Description
WERKWIJZE VOOR HET REGENEREREN VAN Pd-KATALYSATOREN 5
De uitvinding betreft een werkwijze voor het regenereren van Pd-katalysatoren die neerslaan en worden gescheiden van het reactiemengsel na toepassing in een homogeen gekatalyseerde C-C koppelingsreactie.
10 Homogeen gekatalyseerde C-C koppelings- reacties zijn bekend in de literatuur. Voorbeelden daarvan zijn Heck-reacties, Suzuki-reacties en crosscoupling reacties. Hierbij geldt dat de kosten van de katalysator hoog zijn, zodat het van belang is de 15 katalysator te kunnen hergebruiken. In dergelijke reacties worden in de regel liganden toegepast om de Pd-katalysator in oplossing te houden. Een nadeel van toepassing van liganden is echter dat ze veelal duur zijn en dat de opwerking van het reactiemengsel wordt 20 bemoeilijkt. Wanneer geen ligand of weinig ligand ten opzichte van de Pd-katalysator wordt gebruikt in de C-C koppelingsreactie slaat de Pd-katalysator tijdens of na voltooiing van de reactie neer. Gebleken is echter dat wanneer de neergeslagen katalysator, al dan niet op een 25 drager, wordt hergebruikt, de activiteit van de katalysator sterk terugloopt.
Verrassenderwij ze is nu gebleken dat wanneer de Pd-katalysator na afscheiding uit het reactiemengsel wordt behandeld met een broom- of 30 iodiumverbinding de activiteit van de Pd-katalysator wordt hersteld.
Bij voorkeur wordt om het scheiden van de katalysator van het reactiemengsel te vergemakkelijken, tijdens de C-C koppelingsreactie dragermateriaal 35 toegevoegd. Geschikte dragermaterialen zijn 10155 ? n - 2 - bijvoorbeeld de gebruikelijke dragermaterialen voor edelmetaalkatalysatoren of filter-aids,bijvoorbeeld silica, alumina, diatomeeënaarden, in het bijzonder celite(-521) en dicalite.
5 De gewichtsverhouding dragermateriaal ten opzichte van Pd is bij voorkeur 10 tot 100, in het bijzonder 15-25.
Het scheiden van de neergeslagen katalysator en reactiemengsel kan bijvoorbeeld worden 10 bewerkstelligd door filtreren, decanteren of centrifugeren.
Geschikte iodium- of broomverbindingen die kunnen worden toegepast in de werkwijze volgens de uitvinding zijn bijvoorbeeld I2 en Br2.
15 De hoeveelheid toe te passen broom- of jodiumverbinding bedraagt bij voorkeur 0,1-10 mol equivalent, in het bijzonder 1-3 mol equivalent berekend ten opzichte van de hoeveelheid Pd-katalysator.
20 De temperatuur waarbij de behandeling met een jodiumverbinding (of broomverbinding) plaatsvindt is niet bijzonder kritisch en kan bijvoorbeeld kamertemperatuur zijn. In een voorkeursuitvoeringsvorm van de werkwijze volgens de uitvinding wordt de 25 behandeling met de jodium- of broomverbinding in situ uitgevoerd gedurende de volgende reactierun, hetgeen wil zeggen dat de katalysator na de reactie wordt afgescheiden uit het reactiemengsel en als zodanig wordt toegepast in een volgende reactie, waarbij dan 30 bij die volgende reactie de broom- of jodiumverbinding aanwezig is. In dit laatste geval wordt de behandeling .101 552- ! - 3 - met de broom- of jodiumverbinding dus uitgevoerd bij de gekozen reactietemperatuur.
Geschikte oplosmiddelen die desgewenst kunnen worden toegepast in de C-C-koppelingsreactie en 5 bij de behandeling met een broom- of jodiumverbinding zijn bijvoorbeeld water, dipolaire aprotische oplosmiddelen, bijvoorbeeld dimethyl formamide (DMF), N-methylpyrrolidinon (NMP); aromatische koolwaterstoffen, in het bijzonder tolueen; nitrillen, 10 in het bijzonder acetonitril; esters, ethers, of ketonen. Bij voorkeur wordt als oplosmiddel een dipolair aprotisch oplosmiddel of water toegepast.
Onder C-C koppelingsreacies worden in het kader van deze uitvinding verstaan Pd-gekatalyseerde 15 aromatische en olefinische substitutiereacties, zoals bijvoorbeeld beschreven in "Metal-catalyzed Crosscoupling Reactions" P. Diederich and P.J. Stang Eds.

