Abstract
본 발명은 선복화 추출물을 유효성분으로 함유하는 항균용 조성물에 관한 것으로, 메치실린-내성 황색포도상구균에 대해서 항균활성을 갖는 선복화 추출물을 유효성분으로 함유하는 항균용 조성물을 제공하는 뛰어난 효과가 있다.
Description
본 발명은 선복화 추출물을 유효성분으로 함유하는 항균용 조성물에 관한 것이다.
황색포도상구균 (Staphylococcus aureus)은 빈번하게 식중독을 일으키는 주요 원인균이며, 상처를 통한 감염 또는 면역력이 저하되어 있는 환자나 노약자에게 감염되어 질병을 일으킨다. 증식하는 과정에서 과량의 독소를 생성하고, 만성적인 골수염과 축농증을 일으키기도 한다. 특히, 황색포도상구균이 생성하는 장 독소 (enterotoxin)에 의해 식중독 증상이 발생하며, 독소에 오염된 식품을 섭취하여 위나 장에 흡수되면, 구토, 설사, 복통을 일으킨다. 또한, 내열성이 강하고, 독소에 오염된 식품을 가열하여도 파괴되지 않기 때문에 장 독소를 생성하기 위한 수준인 105-106 CFU/g까지 증식하지 않도록 방지하는 것이 필수적이다. 식품의약품안전청에서 보고한 2010년도 집단 식중독 통계현황에 의하면 원인이 밝혀진 국내 집단 식중독 발생 건수 중 황색포도상구균에 의한 발생 건수가 10%가 넘는 것으로 나타났고, 외식 및 단체급식의 발달로 인해 발생 건수에 비해 감염환자의 수가 크게 나타났다.
최근에도 전 세계적으로 오염된 식품 및 식수에 의한 식중독 발생 건수가 증가하고 있으며, 보건 및 산업적으로 심각한 문제를 발생시키는 균주 중에서 항생제에 내성을 갖는 균주의 출현이 계속해서 보고되고 있으며, 그 수가 증가하고 있다. 항생제 내성은 병원에서 처방 받는 항생제의 사용량 및 동물약품 및 사료첨가제로서 사용하는 항생제의 사용량과 관계가 있기 때문에, 국내·외적으로 사용량을 줄이기 위한 노력이 진행되고 있으나, 국내 축·수산업 분야에서 사용하는 항생제 사용량은 유럽과 비교하여 최대 9배 수준에 달한다. 병원에서의 처방률은 2011년 27%로, WHO 권장량보다 높기 때문에 내성 균주에 대한 연구와 대책이 필요한 실정이다.
항생제 내성균주 중 메치실린-내성 황색포도상구균 (MRSA, methicillin-resistant S. aureus)은 β-lactam 계열의 항생제에 대한 친화도가 낮은 세포벽을 가지고 있어서, 황색포도상구균을 제어할 수 있었던 기존 항생제의 효율을 감소시킨다. 이러한 내성의 기전에는 β-lactamase를 과잉 생산하거나, mec (methicillin resistant determinant) A의 발현에 의해 β-lactam 계열의 항생제에 친화도가 낮은 PBP2a (penicillin binding protein 2a) 단백질이 생산되는 경우 및 PBP형 단백질을 PBP2'으로 변형을 유도하는 mecRI의 발현 등을 들 수 있다. 미국질병통제예방센터 (CDC, Centers for Disease Control and Prevention)에서는 미국 내에서 Staphylococcus sp.에 의해 연간 10,800명이 사망하는 것으로 추정하였으며, 그 중에서 5,500명이 메치실린-내성 황색포도상구균에 의해 사망하는 것으로 추정하였다. 미국 내 연구 결과에 따르면 감염에 의한 사망률은 20.7%로 높은 수준이며, 수술환자의 경우 20일 간의 평균 입원기간에 따른 의료비 부담도 증가하는 것으로 나타났다. MRSA는 국내에서도 병원 감염의 가장 흔한 원인균으로 3차 병원에서 발견되는 S. aureus 중 70-80%를 차지하며, 도축장 식육에서도 메치실린을 포함한 다제 내성 S. aureus이 분리되었다. 2011년 국내에서 신고된 MRSA 감염 사례는 3,248건으로 지역사회로의 광범위한 전파가 우려되는 상황이다.
