Description

젤라틴 캡슐
본 발명은 개선된 제약학적 조성물, 특히 연질 젤라틴 캡슐에 넣어진 복용 단위형태의 제약학적 조성물에 관한 것이다.
연질 젤라틴 캡슐(이후로는 간단히 "캡슐"이라칭함)에 넣어진 복용 단위 형태의 제약학적 조성물이 널리 공지되었으며, 이 제약적인 조성물은 기본적으로 글리세린 같은 가소제와 함께 젤라틴을 함유하는 연질 젤라틴 외피로 캡슐화되며, 액체 부형제내에 분산되거나 분산되거나 용해된 하나 이상의 제약학적 활성 재료를 포함하는 "충전물"로 구성된다.
충전물에 사용되는 부형제중의 한 종류로는 약100-600의 분자량을 갖는 폴리에틸렌 글리콜 같은 액체 폴리에틸렌 글리콜이 있다. 테마제팜(Temazepam) 및 로메타제팜(Lormetazepam)같은 벤조 디아제핀 형태의 화합물인 혹종의 제약학적 활성성분은 연질 캡슐내 폴리에틸렌 글리콜로서 투여될때 생체내에서 증진된 효능을 나타내었다.
그러나, 부형제로서 액체 폴리에틸렌 글리콜을 사용할 경우, 그 재료가 흡습성이 있어 캡슐 외피로부터 물을 흡수하게되어 외피를 부서지기 쉬운 상태로 한다는 단점이 수반된다. 가소제가 외피로부터 충전물로 이동함으로써 이러한 취화(脆化)상태는 더욱 축진될 수 있다. 몇달이나 몇해에 걸쳐 일어날 수 있는 이러한 취화의 결과로서, 외피가 탄성을 잃게되어 캡슐의 취급 및 수송시 생기는 기계적 충격에 대한 내성을 잃게된다. 극단적인 경우, 캡슐은 취화 상태가 심하게되거나 또는 기계적 충격을 받아 파괴될 수 있다.
이러한 문제점은 액체 폴리에틸렌 글리콜 부형제를 함유하는 충전물을 갖는 캡슐을 쓸모없게 할 만큼 심각하지 않다. 수천 또는 수백만개의 이러한 캡슐은 파손됨없이 수송되고 취급된다. 그러나 문제는 캡슐이 수십, 수백, 심지어 수천개가 함께 포장되기 때문에 파괴되어, 노출되고, 이 캡슐이 동일 포장내에서 다른 캡슐을 훼손시켜서 결국 많은 수의 캡슐을 쓸모없게 한다는 것이다.
본 발명에 따라, 하나 이상의 소르비탄류와 소르비톨 및 기타 폴리올과의 혼합물을 캡슐 외피에 함침시키고, 동시에 글리세린, 소비톨 또는 프로필렌 글리콜을 캡슐 충전물의 액체 폴리에틸렌 글리콜 부형제의 함침시키므로해서 취화정도가 감소될 수 있음이 발견되었다.
본 발명은 액체 폴리에틸렌 글리콜에 용해되거나 현탁되며 연질 젤라틴 캡슐 외피로 캡슐화되는 하나 이상의 제약학적 활성 재료를 포함하는 제약학적 복용단위 형태를 제공하는데, 여기서 캡슐 외피는 소르비톨과 한개 이상의 소르비판류와의 혼합물을 함유하는 취화 방지용 조성물, 가소제, 및 젤라틴을 포함한다.
특징적으로, 취화방지용 조성물은 25-45중량%의 소르비톨 및 20-30중량%의 소르비탄류(중요한 소르비탄 성분은 1.4-소르비탄)와 함께 물(13-20중량%) 및 0-6중량%의 만니톨과 같은 다가 알코올류를 포함한다.
바람직하게는 폴리에틸렌 글리콜은 또한 글리세린, 소르비톨, 또는 프로필렌 글리콜을 포함한다. 이러한 경우에, 액체 폴리에틸렌-글리콜 충전물에 용해된 글리세린, 소르비톨, 프로필렌 글리콜의 양은 1-20중량%, 바람직하게는 2-12중량%, 더욱 바람직하게는 3-8중량%이다.
외피내의 소르비톨/소르비탄류 혼합물의 양은 4-25중량% 바람직하게는 6-20중량% 더욱 바람직하게는 9-15중량%이다.
상기한 바와같이 소르비톨/소르비탄류 혼합물이 기타 다가 알코올을 함유할 수 있는데 이러한 경우, 소르비톨/소르비탄류/다가 알코올 혼합물중의 소르비톨과 소르비탄류의전체량은 45-75중량%가 적당하며 바람직하기는 55-66중량%이다. 기타 적당한 다가 알코올은 수소첨가된 당류이다.
전체 혼합물은 6중량%이하의 만니톨을 함유하는 것이 적당하며, 바람직하게는 1-4중량%, 더욱 바람직하게는 2-3중량%의 만니톨을 함유한다.
캡슐 외피에 소르비톨/소르비탄류/다가 알코올 혼합물을 함침시키는데 적당한 재료는 글루코스 시럽을 가수분해하고 부분적으로 수소 첨가함으로써 유도된 다가 알코올의 농축 수용액이다. 통상적으로 입수가능한 적당한 재료의 예는 "ANIDRISORB 35/70"이란 상표명으로 시판되는 것이다. 이 생성물은 하기하는 바와같은 분석 특징을 가진다.