Voorbeelden van geschikte C-C koppelings-20 reacties die kunnen worden uitgevoerd in het kader van de uitvinding zijn Heck-reacties, Suzuki-reacties en cross coupling reacties.
Bij een Heck-reactie worden verbindingen van het type RX gesubstitueerd door een olefine waarbij 25 X een leaving group voorstelt en R staat voor optioneel een gesubstitueerde (hetero)aromatische groep, een benzylische groep of een vinylische groep. De Heck-reactie wordt, in aanwezigheid van een palladium verbinding, bij voorkeur in aanwezigheid van een base 30 en bij voorkeur zonder ligand uitgevoerd.
Geschikte voorbeelden van optioneel gesubstitueerde (hetero)aromatische groepen zijn fenyl, 3015590 - 4 - naftyl, pyridyl, pyrrolyl, chinolyl, isochinolyl, furyl, thienyl, benzofuryl, indenyl, pyrimidinyl, pyrazolyl, imidazoyl, p-methoxyfenyl, 6-methoxynaftyl en p-isobutylfenyl. Benzylische groepen zijn 5 verbindingen met formule Ar-C-, waarbij C al dan niet gesubstitueerd kan zijn, met dien verstande dat er op de 1-positie van de substituenten geen H-atoom zit, en Ar staat voor een optioneel gesubstitueerde (hetero)aromatiche groep.
10 De (hetero)aromatische, benzylische en vinylische groep kunnen optioneel gesubstitueerd zijn met 1 of meerdere substituenten, in principe alle substituenten die inert zijn onder de gegeven reactiecondities. Geschikte voorbeelden van dergelijke 15 substituenten zijn een alkyl-, (hetero)aryl-, alkoxy-, cyano-, formyl-, alkylcarbonyl-, carboxyl-, carbamoyl-, nitrogroep of halogeen.
De leaving groep X van de RX verbinding is bijvoorbeeld I, Br, OTf (triflaat), N2+Y' waarin Y een 20 anion, bijvoorbeeld Cl' of BF4'voorstelt, -C(0)-C1, -C(0)-O-C(O)-Ar, -S02C1 of -S02'Na+. Bij voorkeur wordt I of Br als leaving group toegepast.
Als olefine wordt bij voorkeur een verbinding toegepast met formule (1) 25 R1R2C=CHR3 (1) waarin R1, R2 en R3 elk onafhankelijk van elkaar te kiezen zijn in functie van het gewenste eindproduct en 30 zowel electronstuwende, electronzuigend als electronneutrale groepen kunnen voorstellen. Geschikte keuzes voor R1, R2 en R3 zijn : waterstof ; alkylgroepen 101 KRor\ - 5 - met bijvoorbeeld 1 tot 20 koolstofatomen ; alkenyl-groepen met bijvoorbeeld 2 tot 20 koolstofatomen ; (hetero)arylgroepen met bijvoorbeeld 4 tot 50 koolstofatomen ; een carboxylgroep ; alkyl- en 5 arylcarboxylaatgroepen met bijvoorbeeld 2 tot 50 koolstofatomen ; alkyl- en arylacylgroepen met bijvoorbeeld 2 tot 50 koolstofatomen ; een carbamoylgroep of N-gesubstitueerde alkyl- en arylcarbamoylgroepen met bijvoorbeeld 2 tot 50 10 koolstofatomen ; een aminogroep of N-gesubstitueerde alkyl- en arylaminogroepen met bijvoorbeeld 1 tot 50 koolstofatomen ; alkyl- en arylamidogroepen met bijvoorbeeld 2 tot 50 koolstofatomen ; alkoxy- en aryloxygroepen met bijvoorbeeld 1 tot 50 15 koolstofatomen ; cyano ,· nitro ; een halogeen en alkyl-en arylthiogroepen met bijvoorbeeld 1 tot 50 koolstofatomen.
Bij voorkeur is één van de drie substituenten R1, R2, R3 waterstof, met meer voorkeur 20 zijn twee van de drie substituenten waterstof.
