BESCHRIJVING
Titel: Werkwijze voor het verhinderen van replicatie in Cryptosporidium parvum met behulp van ultraviolet licht
GEBIED VAN DE UITVINDING
De onderhavige uitvinding heeft betrekking op een werkwijze voor het verhinderen van de replicatie van Cryptosporidium parvum in water en in het bijzonder op een werkwijze voor het verhinderen van infecties van
Cryptosporidium oöcysten en soortgelijke organismen in : water, gebruikmakend van lage niveau's ultraviolet licht.
ACHTERGROND VAN DE UITVINDING I
Het is in het algemeen welbekend dat het nodig is ~
om xx oöcysten te doden of inactiveren zodat ze niet I
infecteren. Dit is vooral van belang in drinkwater. Een : zo'n methode is het gebruik van ultraviolet ("UV") licht. De stand van de techniek leert, dat een UV dosis i van ten minste 3000 mJ/cm2 vereist is voor inactivering = van Cryptosporidium parvum (Lorenzo-Lorenzo et al., " J. Parasitol. 1993, 79, 67-70) en Giardia muris (E.L. "
Jarol, "Effect of disinfectants on Giardia cysts", CRC
Critical Reviews in Environmental Control, 1988, 18, 1- 28). Snowball en medewerkers (Britse octrooiaanvrage ~ #9416287.2, 8 nov. 1984; Wat. Res. 1995, 29, 2583-2586) ontwikkelden een inrichting die eerst oöcysten van
Cryptosporidium uitfiltreert en ze dan blootstelt aan UV doses van 350-400 mJ/cm2. De octrooipublicatie leert het gebruik van membraanfilters om Cryptosporidium oöcysten te vangen, die vervolgens bestraald worden met een batterij van lage-druk kwiklampen voor een UV dosis
van 350-400 mJ/cm2. Het filter wordt dan in omgekeerde L
richting gewassen naar een tweede filter toe en de z bestraling wordt herhaald. De octrooipublicatie I: vermeldt dat de behandeling de organismen "doodt". _ 1012059 v>" 2 M.J. Lorenzo-Lorenzo, M.E. Area-Mazea, I. Villacorta-Martinez de Maturana en D. Duran-Oreiro, "Effect of Ultraviolet Disinfection of Drinking Water on the Viability of Cryptosporidium parvum Oocysts", J. Parasitol. 1993, 79(1), 67-70. Het artikel rapporteert de verhindering van infectie in muizen na blootstelling gedurende ten minste 150 minuten aan UV uit een (vermoedelijk) lage-druk kwiklamp. Hoewel het artikel niet duidelijk is, kan daaruit worden afgeleid dat de toegepaste UV dosis meer dan 5000 mJ/cm2 was voor het verkrijgen van een meer dan 2 log reductie in infectiviteit. De auteurs beweren dat blootstelling aan UV gedurende 150 minuten of langer de infectiviteit "elimineert", maar ze geven geen ander mechanisme dan dat "UV straling DNA verbreekt doordat het de vorming vari th[y]amine dimeren veroorzaakt, en hoge niveau's kunnen tot celdood leiden". Bij de door hen toegepaste UV doses werden de waargenomen effecten vrijwel zeker door celdood veroorzaakt.
In een artikel van A. Bushnell, W. Clark, J. Dunn en K. Salisbury, "Pulsed Light Sterilization of Products Packaged by Blow-Fill-Seal Techniques”, Pharm. Engin. 1997, Sept./Okt., 74-83, wordt een techniek met UV pulsen beschreven voor het "steriliseren" van oppervlakken die bacteriën, fungi, sporen, virussen, protozoa en oöcysten bevatten. Vermeld werd dat de vereiste UV doses boven 1000 mJ/cm2 liggen. De effectiviteit van de methode werd beoordeeld naar de infectiviteit bij muizen. Bij de gerapporteerde UV doses werden de effecten gedacht te wijten te zijn aan celdood.
In een artikel van R. LaFrenz, "High Intensity Pulsed UV for Drinking Water Treatment", Proc. AWWA WQT Conference, Denver, CO, Nov. 1997, werd een soortgelijk systeem met pulsen beschreven. Hoewel erg weinig details werden beschreven, blijkt dat een muizen-infectiviteitstest werd gebruikt en met een beweerde a 4 I 1012059 ;a 3 100% "inactivering" van Cryptosporidium tot een niveau van 6 log bij energieniveau's van ongeveer 200 mJ/cm2 en hoger. Het artikel beweert dat het gepulseerde UV het "DNA herstelmechanisme" overwint; de toegepaste UV doses zijn echter veel hoger dan vereist zijn met een constante middelbare-druk kwiklamp.
Het is derhalve een doel van de uitvinding om een werkwijze te verschaffen voor het behandelen van water op zulk een effectieve wijze dat Cryptosporidium oöcysten niet kunnen infecteren. Een ander doel van de uitvinding is om een werkwijze te verschaffen waarbij =
ultraviolet licht wordt gebruikt om de Cryptosporidium oöcysten infectie-ineffectief te maken. Weer een ander I
doel van de onderhavige uitvinding is om een werkwijze te verschaffen waarin ultraviolet licht wordt gebruikt, welke werkwijze kosteneffectief is voor het behandelen I
van drinkwater om de mogelijkheid van infectie door r
Cryptosporidium oöcysten te elimineren. :
SAMENVATTING VAN DE UITVINDING T
Algemeen gesproken is gevonden dat het niet nodig r is om pathogenen, zoals Cryptosporidium parvum of ~
Giardia muris met ultraviolet licht te "doden" of 1
"inactiveren" om infectie te verhinderen; er behoeft I
slechts voldoende ultraviolet licht te worden toegepast I
om te verhinderen dat het organisme "repliceert". De werkwijze volgens de onderhavige uitvinding verhindert ~ replicatie (celmitose) door verknoping van het DNA om
infectie te verhinderen. De UV doses die nodig zijn om I
replicatie te verhinderen, zijn ordes van grootte lager Ξ dan nodig zijn voor het "doden" of "inactiveren" van de oöcysten. Dit betekent dat de kosten van UV behandeling ^ ter verhindering van infectie door Cryptosporidium oöcysten aanzienlijk lager zullen zijn.
