Polymere kwatemaire ammoniumverhindingen, werkwijze voor het bereiden daarvan alsmede hun toepassingen.
De uitvinding heeft betrekking op nieuwe iono-5 gene polymere verbindingen met eindstandige tertiaire aminogroepen die toegepast kunnen worden als microbiociden, plantegroeiregula-toren, corrosieremmers, middelen om de bindingssterkte tussen vezels te verminderen zoals bij het vervaardigen van papier, weekmakers, antistatica, middelen om emulsies te breken, en middelen 10 om de aanverfbaarheid en kleurechtheid van textielmaterialen en papier te verbeteren.
De nieuwe ionogene polymere verbindingen volgens de uitvinding hebben de formule 1 op het formuleblad waarin R* ’ een groep met de formule 2, 3 of 4 voorstelt, R een methyl-, 15 ethyl-, propyl-, butyl-, hydroxyethyl- of hydroxypropylgroep voorstelt, R’ dezelfde betekenis heeft als R en wanneer R een methylgroep voorstelt, bovendien een alkylgroep met 6-18 koolstof-atomen en eventueel met 1 of 2 ethenisch onverzadigde bindingen^ of cyclohexylgroep voorstelt, of R en R' samen een pyridyl- of % 20 piperidylgroep vormen, m een geheel getal is van 2-12 en n een oneven getal is van 1-201.
De polymeren volgens de uitvinding worden bereid volgens een tweestaps werkwijze. In de eerste stap worden x molen N,N,N’,N’-tetramethy1-N,N'-bis(3-chloor-2-hydroxypropvl)alkaan-25 diammoniumdichloride, N,N,NT jN’-tetramethyl-NjN'-bisCS-chloor·^- hydroxypropyl)diethyletherdiammoniumdichloride of N,N'-dimethy 1-Ν,Ν'-bis(3-chloor-2-hydroxypropyl)piperaziniumdichloride tot reaktie gebracht met x - 1 molen van een ditertiair amine in water bij verhoogde temperatuur, x is een geheel getal van 2-101.
30 Deze reaktie kan in 1-30 uren bij ongeveer 80-105° C worden uitgevoerd.
In de tweede stap wordt een mol van de in de eerste stap verkregen voorloper tot reaktie gebracht met twee mol van een monotertiair amine in water of een organisch oplos-35 middel bij een temperatuur van ongeveer 25-110° C en gedurende 84018*0 ► ) - 2 - ongeveer 1-30 uren. Geschikte organische oplosmiddelen zijn in water oplosbare lage alkoholen en andere polaire organische oplosmiddelen. Het molekulair gewicht van de voorloper wordt berekend door het molekulair-gewicht van het gebruikte chloorhydroxypropyl 5 gesubstitueerde ditertiaire amine te vermenigvuldigen met x en x - 1 keer het molekulair- gewicht van het tweede ditertiair amine op te tellen.
De eerste stap van de werkwijze berust op de reaktie van een -ditertiair amine met een o<,£0 -dihalogeen- 10 alkylverbinding. Deze reaktie staat bekend als Menschutkin reaktie en wordt toegepast om polymere kwaternaire ammoniumverbindingen met een betrekkelijk laag molekulair-gewicht van in het algemeen ongeveer 50.000 of minder (zogenoemde ionogene polymeren) te bereiden.
15 De ketenlengte van de lineaire polymeren kan als volgt worden geregeld. Wanneer twee molen van de , W -di-halogeenverbinding (x molen) tot reaktie worden gebracht met een mol (x - 1 mol) van het ditertiair amine, ontstaat een polymeer met een zeer korte keten. Wanneer de dihalogeenverbinding 20 wordt aangeduid met A en het ditertiair amine met B kan het polymeer worden aangegeven met A-B-A. Wanneer 5 molen A en 4 molen B met elkaar tot reaktie worden gebracht ontstaat een voorloper A-B-A-B-A-B-A-B-A. Deze algemene regel kan worden toegepast tot een maximum van ongeveer 101 voor A en 100 voor B. Ongeacht het 25 aantal molen A en B dat wordt toegepast, altijd bevat het voor- loperpolymeer aan beide uiteinden een halogeenatoom. Bij de tweede stap waarin de voorloper tot reaktie wordt gebracht met een mono-tertiair amine, ontstaan eindstandige kwaternaire ammoniumgroepen.
