Claims (9)
- (a) 일반식(Ⅱ)의 치환된 인돌-3-아세트산을 리튬화하여 일반식(Ⅲ)의 신톤(Synthon)인 치환된 리듐 1,-디리티오인돌-3-아세테이트를 생성시키고 (b) 신톤(Ⅲ)을 일반식(Ⅳ)의 할로겐화물로 알킬화하여 일반식(Ⅴ)의 치환된 인돌-3-아세트산을 생성하고, (c) 일반식(Ⅴ)의 치환된 인돌-3-아세트산을 에스테르화하여 일반식(Ⅵ)의 상응하는 에스테르를 생성시키고, (d) 일반식(Ⅵ)의 에스테르를 환원시켜 일반식(Ⅶ)의 치환된-벤질트리프토폴을 생성시키고, (e) 일반식(Ⅶ)의 트리프토폴을 일반식(Ⅶ)의 3-옥소-2-알카노산 알킬 에스테르와 반응시켜 일반식(Ⅸ)의 알킬 에스테르의 이성체 A 및 B를 생성시키고, (f) 이성체 A를 알콜 용매로부터 재결정화하여 이성체 A를 이성체 B로부터 분리하고 이성체 A를 가수분해하여 일반식(Ⅰ)의 목적 화합물을 수득하는 단계로 이루어짐을 특징으로 하여 일반식(Ⅰ)의 화합물 및 약제학적으로 허용되는 이의 염을 제조하는 방법.상기식에서, R1은 C1내지 C4의 저급 알킬이고, R2는 수소, 4-할로겐, 2- 및 4-디할로겐, 3-트리풀루오로메틸 또는 4-메톡시이며, R3는 C1내지 C4의 저급 아킬이고, R4및 R5는 수소, C1내지 C6의 저급 알킬 또는 할로겐이며, X는 염소, 브롬 및 요오드로 이루어진 그룹으로부터 선택된 할로겐이다.
- 제 1항에 있어서, R1이 에틸이고, R2,R4및 R5가 수소인 방법.
- 제1항에 있어서, 단계(d)에서 이반식(Ⅵ)의 에스테르의 환원반응을 저급 지방족 알콜을 첨가하면서 THF중의 NaBH4로 수행하는 방법.
- 제1항에 있어서, 단계(e)에서 3-옥소-2-알카노산 알킬 에스테르가 메틸 프로피오닐 아세테이트이며, 반응을 삼불화붕소 에테레이트의 존재하에 수행하는 방법.
- 제1항에 있어서, 단계(e)에서의 일반식(Ⅶ)의 트리프토폴의 톨루엔 용액을 톨루엔중의 3-옥소-2-알카노산 알킬 에스테르와 삼불화붕소 에테레이트의 냉각된 혼합물에 가하는 방법.
- 제1항에 있어서, 단계(f)에서의 이성체 A를 이성체B로부터 분리하는 공정을 이소프로필 알콜로부터 이성체A를 결정화하여 수행하는 방법.
- 제1항에 있어서, 일반식(Ⅴ)의 치환된 인돌-3-아세트산을 NaBH4및 BF3-에테레이트로 환원시켜 일반식(Ⅶ)의 치환된-벤질트리프로토폴을 직접 생성시키는 방법.
- (a)일반식(ⅩⅠ)의 이사틴을 알킬화하여 일반식(ⅩⅢ)의 화합물을 생서시키고, (b) 일반식(ⅩⅢ)의 화합물을 환원시켜 일반식(Ⅶ)의 치환된-벤질트리프리토폴을 생성시키는 단계로 이루어짐을 특징으로 하여 일반식(Ⅰ)의 화합물 및 약제학적으로 허용되는 이의 염을 제조하는 방법.상기식에서, R1은 C1내지 C4의 저급 알킬이고, R2는 수소, 4-할로겐, 2- 및 4-디할로겐, 3-트리풀루오로메틸 또는 4-메톡시이며, R4및 R5는 수소, C1내지 C6의 저급 알킬 또는 할로겐이다,
- 제8항에 있어서, 일반식(ⅩⅢ)의 화합물을 NaBH4및 BF3-에테레이트로 환원시켜 일반식(Ⅶ)의 치환된-벤질트리프로토폴을 직접 생성시키는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
