Abstract
In a method of producing a compound, particularly a protein compound, a genetic vector (50) is provided with a DNA fragment (55) encoding expression of the compound. The vector is introduced into a cell nucleus of a crop cell with plant DNA, whereby the DNA fragment is transferred into the plant DNA and expressed therein. The crop is grown in an at least substantially daylight-free or substantially closed environment (10) in which climate control is provided with which a spatial climate is at least almost completely controlled. Crop development is controlled by a controlled supply of an artificial light spectrum that is offered to the crop and by control of evaporation through the crop. The DNA fragment is expressed in the crop during cultivation and the produced phytocompound is isolated from the crop.
Description
A method of the kind referred to in the preamble leads to a transgenic crop, in which the foreign DNA fragment is fused with the plant's own DNA and is then expressed in vivo in the plant. In many cases this concerns protein compounds for medicinal applications, in which case it is also referred to as “green pharming” or “biopharming” where the crop serves as a “bioreactor”. By using genetic modification and transgenic crops, crops can be stimulated to produce complex protein compounds such as phytohormones and phytoenzymes. This technique heralds a new biotechnological era that paves the way for the production of such complex phyto-compounds that cannot be realized synthetically or only with great difficulty, but are now produced naturally by the transgenic crop.
To this end, a vector selected for this purpose must find a way to the chromosome package in the plant cell. This step is invasive for the plant and the plant often suffers from it, which ultimately damages the yield or can even lead to death. It is also desirable that the in vivo production takes place in a highly reproducible and controllable manner.
The present invention has for its object, inter alia, to provide a method for producing compounds which makes an important contribution thereto.
In order to achieve the intended object, a method of the type described in the preamble according to the invention is characterized in that the crop is grown in an at least substantially daylight-free, at least substantially closed, environment in which a climate control system is provided with which a spatial climate is at least almost completely achieved. is controlled, that a development of the crop is controlled by a controlled supply of an artificial light spectrum that is offered to the crop and by a control of evaporation by the crop, that the DNA fragment is expressed in the crop during cultivation , and that the produced compound is isolated from the crop. It has been found that under the indicated growth conditions and monitoring, practically identical crop development can be achieved over the entire cultivation field. The plants are therefore phenotyically equal and therefore there are in principle no weaker specimens.
Although the invasion of the coding vector in itself could mean an attack on the condition of the plant, the plants grown in this way appear to be in such good and strong condition that they do not seem worth mentioning. Comparable uniformity appears to apply to the production of the intended compound in the crop, calculated over the cultivation field. Both the plant development and the purity and yield of the compound to be obtained are thus largely controllable and reproducible, which is an important factor and condition for further upscaling of the production process.
In that respect, however, a preferred embodiment of the method according to the invention is characterized in that the vector is introduced into a genetically modified Agrobacterium, in particular Agrobacterium
Radiobacter (Agrobacterium Tumefaciens) met een gemodificeerd plasmide dat hetRadiobacter (Agrobacterium Tumefaciens) with a modified plasmid containing the
The agrobacterium hereby carries the intended genetic information (Transfer DNA), which codes for the production of the compound, as part of a modified plasmid that has been artificially incorporated therein for this purpose.
Once in the plant cell, the agrobacterium is able to secrete this DNA segment to allow it to fuse with the plant's own DNA, after which the cell expresses this segment as if it were the (plant's) own genetic information. By using an agrobacterium or comparable micro-organism as a vehicle for transporting the vector into the plant cell, external exposure to it is sufficient, after which natural proliferation starts autonomously. This process is thus highly self-regulating and accurate.
To facilitate the vector's entry into the plant, an incision is usually made in the leaf of the plant. The agrobacterium detects the wound in the plant and finds its way to the plant cell with the intended cell nucleus. However, the incised leaf causes plant damage and the plant suffers to a certain extent. A further preferred embodiment of the method according to the invention aims, inter alia, at avoiding such plant damage and is characterized for that purpose in that the vector is introduced into the crop by placing the crop under vacuum, or at least an underpressure, and exposing it to the vector at a reduced pressure. set. The agrobacterium is thus, for instance, sucked into the plant from a liquid or nutrient medium enriched therewith as soon as the negative pressure around the plant is removed but still prevails in the plant cell. The DNA fragment derived from the plasmid then penetrates into the cell nucleus of the cell and then embeds itself in the plant genome.
