Abstract
본 발명은 적혈구 표면의 혈액형 항원 및 조직특이성 항원 주위에 메톡시폴리에틸렌글리콜(methoxy-polyethylene glycol)의 비독성 폴리머를 공유결합시키는 화학수정(chemical modification)을 통하여, 혈액형 항체 및 조직특이성 항체의 접근을 공간적으로 방해함으로써, 항원-항체 반응에 의한 응집현상이 억제된 적혈구 및 그의 제조방법에 관한 것이다. 화학수정된 적혈구는 사람 또는 동물 유래의 적혈구를 0.9%NaCl 식염수로 희석한 용액의 pH를 9 내지 10으로 유지하면서, 활성화된 메톡시폴리에틸렌글리콜을 첨가하여 전기 적혈구와 반응시키는 방법에 의해 제조된다. 본 발명의 방법으로 화학수정된 적혈구는 혈액형에 관계없이 사용가능하며 충분한 산소전달능력도 갖고 있어, 응급수혈 및 이식장기의 보관 등 적혈구가 사용되는 모든 의학분야에 이용될 수 있다. 또한, 본 발명의 화학수정 적혈구의 제조방법은 혈액형에 관계없이 모든 혈액형 항원에 적용할 수 있고, 매우 단순하고 경제적이어서 차후 대량생산을 위한 반응기의 설계에도 매우 유리하다.The present invention relates to a method for the preparation of blood type antibodies and tissue specific antibodies through chemical modification that covalently attaches non-toxic polymers of methoxy-polyethylene glycol around blood type antigens and tissue specific antigens on the surface of erythrocytes Thereby preventing aggregation due to an antigen-antibody reaction, and a method for producing the same.
