Description
The present invention relates to compounds for treating or preventing osteoarthritis. The compounds alleviate joint pain in a mouse model and inhibit and attenuate the occurrence of post-traumatic osteoarthritis. In addition, the present invention provides a pharmaceutical composition comprising a compound for treating or preventing osteoarthritis. The present invention further provides a method for treating or preventing osteoarthritis by administering the pharmaceutical composition to a patient in need of treatment or prevention of osteoarthritis.
Osteoarthritis (OA) is the most common form of arthritis with a prevalence of about 60% in men and about 70% in women at age 65 or older. Current treatment options for osteoarthritis are limited. These include symptomatic treatment with simple analgesics, nonsteroidal anti-inflammatory drugs (NSAIDs) or intraarticular (IA) injection glucocorticoids and hyaluronic acid (HA) preparations. Non-pharmacological measures range from physical exercise and weight loss to joint lavage fluid and ultimately surgical joint replacement.
In the treatment of osteoarthritis, there is a great unmet need for disease-modifying osteoarthritis drugs that are still unavailable, and for efficacious pain treatments with long lasting effects. Moreover, due to the chronic nature of osteoarthritis disease, which often requires extended duration of treatment, there is a significant unmet need for osteoarthritis treatments that do not have any major side effects. Currently available systemic drugs for relief of osteoarthritis pain, such as non-selective NSAIDs and selective cyclooxygenase 2 (COX-2) inhibitors, are effective in the early to mid-stage of OA, but provide adequate pain relief as the joint deteriorates. It often fails to provide. In addition, NSAIDs cause gastrointestinal complications in a significant number of patients, and COX-2 inhibitors have recently raised concerns about cardiovascular side effects/risks, resulting in the retirement of COX-2 inhibitors Vioxx and Bextra from the US market. . Drugs to treat or prevent osteoarthritis are still in demand by the majority of patients.

Assay Example: Assay of the activity of a compound in a mouse osteoarthritis model
10 week old C57BL/6 mice were used in this study (n = 12 for each group). Anterior cruciate ligament transverse section (ACLT) model was used to simulate pathological conditions in OA. In order to perform ACLT surgery, the joint capsule of the right hind limb of the mouse was split open, and the anterior cruciate ligament (ACL) was transversely cut using a micro-scissor under a surgical microscope. After perfusion with saline to remove tissue debris, the skin incision was sealed.
On the 14th day after surgery, mice were treated according to the scheme shown in Table 1 and FIG. 1.
체적변화유량계(plethysmometer) 모델 600MR(미국 캘리포니아주 소재의 IITC Life Science)을 사용하여 체중 지지 시험을 수행하였다. The blank vehicle contained 5% Pluronic F68 (BASF, Germany) in 10 mM phosphate buffered saline (PBS, NaH 2 PH 4 /Na 2 HPO 4 ) adjusted to pH 7.4. Before each injection and before sacrifice at the end, the leg function was evaluated by weight bearing test. All mice were sacrificed for pathological evaluation and immunohistochemical staining on the 13th day after the initial administration. A weight support test was performed using a plethysmometer model 600MR (IITC Life Science, CA, USA). Briefly, after calibration of the equipment, mice were placed inside the chamber on a scale with their respective hind paws, and allowed to stand for 5-10 seconds until the readings stabilized. The readings reflected the weight on each hind limb. ACLT surgery caused osteoarthritis (OA) and reduced weight on the hind limbs. Results were expressed as the percentage of body weight (% weight support) on the operated leg compared to the non-operated leg on the contralateral side. According to the different treatments, the weight support results at the 13th day after the initial administration are shown in FIG. 2.
The value of% body weight support is expected to be 100 in healthy mice, indicating that the same body weight was carried on both legs. The value of% weight support will be reduced to 50-60 after ACLT surgery because less weight can be placed on the operated leg than on the non-operated leg. Relief of joint pain due to osteoarthritis will be reflected by an increased value of% body weight support.
As can be seen in FIG. 2, no significant difference was observed between the untreated control group and the vehicle control group (P> 0.05), indicating that there is no therapeutic effect of the blank vehicle. On the other hand, intraarticular injection of Compound A showed a dose-dependent effect in improving% body weight support, indicating its effect in alleviating pain caused by OA. It should be noted that 0.01 μmol of Compound A already exhibited a therapeutic effect, in which case this therapeutic effect was significantly increased in% body weight support compared to the untreated control and vehicle control (untreated control: P <0.05, vehicle Control: P <0.0001). Further increase in the dose to 0.05 μmol resulted in a further increase in the% body weight support value (P<0.05 compared to the 1 mM Compound A group), indicating a much stronger therapeutic effect.
It was demonstrated that Compound A significantly relieved joint pain in the mouse ACLT model.
Mouse joints were fixed overnight in 4% paraformaldehyde, dehydrated in increasing concentrations of ethanol, and embedded in paraffin. Sections (5 μm) were cut from paraffin blocks and applied to glass slides. Sections were stained for 5 minutes for proteoglycans with safranin O aqueous solution (0.1%), and then the specimens were mounted.
Pathological changes in the structure and/or appearance of the subchondral bone could be observed in the joints of mice from the untreated control and vehicle control. On the other hand, pathological changes including articular cartilage erosion, proteoglycan loss, and articular cartilage thinning and calcification were attenuated after treatment with Compound A. It was demonstrated that Compound A increased cartilage development and created a pro-chondrogenic environment.