The method according to the invention is particularly suitable for the expression of complex protein compounds which cannot or hardly be synthesized chemically. In that respect, a particular embodiment of the method according to the invention is characterized in that the compound obtained is used in a pharmaceutical comprising a human or animal vaccine, enzyme or hormone. Good experiences have been gained in that respect with a further special embodiment of the method, which is characterized in that one or more tobacco plants (Nicotiana tabacum) are used for the crop.
In de tekening toont:The invention will be further elucidated below on the basis of an exemplary embodiment and an associated drawing. In the drawing shows:
Figuur 1 een uitvoeringsvoorbeeld van een teeltomgeving volgens uitvinding in een eerste stadium;Figure 1 shows an exemplary embodiment of a cultivation environment according to the invention in a first stage;
Figuur 2A-C een schematische weergave van een stap in een uitvoeringsvoorbeeld van de werkwijze volgens de uitvinding; enFigures 2A-C show a schematic representation of a step in an exemplary embodiment of the method according to the invention; and
Figure 3 shows the cultivation environment of figure 1 in an advanced stage.
Incidentally, it should be noted that the figures are purely schematic and are not always drawn to (the same) scale. In particular, some dimensions may be exaggerated to a greater or lesser degree for clarity. Corresponding parts are indicated in the figures with the same reference numeral.
Characteristic of the method according to the invention is that it takes place in a completely closed environment and under accurately controlled and controlled conditions. For this purpose, see Figure 1, the starting point is a daylight-free housing 10 in which a crop 1 is grown under the influence of artificial light 20 in a fully controlled manner. Figure 1 schematically shows a cultivation chamber within such a housing, which can contain a number of such cultivation chambers, also referred to as climate chambers or climate cells, side by side.
Het gaat in dit voorbeeld om de teelt van tabaksplanten {Nicotiana tabacum).Prior to the stage of figure 1, a germination chamber can be provided, but germination can also take place in the chamber shown. This example concerns the cultivation of tobacco plants (Nicotiana tabacum).
The spatial climate within the room 10 is fully controlled by conditioning means (not shown) with which, inter alia, an ambient temperature, air humidity and carbon dioxide content in the room 10 are closely monitored and kept at or around a predetermined value. These environmental factors can thus be adjusted to the stage the crop is in and, for example, imposed differently in a germination phase than in a later growth stage of the crop. If, on the other hand, use is made of a separate germination chamber or cutting chamber, a suitable climate can be continuously maintained therein, after which the crop, after having developed, is transferred to the climate chamber shown for further cultivation therein.
In accordance with the invention, all growth factors that are crucial for phenotypic crop development are controlled and accurately monitored in the climate chamber. This mainly concerns photosynthetically active radiation (PAR) to which the crop is exposed during cultivation and which is supplied entirely by means of artificial light fittings 20 . For this purpose, the artificial light fittings 20 comprise two sets of light-emitting diodes (LEDs) 21,22 which respectively emit radiation {light} in red between 600 and 700 nm and in blue between 400 and 500 nm. This covers the wavelengths that the crop needs for the light reaction of photosynthesis. In addition to the spectrum offered, the intensity of both wavelengths is also adjusted to the crop and a day/night cycle is artificially simulated and imposed.
In addition to the actinic artificial light spectrum to which the crop is subjected, the evaporation by the crop is also artificially imposed and determined. This is based on the insight that a steady sap flow through the crop is indispensable not only for cell development (stretching) but also for effective and efficient photosynthesis. According to the invention, this sap flow is maintained and controlled by means of a delicate interplay of a root temperature imposed on a root system of the crop and a leaf temperature of the leaf system which can be influenced separately from the ambient temperature by means of leaf heating means provided for this purpose.
