Abstract
본 발명은 링커-약물 vc-seco-DUBA 및 이의 중간체의 합성을 위한 개선된 방법, 및 vc-seco-DUBA 링커-약물을 포함하는 항체-약물 접합체를 제조하는 방법에서의 상기 개선된 방법의 용도에 관한 것이다.
Description
본 발명은 링커-약물 vc-seco-DUBA 및 이의 중간체의 합성을 위한 개선된 방법, 및 vc-seco-DUBA 링커-약물을 포함하는 항체-약물 접합체를 제조하는 방법에서의 상기 개선된 방법의 용도에 관한 것이다.
듀오카마이신은 듀오카마이신 A, 듀오카마이신 SA 및 CC-1065를 포함하는 항종양 항생제 패밀리의 구성원이다. 이들은 강력한 항종양 특성으로 알려져 있지만, 일반적으로 극도로 높은 독성 때문에 그 자체로는 사용되지 않는다. 현재, 듀오카마이신은 항체-약물 접합체(ADC)에서 세포독성 약물로서 연구되고 있다.
ADC는, 매우 강력한 세포독성 약물을 암 세포에 특이적으로 유도함으로써 소분자 약물의 잠재적 전신 독성 부작용을 감소시키면서 효능을 향상시켜 암에 효과적인 새로운 치료에 대한 충족되지 않은 큰 필요성을 해결할 수 있는 잠재력을 갖는다.
ADC의 미래의 상업적 성공을 위한 주요 측면 중 하나는 세포독성 약물 및 상응하는 링커-약물 작제물의 합성 방법이며, 여기서 링커 모이어티가 항체와의 접합을 용이하게 하기 위해 세포독성 약물에 부착되며, 이 방법은 산업 규모의 생산에 적합하다.
WO2011/133039의 p. 210, ll. 21-27에서 화합물 18b로서 처음 개시된 하기 식 (I)의 링커-약물 vc-seco-DUBA는 매우 강력한 CC-1065 유사체의 예이다.
항-HER2 항체를 갖는 vc-seco-DUBA의 ADC 트라스투주맙, 즉 SYD985 또는 (vic-)트라스투주맙 듀오카마진은, 몇몇 전임상 연구(M.M.C. van der Lee et al., Molecular Cancer Therapeutics, 2015, 14(3), 692-703; J. Black et al., Molecular Cancer Therapeutics, 2016, 15 (8), 1900-1909) 및 1상 임상 시험(ClinicalTrials.gov NCT02277717)에서 성공적으로 사용되었다. 현재, SYD985 치료는, HER2 양성 국소 진행성 또는 전이성 유방암 환자의 3상 임상 시험에서의 의사의 선택 치료(TULIP; ClinicalTrials.gov NCT03262935)와 직접 비교된다.
링커-약물 vc-seco-DUBA의 합성은 4-단계 공정으로서 WO2011/133039에 기재되어 있다. 50-100 mg 실험실 규모에서 이 과정에 따른 vc-seco-DUBA의 제조는 링커-약물에 단지 21-25%의 전체 수율을 제공하였다. 이 공정의 마지막 두 단계, 즉 단계 3과 단계 4는 전체 공정으로부터의 vc-seco-DUBA의 전체 수율에 중요하며, 단지 약 50%의 총 수율을 나타낸다. 산업 규모에서 이 4-단계 공정의 수율은 훨씬 낮아진다.
따라서, vc-seco-DUBA를 제조하기 위한 개선된 공정이 필요하다. 특히, 수율 및 화학적 순도면에서 효율적이고, 시약 및 반응 조건면에서 비용 효과적이며, 산업적 규모의 생산에 적합한 공정이 필요하다.
본 발명은 산업적 규모로 생산하기에 적합한 공정 조건으로 링커-약물 vc-seco-DUBA 및 이의 중간체의 합성을 위한 개선된 방법으로서, 개선된 수율로 바람직한 vc-seco-DUBA 산물을 제공하는 방법에 관한 것이다.

듀오카마이신은 스트렙토마이세스(Streptomyces) 종의 배양액에서 처음 단리 된 구조 관련 독소 부류이다. 이들은 듀오카마이신 A, 듀오카마이신 SA 및 CC-1065를 포함하는 항종양 항생제 패밀리의 구성원이다. 듀오카마이신은 DNA의 소홈(minor groove)에 결합하여 DNA의 비가역적 알킬화를 일으킨다. 이것은 핵산 구조를 파괴하여 결국 종양 세포 사멸로 이어진다.
