FIELD OF THE INVENTION The present invention relates to organic-inorganic nanofiber composite substrates for trace analysis of materials based on Surface Enhanced Raman Scattering (SERS), more particularly metals derived from various Raman dyes. The present invention relates to a nanofiber composite substrate in which aggregates of nanoparticles are uniformly embedded at various densities inside a nanofiber having crosslinkable polymer hydrogel properties, and a method of manufacturing the same, and an application thereof.
As the environmental pollution problem becomes serious, the need for early detection of environmental pollutants such as various heavy metals and organophosphorus compounds is increasing. In addition, as the threat of terrorism increases in the international community, the need for early detection of various bioterrorism substances such as anthrax and sarin is increasing. Therefore, it is very important to quickly and accurately detect such dangerous substances in the field and to block their diffusion, and the development of trace amount analysis techniques of various chemicals is environmental monitoring, forensic science, and homeland defense. This is a very important issue in the field.
Existing analytical technologies such as environmental pollutants require complicated pretreatment processes, and also require large-scale analysis equipment that is not mobile or very inconvenient.
In this regard, recently, Surface Enhanced Raman Scattering (SERS) has been spotlighted as a method of detecting and biochemically analyzing chemicals due to its high sensitivity.
In this area, research continues on specific structures that can be used as reliable SERS substrates. In order to bring the gold and nanoparticles into the desired shape and shape, attention is drawn to ion beam lithography, electron beam lithography, nanosphere lithography and vacuum evaporation, and methods of gathering small nanoparticles into nanoclusters have been used. These techniques are used to make nanoclusters of gold and silver nanoparticles with highly reproducible SERS.
However, these technologies are expensive and difficult to mass produce. Coating silver nanoparticles with silicon nanowire arrays to produce an economical, stable and reliable SERS substrate [Zhang, M. L., C. Q. Yi, et al. 2008 Applied Physics Letters 92 (19); And Zhang, B. H., H. S. Wang, et al. 2008 Advanced Functional Materials 18 (16): 2348-2355], porous aluminum membranes with gold nanoparticles [Ko, H. and VV Tsukruk 2008 Small 4 (11): 1980-1984.], Hollow gold nanospheres and nanoshells Antibodies conjugated with Lee, S., H. Chon, et al. 2009, Biosensors & Bioelectronics 24 (7): 2260-2263. However, there is a disadvantage that the enhancement effect does not occur effectively because the increase in molecular vibration occurs so that the number of localized plasmon centers decreases.
Currently, substrates available for SERS have various defects in the enhancement and detection of Raman scattering. Raman scattering signals are generally very weak, especially on flat substrates. Weak Raman signals make it difficult to detect and measure Raman scattering signals, thus making it difficult to detect and identify molecular species. Moreover, even though the usable substrate augments the Raman scattering signal, the enhanced Raman scattering signal is usually only in the localized area of the substrate and is not uniform across the substrate surface. The area of the locally enhanced Raman scattering signal is exponentially small compared to the entire area of the substrate surface. Large gaps in the area of the Raman scattering signal and the area of the substrate surface are burdensome to locate and locate the signal and thus to detect and identify molecular species.
Accordingly, the present inventors prepared aggregates of metal nanoparticles induced by Raman dye, and the metal nanoparticle aggregates were crosslinked polymer hydrogel using an electrohydrodynamic (EHD) jetting method. The present invention was completed by confirming that the signal strength for an analyte can be effectively compared and detected.

Hereinafter, the present invention will be described in detail.
The present invention relates to a substrate for use in Surface enhanced Raman scattering (SERS) analysis.
Nnao Lett. 6, 2225-2231). Surface enhanced Raman scattering (SERS) -based detection methods are in the spotlight for high sensitivity analysis. Analyte-containing solutions and immuno-gold nanoparticles with Raman reporters can be added sequentially and specific markers can be quantitatively analyzed by measuring the change in intensity of the characteristic SERS peak of the reporter molecule. When the reporter molecule is adsorbed on a rough metal surface and exposed to excitation light (laser light), electromagnetic and chemical enhancement occurs at the SERS active site of the reporter molecule, known as the hot spot junction, resulting in a significant increase in the SERS signal. Kneipp, J. et al., 2006. Nnao Lett. 6, 2225-2231). This enhancement effect solves the problem of low sensitivity, which is a disadvantage of conventional Raman and fluorescence detection methods.
