Alginate hydrogel is a polysaccharide polymer electrolyte contained in brown algae, which is based on alginate as a support, and is a material that forms a gel by electrostatic attraction when polyvalent ions such as calcium ions (Ca 2+ ) are added. Because it is a natural material that is harmless to the human body, it is also used as a stabilizer for viscous foods such as dental supports, ice cream, and syrup.
Using these properties, a manufacturing method of mixing alginate and a drug for the purpose of delivering medical drugs is being used, and recently, the market for using a substance mixed with alginate and cells for the purpose of delivering therapeutic cells to the inside of the human body. there is a need
Methods for making such alginate beads include a spraying method using a syringe and a spraying device, an ultrasonic mixing method, and an emulsion forming method. The conventional method using a microfluidic chip is characterized in that the alginate bead is manufactured by making a micro mold using a semiconductor process and molding it with a PDMS (polydimethylsiloxane) material.
However, such a conventional method for producing alginate beads can be mainly applied for research purposes and has methodological limitations that cannot mass-produce chips. In addition, since the PDMS material is hydrophobic, there is a problem that the microchannels made to make beads easily stick to and clog the beads, oil components, and air bubbles.
Accordingly, there is a need to develop a manufacturing method capable of solving the problem of microchannel clogging and fluid flow due to frictional force, and mass production of alginate beads.
An object of the present invention is to facilitate the formation of beads by making the channel cross-section into a cylindrical structure rather than the rectangular structure that the conventional PDMS microfluidic chip has, and to minimize frictional force generated while the fluid flows and realize a stable fluid flow, An object of the present invention is to provide a microfluidic device for producing alginate beads encapsulated in cells, in which cells encapsulated alginate beads can be mass-produced in a uniform size through hydrophilic treatment, and the produced beads can be stably discharged.

According to Example 1, while preparing the cell-encapsulated alginate beads, the fluid velocity of the second channel was fixed at 20 μl/min, and the fluid velocity of the first channel was changed from 50 to 300 μl/min, the prepared alginate beads size change was observed. At this time, the size of the outlet at the end of the second inlet was set to 20 μm, 30 μm, and 40 μm, and the change in the bead size was compared. Accordingly, the manufacturing process of the alginate beads and a photograph thereof are shown in FIG. 8, and a graph of changes in channel flow rate and alginate bead size is shown in FIG.
According to this, it can be confirmed that alginate beads of uniform size are produced, and it can be confirmed that the faster the fluid velocity of the first channel, that is, the fluid velocity of oleic acid, the smaller the size of the beads. P100 component is 40-49% isopropanol (Isopropanol), water (water), 1% isopropyl alcohol (Isopropyl Alcohol) (CAS RN: 67-63-0).
First, the P100 was prepared so that the microfluidic device could be completely contained, and after washing, the dried microfluidic device was completely immersed in the prepared P100 for 3 to 4 seconds and then taken out. Thereafter, nitrogen gas (N 2 ) was lightly sprayed to prevent the dry solution from forming thickly in the channel of the microfluidic device, and the result was completely dried. Accordingly, the contact angle of the surface of the hydrophilic surface-treated microfluidic device with respect to water was measured, and the results are shown in FIG. 10 . According to this, it was confirmed that the surface of the microfluidic device was successfully hydrophilized.

In addition, the preparation of the cell-encapsulated alginate beads of Example 1 was tested three times and after 10 days, the number of cells in 30 beads (spheroid), the number of cells per bead, and the viability (%) of cells are shown in the table below. 1 is shown.
According to this, it was confirmed that the growth of the cells was made after the cell encapsulation, and it was confirmed that the cell viability was also shown at a high level of 95% or more.
