Abstract

La invencion se refiere a un metodo para modular la actividad de la poli(ADP-ribosa)polimerasa-1 (PARP-I) en un mamifero que comprende administrar a un mamifero una cantidad efectiva de un compuesto aromatico organico que tiene de 4 a aproximadamente 35 atomos de carbono, en donde el compuesto aromatico organico es capaz de enlazarse a la porcion de la arginina 34 localizada en el dedo de zinc 1 de la enzima PARP-I y en donde el compuesto aromatico organico tiene capacidades donantes de electrones de tal manera que su sistema de electrones p interactuara con la porcion de guanidinio cargada positivamente (cationica) del residuo especifico de la arginina 34 del dedo de zinc 1 de PARP-I y no contiene sustituyentes de benzamida o lactama. En particular, se describen benzopironas sustituidas e indoles sustituidos y sus composiciones farmaceuticas que contienen estos compuestos que modulan la actividad de PARP-I. La invencion tambien se dirige a la composicion de material, equipos y metodos para uso terapeutico y/o profilactico en el tratamiento de enfermedades y trastornos descritos en este documento, por medio de la administracion de cantidades efectivas de estos compuestos. Preferiblemente, las composiciones y metodos proporcionados en este documento inhiben la actividad de la PARP.
