Abstract

Description
본 발명은 암 검출(또는 진단) 및 치료의 분야에 관한 것이다. 구체적으로, 본 발명은 항-EGF 유사 도메인 다중 6 항체(항-EGFL6 항체), 및 항-EGFL6 항체를 사용한 암 검출(또는 진단) 및 치료에 관한 것이다.
암은 신체의 임의의 곳에서 비정상 세포의 조절되지 않은 성장이다. 비정상 세포는 암 세포, 악성 세포, 또는 종양 세포로 불리워진다. 암의 조기 진단은 종종 이러한 질병의 성공적인 치료 또는 치유의 가능성을 증가시킨다.
바이오마커는 암의 검출에서 잠재적인 툴(potential tool)이다. EGFL6은 상피 성장 인자(EGF) 반복 수퍼패밀리 단백질의 구성원이고, EGF-유사 반복 영역 및 인테그린 연관 모티프(integrin association motif; RGD)로 이루어진다. WO2009057849호에는 KLK6, CKS2, IFITMl, SPPl, DPEPl, CSTl, CDH3, ANLN, CXCLl, MELK, CDCAl, CTHRCl, CEACAM6, MMPl, LCN2, HS6ST2, EGFL6 및 CA9로부터 선택된 하나 이상의 유전자의 mRNA 또는 단백질 수준을 측정하는 작용제를 포함하는, 결장암을 위한 진단 조성물이 개시되어 있다.
그러나, EGFL6 단독은 실제로, 임상 암 진단에서 적용되지 않는다. 더욱이, 암 치료를 위한 항-EGFL6 항체를 개발하는 것이 또한 요구되고 있다.

서열번호 7 또는 8의 아미노산 잔기 또는 서열번호 7 및 8 중 어느 하나에 대해 적어도 80% 동일성을 갖는 아미노산 서열을 갖는 변이체로 이루어진 중쇄 상보성 결정 영역 1(H-CDR1); 서열번호 9 또는 10의 아미노산 잔기 또는 서열번호 9 및 10 중 어느 하나에 대해 적어도 80% 동일성을 갖는 아미노산 서열을 갖는 변이체로 이루어진 중쇄 CDR2(H-CDR2); 및 서열번호 11 또는 12의 아미노산 잔기 또는 서열번호 11 및 12 중 어느 하나에 대해 적어도 80% 동일성을 갖는 아미노산 서열을 갖는 변이체로 이루어진 중쇄 CDR3(H-CDR3) 중 적어도 하나를 포함하는 단리된 항-EGFL6 항체 또는 이의 항원-결합 부분으로서, 상기 단리된 항체 또는 이의 항원-결합 부분이 EGFL6에 결합한, 단리된 항-EGFL6 항체 또는 이의 항원-결합 부분을 제공한다.
본 개시의 항체의 특정 구현예는 모노클론 항체, 키메라 항체, 인간화된 항체 및 인간 항체를 포함한다. 일부 구현예에서, 단리된 항-EGFL6 항체는 단쇄 Fv(scFv), IgG, Fab, (Fab)2, 또는 (scFv ')2이다.

상피 성장 인자 유사 단백질 6(EGFL6)의 발현은 암 환자의 혈청에서 평가되었다.
도 3에 도시된 바와 같이, 폐, 결장, 난소, 전립선, 뇌, 자궁 및 유방암 세포주에서 EGFL6의 단백질 발현은 웨스턴 블로트에 의해 측정되었다. EGFL6은 정상 세포주에서 발현되지 않는다. 이러한 결과는, EGFL6이 암에 걸린 환자에서 발견될 수 있으며 EGFL6이 암 마커로서 역할을 할 수 있음을 시사한다.