WO2011/133039는 CC-1065의 듀오카마이신 유도체를 포함하는 하기 식 (I)의 매우 강력한 링커-약물 vc-seco-DUBA(p. 210, ll. 21-27 상 화합물 18b)를 구체적으로 개시한다:
본 발명은 놀랍게 높은 수율로 산업 규모에 성공적으로 적용될 수 있는 vc-seco-DUBA의 개선된 제조 방법에 관한 것이다.
WO2011/133039의 실시예 10에서 vc-seco-DUBA 링커-약물의 화학적 합성은 4-단계 공정으로 기재된다:
상기에서, PNP-Cl은 4-니트로페닐 클로로포르메이트이고, Et3N은 트리에틸아민이고, Boc는 tert-부틸옥시카르보닐이고, TFA는 트리플루오로아세트산이고, CHCl3은 클로로포름이고, DMF는 N,N-디메틸포름아미드이다.
50-100 mg 실험실 규모에서, 이 4-단계 공정은 단지 21-25%의 전체 수율을 보여준다. 산업 규모에서 이 수율은 상당히 낮아진다.
이 4-단계 공정의 낮은 전체 수율은 대부분 마지막 두 단계, 즉 단계 3 및 단계 4의 실험실 규모에서의 단지 약 50%의 낮은 총 수율에 기인할 수 있다. 본 발명자들은 놀랍게도 단계 3에서 산 시약의 변화를 포함하는 변형된 절차가 결정화를 통해 단리될 수 있는 새로운 중간체를 산출하였다는 것을 발견하였다. 예기치 않게, 단계 3의 이러한 변형된 절차 및 새로운 중간체의 사용으로 vc-seco-DUBA의 수율이 상당히 증가한 것으로 밝혀졌다.
통상, 결정화 단계는 산물의 순도를 증가시킬 필요가 있을 때 화학 합성에 도입된다. 그러나, 상당한 양의 산물이 모액에 남아 있기 때문에, 이러한 단계의 도입은 일반적으로 상기 산물의 수율을 감소시킨다. 놀랍게도, 본 발명자들은 상기 기재된 바와 같은 vc-seco-DUBA의 합성에서 결정화 단계를 도입하여 하기 식 (II)의 새로운 중간체를 유도한다는 것은, 순도 증가(94-96%에서 ≥ 99.0%)로 이어졌을뿐만 아니라 vc-seco-DUBA의 수율의 예상치 못한 상당한 증가(53%에서 약 79%)를 보여준다는 것을 발견하였다:
따라서, 제1 실시양태에서, 본 발명은 식 (II)의 화합물에 관한 것이다.
제2 실시양태에서, 본 발명은 하기 식 (III)의 화합물을 1,4-디옥산 중의 염화수소와 반응시켜 식 (II)의 화합물을 형성하는 단계를 포함하는 식 (II)의 화합물의 제조 방법에 관한 것이다:
통상, 식 (III)의 화합물은 1,4-디옥산 중의 10-20 질량% 염화수소와 반응한다. 바람직하게는, 식 (III)의 화합물은 1,4-디옥산 중의 12-18% 염화수소, 더욱 바람직하게는 1,4-디옥산 중의 15% 염화수소와 반응한다. 통상, 식 (III)의 화합물:1,4-디옥산 중의 HCl의 질량비는 1:0.5 내지 1:25의 범위이다. 바람직하게는, 식 (III)의 화합물:1,4-디옥산 중의 HCl의 질량비는 1:1 내지 1:10의 범위이다. 더욱 바람직하게는 1:5 내지 1:10이다.
통상, 염화수소의 양은 식 (III)의 화합물의 양을 초과하는 몰이다. 바람직하게는, 염화수소의 양은 식 (III)의 화합물의 양의 2몰 당량 이상, 더욱 바람직하게는 2 내지 50 당량이다.
바람직하게는, 상기 반응은 물 및/또는 메탄올 중 트리이소프로필실란과 같은 스캐빈저의 존재 하에 일어난다. 상기 물 및/또는 메탄올은 총 용매 질량의 25 질량% 미만, 바람직하게는 15% 미만, 더욱 바람직하게는 10% 미만의 양으로 존재할 수 있다.