의료 및 동물용 항생제 사용을 줄이기 위한 방법으로 합성 항생제에 비해 내성 발생률이 적은 장점이 있는 천연 동·식물 소재로부터 유용 항균소재를 발굴하고, 이용하기 위한 연구가 진행되고 있다. Zoraghi 등은 해양 무척추동물인 Topsentia pachastrelloides로부터 MRSA의 pyruvate kinase를 저해하는 bis-indole 화합물을 분리하였으며 (Zoraghi, R., Worrall, L., See, R. H., Strangman, W., Popplewell, W. L., Gong, H., Samaal, T., Swayze, R. D., Kaur, S., Vuckovic, M., Finlay, B. B., Brunham, R. C., McMaster, W. R., Davies-Coleman, M. T., Strynadka, N. C., Andersen, R. J., and Reiner, N. E. (2011) Methicillin-resistant Staphylococcus aureus (MRSA) pyruvate kinase as a target for bis-indole alkaloids with antibacterial activities. J. Biol. Chem. 286, 44716-44725), Tanaka 등은 엄나무 (Erythrina variegata)의 뿌리로부터 MRSA에 대하여 항균활성을 갖는 신규 isoflavonoids를 분리하였다 (Tanaka, H., Atsumi, I., Shirota, O., Sekita, S., Sakai, E., Sato, M., Murata, J., Murata, H., Darnaedi, D., and Chen, I. Biodivers. 8, 476-482). 중국에서는 약초의학 (TCM; Traditional Chinese Medicine) 산업을 통해 천연 화합물 데이터 베이스를 구축하고 있으며, Zuo 등은 중국의 전통 약용식물로부터 MRSA에 대해서 항균 활성을 갖는 유용소재를 보고하였다 (Zuo, G. Y., Wang, G. C., Zhao, Y. B., Xu, G. L., Hao, X. Y., Han, J., and Zhao, Q. (2008) Screening of Chinese medicinal plants for inhibition against clinical isolates of methicillin-resistant Staphylococcus aureus (MRSA). J. Ethnopharmacol. 120, 287-290). 국내에서도 전통적으로 사용해온 약용식물로부터 MRSA에 대한 항균 소재를 발굴하고, 유용 물질을 분리하는 연구가 진행되고 있다. 한약재로 광범위하게 사용되고 있는 감초 (Glycyrrhiza uralensis)의 MRSA의 내성유발 유전자의 저해활성이 보고되었으며 (Lee, J. W., Ji, Y. J., Yu, M. H., Hwang Bo, M. H., Seo, H. J., Lee, S. P., and Lee, I. S. (2009) Antimicrobial effect and resistant regulation of Glycyrrhiza uralensis on methicillin-resistant Staphylococcus aureus. Nat. Prod. Res. 23, 101-111), 단삼 (Salvia miltiorrhiza), 소나무 (Pinus densiflora), 관중 (Dryopteris crassirhizoma) 등 항 MRSA 활성을 갖는 한약재에 대한 연구 결과가 보고되었다 (Cha, J. D. 선복화는 초롱꽃목 국화과의 여러해살이 풀로서 금불초 (Inula japonica Thunberg) 또는 구아선복화 (Inula britannica Linne)의 꽃을 약용으로 사용하고 있다. 선복화의 정유 (essential oil)에는 quercetin, isoquercetin, caffeic acid, cholorgenic acid, taraxasterol 등 많은 종류의 sterol이 함유되어 있으며, 한의학에서는 진토, 거담, 기관지 경련 이완 및 이뇨 작용이 있다고 하여, 가래가 나오는 증상이나 소화불량 등에 널리 사용한다. 최근에는 선복화로부터 대장암, 유방암 및 자궁 경부암에 대한 항암활성이 보고되었으며, Qin 등 (2008)은 선복화로부터 폐암 세포 및 대장암 세포에 대하여 항암작용을 지닌 물질을 분리하였다. 또한, 항산화, 신경세포 및 간세포 보호 활성을 갖는 선복화의 유용 성분이 보고되었다 (Khan, A. L., Hussain, J., Hamayun, M., Gilani, S. A., Ahmad, S., Rehman, G., Kim, Y. H., Kang, S. M., and Lee, I. J. (2010) Secondary metabolites from Inula britannica L. and their biological activities. 최근 본 발명자들은 대한민국 공개특허 제10-2011-0008483호에 고병원성 조류인플루엔자 H5N1에 항바이러스 활성을 갖는 선복화 추출물을 함유하는 H5N1 감염 예방 및 치료용 조성물을 개시한 바 있다.
그러나, 선복화는 식품공전 상 식품의 제한적 사용 원료로서 사용할 수 있도록 등재되어 있기 때문에 안전성 높은 천연 항균소재로서 유용성이 높은데도 불구하고, 지금까지 메치실린-내성 황색포도상구균 등에 대한 선복화 추출물의 항균활성에 대하여는 보고된 바 없다.