SERS has evolved into a technology that can measure bioimaging and bio signals with high sensitivity at the single molecule level. Raman signals of different molecules adsorbed on nanostructured metal surfaces are very high in the case of silver, gold and copper. When Raman active molecules are adsorbed on the rough metal substrate surface, their SERS signal increases. The reason for this is the electromagnetic and chemical enhancement that occurs at the highly localized plasmon centers of hot spot junctions. Electromagnetism is associated with local surface plasmon resonance associated with rough metal substrates or metal aggregate surfaces. Chemical enhancement, on the other hand, is associated with direct charge transfer or hole-pair excitation processes.
To detect single molecule levels, the SERS signal must be greatly enhanced. Silver colloidal nanoparticle aggregates were first used for SERS detection of single molecules [Nie, S. M. and S. R. Emery 1997 Science 275 (5303): 1102-1106.]. Subsequent studies have shown that silver nanoclusters are best suited for ultrasensitive detection [Kneipp, J., H. Kneipp, et al. 2006, Nano Letters 6 (10): 2225-2231.]. If the Raman reporter molecule is present at the junction of the nanocluster, the signal strength increases tremendously [Chen, J. W., Y. Lei, et al. 2008, Analytical and Bioanalytical Chemistry 392 (1-2): 187-193.]. The distance between the particles, the inherent characteristics of the metal plasmon region, the size and concentration of the nanoparticles and the laser intensity are important parameters controlling signal enhancement.
Research into specific structures that can be used as SERS substrates or substrates continues. In order to bring gold and nanoparticles into the desired shape and shape, attention is drawn to ion beam lithography, electron beam lithography, nanosphere lithography and vacuum evaporation, and methods of gathering small nanoparticles into nanoclusters are used, but these techniques are costly. There is a problem that it is heavy and difficult to mass-produce, and the stability of the nanoclusters in different chemical and biological environments is very important.
In addition, since most surface-enhanced Raman scattering substrates, which are conventionally developed as chemical sensors for chemical microanalysis, have no internal standard signal due to surface-enhanced Raman scattering induced by Raman dyes, There is a difficulty in reproducible trace analysis.
Currently developed surface-enhanced Raman scattering substrates have been limited in the core technology required for trace analysis because of the limited detectable portion.
However, the nanofiber composite substrate for surface-enhanced Raman scattering analysis of the present invention is a new concept substrate that satisfies these requirements, and enables high-speed and high-sensitivity trace analysis of analytes, and realizes chemical stability and data reproducibility.
That is, the present inventors produce aggregates (clusters) of metal nanoparticles induced by various Raman dyes of metal nanoparticles for high speed and high sensitivity trace analysis and biosensing, and the metal nanoparticle aggregates are subjected to electrohydrodynamics. Chemically stabilized by uniform incorporation into various concentrations of nanofibers with polymer (hydrogel) hydrogel properties using electrohydrodynamic (EHD) jetting. By spreading the analyte through the swollen sturucture of the organic-inorganic nanofiber composite substrate, not only high-speed and high sensitivity trace analysis for detecting dangerous drugs, but also chemical stability and data reproducibility were realized.
Therefore, the present invention in one aspect
In the cross-linkable polymer hydrogel nanofibers, aggregates (clusters) of metal nanoparticles by Raman dyes are uniformly distributed, the surface-enhanced Raman scattering analysis nanofiber composite substrate and a method for manufacturing the same It is about.
The substrate of the present invention contains nano clusters, which are aggregates of metal nanoparticles induced by Raman dye, in a uniform density inside a nanofiber having crosslinkable polymer hydrogel properties.
In particular, in order to stabilize organic-inorganic nanofiber composite substrates in aqueous conditions, the polymer hydrogel nanofibers form physically, chemically or photoinitially crosslinks, for example, thermal chemicals. crosslinking or photo-initiated chemical crosslinking that minimizes loss of surface-enhanced Raman scattering signals at high temperatures to form crosslinks and swollen in aqueous solution.