In this example, the leaf heating means comprise separate infrared radiators 30 with which additional infrared radiation can be supplied to the leaf system in addition to the infrared radiation emitted by the artificial light fixture 20. Optionally, the luminaires 20 can also be provided with active cooling of cooling pipes and/or cooling bodies provided therein or therewith, through which a cooling medium circulates in order to reduce the amount of infrared radiation that is emitted.
A system of conduits 40 is provided for the root system, which is in heat-exchanging contact with a cultivation substrate 5 in which the crop is kept. A root temperature can hereby be controlled both upwards and downwards by setting a temperature of a medium circulating through the conduits 40 correspondingly higher or lower with a thermostatic control. The figure assumes cultivation in separate pots that are filled, for example, with topsoil or glass/rock wool as a cultivation substrate. However, hydroponics can also be used instead, in which the root systems of the individual plants are kept in a common pool. In that case, the bath temperature is thermostatically controlled to create the desired root temperature.
During the development of the crop, the necessary nutrients and watering are added to it. All in all, all factors that determine the phenotypic development of the crop are strictly controllable according to this cultivation method and therefore strictly reproducible. This ensures, among other things, that the crop is in an optimal condition after the stage in figure to undergo the subsequent stage, which is shown schematically in figures 2A-2C.
Een voorbeeld van een dergelijke bacterie is een agrobacterieFigure 2A shows a plasmid 50, which is a stable self-contained, usually ring-shaped, DNA structure that can replicate itself in a host cell. Plasmids occur freely in nature, for example in bacteria as an independent DNA segment outside the chromosomal DNA. An example of such a bacterium is an agrobacterium
Agrobacterium Radiobacter shown schematically in Figure 2B and commonly referred to as Agrobacterium Tumefaciens. By means of recombinant DNA techniques, which are assumed to be sufficiently known to the average person skilled in the art, a segment 55 (T-DNA) is placed in the plasmid of figure 2A which codes for the production of the protein compound to be produced. In this example, this concerns an mRNA segment that will be used as a human or animal vaccine.
The plasmid 50 is used as a genetic vector which transfers this T-DNA segment 55 to the plant cell and incorporates it into the plant's own DNA. For this purpose, this plasmid 50 is introduced into the bacterium 60 of figure 2B. Optionally, the plasmid may also be provided with a gene that codes for resistance to antibiotics. The bacteria into which the modified, or at least recombinant, plasmid 50 has been introduced will thus develop a resistance to an antibiotic, while other bacteria will be killed as a result. After this selection, the live bacteria 60 are multiplied on a culture medium 65 and brought to the crop in an autoclave 70, see figure 2C.
Eenmaal bij één plantcel zorgen chemische receptoren in de bacterie 60 ervoor dat het gemodificeerde plasmide 50 naar de celkern van de plant migreert om daar hetThe autoclave 70 provides pump means with which a vacuum, or at least reduced pressure, can be built up and maintained therein. The crop 1 is exposed to the agrobacterium 60 at this underpressure, as a result of which the agrobacterium 60 can penetrate the crop. Once at one plant cell, chemical receptors in the bacteria 60 ensure that the modified plasmid 50 migrates to the cell nucleus of the plant where it
Transfer DNA segment 55 to the plant's own genome. The crop is then transferred to a climate chamber, see figure 3, in which the crop is further developed. The T-DNA that is now part of the plant's own DNA through transcription from the plasmid is now expressed, as a result of which the plant will now produce the intended protein compound. By carefully monitoring and controlling the growth factors, as described above, this further course of cultivation is also highly reproducible and controllable. Not only is a high yield of ingredients obtained in this way, but also a high degree of purity is achieved through such phenotypic control and monitoring. The phyto-compound thus produced in vivo is finally isolated from the crop using separation techniques known per se.
Although the invention has been further elucidated above with reference to only a single exemplary embodiment, it will be clear that the invention is by no means limited thereto.
On the contrary, many variations and embodiments are still possible for an average skilled person within the scope of the invention.