따라서, 본 발명의 목적은 선복화 추출물로부터 메치실린-내성 황색포도상구균에 대해 항균활성을 갖는 항균용 조성물을 제공하는데 있다.
본 발명의 상기 목적은 선복화 추출물의 제조 단계와; 상기 제조된 선복화 추출물의 메치실린-내성 황색포도상구균 (MRSA)에 대한 항균활성 검증 단계와; 선복화 추출물에 의한 MRSA의 형태학적 변화 조사 단계와; 선복화 추출물에 의한 MRSA의 유전학적 항균 메커니즘 조사 단계를 통하여 달성하였다.
본 발명은 메치실린-내성 황색포도상구균에 대해 항균활성을 갖는 선복화 추출물을 유효성분으로 함유하는 항균용 조성물을 제공하는 뛰어난 효과가 있다.

이하, 본 발명의 구체적인 내용을 바람직한 실시예와 실험예를 통하여 상세히 설명한다.

이하, 본 발명의 구체적인 내용을 바람직한 실시예와 실험예를 통하여 상세히 설명한다.
공시재료 : 균주 및 배지
본 발명 공시균주인 메치실린-내성 황색포도상구균 (MRSA, methicillin-resistant S. aureus)는 S. aureus ATCC 33591, ATCC 33593 및 ATCC 33594이며, 각 균주는 20% glycerol stock법을 이용하여 -70℃에서 보관하였으며, tryptic soy agar (TSA, Difco Laboratories, Detroit, MI, USA)와 tryptic soy broth (TSB, Difco Laboratories, Detroit, MI, USA)에서 2-3회 계대하여 사용하였다. 균일한 접종량을 위해 균주의 배양액을 0.5 McFarland standard turbidity에 적용한 후 희석하여 사용하였다.
메치실린-내성 황색포도상구균 (MRSA)에 대한 항균활성 검증용 선복화는 서울특별시 제기동 약재시장에서 건조된 형태로 구입하였으며, 마쇄기를 사용하여 분말형태로 제조하였다.
이하, 본 발명의 구체적인 방법을 실시예와 실험예를 들어 상세히 설명하고자 하지만 본 발명의 권리범위는 이들 예에만 한정되는 것은 아니다.
실시예 1 : 선복화 추출물의 제조
마쇄기를 사용하여 분쇄된 선복화 분말 30 g을 99% 메탄올 300 mL에 침지시킨 후, 24시간 동안 실온에서 정치하여 추출하였다. 추출액을 Whatman No. 2 여과지를 이용하여 여과한 후 감압 하에 40℃에서 농축하였다. 선복화 농축물은 감압 동결건조를 통하여 건조시킨 후 4℃에서 보관하면서 시료로 사용하였다. 상기 선복화 추출물의 제조 과정을 도 1에 나타내었다.
실험예 1 : 선복화 추출물의 항균활성 검증
선복화 추출물의 MRSA에 대한 항균활성을 검증하기 위해서, disc diffusion method를 사용하였다. 즉, TSB에서 24시간 동안 35℃에서 배양된 MRSA의 배양액 (1.5×108 CFU/mL) 100 μL을 20 mL의 TSA를 분주하여 굳힌 plate에 균일하게 도말하였다. 상기 실시예 1에서 제조된 동결 건조된 선복화 추출물을 DMSO (dimethyl sulfoxide)에 50 ㎎/mL의 농도로 용해시킨 다음, 0.45 ㎛ membrane filter를 사용하여 여과하였다. 여과된 시료액 50 μL를 지름 8 ㎜의 paper disc에 흡수시킨 뒤, MRSA의 균이 도말되어 있는 TSA 위에 올려 놓은 후 35℃에서 24시간 동안 배양하였다. 대조군으로 시료를 포함하지 않은 DMSO와 gentamicin (AMRESCO Inc., Ohio, USA)을 사용하였다. 24시간 배양 후 disc 주변에 형성된 저해환 (inhibition zone)의 지름을 측정하여 disc 주변의 저해환 형성 여부를 관찰하였다.
실험결과 선복화 추출물에 의해 생성된 S. aureus ATCC 33591의 저해환 직경은 13-14 ㎜로 관찰되었다. 실험에 사용된 또 다른 MRSA (ATCC 33593, ATCC 33594)에 대해서도 12 ㎜의 저해환이 형성되어 MRSA에 대한 선복화 추출물의 항균활성을 확인할 수 있었다.