Geschikte voorbeelden van olefinen met formule (1) zijn aromatische en alifatische alkenen, bijvoorbeeld etheen, 1,3-butadieen, 1-deceen en styreen ; acrylaten, bijvoorbeeld methylmetacrylaat, n-25 butylacrylaat, 2-ethyl-hexylacrylaat, n-butylmetacrylaat en benzylacrylaat ; olefinische nitrillen, bijvoorbeeld acrylonitril en methacrylonitril ; olefinische amiden, bijvoorbeeld acrylamide en Ν,Ν-dimethylacrylamide ; maleaten, 30 bijvoorbeeld di-n-butylmaleaat ; pyridines, bijvoorbeeld 4-vinylpyridine ; olefinische ethers, bijvoorbeeld cyclohexylvinylether en l-vinyl-2- 1015520 - 6 - pyrrolidon. Ook optioneel gesubstitueerde cyclische olefinen, bijvoorbeeld cyclohexeen, indeen en cycloocteen, waarbij R1 en R3 uit formule (1) samen met de koolstofatomen waaraan ze gebonden zijn een 5 ringstructuur vormen, zijn olefinen die geschikt kunnen worden toegepast in de werkwijze volgens de uitvinding. Bij voorkeur worden alifatische en aromatische alkenen en acrylaten toegepast.
Suzuki-reacties zijn eveneens in de 10 literatuur bekend; bij Suzuki-reacties wordt bijvoorbeeld een organoboorverbinding met formule Ar-B (OR4) -OR5 waarbij Ar in de praktijk veelal staat voor een (hetero) arylgroep en R4 en R5 elk onafhankelijk bijvoorbeeld H of een alkylgroep, bijvoorbeeld met 15 1-8 C-atomen, voorstellen of samen met de 0-atomen waaraan ze gebonden zijn en het B-atoom een ring vormen met 2-5 C-atomen, in contact gebracht met een verbinding met formule R-X, als boven gedefinieerd voor de Heck-reactie.
20 Cross coupling reacties zijn bijvoorbeeld reacties waarbij een organometaalverbinding met formule (R6)TO-M-Qn, waarbij R6 staat voor een alkyl-, vinyl- of arylgroep met 1-18 C-atomen, M staat voor Li, Na, Mg,
Zn, Al, Cu, Zr, Mn, Ni, B, Sn of Si, m = 1, 2, 3 of 4 25 en n = 0, 1, 2, of 3, Q kan in principe elke willekeurige groep zijn. Voorbeelden van R6m-M-Qn zijn R6Li, R6Na, R6MgCl, R6A1(R7)2/ waarbij de groepen R7 elk afzonderlijk dezelfde groepen kunnen voorstellen als R6 met dien verstande dat wanneer R6 een vinyl- of 30 arylgroep voorstelt, R7 gelijk is aan alkyl, R6ZnCl, R6ZnBr, R6B(R8)2, waarbij de groepen R8 elk afzonderlijk dezelfde groepen kunnen voorstellen als R7, R6Sn(C4H9)3, 1015520 - 7 - R6ZrCp2Cl (Cp = cyclopentadienyl) , ReCu, R6C4(CN)Li, R6Cu(CN) Znl, R6Si (CH3) 3, R6SiF3, R6SiF2C6H5< in contact wordt gebracht met een verbinding met formule R-X als boven gedefinieerd.
5 De C-C koppelingsreacties worden bij voorkeur uitgevoerd in aanwezigheid van een base. De base wordt bij voorkeur gekozen uit de groep van tertiare amines, aromatische heterocyclische basen, bijvoorbeeld pyridine(s), alkalimetaal-acetaten, -10 hydroxides, -fosfaten, -carbonaten, en - waterstofcarbonaten. Geschikte keuzes zijn NaOAc, KOAc, K2C03, Na2C03, Cs2C03 en K3P04 of in een aantal gevallen trialkylamines, waarin de alkylgroepen elk onafhankelijk van elkaar bij voorkeur 1 tot 20, in het 15 bijzonder 1 tot 5 koolstofatomen bevatten, bijvoorbeeld Me3N, Et3N, en n-Bu3N.
De C-C koppelingsreactie wordt in de werkwijze volgens de uitvinding uitgevoerd met een Pd-katalysator bij voorkeur zonder ligand. Elke 20 willekeurige, Pd(0)-, Pd(II)-, en Pd(IV)-verbinding kan als katalysator gebruikt worden. Als Pd-katalysator wordt bij voorkeur Pd(0Ac)2, PdCl2, PdBr2, Pdl2,
Na2PdCl4, Na2PdCl6, Pd2(dba)3 (dba=dibenzylideen-aceton) , Pd-clusters, Pd-metaal, optioneel op drager, toegepast. 25 Als dragermateriaal kan bijvoorbeeld alumina, kool, of silica gekozen worden. Extra toevoegen van dragermateriaal, zoals weergegeven in de werkwijze van de uitvinding is dan niet meer nodig.