Through this swollen hydrogel substrate, analytes dissolved in an aqueous solution are diffused and adsorbed onto the surface of the metal nanoparticle aggregates, thereby allowing the analysis of materials based on the surface-enhanced Raman scattering signal induced by the Raman dye. High speed and high sensitivity trace analysis is possible.
At this time, based on the constant intensity of the internal standard signal by the surface-enhanced Raman scattering induced by the Raman dye present in the surface-enhanced Raman scattering substrate of the present invention, Raman for the analyte By analyzing the signal strength comparatively, the analyte can be effectively detected.

Electrohydrodynamic jetting (EHD jetting) is commonly used as an easy technique for the fabrication of polymeric nanofibers and nanoparticles of different shapes and sizes. Manipulation of mainly adjustable parameters can include: 1) viscosity of the polymer solution, electrical conductivity, and surface tension, 2) voltage and distance applied between the two electrodes, and 3) a range of polymeric nanometers to a few nanometers. It includes the flow rate that results in creating the structure. Among other things, electrohydrodynamic co-jetting (EHD co-jetting) is a charge from the vertex of a Taylor cone where the equilibrated laminar flow of the polymer solution is two phases away. -To be jetted in a thin jet stream by charge to charge repulsion.
The crosslinkable polymer (polymer) can be used without limitation as long as it is a hydrophilic polymer.
The polymer of the present invention is a copolymer polymer capable of crosslinking. For example, poly (acrylamide-co-acrylic acid, poly (AAm-co-AA) ], Poly (N-isopropyl acrylamide-co-stearyl acrylate) [Poly (N-isopropyl acrylamide-co-stearyl acrylate), Poly (NIPAm-co-SA)], poly (N-isopropyl acrylamide Group consisting of poly (N-isopropylacrylamide-co-allylamine), poly (NIPAM-co-AA), and poly (NIPAM-co-AA) having an acrylic moiety. Preferred but not limited to In one embodiment of the invention poly (acrylamide-co-acrylate) [poly (AAm-co-AA)] was used.
The "cross-linking" of water soluble polymers is very important for giving resistance to dissolution when exposed to water. Internal molecular crosslinking of the polymer chains helps these nanofibers maintain their original structure in substantially solubilizing environments such as water-soluble conditions, which also allows the nanofibers to act as soft hydrogels containing significant amounts of water. To help. Hydrogels composed of biodegradable polymers are widely used in tissue repair and drug delivery carriers.
Crosslinking of hydrogels can be accomplished through a variety of mechanisms including physical, chemical and photoinitiated crosslinking. Physically or chemically crosslinked nanostructures can be used in a stable and water soluble environment. In particular, the fabrication of multiple compartments of hydrogel nanoparticles and nanofibers relies on the rheological compatibility and dynamic order of different polymer solutions under an externally applied electric field during electrohydraulic co-injection. The maintenance of the hydrodynamic properties of the polymer solution during electrohydraulic co-injection can be the most critical parameter for producing uniform multi-part nanoparticles and nanofibers. Chemical crosslinking in nanofibers requires a crosslinking agent to be added to the polymer solution, which can interfere with rheological properties during the electrohydraulic coinjection process. However, physical crosslinks based on electrostatic and hydrophobic interactions create rheological property constants through electrohydraulic co-injection without crosslinkers.
Therefore, when the metal nanoclusters encapsulated with the crosslinkable polymers (polymers) of the present invention are heated or subjected to ultraviolet rays, crosslinks are formed between the polymers surrounding the metal nanoclusters and thus the metal nanoclusters-polymers are formed. A network is formed. And, due to the formation of a network through such a crosslinking is not dissolved in water.

The nanofiber composite substrate for surface-enhanced Raman scattering analysis of the present invention prepared by the above method is characterized in that agglomerates of metal nanoparticles by Raman dyes are uniformly distributed inside the polymer hydrogel nanofibers.
At this time, it is possible to uniformly distribute the aggregates of the metal nanoparticles at various densities through (i) controlling the type and concentration of Raman dye, (ii) controlling the density of the metal nanoparticle aggregates, and (iii) controlling the electrohydrodynamic spraying.