실험예 2 : 선복화 추출물의 최소저해농도 (MIC, minimal inhibition concentration) 및 최소사멸농도 (MBC, minimal bactericidal concentration) 측정
상기 실험예 1의 diffusion method를 이용하여 저해환의 크기를 기준으로 항균활성을 분석하는 것은 추출물 중의 불용성 항균 물질이 agar 배지에 동일하게 확산되지 않는 난점이 있으므로 천연 소재 추출물의 상대적인 항균활성을 분석하기에는 정확도가 낮다. 따라서, 선복화 추출물의 항균력을 microdilution method를 이용하여 최소저해농도 (MIC) 및 최소사멸농도 (MBC)를 측정하였다. 즉, DMSO에 용해된 선복화 추출물을 멸균된 TSB를 이용하여 2배 희석한 뒤, 96-microwell plate에 100 μL씩 분주한 후, 각 well에 100 μL의 배양액 (약 105 CFU/mL)을 추가하여, 시료의 최종농도를 0.039-2.5 ㎎/mL로 하였다. 37℃에서 24시간 배양한 후 630 ㎚에서 흡광도의 변화를 측정하였으며, 최소저해농도 (MIC)는 630 ㎚에서 흡광도의 변화가 0.05 이하일 때의 농도로 정의하였다. 최소사멸농도 (MBC)는 흡광도의 변화가 0.05 이하의 well (선복화 추출물 0.625-2.5 ㎎/mL을 처리한 well) 에서 100 μL의 배양액을 취한 후 TSA에 도말하여, 균의 성장이 관찰되지 않았을 때의 농도로 정의하였다.
실험결과 하기 [표 1]에 나타낸 바와 같이, S. aureus ATCC 33591에 대한 선복화 추출물의 최소저해농도는 0.625 ㎎/mL로 관찰되었으며, S. aureus ATCC 33593 및 S. aureus ATCC 33594에 대한 선복화 추출물의 최소저해농도는 1.25 ㎎/mL로 확인되었다. 최소사멸농도는 본 발명실험에 사용된 모든 균주에 대해서 2.5 ㎎/mL로 확인되었다.

실험예 3 : 시간에 따른 항균력 측정
선복화 추출물의 MRSA 균주에 대한 시간에 따른 사멸효과를 측정하기 위해서 S. aureus ATCC 33591을 사용하여 관찰하였다. 선복화 추출물에 대한 대조군으로서 DMSO를 사용하였다. 100 μL의 S. aureus ATCC 33591 배양액 (5×106 CFU/mL)을 추출물 (2.5 ㎎/mL)을 포함한 10 mL의 TSB에 접종한 후, 35℃에서 24시간 동안 배양하면서 균의 생장을 관찰하였다. 접종 후 0, 4, 8, 12 및 24 시간에 배양액으로부터 100 μL를 취하여 희석한 후 TSA에 도말하여 35℃에서 24시간 동안 배양한 후 균의 집락을 계수하였다.
실험결과 도 2에 나타낸 바와 같이, S. aureus ATCC 33591은 배양 12시간 만에 9 log CFU/mL까지 성장한 반면, 최소사멸농도인 2.5 ㎎/mL에서는 균의 성장이 저해되었으며, 배양 24시간 후에는 완전히 사멸되었다.
실험예 4 : 선복화 추출물에 의한 균의 형태학적 변화 조사
선복화 추출물에 의한 MRSA의 형태학적 변화를 관찰하기 위해서, S. aureus ATCC 33591을 선복화 추출물에 처리한 후 주사전자현미경을 이용하여 관찰하였다. 즉, S. aureus ATCC 33591을 0.625 ㎎/mL의 농도로 선복화 추출물을 함유하는 TSB에 접종하여 37℃에서 24시간 배양한 후 배양액을 원심분리하고, 균체를 PBS로 2-3회 세척하였다. 세척된 균체에 2.5% glutaraldehyde 용액을 투입한 후 4℃에서 균을 고정시켰다. 고정된 균체를 60-100% ethanol을 순차적으로 이용하여 탈수시킨 후 데시케이터에서 건조하였다. 건조된 균체를 금 (gold)으로 코팅하고, 주사전자현미경 (SNE-3000M, SEC Co., Suwon, Korea)으로 관찰하여, 선복화 추출물에 의한 MRSA의 형태학적 변화를 검증하였다.
실험결과 도 3에 나타낸 바와 같이, 선복화 추출물을 처리한 TSB에서 배양한 균의 성상을 추출물을 처리하지 않은 TSB에서 배양한 균의 성상과 비교하였을 때, 세포의 표면이 거칠고, 수축되어 있으며, 막의 파괴로 인해 사멸된 세포들이 관찰되어 선복화 추출물에 의한 균의 사멸을 확인하였다.