식 (III)의 화합물은, 예를 들어 문헌[R.C. Elgersma et al., Molecular Cancer Therapeutics, 2015, 12(6), 1813-1835]에서 기재된 바와 같이, 하기 식 (IV)의 화합물을 4-니트로페닐 클로로포르메이트와 반응시켜 하기 식 (V)의 화합물을 형성한 후, 1-히드록시벤조트리아졸 수화물의 존재 하에 하기 식 (V)의 화합물을 하기 식 (VI)의 화합물과 반응시켜 식 (III)의 화합물을 형성함으로써 제조될 수 있다:
통상, 식 (IV)의 화합물과 4-니트로페닐 클로로포르메이트의 반응은 0 내지 20℃의 온도에서 수행된다. 바람직하게는, 온도는 0 내지 10℃, 더욱 바람직하게는 0 내지 6℃, 더욱 더 바람직하게는 2 내지 6℃, 가장 바람직하게는 3 내지 5℃이다.
식 (V)의 화합물의 제조에 사용하기에 적합한 용매는, 비제한적으로, 유기 용매, 바람직하게는 비양성자성 용매, 더욱 바람직하게는 극성 비양성자성 용매이다. 바람직한 용매는 에테르 용매, 아미드 용매 또는 이들의 혼합물이다. 특히 바람직한 용매는 테트라히드로푸란(THF), N,N-디메틸아세트아미드(DMA) 또는 이들의 혼합물이다. THF와 DMA의 혼합물이 가장 바람직하다.
식 (V)의 화합물의 제조에 사용하기에 적합한 염기는 유기 염기, 예를 들어 3차 아민이다. 특히 적합한 염기는 Et3N이다.
통상, 식 (V)의 화합물과 식 (VI)의 화합물의 반응은 0 내지 20℃의 온도에서 수행된다. 바람직하게는, 온도는 0 내지 10℃, 더욱 바람직하게는 4 내지 10℃이다.
식 (III)의 화합물의 제조에 사용하기에 적합한 용매는, 비제한적으로, 유기 용매, 바람직하게는 비양성자성 용매, 극성 용매 또는 이들의 혼합물이다. 바람직한 용매는 에테르 용매, 아미드 용매 또는 이들의 혼합물이다. 특히 바람직한 용매는 THF, DMA 또는 이들의 혼합물이다. THF와 DMA의 혼합물이 가장 바람직하다.
식 (IV)의 화합물은 종래 기술에 공지된 임의의 적합한 방법, 예를 들어 WO2015/185142의 실시예 6a에 기재된 방법에 의해, 또는 이와 유사한 방법으로 제조될 수 있다.
다른 실시양태에서, 본 발명은 vc-seco-DUBA를 제조하기 위한 식 (II)의 화합물의 용도에 관한 것이다.
또 다른 실시양태에서, 본 발명은 식 (II)의 화합물을 하기 식 (VII)의 화합물과 반응시키는 vc-seco-DUBA의 제조 방법에 관한 것이다:
예기치 않게, WO2011/133039의 실시예 10에서 사용된 트리에틸아민(Et3N) 대신 N,N-디이소프로필아민(DIPEA)을 염기로 사용하여 식 (II)의 화합물을 식 (VII)의 화합물과 반응시킬 때 vc-seco-DUBA의 수율이 더욱 더 증가되었다. 통상, 식 (II)의 화합물:DIPEA의 몰비는 1:1 내지 1:15의 범위이다. 바람직하게는, 비는 1:1 내지 1:10, 더욱 바람직하게는 1:2 내지 1:7, 더욱 더 바람직하게는 1:3 내지 1:5의 범위이고, 가장 바람직하게는 비는 약 1:4이다.
통상, 식 (II)의 화합물과 식 (VII)의 화합물의 반응은 0 내지 20℃의 온도에서 수행된다. 바람직하게는, 온도는 0 내지 10℃, 더욱 바람직하게는 0 내지 5℃이다.
vc-seco-DUBA를 제조하기 위해 식 (II)의 화합물과 식 (VII)의 화합물의 반응에 사용하기에 적합한 용매는, 비제한적으로, 유기 용매, 바람직하게는 비양성자성 용매, 더욱 바람직하게는 극성 비양성자성 용매이다. 바람직한 용매는 에테르 용매, 아미드 용매 또는 이들의 혼합물이다. 특히 바람직한 용매는 THF, DMA, N,N-디메틸포름아미드(DMF) 또는 이들의 혼합물이다. DMA가 가장 바람직하다.
