Abstract

La presente invención se refiere a variantes de glucoamilasa que presentan una mayor actividad en almidón de partida en comparación con la glucoamilasa descrita como SEQ ID NO: 2, que comprende una o más modificaciones en el dominio catalítico y/o una o más modificaciones en el dominio de unión a almidón, seleccionadas de: a) al menos una, preferiblemente al menos dos, preferiblemente al menos tres, preferiblemente al menos cuatro de: V18M, T43K, N112L, T116R, A117Q, G120S, A271F, Y295W, Q318Y; y/o b) introducir al menos tres, preferiblemente al menos cuatro sustituciones seleccionadas del grupo: S458C, S458SCGG, S458SGGC, A493V, A518K, E520Q, N527M, S540K,R, S(G)546P, T(V)549W, N503R, N539R+ I541Y+ T543V+ A545S+ S546GCGV+ G547S+ S548T+ T549A. La presente invención también se refiere a polinucleótidos que codifican las variantes; a constructos de ácido nucleico, vectores y células hospedadoras que comprenden los polinucleótidos; y a métodos para usar las variantes.