De eigenschappen van de polymeren volgens de uitvinding en varia-30 ties in hydrofiele en hydrofobe eigenschappen worden door de aard van het tertiair amine en de ketenlengte van de voorloper bepaald.
De in de eerste stap gebruikte Ν,Ν,Ν',Ν'-tetra- methyl-N,N?-his (3-chloor-2-hydroxypropyl)alkaandiammoniumdichloriden 35 worden bereid door reaktie van dihydrochloridezouten van N,N,NT,NT- 8401968 i i' - 3 - tetramethyl- Οζ, (O ,-alkaandiaminen met in de alkaangroep 2-12 koolstofatomen, met 2 mol epichloorhydrin. De andere X, ω-di-chloordikwaternaire ammoniumchloriden worden op dezelfde wijze bereid uit dilxydrochloridezouten van respektievelijk 2,2’-oxobis-5 (Ν,Ν-dimethylethanamine) en 1,4-dimethylpiperazine. Elk ditertiair amine dat wordt gebruikt om de 0( , -dichloordikwaternaire ammo-niumverbindingen te bereiden, wordt dan als de vrije base tot reaktie gebracht met de 5^, ^-dichloorresten. Daarbij kunnen verschillende ditertiaire aminen worden gebruikt. Het c{, -dichloor-10 derivaat kan dus eerst met een ditertiair amine tot reaktie gebracht en daarna met een ander ditertiair amine of met mengsels van ditertiaire aminen om de voorloper te verkrijgen.
De in de tweede stap gebruikte monotertiaire aminen om de ionogene polymere voorlopers uit de eerste stap te 15 termineren, omvatten alifatische , alicyclische, alkylaromatische, aromatische en heterocyclische aminen .De alifatische groepen kunnen een of meer ethenisch onverzadigde bindingen bevatten en door een of meer hydroxylgroepen zijn gesubstitueerd. Als voorbeelden van deze aminen kunnen worden genoemd N,N-dimethylmethana-20 mine (trimethylamine), Ν,Ν-diethylethanamine (triethylamine), N,N-dimethyl-l-octadecanamine(dimethylstearylamine), N,N-dimethyl- 1-octadecenamine (dimethyloleylamine), N,N-dimethyl-l-dodeeanamine (dimethyllaurylamine), N,N-dimethyl-l-tetradecanamine (dimethyl-myristylamine), N,N,-dimethyl-1-hexadecanamine (dimethylpalmityl-25 amine), N-methyl-N-octadecyl-l-octadecanamine (methyldistearyl-amiiie), N-decyl-N-methyl-l-decanamine (didecylmethylarnine), methyldicocoamine, methyl-gedihydrogeneerd talkamine, l-chloor-3-(dimethylamino)-2-propanol, pyridine, 2,2’-2''-nitrilotris(ethanol)- ‘ (triethanolamine), 2-(dimethylamino)ethanol, 1,1',1 *'-nitrilotris-30 2-propanol(triisopropanolamine), Ν,Ν-bis(I-methylethyl)-2-propana-mine, Ν,Ν-dimethylcyclohexylamine, en N-methylpiperidine.
Polymere verbindingen worden toegepast in de pulp- en papierindustrie, in de textielindustrie en voorhhet behandelen van afvalwater voor de bovenaangegeven doeleinden.
35 Maar van de bekende polymere verbindingen voldoet er geen enkele 840196« I f - 4 - geheel. Sommigen zijn weliswaar geschikt als retentiehulpmiddel en flocculatiemiddel maar missen de andere gewenste eigenschappen.