In addition, the nanofiber composite substrate for surface-enhanced Raman scattering analysis of the present invention is not difficult to manufacture the manufacturing cost is also economical.
The dye (Raman dye) induced in the aggregate of metal nanoparticles of the desired cluster size is stabilized by protein coating and subsequently encapsulated using a polymer (polymer).
As described above, the polymeric polymers are biohybrid, non-toxic and hydrogel polymers, and the encapsulation uses electrohydraulic injection techniques. The polymer swells in a water-soluble environment and becomes a soft matrix to hold metal nanoclusters well. Electrohydrodynamic jettig (EHD injection) is a technique that is well used to encapsulate many inorganic nanoparticles inside various polymers [Yoshida, M., KH Roh, et al. 2009, Advanced Materials 21 (48): 4920]. After EHD injection, the polymer polymer is thermally or light cured to stabilize the nanoprobe placed in aqueous media. Crosslinked polymers with Raman active centers therein can be used as sensitive SERS substrates for biosensing and bioimaging.
On the other hand, the present invention relates to a material analysis method using the nanofiber composite substrate for surface-enhanced Raman scattering analysis in another aspect.
There is no limit to the substance to be analyzed. For example, dipicolinic acid (DPA), malachite green (MG), and the like can be usefully detected and analyzed as a dangerous substance such as a carcinogen. In one embodiment of the present invention, malachite green (MG) was used as an analyte.
Malachite green is a green industrial pigment and antibacterial agent that has been used in aquaculture for parasites, fungi and bacterial infections, but since its use in the United States in 1991, malachite green has been found to be highly toxic to fish. In 2002, Europe and China also regulated the use of malachite greens for edible fish in 2002. Korea also regulates toxic chemicals.
In particular, the surface-enhanced Raman scattering analysis nanofiber composite substrate itself has a Raman dye used in the manufacture of metal nano-cluster, so that its internal standard signal exhibiting a constant intensity by surface-enhanced Raman scattering (Internal standard) On the basis of the signal, there is an advantage that can be effectively detected by comparing the Raman signal strength for the analyte.
Conventionally, the surface-enhanced Raman scattering substrate itself, which is mostly developed as a chemical sensor for chemical microanalysis, does not have an internal standard signal due to surface-enhanced Raman scattering induced by Raman dye. Although reproducible trace analysis was difficult, the present invention solves this problem since an internal standard signal exists.
In one embodiment of the present invention, malachite green (MGITC), a carcinogenic substance dissolved in a small amount in solution, is diffused through the organic-inorganic nanofiber composite substrate, which is a swollen hydrogel, to surface a metal nanoparticle aggregate. Malachite green was detected and analyzed by observing surface-enhanced Raman scattering spectra according to its concentration, which appeared by adsorption to.
In particular, Raman signal strength with increasing concentration of malachite green, compared to the constant internal standard signal intensity due to surface-enhanced Raman scattering induced by RBITC Raman dye present on the surface-enhanced Raman scattering substrate itself Quantitative evaluation was performed by observing changes in.
The figure shows a schematic diagram for trace analysis of MGITC according to SERS technology according to an embodiment of the present invention. Crosslinked polymeric nanofibers include hydrogel properties, allow dye molecules to penetrate the matrix, and allow them to be absorbed on silver nanoclusters. The incoming laser beam interacted with the staining molecules absorbed on the silver nanoparticle aggregates, generating respective Raman signals from the substrate before and after the MGITC solution immersion.
On the other hand, according to the method of the present invention, the MGITC concentration range of 0.5-6 ppm and the SERS signal have a linear linear relationship, and the limit concentration of the analyte that can be detected is defined as a limit of detection (LOD). To date, according to the embodiment LOD is possible up to about 0.5 ppm (part per million), it can be useful for the detection of a wide range of target substances present in trace amounts. Liquid chromatography mass spectroscopy (LC-MS) is a typical conventional technique used as a major microanalysis method, and the LOD of SGI-based MGITC analytes has been reported to about 1-2 ppb (part per billion) to date. However, detection limits corresponding to the LOD of LC-MS are possible.
