Abstract
본 발명은 대장균에서 발현된 유전자 재조합 닭 성장 호르몬의 정제 방법에 관한 것으로, 닭 성장 호르몬이 봉입체로 발현된 대장균을 파쇄하여 봉입체를 수득하고, 상기 봉입체를 세척하고, 강알칼리와 요소로 용해하고 산화시킨 후, 한외여과 및 음이온 교환 크로마토그래피함으로써 재조합 닭 성장 호르몬을 고순도로 효율적으로 정제할 수 있다.

Claims (6)
- 닭 성장 호르몬이 봉입체로 발현된 대장균을 파쇄하여 봉입체를 수득하고, 상기 봉입체를 세척하고, 강알칼리와 요소로 용해하고 산화시킨 후, 한외여과 및 음이온 교환 크로마토그래피하는 단계를 포함하는 대장균에서 발현된 닭 성장 호르몬의 정제 방법.
- 제1항에 있어서, 상기 강알칼리가 수산화나트륨인 것을 특징으로 하는 방법.
- 제2항에 있어서, 강알카리를 가하여 단백질 용액의 pH를 11이상으로 조정하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 요소의 농도가 4 내지 8M인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 한외여과가 글리신을 포함하는 완충용액으로 수행되는 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 음이온 교환 크로마토그래피에서 글리신을 포함하는 0.0 내지 0.2M 선형 농도 구배의 염화나트륨 완충용액으로 닭 성장 호르몬 단백질을 용출시키는 것을 특징으로 하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
