세균에 대한 항균력 테스트에서는 미국약전(USP)에서 지정한 세균 Escherichia coli ATCC 8739, Pseudomonas aeruginosa ATCC 9027 및 Staphylococcus aureus ATCC 6538를 이용하였다. 세균은 TSB 배지에 2회 계대 배양한 후 실험에 사용하였다.In the antibacterial activity test, bacteria Escherichia coli ATCC 8739, Pseudomonas aeruginosa ATCC 9027, and Staphylococcus aureus ATCC 6538 designated by the United States Pharmacopoeia (USP) were used. 0.8% NaCl 수용액에 항균용 조성물로서 루테린 단독 또는 루테린과 방부 화합물을 각각의 농도로 첨가하였고, 이때 최종 루테린 농도는 0.5 내지 1.5 mM이었다. 이후 세균을 107 cfu/ml가 되도록 접종하여 37℃에서 3일간 보관 뒤, TSA 배지에 100 uL를 도말하였다. 도말한 시료는 37℃에서 배양하여 균락(colony) 형성 유무를 확인하였다. 그 결과는 하기 표 2에 나타내었다.Reuterin alone or reuterin and a preservative compound were added at respective concentrations as an antibacterial composition to a 0.8% NaCl aqueous solution, and the final reuterin concentration was 0.5 to 1.5 mM. 상기 표 2를 참조하면, 각 방부 화합물을 단독 사용한 경우에는 3종의 세균에 대해 항균 활성이 없었으나, 루테린을 첨가할 경우 항세균 활성이 나타나며, 루테린을 단독 사용한 경우에 비해 저농도로 우수한 항세균 활성을 발휘하는 것을 확인하였다.Referring to Table 2 above, when each preservative compound was used alone, there was no antibacterial activity against the three types of bacteria, but when reuterin was added, antibacterial activity was shown, and the antibacterial activity was excellent at a low concentration compared to when reuterin was used alone. 2-1. 화장료 조성물의 제조2-1. Preparation of cosmetic compositions
화장료 조성물에서의 방부 화합물 및/또는 루테린의 항균력을 확인하기 위해 하기 표 3 및 4와 같이 로션 및 시트마스크 제형의 화장료 조성물을 제조하였다.To confirm the antibacterial activity of the preservative compound and/or reuterin in the cosmetic composition, cosmetic compositions in lotion and sheet mask formulations were prepared as shown in Tables 3 and 4 below.
2-2. 화장료 제형에서의 항균력 평가2-2. Evaluation of antibacterial activity in cosmetic formulations
화장료 조성물에서의 항균력 평가를 위해 상기 제형들을 이용하여 테스트를 진행하였다. 항균력 테스트에서는 미국약전(USP)에서 지정한 세균 및 진균을 이용하였다 (Escherichia coli ATCC 8739, Pseudomonas aeruginosa ATCC 9027, Staphylococcus aureus ATCC 6538, Candida albicans ATCC 10231 및 Aspergillus niger ATCC 16404)를 이용하였다. 세균과 C. albicans는 각각 TSB와 SDB 배지에 2회 계대 배양한 후 실험에 사용하였고, A. niger는 SDA 배지에 도말하여 수득한 포자를 실험에 사용하였다.To evaluate the antibacterial activity of cosmetic compositions, tests were conducted using the above formulations. 각각의 화장료 조성물에 세균의 경우 107 cfu/ml가, 진균의 경우 105 cfu/ml가 되도록 접종한 후 각각 37℃와 30℃에서 3일간 보관하였다. 100 uL의 시료를 900 uL의 Letheen 배지에 혼합한 뒤 세균의 경우 TSA에, 진균의 경우 SDA에 각각 도말하였다. 도말한 시료는 각각 37℃와 30℃에서 3일간 배양하여 균락(colony) 형성 유무를 확인하였다. 그 결과는 하기 표 5에 나타내었다.Each cosmetic composition was inoculated to a concentration of 10 7 cfu/ml for bacteria and 10 5 cfu/ml for fungi and then stored at 37°C and 30°C for 3 days, respectively. 100 uL of sample was mixed with 900 uL of Letheen medium and then plated on TSA for bacteria and SDA for fungi. 2-3. 화장료 제형에서의 항균력 유지 평가2-3. Evaluation of antibacterial activity maintenance in cosmetic formulations
화장료 제형에서의 항균력이 유지되는지 확인하기 위해 '2-2. 화장료 제형에서의 항균력 평가'에서 사용한 세균 또는 진균 접종 시료 (화장료 2 ~ 5 및 7 ~ 10)를 이용하여 각각의 균주를 누적 접종하여 항균력이 유지되는지를 확인하였다. 세균의 경우 3종의 균 (E. coli, P. aeruginosa 및 S. aureus)을 혼합하여 107 CFU/ml이 되도록 접종하였고, 진균의 경우 C. albicans 및 A. niger를 각각 105 CFU/ml이 되도록 접종하였다. 각 균주의 접종은 7일 마다 동일한 시료에 접종을 하였으며, 접종이 완료된 시료는 매 접종 3일차에 100 uL의 시료를 900 uL의 Letheen 배지에 혼합한 뒤 TSA와 SDA에 각각 도말하여 균락 형성 유무를 확인하였다. 그 결과는 하기 표 6에 나타내었다.To check whether the antibacterial activity of the cosmetic formulation is maintained, '2-2.
