Abstract
본 발명은 식물내 이종 유전자 발현을 위한 프로모터로서 유용한 분리된 비-암호화 뉴클레오티드 서열에 관한 것이다. 본 발명은 또한 분리된 뉴클레오티드 서열을 포함하는 벡터 및 식물 세포에 관한 것이다. 옥수수로부터의 AHAS 프로모터는 도입된 유전자를 높은 레벨로 여러 식물 조직에서 발현시키기 위해 사용된다. 옥수수의 als1 및 als2 유전자로부터의 프로모터를 클로닝하고 서열분석한 다음, 이들 유전자로부터의 프로모터 영역을 플라스미드 5'내 리포터 유전자 β-글루쿠로니다제(GUS)에 도입시킨다. 두 프로모터 단편은 XI그2 옥수수주로부터 기원한다. als1 프로모터 단편은 약 1400 염기쌍 길이인 반면, als2 프로모터 단편은 819 염기쌍을 포함한다.

제1도는 옥수수 XA17로부터의 AHAS 프로모터 서열 ALS1을 도시한다, 제2도는 옥수수 XA17로부터의 AHAS 프로모터 서열 ALS2을 도시한다. 제3도는 옥수수 XI12로부터의 AHAS 프로모터 서열 ALS2을 도시한다. 제4도는 pcD221B(als2-프로모터-GUS-ocs 터미네이터 플라스미드) 또는 pAC400(CaMV 35S-GUS-ocs 터미네이터 플라스미드)으로 형질전환한 후 블랙 멕시칸 스위트 옥수수 세포 또는 벼 현탁 세포의 원형 질체의 일시 분석으로부터 β-글루쿠로니다제 활성을 나타내는 막대 그래프이다. 제5도는 pCD221B(als2-프로모터-GUS-ocs 터미네이터) 또는 pAC400(CaMV 35S-GUS-ocs 터미네이터 )으로 형질전환 후 안정하게 형질전환된 블랙 멕시칸 스위트 세포주에서 β-글루쿠로니다제 활성을 나타내는 막대 그래프이다.

- 식물내 유전자 발현을 위한 AHAS 프로모터를 포함하는 분리된 뉴클레오티드 서열.
- 제1항에 있어서, 도1의 서열(서열 1)을 포함하는 뉴클레오티드 서열.
- 제1항에 있어서, 도2의 서열(서열 2)을 포함하는 뉴클레오티드 서열.
- 제1항에 있어서, 도3의 서열(서열 3)을 포함하는 뉴클레오티드 서열.
- 제1항 내지 4항 중 어느 한 항에 있어서, 식물이 단자엽 식물임을 특징으로 하는 뉴클레오티드 서열.
- 제5항에 있어서, 단자엽 식물이 옥수수임을 특징으로 하는 서열.
- 제1항 내지 5항 중 어느 한 항의 뉴클레오티드 서열을 포함하는 형질전환 벡터.
- 제7항의 벡터를 포함하는 식물 세포.
- 제1항 내지 5항중 어느 한 항의 뉴클레오티드 서열을 포함하는 성숙 식물.
- 제1항 내지 4항중 어느 한 항의 뉴클레오티드 서열의 제어하에 식물내에 이종 유전자를 발현시키는 것으로 이루어지는, 식물내 높은 레벨로 및 식물의 상이한 조직에서의 이종 유전자의 발현 방법.