In the present invention, (i) controlling the type and concentration of Raman dye, (ii) controlling the density of the metal nanoparticle aggregates, and (iii) controlling the electrohydrodynamic spraying to uniformly distribute the aggregates of the metal nanoparticles at various densities to about 1 ppb or less. It is possible to overcome the detection limit corresponding to the LOD of LC-MS.
As described above, the present invention uses a nanofiber composite substrate for surface-enhanced Raman scattering analysis, characterized in that agglomerates of metal nanoparticles by Raman dye are uniformly distributed in the crosslinkable polymer hydrogel nanofibers. The present invention relates to a method for effectively detecting and comparing a trace amount of a substance.

The results are shown in FIG.
2 (A) shows absorbance spectra collected at different time intervals during silver nanoparticle aggregate formation. As individual particle sizes increased with increasing incubation time, they had large and narrow absorbance peaks at 400 nm. The major absorbance peak at 400 nm shifted to a longer wavelength up to 429 nm in 60 minutes. Additional peaks observed at 649 nm at 50 minutes indicate cluster formation. RBITC had a characteristic peak at 555 nm.

And, the transmission electron microscope confirmed the diameter and shape of single silver nanoparticles and silver nanoclusters (AgNCs).

FIG. 2 (C) shows that the clusters range in size from 60 to 100 nm after 60 minutes of incubation time with a TEM image, and FIG. 2 (D) shows that single silver seed particles range in size from 6 to 13 nm. As the incubation time increased, the individual particle size also increased, and the coalescence between the single particles also resulted in the formation of silver nanoparticle aggregates which were uniquely observed in the UV-Vis spectrum.
Raman signal measurement
In addition, Raman signals were measured during the production of AgNCs.
Figure 2 (E) shows the increasing Raman signal of the silver nanoparticle aggregates over the course of the aggregation reaction time. Maximum Raman signal was obtained after 60 minutes of incubation time. Very long incubation times had an adverse effect on signal strength, probably due to larger cluster formation.
2 (F) shows intensity plots of two major Raman peaks over RBITC spectra versus incubation time. There was no noticeable difference in the position of the peak among all the spectra collected at different time intervals.
The RBITC concentration was optimized so that the cluster had a maximum Raman signal after growing to a size of 60-100 nm. Isothiacinate dyes such as RBITC and MGITC have a strong aggregation tendency compared to other dyes. The optimal dye concentration of 7.5 μM RBITC over 60 min incubation time was found to produce the maximum SERS signal.
Colloidal stability of silver nanoparticles is confirmed by the capping of negatively charged citrate ions around the silver nanoparticles. This relies on the principle that the aggregation mechanism of silver nanoparticles by Raman dye decreases as the electrostatic repulsion between nanoparticles by anions decreases as the Raman dye is absorbed on the surface of the metal nanoparticles. The size of the individual particles increases due to the additional deposition of silver and the dye molecules have a high probability of being captured at the junction of the particles to be merged.
This kind of structure is associated with high Raman signals due to the high electromagnetic fields present at the spot. The small silver seed nanoparticles had very low Raman intensity, as clearly shown in the graph of increasing nanoparticles until the maximum particle size of individual nanoparticles 25-30 nm and average aggregate (cluster) size 76 nm. After a long incubation period of 80-90 minutes, the aggregate (cluster) size was quite large and precipitated quickly resulting in a decrease in signal strength.
A useful meaning of aggregate (cluster) formation is to increase the turbidity of the mixture, the appearance of added peaks in the UV-Vis spectrum, and the maximum Raman signal. AgNCs in the polymer nanofibers are encapsulated during electrospinning and the polymer crosslinks by exposure to UV light.
To confirm this, the obtained fibers were exposed to water and observed by CLSM imaging in a swollen state.
As a result, as shown in FIG. 3, the Raman signal was very stable and retained their original characteristic peaks before and after optical crosslinking.
There was no significant reduction in the signal after UV crosslinking as compared to the decrease in the signal in thermal crosslinks which resulted in a significant decrease in Raman signal by desorption of the dye at high temperatures, for example at high temperatures.
