RNA를 추출할 때, genomic DNA의 오염을 방지하기 위하여 Quiagen 회사의 DNase 처리하는 과정도 추가하였다. 전체 RNA는 Bio-Rad사의 nanodrop을 이용하여 정량한 후 1.5 μg의 RNA를 주형으로 사용하여 Thermo scientific 회사의 random hexamer primer와 M-MLV 역전사효소를 이용하여 cDNA를 만들었다. 역전사반응은 Bio-Rad사의 thermal cycler 기기를 이용하여 수행하였다. Quantitative real-time PCR 분석을 위하여 Biosystems사의 Primer Express Software (version 3.0)를 이용하여 primer를 디자인하였다. OmpA 및 16S rRNA의 PCR 수행을 위한 프라이머 서열은 다음과 같다. OmpA (sense primer: 5′- TTG CAC TTG CTA CTA TGC TTG TTG-3′, anti-sense primer: 5′-TGG CTG TCT TGG AAA GTG TAA CC-3′); 16S rRNA (sense primer: 5′-ACT CCT ACG GGA GGC AGC AGT-3′, anti-sense primer: 5′-TAT TAC CGC GGC TGC TGG C-3′). 실시예 5: 균막 형성 억제능Example 5: Biofilm formation inhibitory ability
OmpA 단백질은 병원성 세균의 균막 형성, 세포부착 및 침입, 세포사 유도 등 다양한 기능을 하는 단백질이다. 따라서 OmpA 단백질이 제대로 발현되지 않는다면 이러한 병원성도 줄어들어야 항병원성 약물로 활용할 수 있다. 따라서 본 발명자들은 OmpA의 프로모터를 저해하는 본 발명의 223604와 195925 화합물이 세균의 균막 형성을 억제하는지 여부를 평가하였다.OmpA protein is a protein that performs various functions such as formation of biofilm of pathogenic bacteria, cell adhesion and invasion, and induction of cell death. 구체적으로 Luria-Bertani(LB) broth에서 하룻밤 배양한 reporter strain을 파장 600 nm에서 흡광도(optical density, OD)에서 2.0이 되도록 조정한 후 LB broth에 200배 희석하였다. 희석된 균액 297 μl를 5 ml의 polystyrene tube에 넣은 후 화합물을 최종농도가 2.5 μM이 되도록 3 μl 첨가하였다. 음성 대조군으로는 화합물의 용매로 사용된 DMSO 3 μl를 첨가하였다. 균액은 30oC에서 24시간 정치배양하였다. 배양이 끝난 균액은 600 nm 파장에서 흡광도를 측정하여 부유 세균의 성장을 확인한 다음, 부유세균은 제거하고 튜브를 300 μl의 PBS로 1회 세척하였다. 균막을 형성한 세균은 0.1%의 crystal violet 용액으로 실온에서 15분간 염색하고 3차 증류수를 이용하여 2회 세척한 후, 95% 에탄올로 균막형성 세균을 튜브로부터 모두 추출한 후 570 nm의 파장에서 흡광도를 측정하여 600 nm에서 측정한 흡광도로 나누어서 균막 형성능을 측정하였다.
