Description
The present invention relates to a novel peptide for detecting interleukin 1 beta and its use.
Interleukin 1beta, a type of cytokine encoded by the IL 1B gene, is a pro-inflammatory protein that plays an important role in innate host defense by initiating the acute phase response to infection and damage. Interleukin 1 beta is expressed by monocytes, macrophages, and dendritic cells and activates receptors in a paracrine manner, leading to the expression of proinflammatory gene products. Interleukin 1 beta is most often secreted in its active form, but under some conditions it may be secreted as an uncleaved, inactive protein. Activation of the inflammasome, a multi-protein complex that responds to pathogens, stress conditions, and danger signals, converts the inactive proInterleukin 1beta to interleukin 1beta, which becomes activated and participates in the inflammatory response. . As a key mediator of the inflammatory response, it is essential for host response and resistance to pathogens. It is also known as a complex cytokine network because it plays a dual role as an inhibitor of gastric acid secretion while aggravating damage in chronic diseases and acute tissue damage. It has been found to be related to various inflammatory side effects, including Crohn's disease, inflammatory bowel disease such as ulcerative colitis, and Alzheimer's disease, and rapid and accurate detection of interleukin 1 beta is recognized as an important factor in clinical care. It is becoming.

Hereinafter, the present invention will be described in detail.
The present invention relates to a peptide capable of specifically detecting interleukin 1 beta, and provides a peptide for detecting interleukin 1 beta comprising part or the entire amino acid sequence shown in SEQ ID NO: 1 below. .
YVHHYSTGQMFN(서열번호 1).YVHHYSTGQMFN (SEQ ID NO: 1).
The amino acid sequence of the peptide according to the present invention, represented by SEQ ID NO: 1, is a linear model with 12 random amino acids at the end of the gene that produces pIII, a type of coat protein, in the genome of M13 bacteriophage. Ph.D.-12 (Ph.D.-12), a bacteriophage random peptide library designed to express hundreds of millions of different peptides obtained by artificially inserting gene sequences to express peptides and then infecting E.coli. The sequence that binds to interleukin 1 beta was discovered using New England BioLab.
The M13 bacteriophage is a nano-sized material that infects only E. coli of a specific strain, and there has been no report to date on the possibility of it mutating and infecting the human body. In particular, it was approved by the FDA in 2006 and is being used as an additive to prevent bacterial contamination in instant foods. It is attracting attention not only as an alternative that can solve the problem of antibiotic resistance, but also as a biocompatible nanomaterial that is harmless to the human body. In addition, when using the bacterial bacteriophage, it can be easily and quickly fixed to the surface of the transducer through a chemical linker using the robust and heat-stable characteristics of the bacteriophage, making it easy to bind to biomolecules, which is a great advantage in sensor development. In addition, the manufacturing process is simple, contributing to improved productivity and stability, and has the advantage of being able to utilize various genetic engineering and chemical modifications.
The peptide according to the present invention is a low-molecular-weight peptide and has the advantage of being three-dimensionally stable due to its small size, easily passing through mucous membranes, and being able to recognize molecular targets even deep in the tissue. In addition, the small molecule peptide according to the present invention is relatively simple to mass produce compared to antibodies and has little toxicity. In addition, the low-molecular-weight peptide according to the present invention has the advantage of high binding affinity to the target substance and does not undergo denaturation even during heat/chemical treatment. Additionally, because the molecule size is small, it can be used as a fusion protein by attaching it to another protein.
대표적인 방법으로는 액체 또는 고체상 합성, 단편 응축, F-MOC(9-fluorenylmethoxycarbonyl) 또는 T-BOC(tert-butyloxycarbonyl) 화학법이 포함되나, 반드시 이에 제한되는 것은 아니다. Peptides according to the present invention can be prepared by chemical synthesis known in the art. Representative methods include, but are not necessarily limited to, liquid or solid phase synthesis, fragment condensation, and 9-fluorenylmethoxycarbonyl (F-MOC) or tert-butyloxycarbonyl (T-BOC) chemistry. Additionally, the peptide of the present invention can be produced by genetic engineering methods. First, a DNA sequence encoding the peptide is constructed according to a conventional method. DNA sequences can be constructed by PCR amplification using appropriate primers. Alternatively, DNA sequences may be synthesized by standard methods known in the art, such as using automated DNA synthesizers (e.g., those sold by Biosearch or Applied Biosystems). The constructed DNA sequence is a vector containing one or more expression control sequences (e.g., promoter, enhancer, etc.) that are operably linked to the DNA sequence to control the expression of the DNA sequence. and transform the host cell with the recombinant expression vector formed therefrom. The resulting transformant is cultured under appropriate media and conditions to express the DNA sequence, and a substantially pure peptide encoded by the DNA sequence is recovered from the culture. It is clear to those skilled in the art that biological functional equivalents that can be included in the range of the peptide for detecting interleukin 1 beta of the present invention will be limited to those that include mutations in the amino acid sequence that exhibit biological activity equivalent to the peptide of the present invention.
