyttelse av celluloseråstoffer.
Oppfinnelsen skal nå beskrives ved hjelp av et eksempel på en foretrukket utførelse.
Eksempel
Utgangsstoffet ble fremstilt ved å suspendere 12,5 g papirmasse (80 vekt-% celluloseinnhold) i 100 g vann inneholdende i oppløst form 0,25 g asparagin som nitrogenkilde,
0,1 g kaliumhydrogenfosfat (KI^PO^), 0,3 g magnesiumsulfat (MgSO^•71^0) og 0,02 g gjærekstrakt. Den dannede blanding ble innstilt på pH 4,0 ved tilsetning av acetatpuffer og temperatursterilisert. Til denne blanding satte man 1 g raffinert teknisk cellulase og to inokuleringsløkker av Saccharomyces cerevisiae mycel direkte fra skrå-agar, og blandingen ble omsatt anaerobt ved ca. 30°C i 96 timer.
Ved analyse på dette tidspunkt fant man at det var dannet
2 g etanol i blandingen. Etter at reaksjonen fortsatte ytterligere 96 timer, hadde man totalt 4 g etanol.
Sammenligningsforsøk
For sammenligningsformål gjennomførte man etanol-produksjon etter den tidligere kjente to-trinns-metode.
Til en blandingssuspensjon av papirmasse i vann i ovennevnte mengder tilsatte man 1 g av den samme raffinerte, tekniske cellulase og blandingen ble omsatt ved 40°C i 9 6 timer. Mengden reduserende sukker i den forsukrede væske var 5 g, hvorav 60% (3 g) var cellobiose og resten, 2 g, glukose. Etter fortsatt reaksjon i 96 timer fant man ingen forandring
i mengden produsert sukker. Den forsukrede væsken ble blandet med samme nitrogenkilde, fosfat og andre uorganiske salter, som ovenfor, og varmesterilisert. Saccharomyces
cerevisiae ble tilsatt og gjæringsmediet satt til gjæring. Alkoholmengden etter avsluttet gjæring var under 1 g.
Siden glukoseinnholdet i den forsukrede væske som utgangspunkt for alkoholfermentering var 2 g, ville det teoretiske maksimalutbytte av etanol være 1 g.
Man ser altså av denne sammenligning at foreliggende oppfinnelse gjør det mulig å produsere etanol i vesentlig større utbytte enn etter den kjente to-trinns-metode.

- Fremgangsmåte for fremstilling av etanol fra celluloseholdige materialer, hvorved cellulosen enzymatisk forsukres til glukose med en cellulase, og glukosen for-gjæres til etanol under anaerobe forhold med en mikroorganisme av arten Saccharomyces cerevisiae, karakterisert ved at man samtidig omsetter cellulosen, cellulasen og mikroorganismen.
