Abstract
Description
Korte aanduiding: Werkwijze ter bereiding van cyclische secundaire amineverbindingen, cyclische secundaire amine verbindin gen en toepassingen hiervan.Short designation: Process for preparing cyclic secondary amine compounds, cyclic secondary amine compounds and uses thereof.
The present invention relates to a process for the preparation of cyclic secondary amine compounds. More particularly, the present invention relates to novel cyclic secondary amine compounds, especially pyrrolidine type cyclic amines, as well as to their use.
The preparation of cyclic secondary amine compounds is known per se from US patent 2,580,738. According to this US patent, N-substituted-2,5-dialkyl-2,5-dicyanopyrrolidines are prepared via a so-called double strecker reaction of a 1,4-diketone compound with a primary amine and two equivalents of cyanide. Specifically, acetonyl acetone and two equivalents of hydrogen cyanide are first reacted and then the primary amine compound is added to form the dicyanopyrrolidines. However, experiments have shown that the exact order of addition of the different reactants is not critical, meaning that in each embodiment the desired N-substituted-2,5-dimethyl-2,5-dicyanopyrrolidines are formed. The method described in this US patent ensures that CN substituents are present at the 2,5 positions of the pyrrolidine ring. In addition, at the 3,4-positions of the pyrrolidine ring, only hydrogen atoms are shown as substituents.
The preparation of cyclic secondary amine compounds is also known from U.S. Patent 4,093,586. This US patent is based on the preparation method described in US-A-2,580,738 discussed above, however, according to US-A-4,093,586, an additional reaction is performed on the 2.5-positions of the pyrrolidine ring. In particular, U.S. Pat. No. 4,093,586 discloses cyclic secondary amine compounds wherein the substituents present at the 2,5-positions of the pyrrolidine ring are an alkyl group and a CO 2 R wherein R is an alkyl or phenethyl group. As the substituents present at the 3,4 positions in the pyrrolidine ring, only hydrogen is mentioned.

Experiments conducted by the present inventors have shown that both the yield and stability of the synthesized 2,5-dicyanopyrrolidines, as well as the reaction rate and stereochemistry, were affected by steric factors. A wide applicability of the above-described method is as yet prevented by the limited success in the conversion to esters of both nitrile groups, so that ultimately only cis-2,5-dimethoxycarbonyl-2,5-dimethylpyrrolidine-N-oxyl could be prepared.
A general method of preparing cyclic secondary amine compounds and their nitroxide derivatives is the route of the conjugated addition of a nitro compound to an α, β-unsaturated ketone compound to form a κ-nitrocarbonyl compound, followed by reductive cyclization to form the cyclic nitron compound. The latter compound is then treated with a Grignard reagent, followed by oxidation to form pyrrolidines-based nitroxides, as described in J.Am.Chem.Soc. 1951, 73, 1276 and J.Am.Chem.Soc. 1952, 74, 3664. The method is particularly suitable for the synthesis of nitroxides with simple alkyl or aryl substituents and is often accompanied by a lack of control of the stereochemistry and low yields in the final reaction steps.
It is therefore desirable to develop a new method of preparing cyclic secondary amine compounds and the corresponding nitroxide derivatives which offers great potential in preparing pyrrolidine compounds having a variety of substituents at the 2.5 positions and with control over stereochemistry. In addition, it is desirable to control the stereochemical outcome of the reaction by an appropriate choice of reaction conditions. In addition, there is a need for a new method of preparing cyclic secondary amine compounds and the corresponding nitroxide derivatives, which method uses commercially available and attractive raw materials and can be performed in a few steps with high selectivity and reproducibility.

The 1,3-dipolar cycloaddition with the compound represented by formula (2) is in certain embodiments carried out as a thermal cycloaddition. As such a suitable compound, for example, LiBr or AgOAc are used. Tertiary amines can, inter alia, be used as a suitable base. In general, Lewis acid / complexing metal salts can be used on imine-lone pair and / or R, and / or R3.
De onderhavige uitvinding heeft verder betrekking op 15 de nieuwe cyclische secundaire amineverbindingen weergegeven volgens conclusie 3.The present invention further relates to the new cyclic secondary amine compounds shown in claim 3.
Bovendien verschaft de onderhavige uitvinding een werkwijze ter bereiding van cyclische secundaire amineverbindingen 35 en de derivaten hiervan onder toepassing van in de handel verkrijgbareAlthough oxidized cyclic secondary amine compounds corresponding to the nitroxide derivative of formula 4 are known from international patent application WO 97/46593, it is not known from this international patent application by which method such compounds are prepared. As suitable groups for substituents R 1 - R 4 are mentioned in this international patent application: halogen atoms, such as chlorine or bromine, saturated or unsaturated linear, branched or cyclic hydrocarbon groups with 1 to 10 carbon atoms, such as alkyl, cycloalkyl or phenyl, ester - or alkoxyl group, phosphonate group, represented by -P (0) (0R) 2, wherein R is a saturated aliphatic radical with a carbon number of 1-3; R5 - R8 have, in addition to the same meaning as Rj - R4, the meaning of a hydrogen atom, a hydroxyl or acid group. Although in this international patent application a number of groups R 1 - R 8 30 correspond to the present groups R 1 - R 8, it should be clear that the choice with regard to Rt - R 4 according to the present invention is wider than can be assumed from the international patent application.
