Description
The present invention relates to a solid-state reaction chip and a measuring method using the same.
It is known that type I allergies that cause diseases such as bronchial asthma, allergic rhinitis, urticaria, or anaphylactic shock are caused by excessive reactions of specific antigens (allergens) and IgE antibodies thereto. At each clinical site, such as a hospital, tests are conducted to detect IgE antibodies contained in the test fluid in identifying causative allergens that are the cause of allergic diseases. At present, as a method for detecting an allergen-specific IgE antibody contained in a test solution, CAP using immunofluorescence as a measurement principle and MAST using immunochemiluminescence as a measurement principle are used.
Conventionally, in order to efficiently detect a measurement target substance such as a nucleic acid, a protein or a glycolipid contained in a test solution, a binding substance having a specific binding ability to the measurement target substance is immobilized on the solid phase, A solid-state reaction chip is used in which a substance to be bound bound to a binding substance is detected. Solid-state reaction chips are also called biochips in the field of biochemistry. For example, mRNA chips, cDNA chips, micro PCR chips, protein chips, and the like are known.
In recent years, immunoassay microchips using an antigen-antibody reaction have been proposed for the diagnosis of allergic diseases. For example, Patent Document 1 discloses an immunoassay microchip having a plurality of allergen antigens as independent, i.e., A biochip analysis method is disclosed in which a biochip is used to automate the process of detecting the reaction between a test solution and an antigen after collection of a test solution and to obtain a measurement result quickly.
The CAP of the prior art is, in principle, a single-item method in which measurement is carried out for each type of allergen. Since a test fluid is required for each measurement, the physical burden on the subject is large and the throughput related to the measurement is also poor. On the other hand, MAST is a multi-item method in which, for example, about 30 kinds of allergens are measured at one time. Although the number of simultaneously measurable allergens is larger than that of the single item method, the operation is troublesome and requires about 5 hours until the measurement result is obtained.
In addition, in the technique described in Patent Document 1, a suction nozzle for sucking each liquid supplied from each nozzle such as a cleaning nozzle, an antibody nozzle, and a reagent nozzle is required, and time is required until suction of each liquid is completed In addition, there is a possibility that the suction force is insufficient due to the poor state of the suction pump, or the attachment attached to the suction nozzle adheres to the bio chip, resulting in contamination.

As described above, the

In the measurement method according to the present embodiment, an example in which an IgE antibody that specifically binds to an allergen is used will be described. The allergen immobilized as a binding substance in the
Immobilization of the allergen to the
The physiologically active substance having the specific binding activity to the IgE antibody as the substance to be measured and having the enzyme activity is not particularly limited, and examples thereof include alkaline phosphatase, beta -galactosidase, glucose oxidase , An anti-IgE antibody labeled with urease, creatinine kinase, myocyte, glucose-6-phosphate dehydrogenase, peroxidase or the like is preferably used. In this embodiment, an example using HRP-labeled antibody labeled with anti-IgE antibody with Western wasabi peroxidase (HRP) will be described. When an HRP-labeled antibody is used, the resulting signal can be obtained as a colorimetric signal or a chemiluminescence signal depending on the substrate. As the substrate used for the generation of the colorimetric signal, for example, tetramethylbenzidine and derivatives thereof, o-phenylenediamine, triarylmethanes, imidazole leuco dye and the like can be used. Examples of the production of the chemical luminescence signal include acridinium salts, dioxetanes, luciferin, lucigenin, and oxalyl chloride.