These amino acid mutations are made based on the relative similarity of amino acid side chain substitutions, such as hydrophobicity, hydrophilicity, charge, size, etc. Analysis of the size, shape and type of amino acid side chain substitutions shows that arginine, lysine and histidine are all positively charged residues; Alanine, glycine and serine have similar sizes; It can be seen that phenylalanine, tryptophan and tyrosine have similar shapes. Therefore, based on these considerations, arginine, lysine and histidine; Alanine, glycine and serine; And phenylalanine, tryptophan, and tyrosine can be said to be biologically equivalent in function.
Each amino acid is assigned a hydrophobicity index based on its hydrophobicity and charge: isoleucine (+4.5); Valine (+4.2); leucine (+3.8); phenylalanine (+2.8); Cysteine/Cysteine (+2.5); Methionine (+1.9); Alanine (+1.8); Glycine (-0.4); threonine (-0.7); Serine (-0.8); Tryptophan (-0.9); Tyrosine (-1.3); Proline (-1.6); histidine (-3.2); glutamate (-3.5); Glutamine (-3.5); Aspartate (-3.5); Asparagine (-3.5); Lysine (-3.9); and arginine (-4.5).
The hydrophobic amino acid index is very important in imparting the interactive biological function of peptides. It is a known fact that similar biological activity can be maintained only when substituted with an amino acid having a similar hydrophobic index. When introducing a mutation with reference to the hydrophobicity index, substitution is made between amino acids showing a difference in hydrophobicity index, preferably within ±2, more preferably within ±1, and even more preferably within ±0.5.
Meanwhile, it is also well known that substitution between amino acids with similar hydrophilicity values results in peptides with equal biological activity. As disclosed in U.S. Patent No. 4,554,101, the following hydrophilicity values are assigned to each amino acid residue: arginine (+3.0); Lysine (+3.0); Asphaltate (+3.0±1); glutamate (+3.0±1); serine (+0.3); Asparagine (+0.2); Glutamine (+0.2); Glycine (0); threonine (-0.4); Proline (-0.5±1); Alanine (-0.5); histidine (-0.5); Cysteine (-1.0); Methionine (-1.3); Valine (-1.5); leucine (-1.8); isoleucine (-1.8); Tyrosine (-2.3); Phenylalanine (-2.5); Tryptophan (-3.4).
When introducing a mutation with reference to the hydrophilicity value, substitution is made between amino acids that show a difference in hydrophilicity value, preferably within ±2, more preferably within ±1, and even more preferably within ±0.5.
(H. Neurath, R.L.Hill, The Proteins, Academic Press, New York, 1979). 가장 통상적으로 일어나는 교환은 아미노산 잔기 Ala/Ser, Val/Ile, Asp/Glu, Thr/Ser, Ala/Gly, Ala/Thr, Ser/Asn, Ala/Val, Ser/Gly, Tyr/Phe, Ala/Pro, Lys/Arg, Asp/Asn, Leu/Ile, Leu/Val, Ala/Glu, Asp/Gly 간의 교환이다.Amino acid exchanges in peptides that do not overall alter the activity of the molecule are known in the art (H. 실시예 1. Discovery of peptides specific for interleukin 1 beta
(Cat. No. ILB-H4110) was purchased from Acro, biotinylated, and stored in an ultra-low temperature refrigerator until use.
In addition, Ph.D.-12 (New England BioLab) was used as a bacteriophage random peptide library (phage display peptide library). This is done by artificially inserting a gene sequence to express a linear peptide with 12 random amino acids at the end of the gene that produces pIII, a type of coat protein, in the genome of M13 bacteriophage, and then inserting the gene sequence into E. coli. It is composed of recombinant bacteriophages that express hundreds of millions of different peptides obtained by infecting .
(Streptavidin High Binding Capacity Coated 96-Well Plates (Thermo scientific, Cat. no. 15501))을 사용했다. A total of four pannings were performed using Ph.D.-12, and panning was performed using streptavidin-coated plates (Streptavidin High Binding Capacity Coated 96-Well Plates (Thermo scientific, Cat. no. 15501)) was used. Panning was performed as follows.
First, 1 μL of the M13 library (concentration: 1.0 Then, it was stirred at 100 rpm for 1 hour at room temperature. When the agitation of the target protein, biotinylated interleukin 1 beta and M13 library mixture (phage-protein complex), was completed, the streptavidin-coated plate was repeated three times using 0.1 M phosphate buffered saline (PBS). Washed. Afterwards, 100 μL of the biotinylated interleukin 1 beta and M13 library mixture was added to the plate and reacted at 110 rpm for 10 minutes. Afterwards, 1 μL of biotin (concentration: 0.1 mM) was added and blocked for 5 minutes, and then washed 10 times repeatedly using 200 μL of PBST (0.1% Tween 20 added to 0.1 M PBS). did. Finally, the bacteriophage that specifically binds to interleukin 1 beta was eluted in 100 μL of 0.2 M Glycine-HCl (pH 2.2) 1 mg/mL BSA (Bovine Serum Albumin) solution, and neutralization was performed. 15 μL of Tris-HCl (pH 9.0) was added.