Polymeren van het ionogene type die worden bereid door reaktie van ditertiaire aminen met dihalogeenverbindingen 5 zijn typisch produkten met een betrekkelijk laag molekulair-gewicht. Deze verbindingen kunnen doeltreffend worden gebruikt voor het bestrijden en beheersen van mieroorganismen, maar als flocculatiemiddel of plantegroeiregulator hebben zij hun beperkingen. De meest veelzijdige kationogene polymeren zijn poly-10 ethyleeniminen die in verschillende molekulair-gewichttrajekten kunnen.worden bereid door verschillende katalysatoren te kiezen en gebruik te maken van dwarsverknopingsmiddelen. Maar geen enkel polyethyleenimine is een goed bestrijdingsmiddel tegen micro-organismen. Daar komt bij dat voor het bereiden van polyethyleen-15 iminen het uiterst giftige ethyleenimine nodig is, dat in de laatste tijd als carcinogeen is beschreven. Aan het behandelen van ethyleenimine op technische schaal zijn dan ook van overheidswege allerlei beperkingen opgelegd.
De degraderende werking van microorganismen 20 op organische materialen is algemeen bekend. Eliminatie of groei-remming van algen, bacteriën en fungi vormt dan ook het onderwerp van een groot aantal researchprojekten en octrooischriften. Kwaternaire ammoniumverbindingen hebben toepassing gevonden voor het behandelen van industrieel koel-water en zwembaden.
25 De kationogene polymeren volgens de uitvinding zijn werkzaam tegen algen, bacteriën en fungi in watersystemen, zelfs wanneer zij in zeer lage concentraties worden toegepast.
Een zeer belangrijk aspekt van de uitvinding, zo niet het meest belangrijke, is dat de onderhavige verbindingen 30 opmerkelijke eigenschappen als plantegroeiregulatoren vertonen.
In dit verband is het volgende van belang. In 1975 heeft de National Academy of Sciences erop gewezen dat de toepassing van plante-groeiregulatoren er de oorzaak van kan zijn dat de opbrengst in de landbouw zo enorm sterk is toegenomen. Plantegroeiregulatoren 35 worden toegepast om gewassen te modificeren door wijziging van de 8 4 0 1 9 6 8 - 5 - i i snelheid, het patroon of beide van hun respons op interne en externe faktoren die alle ontwikkelingsstadia vanaf ontkieming via vegetatieve groei, reproduktieve ontwikkeling, rijping en veroudering, alsook conserving na de oogst, beheersen.
5 Plantegroeiregulatoren die geen voedingsstoffen zijn, bestaan gewoonlijk uit natuurlijke of synthetische verbindingen, en worden rechtstreeks op een gewas aangehracht om door wijziging van de ontwikkeling of struktuur op een of andere gunstige manier de opbrengst te verhogen , de kwaliteit te verbeteren 10 of het oogsten te vergemakkelijken.
Het is gebleken, dat wanneer de verbindingen volgens de uitvinding worden aangebracht op katoen, peulvrucht-gewassen in het bijzonder sojaboon, en andere gewassen, zij een verscheidenheid aan plantegroeiregulatorresponsies teweeg brengen.
15 Als plantegroeiregulatoren kunnen de verbindingen volgens de uitvinding zonder meer of in combinatie met een adjuvans in vaste of vloeibare vorm worden toegepast. Plantegroeiregulator-preparaten worden bereid of vervaardigd door het werkzaam bestanddeel te mengen met een adjuvans met inbegrip van verdunningsmidde- 20 len, verdikkingsmiddelen, dragers en conditioneringsmiddelen. Het werkzaam bestanddeel kan dus worden toegepast met een adjuvans zoals een organische vloeistof, water, bevochtigingsmiddel, dis-pergeermiddel, emulgator of een passende combinatie daarvan.
Geschikte vloeibare verdunningsmiddelen omvatten 25 bijvoorbeeld water, alkoholen en glycolen.