Er is gevonden dat wanneer biologische organismen “ aan ultraviolet licht (UV) in het gebied van 200-300 nm “ 1012059 _ 4 worden blootgesteld, het UV door DNA, RNA en eiwitten geabsorbeerd kan worden. Absorptie van eiwitten kan tot openbreken van celwanden en dood van het organisme leiden. Het is bekend dat absorptie door DNA of RNA (specifiek door thymine basen) verknoping van de DNA of RNA dubbele helix strengen veroorzaakt door de vorming van thymine dimeren. Als genoeg van deze verknopingen in DNA worden gevormd, kan het DNA zich niet in twee strengen scheiden en daardoor kan de cel in mitose niet repliceren. Cellen die niet kunnen repliceren, kunnen nie't infecteren. De onderhavige uitvinding gebruikt aanzienlijk lagere UV doses voor het bereiken van de toestand van gehinderde replicatie, welke doses veel lager zijn (in ordes van grootte) dan de doses die vereist zijn om vernietiging van de oöcystcellen te veroorzaken.
De onderhavige uitvinding gebruikt een brede-band (200-300 nm) middelbare-druk UV lamp om de preventie te realiseren. De vereiste dosis kan zo laag zijn als 10 mJ/cm2. Doses boven 30 mJ/cm2 geven meer dan 4,5 log verwijdering, gemeten naar muizen-infectiviteit. De dosisniveau's zijn dus significant lager dan eerder werden gebruikt, wat tot gevolg heeft dat significant lagere stroomniveau's nodig zijn om de resultaten te bereiken. De werkwijze verschaft derhalve een : aanzienlijke stijging van de kosten-effectiviteit van UV reductie in Cryptosporidium oöcysten infectie in drinkwater. Andere voordelen zullen duidelijk worden uit de hiernavolgende gedetailleerde beschrijving van een momenteel geprefereerde uitvoeringsvorm van de uitvinding.
BEKNOPTE FIGUURBESCHRIJVING

MOMENTEEL GEPREFEREERDE UITVOERINGSVORM .
Pilottest uitdagingen van Cryptosporidium parvum en Giardia muris werden uitgevoerd in een 111 L (29,4 gal) UV reactor, die 6 x 1 kW Rayox middelbare-druk UV lampen, horizontaal gemonteerd in een torentype reactor, bevatte. De organismen werden voor de reactor stroomopwaarts van een statische menger geïntroduceerd en achter de reactor op 1 micrometer filters verzameld. De totale stroomsnelheid tijdens de test bedroeg ongeveer 215 gpm (814 L/minuut). De “ organismen werden uit het filter geëxtraheerd en geconcentreerd. Vier van de Crypto monsters werden aan muizen gegeven. Deel van het concentraat werd achter- - gehouden voor in vitro tests.
De UV doses werden berekend uit de gemiddelde bestraling (bepaald uit een geavanceerd mathematisch model van de reactor) maal de verblijftijd in de reactor (ongeveer 8,3 sec). De dosis werd gevarieerd door een of twee lampen aan te zetten en op laag of hoog vermogen.
Samenvatting van de resultaten In vitro tests
Er werden twee in vitro tests gebruikt: vitale kleurstoffen en excystatie. Er werd vastgesteld dat de procescontrole middelbare levensvatbaarheidswaarden 71,7% bedroegen in de vitale kleurstoffen test. De voor normalisatie van deze levensvatbaarheidswaarden op 100% benodigde factor werd berekend door 100 te delen door 71,7 = 1,39. Na blootstelling van oöcysten aan verschillende UV doses, werden hun feitelijke levensvatbaarheidswaarden bepaald (Tabel 1) en vervolgens 1012059 6 genormaliseerd door elke levensvatbaarheidswaarde te vermenigvuldigen met de berekende factor (1,39). Op gelijke wijze werden Cryptosporidium en Giardia procescontrole excystatiewaarden op 100% genormaliseerd en werd hun respectievelijke normalisatiefactor gebruikt om de feitelijke excystatiewaarden voor organismen, die aan UV waren blootgesteld, bij te stellen. Deze resultaten worden berekend als het percentage levensvatbaarheid voor een UV test gedeeld door het percentage levensvatbaarheid voor de procescontrole. Aldus te werk gaande veranderen de 1evensvatbaarheids-factoren (in procent) voor in vitro tests in die welke - in onderstaande tabel worden getoond.

1. Werkwijze voor de preventie van Cryptosporidium oöcysten. omvattende bestralen van water met een continue brede band ultraviolet licht in doses van ongeveer 10 mj/cm2 tot ongeveer 175 mJ/cm2.
2. Werkwijze volgens conclusie 1, waarin de genoemde brede band een frequentie is van 200 tot 300 nm onder toepassing van een UV lamp.
3. Werkwijze volgens conclusie 1 of 2 waarin de genoemde doses ongeveer 20 mJ/cm2 tot ongeveer 30 mJ/cm2 bedraagt. 1012059 -
