Abstract
La presente descripción proporciona composiciones y métodos para expandir células T o linfocitos infiltrantes de tumores (TIL) in vitro. Las células alimentadoras K562 diseñadas para expresar una molécula coestimuladora (p. ej., ligando 41BB (41BBL)) y la interleucina 21 (IL21) o la interleucina 7 (IL7) se pueden usar en un paso de protocolo de expansión rápida (REP) para expandir las células T o TIL. Por lo tanto, en el presente documento se proporciona un cultivo que comprende TIL o células T y células alimentadoras K562 modificadas. Las células T pueden modificarse para expresar un receptor de antígeno quimérico (CAR) o un receptor de células T (TCR) o los TIL pueden modificarse para expresar IL15 unida a la membrana (mbIL15).