바람직한 실시양태에서, 공정은 1-히드록시벤조트리아졸 수화물의 존재 하에 수행된다. 통상, 식 (II)의 화합물:1-히드록시벤조트리아졸 수화물의 몰비는 1:1 내지 1:10의 범위이다. 바람직하게는, 비는 1:1 내지 1:7, 더욱 바람직하게는 1:2 내지 1:5, 더욱 더 바람직하게는 1:2 내지 1:3의 범위이고, 가장 바람직하게는 비는 약 1:2.5이다.
본 발명은 또한 하기 식 (VIII)의 vc-seco-DUBA ADC의 제조 방법에 관한 것이며, 여기서 vc-seco-DUBA 링커-약물 화합물은 상기 기재된 바와 같은 본 발명에 따른 방법으로 제조된다:

vc-seco-DUBA 링커-약물을 항체에 접합시키는 것은 예를 들어 WO2011/133039, WO2015/177360 및 WO2017/137628에 기재된 바와 같이 수행될 수 있다.
야생형 ADC는 사슬간 디설피드 결합의 환원을 통해 생성된 시스테인 측쇄의 유리 티올을 통해 링커-약물을 항체에 접합시킴으로써 생산된다. 제조는 용매 노출된 사슬간 디설피드의 부분 환원 이후 말레이미드 함유 링커-약물로 생성된 티올의 변형을 수반한다. 시스테인 부착 전략은 환원된 디설피드당 최대 2개의 약물을 생산한다. 대부분의 인간 IgG 분자는 4개의 용매 노출된 디설피드 결합을 가지므로 항체당 0-8개의 약물 범위가 가능하다. 항체당 약물의 정확한 수는 디설피드 환원 정도 및 후속 접합 반응에 사용되는 링커-약물의 몰 당량의 수에 의해 결정된다. 모든 4개의 디설피드 결합의 완전한 환원은 항체당 8개의 약물을 갖는 균질한 작제물을 형성하는 반면, 부분적인 환원은 통상 항체당 0, 2, 4, 6 또는 8개의 약물을 갖는 이종 혼합물을 생산한다.
부위 특이적 ADC는 돌연변이된 항체의 적합한 위치에서 조작된 시스테인 잔기의 측쇄를 통해 링커-약물을 항체에 접합시킴으로써 생산된다. 조작된 시스테인은 일반적으로 시스테인 또는 글루타티온과 같은 다른 티올에 의해 캡핑되어 디설피드를 형성한다. 이러한 캡핑된 잔기는 약물 부착이 일어날 수 있기 전에 캡핑해제(uncapping)되어야 할 필요가 있다. 조작된 잔기에 대한 약물 부착은, 고유의 사슬간 및 돌연변이 디설피드를 환원시킨 다음, CuSO4 또는 디히드로아스코르브산과 같은 온화한 산화제를 사용하여 고유의 사슬간 시스테인을 재산화시키고, 이어서 캡핑되지 않은 조작된 시스테인과 링커-약물의 표준 접합에 의해 달성되거나, 또는 사슬간 디설피드 결합보다 더 높은 비율로 돌연변이 디설피드를 환원시키는 온화한 환원제를 사용하여, 링커-약물과 함께 캡핑되지 않은 조작된 시스테인의 표준 접합에 의해 달성된다. 최적 조건 하에서, (하나의 시스테인이 mAb의 중쇄 또는 경쇄에 조작된 경우) 항체당 2개의 약물(즉, 약물 대 항체 비, DAR은 2)이 부착된다.
바람직한 실시양태에서, 본 발명에 따라 사용하고자 하는 항체는 항-HER2 항체이며, 더욱 바람직하게는 항-HER2 항체 트라스투주맙이다.
