실험예 6. 항염증 효능 평가Experimental Example 6. Evaluation of anti-inflammatory efficacy
실시예 1의 글라이신을 이용한 수염틸란드시아 추출물의 항염증 효능을 평가하기 위하여, 인터루킨-1α (IL-1α) 시험을 하였다.To evaluate the anti-inflammatory efficacy of the Tillandsia extract using glycine in Example 1, an interleukin-1α (IL-1α) test was performed.
구체적으로, 대식세포 RAW264.7(ATCC)는 혈구계(hemacytometer)를 이용하여 계수한 후, 24 웰 플레이트에 2 Х 105 세포/웰 농도로 분주하였다. 24시간 동안 DMEM 배지(10% 우태아혈청 및 5% 페니실린/스트렙토마이신 포함)에서 배양한 후, LPS 1 ug/ml (Sigma L2630)과 실시예 1의 수염틸란드시아 글라이신 첨가 추출물(시험물질) 또는 수염틸란드시아 열수추출물(비교물질)을 각각 다양한 농도(2.5%(w/w), 5%(w/w), 10%(w/w))로 각 웰에 첨가하고 37℃, 5% CO2 배양기에서 24시간 동안 배양하였다. 24시간 시료를 처리 후 배양액을 회수하고 인터루킨-1α ELISA (R&D systems MLA00)를 진행하였다. ELISA는 제조사에서 제공한 프로토콜을 이용하였다. 실험의 양성대조군으로 L-NMMA (N-Methylarginine) 100 uM (Sigma M7033)을 처리하여 검증하였다. IL-1α 시험 결과는 도 6에 나타내었다.Specifically, macrophages RAW264.7 (ATCC) were counted using a hemacytometer and then dispensed at a concentration of 2 Х 10 5 cells/well in a 24-well plate. Cultured for 24 hours in a % CO 2 incubator. After processing the samples for 24 hours, the culture medium was collected and interleukin-1α ELISA (R&D systems MLA00) was performed. ELISA used the protocol provided by the manufacturer. As a positive control for the experiment, 100 uM (Sigma M7033) of L-NMMA (N-Methylarginine) was treated and verified. The IL-1α test results are shown in Figure 6.