De plantegroeiregulatorpreparaten volgens de uitvinding bevatten gewoonlijk een of meer oppervlakteaktieve stoffen in een voldoende hoeveelheid om het preparaat gemakkelijk dispergeerhaar in water te maken.
30 Uit laboratoriumproeven beschreven in de onder staande voorbeelden LXVIII en LXIX blijkt dat de verbindingen volgens de uitvinding als plantegroeiregulatoren werkzaam zijn omdat zij de ontwikkeling van etheen en zuurstof en de kooldioxyde-fixatie gunstig beïnvloeden.
35 Hoewel bij toepassing als plantegroeiregulatoren 84 0 1 9 6 9 3 $ - 6 - gunstige resultaten worden bereikt met zo weinig als 70 g/ha en zo veel als 2240 g/ha, worden de beste resultaten in het algemeen bereikt met ongeveer 140-1120 g/ha verhinding volgens de uitvinding. Met meer dan 2240 g/ha wordt doorgaans geen verbetering 5 bereikt terwijl de kosten toenemen. Het is duidelijk dat de optimale hoeveelheid afhankelijk is van velerlei faktoren zoals de populatiedichtheid, het type of de variëteit van de gewassen, en de doeltreffendheid waarmee de verbindingen worden opgebracht.
Als plantegroeiregulatoren kunnen de verbin-10 dingen volgens-de uitvinding met goed gevolg op allerlei soorten planten worden toegepast. Bijzonder waardevol zijn zij voor belangrijke landbouwgewassen zoals katoen, granen, peulvruchten en talrijke groente- en fruitverschaffende gewassen. Ook kunnen ! zij worden toegepast op siergewassen, kamerplanten en andere 15 planten voor hoofdzakelijk of alleen decoratieve doeleinden.
De ionogene polymeren volgens de uitvinding zijn oplosbaar in water of andere polaire oplosmiddelen zoals alkoholen, glycolen en dimethylformamide. Voor het bestrijden of beheersen van microorganismen geschikte concentraties variëren van 0,5-500 20 dpm. berekend op het gewicht van het te behandelen water. Voor het bestrijden van corrosie in waterige systemen zijn concentraties van 0,5-500 dpm. berekend op het gewicht van het te behandelen water geschikt, bij voorkeur concentraties van 0,5-50 dpm.
Als middel om vezels in cellulosepulp van elkaar los te maken, 25 althans de bindingssterkte tussen de vezels te verminderen, worden de ionogene polymeren volgens de uitvinding gebruikt in hoeveelheden van 0,1-2,0 gew.% berekend op het droge vezelgewicht.
Het zachtmaken van textiel, papier of cellulose-vezelvellen wordt met de polymeren volgens de uitvinding bereikt 30 in hoeveelheden van 0,1-1,0 gew.% berekend op het droge gewicht van het te behandelen materiaal.
Als middel om olie-in-water of water-in-olie emulsies te breken, zijn concentraties van 0,5-500 dpm. berekend op het gewicht van de emulsies geschikt.
35 Geschikte concentraties voor toepassing van de 8401958 0 i - 7 - polymeren volgens de uitvinding als antistatica voor textiel-weefsels, kunststoffen of papier bedragen 0,1-2,0 gew.% berekend op het te behandelen materiaal.
Om de aanverfbaarheid en kleurechtheid van textiel-5 materialen en papier te verbeteren, worden de polymeren volgens de uitvinding geschikt toegepast in concentraties van 0,05-1,0 gew.% berekend op het droge textielweefsel of droge papier.
Hoewel de uitvinding op de eerste plaats betrekking heeft op nieuwe ionogene polymeren, maken ook methoden voor 10 het regelen van de groei en ontwikkeling van gewassen, en voor het bestrijden en beheersen van algen, bacteriën en fungi in waterige systemen deel uit van de uitvinding.