하나의 특정 실시양태에서, 본 발명은 하기 식 (IX)의 트라스투주맙 vc-seco-DUBA ADC의 제조 방법에 관한 것이며, 여기서 vc-seco-DUBA 링커-약물 화합물은 전술한 바와 같이 본 발명에 따른 방법으로 제조된다:

메틸CBI-아자인돌-벤즈아미드-MOM(1)(1.0 g, 1.75 mmol)을, 트리에틸아민(Et3N)(0.55 g, 4.94 mmol)의 존재 하에 0℃의 온도에서 약 1.5시간 동안 테트라히드로푸란(THF)(4.5 g) 및 N,N-디메틸아세트아미드(DMA)(3.0 g)의 혼합물 중에서 4-니트로페닐 클로로포르메이트(PNP-Cl)(0.43 g, 2.12 mmol)와 반응시켜 6℃까지 가온시켰다. 메틸CBI-아자인돌-벤즈아미드-MOM-PNP(2)를 포함하는 슬러리를 얻었다.
단계 2에서, tert-부틸(2-((2-(2-히드록시에톡시)에틸)아미노)에틸)(메틸)-카바메이트(3)(0.58 g, 2.19 mmol)를 DMA(1.7 g)에 용해시키고 1-히드록시벤조트리아졸 수화물(HOBt)(0.35 g, 2.28 mmol)을 첨가하였다. 이 수득된 용액을 4℃의 온도에서 1.5시간 동안 슬러리와 반응시켜 10℃까지 가온시켰다.
반응 완료 후, 에틸 아세테이트(EtOAc)(8.8 g)를 반응 혼합물에 첨가하고 용액을 염수(11.3 g), 포화 중탄산나트륨 용액(3.8 g)으로 세척하고 다시 염수(3.8 g)로 세척하였다. 유기층을 분리하고 탄소 여과로 정제하였다. 용매를 회전식 진공 증발기에서 증발시켰다. 수득된 메틸CBI-아자인돌-벤즈아미드-MOM-Boc-에틸렌디아민-D(4)를 아세톤(20 g)에 용해시키고, 결국 탄소 여과에 의해 다시 정제하였다.
미정제 산물을 실리카겔 컬럼 크로마토그래피로 정제하여, 이를 이동상(DCM:MeOH = 97:3 내지 94:6)으로 용리시켰다. 합한 산물 분획을 농축시키고 진공 건조시켜 메틸CBI-아자인돌-벤즈아미드-MOM-Boc-에틸렌디아민-D(4)(1.27 g, 1.48 mmol; 84% 수율, 93.82% 순도)를 수득하였다.

seco
-DUBA의 제조
메틸CBI-아자인돌-벤즈아미드-에틸렌디아민-D 히드로클로라이드(5)의 제조
메틸CBI-아자인돌-벤즈아미드-MOM-Boc-에틸렌디아민-D(4)(1.27 g, 1.48 mmol)의 메톡시메틸(MOM) 및 tert-부틸옥시카르보닐(Boc) 기를, 스캐빈저(트리이소프로필 실란(0.63 g), 물(0.4 g) 및 메탄올(0.3 g))의 존재 하에 1,4-디옥산(7.5 g) 중의 15% 염화수소(HCl)로 제거하였다. 메틸CBI-아자인돌-벤즈아미드-에틸렌디아민-D 히드로클로라이드(5)는 반응 용액으로부터 황색 고체로서 결정화되었다.
수득된 황색 고체를 여과하고, 아세톤으로 세척하고 질소 및 진공을 사용하여 필터 상에서 건조시켜 순수한 산물(5)(1.0 g, 1.33 mmol; 90% 수율, ≥ 90% 순도)을 제공하였다.
vc-seco-DUBA의 제조
메틸CBI-아자인돌-벤즈아미드-에틸렌디아민-D 히드로클로라이드(5)(1.0 g, 1.33 mmol)를, N,N-디이소프로필아민(DIPEA)(0.65 g, 5.10 mmol) 및 HOBt(0.47 g, 3.16 mmol)의 존재 하에 0℃의 온도에서 암실에서 1.5시간 동안 DMA(17.8 g) 중 말레이미드-OEG2-val-cit-PABA-PNP(6)(0.98 g, 1.29 mmol)와 반응시켜 5℃까지 가온시켰다. 반응 혼합물을 23 내지 25℃(50 내지 60분)의 온도에서 물(201.1 g)에 적가하고 vc-seco-DUBA 미정제 산물의 침전물을 수득하였다. 30분 동안 교반한 후, 침전된 미정제 산물을 압력 필터에서 여과하였다. 필터 케이크를 물로 완전히 세척하고 진공 및 약간의 질소 흐름 하에 필터에서 건조시켰다.