De hoeveelheid toe te passen Pd-katalysator 30 is niet kritisch: bij voorkeur wordt een hoeveelheid van 0,01 - 5 mol%, met meer voorkeur van 0,1 - 2 mol%, 1015520 - 8 - met de meeste voorkeur 0,2 - 1 mol% toegepast, berekend ten opzichte van de hoeveelheid RX.
De temperatuur waarbij de C-C koppelings-reactie bij voorkeur wordt uitgevoerd wordt in 5 belangrijke mate bepaald door de keuze van de R-X verbinding en ligt bij voorkeur tussen 0 en 200°C, bijvoorbeeld voor R-I tussen 50 en 100°C en voor R-Br tussen 100 en 150°C.
De uitvinding wordt verder toegelicht aan 10 de hand van de voorbeelden zonder evenwel daardoor te worden beperkt.
Voorbeelden 15 Definities
Opbrengst = Ceinde/D0 * 100 % (Y)
Conversie = (Do-De)D0 * 100 %
Selectiviteit = (opbrengst/conversie) * 100 % waarbij: 20 Ceinde = aantal mol produkt dat aan het einde van de reactie gevormd is.
D0 = aantal mol RX aan het begin van de reactie.
De = aantal mol RX aan het eind van de reactie.
25 Voorbeeld I
Regenereren van de Pd-katalysator met I2 in de Heek reactie van fenyljodide en n-butylacrylaat
In een Schlenkvat van 50 ml werd afgewogen: Pd(0Ac)2 (10,5 mg ; 0,047 mmol, 0,24 mol% t.o.v.
30 fenyl jodide) en 200 mg Celite. 25 ml NMP, en een magnetische roerder werden toegevoegd en het Schlenkvat werd afgesloten met een rubber septum. Onder stikstof t015520 - 9 - atmosfeer werd vervolgens al roerende m.b.v. een injectiespuit toegevoegd: Et3N (2,20 gr, 21,8 mmol), dihexylether (2,0 mmol) als interne standaard voor GC-analyse, en fenyljodide (4,1 gr, 20,1 mmol). Het 5 reactiemengsel werd verwarmd tot 70°C, en m.b.v. van een injectiespuit werd n-butylacrylaat (3,25 gr, 25,3 mmol) toegevoegd. Na een reactietijd van 50 minuten bij 70°C was volgens de GC-analyse de conversie > 95%, de opbrengst > 90%, en de selectiviteit > 95%.
10 Het experiment werd na 90 minuten stopgezet, en het reactiemengsel werd m.b.v. een centrifuge gescheiden van de neergeslagen Pd op de Celite. Het grijze residue werd met 25 ml N-methylpyrolidinon (NMP) teruggevoerd naar het 15 Schlenkvat en geheractiveerd met I2 (25 mg, 0,099 mmol, 2,1 molequivalent t.o.v. Pd(OAc)2). Het Schlenkvat werd weer afgesloten met een rubber septum en onder stikstof atmosfeer werden de benodigde reagentia voor de Heek reactie op dezelfde manier en in dezelfde volgorde 20 toegevoegd als hierboven vermeld.
, Na een reactietijd van wederom 50 minuten bij 70°C was volgens de GC-analyse de conversie > 95%, de opbrengst > 90%, en de selectiviteit > 95%.
Het experiment werd na 90 minuten 25 stopgezet, en het reactiemengsel werd m.b.v. een centrifuge gescheiden van de neergeslagen Pd op de Celite.
De Heek reactie van fenyljodide en n-butylacrylaat kon via deze werkwijze tot minimaal 7 30 keer met nagenoeg dezelfde conversies, opbrengsten en selectiviteiten uitgevoerd worden.

Het zelfde experiment, waarbij het grijze 10 residue met de neergeslagen Pd-katalysator op Celite steeds met 25 ml methyl-t-butylether (MTBE) werd gewassen, alvorens te heractiveren met jood, gaf dezelfde resultaten.

Het zelfde experiment, waarbij het grijze residue met de neergeslagen Pd-katalysator op Celite steeds met 2 molequivalenten broom in plaats van jood 5 werd geheractiveerd, gaf dezelfde resultaten.