De hierna volgende voorbeelden en proeven dienen uitsluitend ter toelichting op de uitvinding zonder de conclusies 15 te beperken.
Voorbeeld I
Bereiding van N,N,NT,N>-tetramethyl-N,N,t-bis(3-chloor-2-hydroxy-propy1)ethaandiammoniumdichloride.
20 (Voorloper A)
In een 1 1 vierhalsrondbodem voorzien van een terugvloeikoeler, mechanische roerder, thermometer en druppel-25 trechter werd 187,8 g (1,0 mol) 61,9 %-ige waterige oplossing van N,N,N’,Nf-tetramethyl-l,2-ethaandiamine gebracht. De oplossing werd met een ijs-waterbad gekoeld en 197,1 g (2 mol) 37 %-ig zoutzuur werd toegevoegd met een zodanige snelheid dat de temperatuur beneden 45° C bleef. Aan dit mengsel werd onder krachtig 30 roeren 185,0 g (2,0 mol) epichloorhydrin langzaam toegevoegd zodanig dat de temperatuur niet hoger werd dan 45° C. Toen alles was toegevoegd, werd de temperatuur verhoogd en 30 minuten tussen 60 en 70° C gehouden. Aldus werd een 65,7 %-ige waterige oplossing van de titelverbinding verkregen.
35 Een evenmatige hoeveelheid van de waterige oplos- 0 4 0 ' ~ ~ » & - 8 - sing werd met vier keer zijn volumehoeveelheid aan aceton behandeld. Daarbij ontstond een kleverig neerslag waarvan de bovenstaande vloeistof werd afgeschonken. Het neerslag werd opgelost in methanol en de gevormde oplossing werd verdund met aceton. Het daarbij 5 gevormde neerslag werd afgefiltreerd en onder verminderde druk gedroogd boven waarbij een zeer hygroscopische witte vaste stof werd verkregen .

25 Aan een onder heftig roeren refluxende oplossing van 50 g (0,12 mol) Kemamine T-6501 (methyldicocosamine met een gemiddeld molekuulgewicht van 415,5 en afkomstig van Humko Sheffield Chemical Co.) in 250 ml aceton werd 32,5 g (0,06 mol) 69,3 %-ige waterige oplossing van het produkt uit voorbeeld I langzaam toe-30 gedruppeld. Nadat alles was toegevoegd werd het refluxen onder roeren nog 5 uren voortgezet. Na afloop werd de aceton afgedestilleerd. Het olieachtige residu, dat 87,9 gew.% van het gewenste polykwaternaire ammoniumzout bevatte, werd naar een houder overgebracht. Bij afkoelen daarvan ontstond een witte, spekachtige 35 halfvaste stof met een beperkte oplosbaarheid in water.

Aan 40,0 g (0,0774 mol) gesmolten Kemamine T-9701 (gedydrogeneerd methyltalkamine met een gemiddeld molekulair-ge-5 wicht van 536,8 en afkomstig van Humko Sheffield Chemical Co.) werd 20,9 g (0,0387 mol) van de voorloper A oplossing uit voorbeeld I langzam en onder heftig roeren toegevoegd bij een temperatuur tussen 80 en 95° C. Na 16 uren roeren bij deze temperatuur werd het reaktiemengsel afgekoeld waarbij een polykwaternair ammonium-10 zout werd verkregen als een witte spekachtige halfvaste stof met een gehalte vaste stof van 89,5 gew.%.