소르비톨/소르비탄류 혼합물에 부가하여, 캡슐외피의 젤라틴 재료는 또한 글리세린, 프로필렌 글리콜 또는 소르비톨(소르비톨/소르비탄류 혼합물에 이 소르비톨을 부가)과 같은 가소제를 함유할 수 있으며 이것은 10-40중량%, 바람직하게는 25-45중량%의 양으로 존재한다. 또한 외피 재료는 착색제(안료 도는 염료) 및 산화제 또는 방부제(예컨대 포타슴 소르베이트 및 에틸, 메틸, 프로필 파라벤류)와 같은 기타 통상적인 성분을 함유할 수 있다.
본 발명 조성물의 제약학적 활성 성분은 경구 투여할 수 있는 여러가지 제약학적 재료일 수 있다. 재료가 액체 폴리에틸렌 글리콜 부형제내에서 불충분하게 용해될 경우, 충전물은 물 또는 에탄올과 같은 공-용매 또는 분산제를 함유할 수 있다. 그러나, 바람직하게 본 제약학적 재료는 상기에 언급한 벤조디아제핀류 형태의 화합물같은 액체 폴리에틸렌 글리콜내에 가용성인 재료이다.
본 발명의 이해를 돕기위해 다음 실시예가 상술된다.
하기의 충전 조성물 및 외피 조성물을 갖는 연질 젤라틴 캡슐을 제조하였다.

두개 배취(batch)의 캡슐을 제조하였는데, 첫번째 베취(실시예 5)는 캡슐당 10mg의 레마제팜을 함유하며 두번째 배취(실시예 6)는 캡슐당 20mg의 테마제팜을 함유하였다.
비교할 목적으로, 다음의 것(중량 %)으로 구성된 통상적인 외피 제조용 조성물을 사용하지만 충전물은 동일한 것을 사용하여 젤라틴 캡슐을 제조하였다.

다시 두개 배취의 캡슐을 제조하였는데 첫번째 배취(비교 실시예 1)는 캡슐당 10mg의 테마제팜을 함유하며 두번째 배취(비교 실시예 2)는 캡슐달 20mg의 테마제팜을 함유하였다.
캡슐 배취를 수개월간 20℃로 저장하고, 해리스 경도 실험기를 사용하여 배취 샘플의 경도를 간격을 두어 시허했다. 기본적으로, 상기 기기는 캡슐을 자동적으로 상승하는 플래트폼(platform)과 로드셀(load cell)에 부착된 플런저(plunger) 사이에서 20초동안 압축한다. 그러므로, 캡슐을 시험하기 위해서, 캡슐을 플래트폼에 수평으로 놓아 플런저와 플래트폼 둘다에 접촉되도록한다. 시험하는 동안, 플래트폼은 자동적으로 상승하며 로드 셀 표시기는 압축력에 대한 캡슐의 저항 값을 나타낸다. 20초 동안의 시험이 끝난뒤 표시된 값은 시험한 캡슐의 경도를 나타낸다.
실제로, 경험상의 문제로서, (상기 기술된 바와같이 측정된) 12뉴톤보다 큰 경도에서 캡슐이 취화에의해 파괴될 수 있음을 알았다.

이같은 결과로 부터 본 발명의 캡슐은 비교 캡슐보다 일반적으로 낮은 초기경도를 지니며, 캡슐의 경도가 12 또는 이보다 작을 때를 참작하면 본 발명 캡슐의 유효저장수명이 통상적인 비교 캡슐의 유효저장 수명보다 적어도 2배 이상 긴것을 알 수 있다.

두개 배취의 캡슐을 제조하였는데 한개는(실시예 7)캡슐당 10mg 테마제팜을 함유하며 다른 하나는(실시예 8) 캡슐당 20mg의 테마제팜을 함유하였다.
배취를 세개의 군으로 나누어 각기 군을 20℃, 30℃, 40℃의 온도에서 저장하였다. 캡슐정도를 실시예 5 및 6에서 기술된 바와같이 측정하여 다음 표2에 표시된 결과를 얻었다.

Claims (5)

  1. 소르비톨 25-45중량% 및 한개 이상의 소르비탄류 20-3-중량%와 함께 물 및 기타 다가 알코올류를 포함하는 취화(脆化) 방지용 조성물 및 젤라틴, 가소제를 함유하는 연질 젤라틴 캡슐내에서, 캡슐화되며 액체 폴리에틸렌 글리콜내에 용애 또는 현탁되는 한개 이상의 제약학적 활성재료를 함유하는 제약학적 복용단위 형태.
  2. 제1항에 있어서, 취화방지용 조성물은 만니톨을 6중량%까지를 함유하는 복용단위 형태.
  3. 제1항에 있어서, 외피는 소르비톨 소르비탄 혼합물을 4-25중량% 함유하는 복용단위 형태.
  4. 제1항에 있어서, 폴리에틸렌 글리콜은 또한 글리세린, 소르비톨 또는 프로필렌 글리콜을 함유하는 복용단위 형태.
  5. 제4항에 있어서, 글리세린, 소르비톨 또는 프로필렌 글리콜은 1-20중량%로 폴리에틸렌 글리콜내에 존재하는 복용단위 형태.