Voorbeeld IV
Het experiment als weergegeven in voorbeeld III werd herhaald, waarbij Silica in plaats van Celite 10 werd gebruikt. Dezelfde resultaten werden bereikt.

De werkwijze volgens voorbeeld I werd herhaald gebruikmakend van verschillende arylhalogenides en/of olefinen. De reactieomstandigheden en de resultaten van de eerste 2 20 runs zijn weergegeven in Tabel 1.
Voorbeeld VIII
De werkwijze volgens voorbeeld I werd herhaald gebruikmakend van K2CO3 (1,76 gr, 12,8 mmol) en 25 n-Bu4NBr (1,70 gr, 5,3 mmol) in plaats van Et3N, en met water (29 ml) als oplosmiddel in plaats van NMP. Na een reactietijd van 120 minuten bij 80°C was volgens de GC-analyse de conversie > 95%, de opbrengst > 90%, en de selectiviteit > 95%.
30 De neergeslagen Pd op het Celite werd afgescheiden, geheractiveerd met jood (32,7 mg, 0,13 mmol, 2,75 molequivalenten t.o.v. Pd) en opnieuw J015520 - 12 - gebruikt in de Heck reactie van fenyljodide en n-butylacrylaat zonder aan activiteit in te boeten.

Voorbeeld IX
Het regenereren van Pd-katalysator met jood in de Suzuki koppeling en de cross coupling reactie
In een Schlenkvat van 50 ml werd afgewogen: 5 Pd(0Ac)2 (19 mg ; 0,085 mmol, 0,5 mol% t.o.v, 4-jodoanisool) en 300 mg Celite. 27 ml NMP, en een magnetische roerder werden toegevoegd. Het reactiemengsel werd verwarmd tot 70°C en onder stikstof atmosfeer werd vervolgens al roerende toegevoegd: Et3N 10 (2,20 gr, 21,8 mmol), dihexylether (2,0 mmol) als interne standaard voor GC-analyse, 4-jodoanisool (4,15 gr, 17,7 mmol) en fenylboorzuur (2,14 gr, 17,5 mmol). Het Schlenkvat werd afgesloten met een rubber septum en verwarmd naar 130°C. Monstername gevolgd door GC-15 analyse laat de vorming van 2 producten zien: het suzukiproduct 4-fenylanisool, en het cross coupling product 4,4'-dimethoxybifenyl. Na een reactietijd van 16 uur bij 130°C was alle 4-jodoanisool omgezet en werd de reactie stopgezet.
20 Het reactiemengsel werd m.b.v. een centrifuge gescheiden van de neergeslagen Pd op de Celite. Het grijze residue werd met 25 ml diethylether gewassen, gescheiden, en met N-methylpyrolidinon (NMP) teruggevoerd naar het Schlenkvat en geheractiveerd met 25 jood (41,8 mg, 0,165 mmol, 1,9 molequivalent t.o.v.
Pd(0Ac)2) · Onder stikstof atmosfeer werden de benodigde reagentia voor de volgende run op dezelfde manier en in dezelfde volgorde toegevoegd als hierboven vermeld. In de tweede run werd dezelfde selectiviteit en activiteit 30 waargenomen als in de eerste run.

Claims (8)
1. Werkwijze voor het regenereren van een Pd-katalysator die neerslaat en wordt gescheiden van 5 het reactiemengsel na een homogeen gekatalyseerde C-C koppelingsreactie, met het kenmerk, de Pd-katalysator vervolgens wordt behandeld met een broom- of jodiumverbinding.
2. Werkwijze volgens conclusie 1, waarbij de 10 behandeling in situ wordt uitgevoerd in een volgende reactierun.
3. Werkwijze volgens conclusie 1 of 2 waarin als broom- of j odiumverbinding Br2 of I2 wordt toegepast.
4. Werkwijze volgens een der conclusies 1-3, waarbij de hoeveelheid broom- of jodiumverbinding ligt tussen 1 en 3 molequivalent berekend ten opzichte van de hoeveelheid Pd-katalysator.
5. Werkwijze volgens een der conclusies 1-4, met het 20 kenmerk, dat de C-C koppelingsreactie wordt uitgevoerd in aanwezigheid van dragermateriaal.
6. Werkwijze volgens een der conclusies 1-5, waarbij de C-C koppelingsreactie een Heck-reactie is.
7. Werkwijze volgens een der conclusies 1-5, waarbij 25 de C-C koppelingsreactie een Suzuki-reactie is.
8.
