Abstract

La presente invención proporciona nuevas herramientas para controlar las modificaciones de post-traducción de polipéptidos recombinantes. Estas herramientas son péptidos de señal particulares que permiten la activación de polipéptidos recombinantes durante su síntesis en una célula huésped para compartimientos sub-celulares específicos y un diseño específico de dichos polipéptidos recombinantes dentro de dichos compartimientos sub-celulares. Estos péptidos de señal son SEC ID No. 1 a SEC ID No. 31 y se describen aquí. La presente invención se refiere, por lo tanto, también a un procedimiento para producir un polipéptido recombinante, en particular a un polipéptido modificado de manera de post-traducción, que comprende los pasos de transfectar o transformar una célula con al menos un vector de ácido nucleico que codifica una proteína recombinante, la cual es el polipéptido que antes fe modificado de manera de post-traducción o una proteína recombinante diferente a dicho polipéptido. La proteína recombinante comprendiendo una señal de péptido de acuerdo con la presente invención; desarrollar la célula transfectada; y cosechar el polipéptido modificado de manera post-traducción; en donde, cuando dicha proteína recombinante es diferente al polipéptido, el método también comprende un paso de transfectar dicha célula con por lo menos un vector que codifica dicho polipéptido. La presente invención permite ventajosamente, por ejemplo, incrementar el rendimiento de producción de un polipéptido recombinante, para evitar la inmunogenicidad de polipéptidos recombinantes y para obtener polipéptidos recombinantes terapéuticamente activos que la copia exacta de su contraparte natural. Esta invención se refiere particularmente al campo de reorientación de plantas hechas farmacéuticos (PMP).