실시예 4 동물 모델에서 대장암 세포주 HCT116의 세포 이동 억제 및 항-EGFL6 단쇄 항체 S12 및 E5에 의한 암 전이의 억제에 대한 단쇄 항체의 평가
세포 침투 이동 분석은 대장암 세포주 HCT116의 세포 이동을 억제하는 능력에 대해 단쇄 항체를 평가하기 위해 수행되었다. 도 4(A)에 도시된 바와 같이, 항-EGFL6 단쇄 항체는 대장암 세포주 HCT116에 첨가되었고, 세포 배양 조건 하에서 암 세포의 성장을 억제한다. 결과는, 항체 Ab4, Ab5 (E5) 및 Ab10이 암세포 이동을 억제하는 데 효과적임을 나타내었다.
CT26 동종이식 마우스 모델에서, 폐로의 대장암 세포 CT26의 전이를 억제하는 능력은, 마우스에 항-EGFL6 단쇄 항체(S12 및 E5) 및 소분자 약물 파클리탁셀이 투여된 후 관찰되었다. 항-EGFL6 단쇄 항체 S12 및 E5는 3일에 한번씩 15 mg/kg의 용량으로 투여되며, 파클리탁셀은 4일에 한번씩 20 mg/kg의 용량으로 투여되었다. 도 4(B)에 도시된 바와 같이, 3일에 한번씩 15 mg/kg의 용량으로 2개의 항-EGFL6 단쇄 항체로 치료된 그룹은 4일에 한번씩 20 mg/kg의 용량으로 파클리탁셀로 치료된 대조군과 비교하여 종양 캡슐의 형성을 효과적으로 억제할 수 있다.
실시예
5
단쇄 항체 E5 및 S12에 의한 생체내에서 혈관신생 억제
5주령 누드 마우스는 혈관신생을 유도하기 위해 500 ㎕의, EGF와 혼합된 마트리겔로 피하 주사되었다. 누드 마우스는 하기 4개의 그룹으로 나누어졌다: (1) 음성 대조군(기초)으로서의 마트리겔 단독; (2) 양성 대조군(대조군)으로서 EGF와 혼합된 마트리겔(150 ng/mL); (3) EGF-유도 조건 하에서 15 mg/kg의 용량으로 단쇄 항체 E5로 치료된 그룹; 및 (4) EGF-유도 조건 하에서 15 mg/kg의 용량으로 단쇄 항체 S12로 치료된 그룹. 각 그룹은 3마리의 누드 마우스를 가지며, 단쇄 항체 E5 및 S12는 1주일 동안 꼬리 정맥 주사에 의해 1주일에 3회 투여되었다. 누드 마우스는 8일째에 희생되었다. 마우스 복부에서의 기질 마이셀은 그라인딩 로드에 의해 제거되고 균질화되고, 이후에, 헤모글로빈 함량을 측정하였다.
도 5에 도시된 바와 같이, 혈관신생은 마트리겔이 단독으로 주사된 음성 대조군(기초)에서 나타나지 않았다. 음성 대조군(기초)과 비교하여, 혈관신생은 24시간 동안 EGF와 혼합된 마트리겔이 주사된 양성 대조군(대조군)에서 나타났다. 도 5에 도시된 바와 같이, 항-EGFL6 단쇄 항체 E5 및 S12 둘 모두로 치료된 그룹이 혈관신생을 억제하는 효과를 나타낸다는 것이 명백하였다. 또한, 누드 마우스는 희생되었으며, 마우스 복부에서의 기질 마이셀이 제거되었으며, 헤모글로빈 함량이 측정되었다. 도 5에는 항-EGFL6 단쇄 항체 E5 및 S12로 치료된 누드 마우스의 기질 마이셀에서의 헤모글로빈 함량이 누드 마우스 독립체의 관찰과 관련하여 더 낮은 값을 나타낸다는 것이 예시되었다. 또한, 도 5에 도시된 바와 같이, 항-EGFL6 단쇄 항체 E5 및 S12는 혈관신생을 억제하는 데 유의미한 효과를 나타내었다. 항-EGFL6 단쇄 항체 E5 및 S12로 치료된 그룹의 헤모글로빈 함량은 음성 대조군(기초(Basal))의 함량과 거의 동일하였다. 이러한 결과는 항-EGFL6 단쇄 항체 E5 및 S12가 EGFL6을 중화시키는 능력을 가지고 이에 따라 혈관신생을 억제하는 효과를 달성함을 나타낸다.