5 (Voorloper I)
In een 5 1 vierhalsrondbodem voorzien van een terugvloeikoeler, mechanische roerder, thermometer en druppel-trechter werd 3198,5 g (5,0 mol) 47,6 %-ige waterige oplossing 10 van 3,4-dimethylpiperazine gebracht. Onder koelen van de oplossing met. een ijs-waterbad werd 985,5 g (10,0 mol) 37 %-ig zoutzuur toegevoegd met een zodanige snelheid dat de temperatuur beneden 45° C bleef. Aan de gevormde oplossing van 1,4-dimethylpiperazine-dihydrochloride werd 925,2 g (10,0 mol) epichloorhydrin langzaam 15 en onder heftig roeren toegevoegd zodat de temperatuur niet boven 45° C kwam. Toen alles was toegevoegd werd het reaktiemengsel eerst 30 minuten bij een temperatuur tussen 60 en 70° C verwarmd en daarna 1 uur bij 100° C. Aldus werd een 59,8 %-ige oplossing van N,N'-dimethyl-N,NT-bis(3-chloor-2-hydroxypropyl)piperazine-20 dichloride verkregen.

Van de in voorbeeld XXX bereide 59,8 %-ige 25 waterige oplossing van NjN'-dimethyl-NjN’-bis(3-chloor-2-hydroxy-propyl)piperaziniumdichloride (voorloper I) werden verschillende hoeveelheden met 1,4-dimethylpiperazine in de in onderstaande tabel E aangegeven molverhouding 18 uren onder heftig roeren ge-refluxd. 5 (Voorloper M)
In een 2 1 vierhalsrondbodem voorzien van een terugvloeikoeler, mechanische roerder, thermometer en druppeltrech-ter werden 160,2 g (1,0 mol) 2,2’-oxybis(N,N-dimethylethanamine) 10 en 293,9 g water gebracht. Onder koelen van het mengsel met een ijs-waterhad en heftig roeren werd 197,1 g (2,0 mol) 37 %-ig zoutzuur langzaam toegevoegd zodat de temperatuur van het reaktie-mengsel beneden 45° C bleef. Aan de gevormde 2,2’-oxybis(N,N-di-methylethaanamine)dihydrochlorideoplossing werd 185,0 g (2,0 mol) 15 epichloorhydrin langzaam en onder roeren toegevoegd zodat de temperatuur niet hoven 45° C kwam. Toen alles was toegevoegd werd het refluxen nog 6 uren voortgezet. Aldus werd een 50 %-ige waterige oplossing van de titelverbinding verkregen.
20 Voorbeeld Lil en LUI
Van de in voorbeeld LI bereide 50,0 %-ige waterige oplossing van N,N,N’jN'-tetramethyl-NjN'-bis(3-chloor-2-hydroxy-propyl)diethyletherdiammoniumdichloride (voorloper M) werden 25 verschillende hoeveelheden met 2,2f-oxybis(N,N-dimethylethaan-amine) in de in onderstaande tabel J aangegeven molverhouding 6 uren onder heftig roeren gerefluxd. Het gehalte vaste stof van de als waterige oplossingen verkregen polykwatemaire ammonium-zouten is eveneens in onderstaande tabel J vermeld.

10 Van de in de voorgaande voorbeelden bereide nieuwe verbindingen werden er enkele onderzocht op hun anti-bacteriele werking. Het gebruikte microorganisme was Enterobacter aerogenes. De gevolgde methode was de volgens het Amerikaanse octrooischrift 4.054.542 gemodificeerde methode beschreven in het 15 Amerikaanse octrooischrift 2.881.070. De concentratie in dpm.
nodi? om 80 % of meer sterfte onder Enterobacter aerogenes in een basaal medium met verschillende pHTs na 18 uren teweeg te brengen, is in onderstaande tabel N aangegeven.

De in voorbeeld XVI bereide verbinding werd onderzocht op zijn groeiremmende werking van fungi. De gebruikte schimmels waren Chaetomium globosum en Penicillium roquefort!.
De gevolgde methode was dè volgens het Amerikaanse octrooischrift 15 4.054.542 gemodificeerde methode beschreven in het Amerikaanse octrooischrift 3.356.706. De groei van beide schimmels werd door de verbinding uit voorbeeld XVI in een concentratie van 8 dpm. geheel geremd.