Relationship
Based on the results of previous experiments on incubation time, crosslinking type, temperature, etc., Raman signals can be adjusted to various parameters, such as the number of parameters that can be easily adjusted by controlling the amount of AgNCs in the polymer nanofibers or the number of hot spots. It can be expected that it can be adjusted by.
Therefore, the SERS signal characteristics according to various parameters were investigated.
Another mechanism that occurs simultaneously in nanofibers is the partial hot fusion of single nanoparticles at the binding sites where the dye molecules are trapped, which is a potential hotspot where SERS signals are enhanced because of the large number of dye molecules located at the binding sites. Such dye aggregation is useful to achieve high signal enrichment. Therefore, the experiment was utilized for temperature, incubation time, dye concentration, temperature and amount of silver ions added.
5-1 Relationship with temperature
First, it can be seen that the SERS signal has an adverse effect at high temperatures.
Although other dyes have different stability as a signal at high temperatures, compared to other dyes such as R6G and CV, for dyes containing isothiocyanate groups, the dyes of the metal-sulfur bond between the isothiocyanate group and the metal particles It is stabilized due to bond formation. The low signal is thought to be due to the desorption of the dye from the silver nanoparticles which reduces the SERS effect. Most organic dyes have a stable chemical structure at 175 ° C. (thermal crosslinking temperature), eliminating the possibility of dye degradation.
However, photocrosslinking of nanofibers with Raman active centers inside was achieved under moderate conditions that prevented significant degradation of the SERS signal.
Successful polymerization of P (AAm-co-AA) and crotonyl chloride was confirmed from the H 1 NMR spectrum.
Pure P (AAm-co-AA) showed no characteristic peaks of double bonds in crotonyl chloride at 6.640, 6.957,6.974, 6.977,6.996,7.013 and 7.030, which was also seen in the modified polymer as shown in FIG. .
5-2. 5-2. AgNCsAgNCs 양과의 관계 Relationship with sheep
In addition, Raman signal intensity varied depending on the amount of AgNCs in the polymer nanofibers, while maintaining other unchanging parameters such as spinning time, dye concentration, cluster size and individual silver particle size.
In particular, as shown in FIG. 4, the relationship between Raman intensity and AgNCs amount was shown in a linear graph. Therefore, it can be predicted that the introduction of various amounts of AgNCs can regulate the SERS signal.
Plots of Raman intensity of specific peaks in the RBITC spectrum with respect to the amount of AgNCs (silver nanoparticle aggregates) encapsulated in the nanofibers are shown in FIG. 4 (C, D).
1645.9 cm −1 was selected as the reference peak from the RBITC spectrum and the signals at different AgNCs concentrations were compared. It was also found that by introducing different amounts of AgNCs in the polymer nanofibers, the desired Raman signal could be easily controlled.
5-3. Relationship to solvent
The Raman signal stability of AgNCs encapsulated in nanofibers was then analyzed by immersing these fiber mats in different solvents.
The change in Raman signal intensity in other solvents, ie DI water, acetone and ethanol, is shown in FIG. 5 (A).
The signal was stable and no noticeable degradation following exposure to this solvent was seen. It is considered that due to the crosslinking of the polymer shell by UV irradiation, decomposition of the silver nanoclusters that stabilize the SERS substrate is prevented.
5-4. Reproducibility of the signal
On the other hand, one of the most important forms of SERS substrate is the reproducibility of the SERS signal, especially for quantitative purposes. Raman spectra were collected at 14 randomly selected spots on the surface of the nanofiber substrate to confirm signal reproducibility.
These results are shown in FIG. 5 (B).
The intensity of the main peak is indicated in other spots. Significantly reproducible signals appeared with small deviations from the mean values.
And salt concentration
In addition, signal stability at different pH values was analyzed in the range of 1.5-11.0 by changing the pH value using 1.0 M NaOH and 0.5 M HCl at room temperature.
As a result, as shown in Fig. 5C, the Raman signal intensity of the encapsulated Raman active silver nanoparticle aggregate was stable over a wide range of pH. Crosslinked polymer networks, covering silver nanoparticle aggregates, protect the inherent properties of these substrates.
The signal intensity of the substrate was then examined over the salt concentration. A 1.0 M NaCl aqueous solution was used as the source of chloride ions.