일반적으로, 천연소재에 함유된 유용 항균물질들은 세포 표면에 결합하여, 세포막을 파괴시킨다. 즉, 천연항균 물질들은 프로톤 구동력 (proton motive force), 호흡연쇄작용 (respiratory chain) 및 전자전달체계 (electron transfer process)를 저해하여, 결과적으로 증식 및 생존에 필요한 DNA, RNA, 단백질, 지질 및 당의 합성을 저해함으로써 항균작용을 나타낸다.
실험예 5 : 선복화 추출물의 유전학적 항균 메커니즘 조사
선복화 추출물에 의한 MRSA의 유전학적 항균 메커니즘을 분석하기 위해서 S. aureus ATCC 33591 (약 109 cells)을 선복화 추출물이 각 0.25 ㎎/mL 및 2.5 ㎎/mL의 농도로 용해되어 있는 10 mL의 PBS에 접종한 후 24시간 동안 35℃에서 배양하였다. 배양 종료 후 원심분리기를 이용하여 균체를 세척 및 회수한 후, RNA isolation kit (EzWayTM Total RNA Isolation Kit, KOMA BIOTECH, Seoul, Korea)을 사용하여 RNA를 추출하였다. 상기 추출한 RNA 시료에 PCR buffer, Taq, dNTP, primer 및 cDNA를 첨가하여 RT-PCR를 실시하였다. 본 발명 PCR에 사용된 primer의 염기서열은 다음과 같다: mecA (F: 5'-ATGAGATTAGGCATCGTTCC-3', R: 5'-TGGATGACAGTACCTGAGCC-3'), mecI (F: 5'-CTGCAGAATGGGAAGTTATG-3', R: 5'-ACAAGTGAATTGAAACCGCC-3'), mecRI (F: 5'-AAGCACCGTTACTATCTGCACA-3', R: 5'-GAGTAAATTTTGGTCGAATGCC-3'), femA (F: 5'-CATGATGGCGAGATTACAGGCC-3', R: 5'-CGCTAAAGGTACTAACACACGG-3'). PCR 조건은 initial denaturation 과정으로 95℃에서 15분 간 반응시켰다. 95℃에서 5분 동안 denaturation, 60℃에서 1분 간 annealing, 72℃에서 1분 간 extension 과정을 1 cycle로 하여 30회 시행하였다. Final extension으로 72℃에서 10분 동안 처리한 후 1% agarose gel을 이용하여 전기영동을 실시하였으며, marker와의 비교를 통해 femA, mecA, mecRI 및 mecI의 발현 정도를 확인하였다.
MRSA 세포벽의 생산효소인 penicillinase와 PBP 2a 전사 관련 단백질인 mecA의 발현에 대한 선복화 추출물의 저해활성을 조사하기 위해서 S. aureus ATCC 33591을 사용하여 실험한 결과, mecA의 발현이 농도 의존적으로 감소하는 것을 관찰하였다 (도 4). 대조군과 비교하였을 때, MBC (2.5 ㎎/mL)에서 PBS 2'을 유도하는 mecRI의 발현이 억제된 반면, PBP 2' 생산을 억제하는 mecI의 발현은 증가한 것으로 나타났다. 따라서, 선복화 추출물이 PBP 단백질의 PBP 2'으로의 전환하는 경로에 관여함으로써 MRSA에 대하여 항균활성을 갖는 것으로 판단되었다.
이상에서 설명한 바와 같이, 본 발명은 메치실린-내성 황색포도상구균에 대해 항균활성을 갖는 선복화 추출물을 유효성분으로 함유하는 항균용 조성물을 제공하는 뛰어난 효과가 있으므로 천연항생제 산업 상 매우 유용한 발명인 것이다.
Claims (4)
- 선복화 추출물을 유효성분으로 함유하는 항균용 조성물.
- 제1항에 있어서, 상기 선복화 추출물은 메탄올로 24시간 동안 실온에서 추출한 것이 특징인 항균용 조성물.
- 제1항 내지 제2항 기재의 선복화 추출물은 메치실린-내성 황색포도상구균에 대해서 항균활성을 갖는 것이 특징인 항균용 조성물.
- 제3항에 있어서, 메치실린-내성 황색포도상구균은 S. aureus ATCC 33591, ATCC 33593 또는 ATCC 33594 균주 중의 어느 하나인 것이 특징인 항균용 조성물.