20 Voorbeeld LXVI
Van de in de bereidingsvoorbeelden bereide verbindingen werden er enkele onderzocht op hun groeiremmende werking aan algen. De gebruikte algen waren Chlorella pyrenoidosa, 25 Chlorococcum hypnosporum en Phormidium inundatum. De gevolgde methode was die volgens het Amerikaanse octrooischrift 3.771.989 voorheeld 2. Na 28 dagen werd de omvang van de algengroei vastgesteld en uitgedrukt in cijfers van 0 t/m 4 voor respektievelijk geen groei (0), zeer slecht, schraal of twijfelachtig (1), 30 slecht (2), goed (3), en uitstekend (4). De concentratiaverbinding in dpm. waarmee na 28 dagen een 0 (geen groei) kon worden toegekend, is in onderstaande tabel 0 aangegeven.

Voorbeeld LXVII
Van enkele verbindingen bereid in de Bereidings- 25 voorbeelden werd het vermogen onderzocht om de bindingssterkte tussen houtvezels te verminderen. Het gebruikte vezelmateriaal was nat gebleekt naaldhout Kraftpulp in de vorm van een waterige brij met een concentratie van 0,5 gew.%. Van de pulp werden met behulp van een met de hand bedreven laboratoriumapparaat matjes 2 30 van 20 x 20 cm en met een gewicht van 120 g/m vervaardigd. Na persen en drogen volgens standaardprocedure werd het effekt van de proefverbindingen op de vermindering van de bindingssterkte tussen de vezels bepaald met behulp van een Scott Internal Bond Tester zoals beschreven in TAPPI UM-403. De bindingsfaktor 35 werd als volgt berekend: §4 01S 6 8 -22- bindings sterkte tussen vezels in be- r . ,. . handelde pulpmat „ inn bindingsfaktor---:—-r τ—— -*--ï—:- .x 100.

Dus hoe kleiner de bindingsfaktor, hoe groter het effekt op de vermindering van de bindingssterkte tussen de vezels in de behandelde pulpmat. De resultaten zijn in onderstaande 10 tabel P aangegeven.

Voorbeeld LXVIII
De verbindingen uit de voorbeelden XLVI, XLVII, XLVIII en L werden onderzocht op hun plantegroeiregulerend vermo-2Q gen. De verbinding uit voorbeeld L vertoonde een uitgesproken positief effekt op de toename in etheenontwikkeling bij sojaboon en katoen en bleek bovendien een uitstekend middel om vruchten te doen rijpen, de katoenzaaddoos te openen en bloei te induceren. De andere verbindingen daarentegen hadden een negatief effekt op de etheenontwikkeling en deden de bloei, rijping en groei ver- 8401968 -23- minderen.
Daarnaast werd de invloed van de bovengenoemde verbindingen op de kooldioxydefixatie en zuurstofontwikkeling 14 onderzocht. Het percentage zuurstofontwikkeling en de CO^-5 opname door bladschijfjes werden als volgt bepaald:
Katoenr en sojaboonzaden afkomstig van een enkele ouderplant werden ontkiemd in met leidingwater bevochtigd vermi-culiet in een klimaatkamer. Acht tot tien dagen na de ontkieming werden met een kurkeboor acht schijfjes met een diameter van 1 cm 10 uitgesneden uit op het oog gelijkwaardige, volledig geexpandeerde bladeren, en in een 30 ml glasflesje gebracht. Meteen werd een kleine hoeveelheid ontgast 0,1 M fosfaatbuffer (pH 6,4) aan elk flesje toegevoegd.De buffer werd in de bladschijfjes geïnfiltreerd door van tijd tot tijd een vacuum van 50-75 mm kwik aan te leggen. 15 Na voorzichtig afschenken werd een verse hoeveelheid van 25,0 ml van dezelfde buffer toegevoegd en werd in elk flesje een magnetisch roerstaafje gebracht. Vervolgens werd elk flesje voorzien van een rubberstop waardoorheen een Clark zuurstofelektrode was gestoken. De stoppen waren voorzien van een groef langs de buiten-20 kant om luchtbelletjes te laten ontsnappen. De flesjes met de elektrode werden in een watermantel uit Plexiglas geplaatst waarna met roeren werd begonnen. Na 10 minuten equilibreren in dimlicht werden aan beide kanten van de watermantel koel-witte en fluorescerende lampen ontstoken. Deze handelingen werden uitge-25 voerd aan 4 flesjes met elektrode tegelijk.