The nanofibers were prepared by electrospinning directly on a stain-free steel substrate for 5 minutes, and the diameters of the nanofibers were in the range of 220-260 nm.
Other polymer concentrations of 7.5% w / v, 10.0% w / v, 12.5 w / v% and 15.0 w / v% were used to make good fiber size and uniformity.
Lower polymer concentrations showed a mixture of nanofibers and nanoparticles along the beads on the string structure, and as the polymer concentration increased, the relative amount of fibers increased.
In addition, particles and beads on the string structure were removed in experiments using polymer concentrations of 12.5 w / v% or higher. Using high polymer concentrations, the average diameter of the nanofibers was increased by decreasing the distance on the electrode and increasing the flow rate.
6-2. 제타 6-2. zeta 포텐셜Potential
In addition, the adsorption of the dye on the surface of the nanoparticles resulted in a significant decrease in the zeta potential of silver colloids, a force that induces cluster formation (data not shown).
6-3. Crosslink
In addition, cross-linked polymer nanofiber mats were identified in a dry and swelled state using a confocal laser scanning microscope.
6 (E) is a direct image of the polymer nanofibers encapsulating AgNCs in a dry state under a microscope.
6 (F) is an image of the fiber swollen in aqueous solution after successful crosslinking of the polymer. The nanofibers were optically crosslinked and resuspended in DI water after sonication for 1 minute. The fibers swelled after immersion in water to form a soft matrix to hold AgNCs. Such hydrogel properties will be very useful for many biological devices.
Varying amounts AgNCsAgNCs 도입 Introduction
In addition, field emission scanning transmission electron microscopy was used to confirm successful encapsulation of RBITC-based AgNCs in nanofibers at various densities.
Raman signal increase with increasing amount of AgNCs in nanofibers was consistent with increased density of AgNCs in polymeric nanofibers identified by TEM images as shown in FIG. 7. That is, it can be seen that the SERS signal can be regulated by introducing various amounts of AgNCs into the nanofibers.
Green Isothiocyanate Trace Detection
Other solutions of malachite green isothiocyanates with varying concentrations ranging from 0.5 ppm to 6.0 ppm were prepared in a water and ethanol mixture in a volume ratio of 3: 1. Nanofiber mats placed on aluminum foil were cut to size and immersed in MGITC solution for 30 minutes. The mat was washed thoroughly three times with DI water and used for Raman spectr analysis.
Raman spectra were collected for 5 seconds exposure time under 633 nm illumination of the He / Ne laser. All samples were analyzed under the same conditions of the Ramanscope. The captured spectra were corrected and used for quantitative analysis. Prior to use for analyte detection, uniform SERS signals were identified at various spots on the substrate.
SERS spectra collected after immersion of the nanofiber mat in solutions of various concentrations of analytes were compared with the original spectra of the nanofiber mat. Each sample was tested for at least 5 scan cycles and the resulting SERS spectra were averaged. Raman intensity at 1621 cm-1, the inner Raman dye, 1646cm generated from RBITC - consistent with peak for analysis in the first water. The ratio of the internal analyte peak intensity to the intensity of the internal peak is indicated for increasing concentrations of analyte solution.
MGITC, which is water soluble in an ethanol-water mixture, is easily adsorbed onto the surface of the nanoclusters, which diffuse into the nanofibers and produce high SERS signals. Raman signals generated from MGITC after diffusion into polymer nanofibers were measured and shown for various concentrations. Sensitively up to 0.5 ppm concentration of MGITC in aqueous solution. RBITC peak was used as internal standard.
As a result, as shown in FIG. 8, the MGITC peak intensity increased with concentration, and it was confirmed that a linear relation exists between the concentration and the Raman intensity.
RBITC Characteristics Keeping the area under the peak constant in the analysis means good stability without severe diffusion of RBITC in solution. The constant area under the RBITC peak can be compared to the area under increasing MGITC peaks and was used as an internal standard to eliminate errors caused by the intensities of the different peaks in the MGITC spectrum under different conditions. To obtain high reproducible quantitative results, it was found that instead of using only one analyte as a quantitative tool, the ratio of increasing analyte SERS signal to a constant internal signal is important.
