Abstract
Description
본 발명은 신규한 락토바실러스 플란타룸, 이의 제조방법 및 이를 이용한 대장균성설사병 예방 또는 치료용 약학 조성물에 관한 것이다. The present invention relates to a novel Lactobacillus plantarum, a preparation method thereof, and a pharmaceutical composition for preventing or treating E. coli diarrhea.
2008에 국립축산과학원에서 발행한 자료인 '소의 최근 5년간 사육두수 추이 및 질병발생현황(2001~2008년간 질병 발생 소 11,533두 분석자료)'에 의하면, 조사기간 동안 유방염(Mastitis)이 7,440마리로 가장 빈번하게 발생하였고, 그리고 대장균성설사병(colibacillosis)이 1,320마리, 살모넬라증(salmonellosis)이 110마리로서 두 소화기성 질병 또한 많은 비중을 차지하였다. 신생 한우의 폐사율은 5~6%정도이며, 신생우의 설사가 다발적으로 발생하는 경우에는 폐사율이 50%를 넘기도 한다. 성장 시기별 발병 비율을 보면 포유기(1~60일령) 중 질병발병률은 70.9%이고 그중 폐사율은 16.1%, 육성기(61~180일령) 중 질병발병률은 27.9%이고 그 중 폐사율은 2.3%이다(고영두, 2006).According to the data published by the National Institute of Animal Science in 2008, the number of heads of cattle breeding and disease outbreaks in the last five years (analytical data from 11,533 cows in 2001-2008) was 7,440 mastitis during the survey period. 동물 분변에서 선발한 유산균 중에서, 내산성 (acid-resistance), 내담즙성 (bile-resistance), 항균력 (antagonism against pathogens)이 좋다고 판명된 락토바실러스 살리바리우스 29 (Lactobacillus salavarius 29), 페디오코커스 아시딜랙티아 175 (Pediococcus acidilactia 175), 락토바실러스 플란타룸 177 (Lactobacillus plantarum 177)을 육종 대상 유산균으로 선발하였다. 육종 대상 유산균은 API TEST, 16s gene sequencing, 그람염색 (gram staining)을 하여 동정 (identification)을 하였다.

병원균에 대한 유산균의 항균력을 정량하는 방법이다. 실험방법은 우선 유산균이 자랄 수 있는 배지인 MRS 아가 (agar)에 한 플레이트에 약 10개의 유산균을 spreading 하였다. 그 다음 병원균이 섞인 아가 (agar)를 그 위에 부어 일정 시간 배양하였다. 그러면 MRS agar에는 유산균 콜로니가 형성되고, 병원균이 섞인 아가를 처리한 층에는 병원균 론 (pathogen lawn)을 형성하였다. 유산균은 항균물질을 분비하기 때문에 유산균 콜로니 (colony) 주변은 병원균이 자라지 못하여 투명환 (clear zone)을 형성하였다. (도 3). 그 투명환 (clear zone)의 지름을 측정하여 항균력을 정량화하였다. 이 연구에서는 투명환 (clear zone)의 절대값이 아닌, 유산균 콜로니 (colony) 지름에 대한 투명환 (clear zone)의 지름의 비인 CC 비율 (clear zone diameter/colony diameter)를 측정하여 좀 더 정확하게 항균력을 정량화하였다 (도 2).

야생형 (Wild type) 유산균의 목적 병원균에 대한 항균력을 정량화하기 위하여 병원균-탑아가분석 (pathogen-topagar-assay)을 수행하여, 항균력 비교표 (antipahogenic ability standard index)를 작성하였다. 항균력 비교표를 분석한 결과 락토바실러스 플란타룸 177 (Lactobacillus plantarum 177)과 페디오코커스 아시딜랙티아 175 (Pediococcus acidilactia 175)가 락토바실러스 살리바리우스 29 (Lactobacillus salavarius 29)보다 항균력이 더 높았고 높은 것 둘 중에서는 락토바실러스 플란타룸 177 (Lactobacillus plantarum 177)이 약간 더 높았다. 이 결과를 토대로 하여 자외선 돌연변이 육종 대상균으로 락토바실러스 플란타룸 177 (Lactobacillus plantarum 177)을 선정하였다.

MRS 아가 플레이트 (MRS agar plate)에 락토바실러스 플란타룸 (Lactobacillus plantarum)를 spreading을 한 후, 살균램프 (germicidal lamp)를 이용하여 UV를 조사 (irradiation)시켰다. 그리고 나서 37℃ 배양기에서 배양하여 형성된 콜로니 (colony)들을 수합하였다 (도 1).

100㎕의 PB (Protoplast buffer; 10 mM Tris HCl, pH 6.3, 20 mM CaCl2, 0.5 M sucrose)를 넣어, 희석한 다음 900㎕의 60 % PEG6000을 넣고, 상온에서 30분간 배양 (incubation)하여 원형질체 융합 (protoplast fusion)을 형성시켰다. 음전하를 띄는 PEG는 2가 양이온인 칼슘과 이온결합을 하고 결국 음전하 또는 양전하를 띄는 세포막 (plasma lemma)의 성분과 결합하였다. 그 다음에 PEG는 세포막 성분 (plasma lemma components)에 결합하여 세포막 성분 (plasma lemma components)를 뽑아내면서, 세포막 기관 (plasma lemma organisation)을 분산시키면서, 원형질체 (protoplast)들 간의 융합 (fusion)을 유도하였다.