Met deze inrichting is het mogelijk om aspekten van bladaktiviteit te onderzoeken door de zuurstofontwikkeling en netto kooldioxydeopname te bepalen. Gebruikmakend van dezelfde inrichting kon ook een indruk worden verkregen over de schijn- 30 bare kooldioxydeopname door onmiddellijk na afsluiten van elk 14 flesje 0,5 ml van een 0,1 millicurie per mol NaH COgOplossing in de buffer te injekteren. Na afloop van de zuurstofbepaling werden de bladschijfjes afgespoeld met leidingwater en in cen-trifugebuizen gebracht die 2 ml 5 gew./vol. %-ig trichloorazijn-35 zuur in ethanol en 0,1 g geaktiveerde houtskool bevatten, en bedekt 8401968

-24- met Parafilm. Na 48 uren incuberen bij 21-24° C werden de monsters 10 minuten gecentrifugeerd bij een relatieve centrifugekracht van 1000 of 13.000 g. Van de bovenstaande vloeistof werd 0,1 ml in een telflesje gebracht dat 20 ml scintillatievloeistof (4,0 g ΡΡ0 5 (2,5-difenyloxazool), 0,1 g POPOP (1,4-bis/~2-(5-fenyloxazolyl)7- benzeen), in 700 ml tolueen en 300 ml absolute ethanol) bevatte.

De telresultaten werden geïnterpreteerd als rest- C aktiviteit gefixeerd in ethanolische TCA-stabiele vormen. Het effekt als . . 14 plantegroeiregulatoren op de zuurs tof ontwikkeling en {^“fixatie 10 werd bepaald door deze verbindingen in een hoeveelheid van 0,25 ml aan het begin van de 10 minuten durende equilibratie toe te voegen aan het reaktiemedium. De resultaten zijn in onderstaande tabel Q vermeld.

Voorbeeld LXIX
De verbindingen uit de voorbeelden XLVI, XLVII, 35 XLVIII en L hebben een verhogende werking op de kooldioxydefixatie 8401988 -25- en zuurstofontwikkeling en daarmee een positief effekt op de voedselopname door het gewas zoals stikstofassimilatie voor de groei en kaliumassimilatie voor de vruchtrijping.
De invloed van de bovengenoemde verbindingen op 5 de etheenontwikkeling werd als volgt bepaald:
Bladschijfjes zoals verkregen in voorbeeld LXVIII werden ongeveer 20 minuten in een oplossing van de proefverbindingen ondergedompeld waarna zij met gedeioniseerd water werden afgespoeld en na snel afvloeien in 4 ml flesjes werden gebracht die met een jQ septum werden afgesloten. Na respektievelijk 1, 8 en 24 uren' werden monsters getrokken voor het bepalen van de etheenontwikkeling. Elke keer nadat een monster was getrokken werden de flesjes gespoeld met stikstof alvorens ze weg te zetten voor een volgende bemonstering. De ontwikkelde hoeveelheid etheen werd bepaald door 15 1 ml monsters uit de flesjes te injekteren in een Perkin-Elmer gaschromatograaf voorzien van een ongeveer 180 cm PoraPak Q, 0,149-0,177 kolom. Als referentie werd de detektorrespons op standaardetheen gebruikt. De hoeveelheid ontwikkeld etheen werd 2 -1 omgerekend in ml etheen/1 cm uur . De positieve of negatieve 20 veranderingen ten opzichte van de niet-behandelde gewassen, in procent, zijn in onderstaande tabel R vermeld.