vc-seco-DUBA 미정제 산물을 먼저 저압 플래쉬 크로마토그래피(정지상 - 실리카겔 0.040 내지 0.063 mm; 이동상 - 디클로로메탄:메탄올 = 90:10)에 적용하였다. 적합한 분획(vc-seco-DUBA의 UPLC-IN 순도 ≥ 90%)을 플라스크에 수집하고, 여과하고 증발시켰다. 예비 크로마토그래피(정지상 - 실리카겔 0.015 내지 0.040 mm; 이동상 - 디클로로메탄:메탄올 = 90:10 내지 85:15)에 의해 추가 정제를 수행하였다. 적합한 분획(vc-seco-DUBA의 UPLC-IN 순도 ≥ 90%)을 플라스크에 수집하고 용매를 DMA로 전환시켰다. 최대 온도 25℃에서 농축을 수행하였다. 농축된 용액을 합하고 0.2 μm 필터를 통해 여과하고 물에 첨가하여 순수한 vc-seco-DUBA를 미세한 황색 분말로서 침전시켰다(수율: 35-45%; 순도: ≥ 99.0%).
산물을 여과하고 물로 세척하고 질소 및 진공을 사용하여 필터에서 최대 25℃의 온도에서 건조시켰다.
비교예 - vc-
seco
-DUBA의 제조
vc-seco-DUBA의 합성은 WO2011/133039의 실시예 10에 기재된 절차에 따라 수행된다.
단계 1
메틸CBI-아자인돌-벤즈아미드-MOM-Boc-에틸렌디아민-D(4)(0.1 mmol)를 클로로포름(CHCl3)(6 ml)에 현탁시키고 얼음에서 냉각시켰다. 2 ml의 산(트리플루오로아세트산(TFA) 또는 1,4-디옥산(7.5 g) 중의 15% HCl)을 첨가하고 혼합물을 3시간 동안 교반하였다. 이어서, 혼합물을 진공 농축시켰다.
단계 2
잔류물을 N,N-디메틸포름아미드(DMF)(4 ml)에 용해시키고, 용액을 얼음에서 냉각시키고 말레이미드-OEG2-val-cit-PABA-PNP(6)(0.13 mmol) 및 염기(1 mmol, Et3N 또는 DIPEA)를 첨가하였다. 혼합물을 2시간 동안 교반하고, 진공 농축시키고, 잔류물을 컬럼 크로마토그래피(SiO2, 디클로로메탄:메탄올, 1:0 내지 8:2)로 정제하였다.
상기 절차는, 단계 1에서 메틸CBI-아자인돌-벤즈아미드-MOM-Boc-에틸렌디아민-D(4)의 MOM 및 Boc 기를 제거하기 위해 1,4-디옥산 중의 HCl 또는 종래 기술의 산 TFA를 사용하고 단계 2에서 메틸CBI-아자인돌-벤즈아미드-에틸렌디아민-D 히드로클로라이드(5) 및 말레이미드-OEG2-val-cit-PABA-PNP(6)의 커플링 반응을 촉진하기 위한 DIPEA 또는 종래 기술의 염기 Et3N을 사용하여 수행됨으로써 vc-seco-DUBA의 제조 효율에 대한 산 및 염기 선택의 영향을 결정하였다.
하기 표는 vc-seco-DUBA의 수율을 나타낸다.
단계 1에서 TFA 대신 1,4-디옥산 중의 HCl을 사용함으로써 HPLC에 의해 측정된 vc-seco-DUBA의 전체 수율은 25.8% 증가하였다. 단계 2에서 Et3N 대신 DIPEA를 사용함으로써 HPLC에 의해 측정된 vc-seco-DUBA의 전체 수율은 23.6% 감소하였다. 그러나, 단계 1에서 1,4-디옥산 중의 HCl을 사용하고 단계 2에서 DIPEA를 사용함으로써 HPLC에 의해 측정된 vc-seco-DUBA의 전체 수율은 29.8% 증가하였다.

- 제9항에 있어서, 식 (I)의 화합물을 항-HER2 항체 트라스투주맙에 접합시키는 것인 방법.