An amplification reaction was performed to use the four bacteriophage candidates selected through Example 1 in an ELISA (enzymed-linked immunosorbent assay) experiment. Each bacteriophage stock was placed in LB medium together with pre-cultured E. coli ER2738 and cultured at 37°C at 210 rpm for 4-5 hours. This was centrifuged to recover the supernatant, 20% PEG (polyethylene glycol)/2.5M NaCl was added, left in a 4°C refrigerator for 15 hours, and then centrifuged again. Then, the precipitate was obtained, dissolved in PBS, and then subjected to bacteriophage titration, and the concentration of each bacteriophage (PFU/mL: Plaque forming units/mL) was calculated using Equation 2 below:
2][Equation 2]
Bacteriophage concentration (PFU/mL) = Plaque number x dilution factor x inoculum converted to mL
그 후 PBST로 6회 반복하여 세척한 후, 항-M13-HRP(Anti-M13-HRP) 항체와 ABTS(2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid))를 이용하여 405nm에서 흡광도(OD)값을 측정하였다. Afterwards, the upper layer solution was removed, and 200 μL of blocking solution (5% BSA in NaHCO 3 , pH 8.6) was added and reacted at 4°C for 1 hour and 30 minutes. Afterwards, after repeated washing with PBST 6 times, each selected bacteriophage was treated at a concentration of 10 12 PFU/mL and stirred at a speed of 100 rpm for 1 hour at room temperature. After washing with PBST repeatedly 6 times, using anti-M13-HRP antibody and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) The absorbance (OD) value was measured at 405 nm.

Example 5. Confirmation of immobilization on gold chip
It was confirmed whether the selected IL-1beta 12mer 4-19 could be immobilized on a gold working electrode using CHI 660E (Electrochemical Workstation, Ch Instruments Inc, USA) equipment. The analysis was conducted at room temperature, and disposable gold chips were used.
For this purpose, cyclic voltammetry (CV), a type of potentiometric electrochemical measurement, was performed. The CV refers to the thermodynamics and kinetics of the electrochemical reaction of materials that occur near the electrode surface due to the change in current value when a voltage is injected at a high speed to the working electrode in the presence of a chemical crystal capable of oxidation/reduction reaction in the electrolyte. This is an analysis method that allows you to check parameters.
First, to polish the gold chip, it was immersed in ethanol for 5 minutes and then washed with distilled water. Accordingly, 250 μL of Piranha solution prepared by mixing sulfuric acid (H 2 SO 4 ) and hydrogen peroxide (H 2 O 2 ) in a 7:3 ratio was applied, left at room temperature for 8 minutes, and then washed again for the third time. Washed with distilled water.
Afterwards, the gold chip was immersed in 11-MUA (11-Mercaptoundecanoic acid) (concentration: 1 mM), which has a thiol group (-SH) at one end, and stirred at room temperature for 14 hours to fix the MUA on the gold chip. After that, it was thoroughly washed with tertiary distilled water and fixed in a gold chip dedicated cell. Afterwards, 75 μL of EDC (1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride) and NHS (N-Hydroxysuccinimide) (concentration ratio: 800mM:200mM) were added to the gold chip on which MUA was immobilized and incubated for 1 hour. It was stirred at room temperature for a while. At this time, the carboxyl group of MUA and EDC react first to form an intermediate (O acylisourea intermediate), and then NHS reacts to form NHS ester. The gold chip with the desired functional group activated as described above was washed with distilled water and 75 μL of IL-1beta 12mer 4-19 bacteriophage (concentration: 1.0×10 12 PFU/mL) was added and stirred at room temperature for 1 hour. Afterwards, the gold chip was washed with tertiary distilled water and an electrolyte solution (K 4 [Fe(CN) 6 ] 4- 2.5mM+K 3 [Fe(CN) 6 ] 4- 2.5mM in 1M KNO 3 ) was added. Afterwards, the change in current according to the scan speed was confirmed through CV with the counter electrode and reference electrode to determine whether IL-1beta 12mer 4-19 according to the present invention was immobilized on the gold chip activated with a functional group. Confirmed.
The surface adsorption rate was checked to confirm whether IL-1beta 12mer4-19 according to the present invention was well immobilized on the surface of the gold chip treated with MUA-EDC/NHS as described above. This was confirmed by varying the scan speed from 50 to 40 mV/s and measuring the change in peak current according to speed (see left graph in Figure 4). In addition, the anode (ipa) and cathode (ipc) peak current densities according to the scan speed were also confirmed (see the graph on the right side of Figure 4.) The anode (ipa) and cathode (ipc) peak current densities according to the scan speed are proportional to the potential sweep speed. It was observed that