실시예 6 동물 모델에서 항-EGFL6 인간화된 IgG 항체에 의한 생체내에서 종양 성장의 억제
닭 scFv 항체의 가변 영역의 인간화는 종래 연구에서 적용된 알고리즘에 의해 생성된 분자 모델의 도움으로 수행될 것이다[Tsurushita et al., 2004; Zilber et al., 1990]. 간단하게, 항-EGFL6 scFv 항체의 CDR에 대한 수용체로서 사용되는 인간 V 영역 프레임워크는 서열 상동성을 기초로 하여 선택될 것이다. CDR 구조의 적절한 형성을 위해 중요한 3차원 모델로부터 예측된 인간화된 V 영역에서의 아미노산 잔기는 닭 항-EGFL6 scFv 항체의 상응하는 잔기로 치환될 것이다. 다른 방법은 인간화된 항-EGFL6 항체의 구조를 추가로 미세-조정하기 위해 결합되고 이용될 것이다[Ewert et al., 2003; Sidhu et al., 2004].
2개의 항-EGFL6 인간화된 IgG 항체(S12 및 E5)는 이종이식 마우스에서 대장암 세포 HCT116, 비-소세포 폐암 A549, 삼중 음성 유방암 세포 MDA-MB-231 및 교모세포종 암 세포 U87의 성장을 억제하는 능력을 평가하였다. 마우스는 1주일에 2회 명시된 용량 및 정맥 주사로 투여되었다. 초기 종양 크기는 약 200 ㎣이었다. 실험의 종료 후에, 종양 성장 억제의 비율(%TGI)은 대조군과 비교하여 계산되었다. 도 6(A)에 도시된 바와 같이, 인간화된 IgG 항체 S12 및 E5를 갖는 두 그룹 모두는 종양 성장(대장암 세포 HCT-116)을 억제하는 능력을 갖는다. S12를 갖는 그룹의 %TGI는 7.5%이며, E5 항체를 갖는 그룹은 36.2%이다. 도 6(B)에 도시된 바와 같이, 인간화된 IgG 항체 E5는 비-소세포 폐암 A549에서 종양 성장을 억제하는 능력을 갖는다. E5 항체(10 mg/kg)를 갖는 그룹의 %TGI는 14.3%이며, E5 항체(20 mg/kg)를 갖는 그룹은 80.8%이다.
도 7(A)에 도시된 바와 같이, 인간화된 IgG 항체 E5를 갖는 그룹은 종양 성장(삼중 음성 유방암 세포 MDA-MB-231)을 억제하는 능력을 갖는다. E5 항체(20 mg/kg)를 갖는 그룹의 %TGI는 65.1%이며, E5 항체(40 mg/kg)를 갖는 그룹은 65.4%이다. 도 7(B)에 도시된 바와 같이, 단쇄 항체 E5를 갖는 그룹은 종양 성장(교아종 암 세포)을 억제하는 능력을 갖는다. E5 항체(20 mg/kg)를 갖는 그룹의 %TGI는 36.5%이다.
인간 비-소세포 폐암 A549 이종이식 모델에서 파클리탁셀과 조합된 인간화된 IgG 항체 E5의 항암 활성이 추가로 수행되었다. 파클리탁셀(5 mg/kg), E5 IgG(10 mg/kg) 및 E5 IgG(10 mg/kg)와 조합한 파클리탁셀(5 mg/kg)은 마우스에 정맥 주사되었다. 도 8에 도시된 바와 같이, 항체 E5 IgG를 갖는 그룹은 종양 성장을 억제하는 능력을 가지며, E5 IgG와 조합한 파클리탁셀은 종양의 억제에서 예상치 못한 효능을 갖는다. E5 항체(10 mg/kg)를 갖는 그룹의 %TGI는 14.3%이며, E5 IgG와 조합한 파클리탁셀을 갖는 그룹은 61.8%이다.
