Description
The present invention relates to a method for deuterating aromatic compounds by hydrogen-deuterium exchange reaction, which replaces hydrogen with deuterium at a high substitution rate even for aromatic compounds with low solubility without using heavy water sources other than heavy water or minimizing the actual amount used. It relates to a method for deuterating aromatic compounds.
Although isotope-labeled substances are physically and chemically very similar to unlabeled standard substances, they can be quantified using mass differences, so they can be used as labeled compounds in the investigation of the mechanisms of chemical reactions, metabolic pathways in the human body or animals, and environmental investigations. It is being used usefully. Recently, it has been reported that replacing hydrogen in aromatic compounds used as light emitting materials in organic light emitting diodes (OLEDs) with deuterium improves the stability of the devices at high current densities and extends their lifespan. Since the atomic mass of deuterium is twice that of hydrogen, compounds substituted with deuterium have lower zero point energy, vibration energy, and ground state energy compared to compounds bonded with hydrogen. Additionally, because the van der Waals radius of deuterium is smaller than that of hydrogen, the C-D bond is shorter and stronger than the C-H bond. Therefore, the central molecular volume decreases, which reduces the electrical polarizability and weakens the interactions between molecules. These characteristics are effective in increasing OLED lifespan and driving characteristics by maintaining the thin film in an amorphous state.
Methods for deuterating aromatic compounds, which are the light-emitting materials of organic light-emitting devices, can be broadly divided into two types depending on the deuteration step. The first method is to prepare a deuterated final product from commercially available or available deuterated starting materials or deuterated intermediates. However, the types of commercially available deuterated starting materials or intermediates are limited, and even when available, they are very expensive. Therefore, yield decreases due to by-products or unreacted products generated during the process up to the synthesis of the final material and losses during separation of the final material. Considering this, it is not economical in most cases. The second method is to synthesize the non-deuterated final material and then produce the deuterated final material through a hydrogen-deuterium (H/D, hydrogen-deuterium) exchange reaction in the final step.
The hydrogen-deuterium exchange reaction is achieved by treating a non-deuterated compound with a solvent that acts as a deuterium source. Since the hydrogen-deuterium exchange reaction of aromatic compounds generally has a slow reaction rate, it is carried out using a catalyst. As a catalyst, an acid catalyst or a metal catalyst is generally used. Examples of the acid catalyst include Bronsted acids such as D 3 PO 4 /BF 3 , DCl, D 2 SO 4 , AcOD, CF 3 CO 2 D or Lewis acids such as AlBr 3 , EtAlCl 2 and MoCl 5 . Publication Patent No. 10-2022-0027786 reports the use of organic compounds such as anhydride or sulfonic anhydride, which are hydrolyzed by heavy water to produce acid, as catalysts, and Publication Patent No. 10-2021-0081970 Ho proposed using Ti, Hf, and Zr-based catalysts to solve the problem that AlCl 3 , a Lewis acid, reacts violently with moisture, making handling and use difficult and causing side reactions. However, if the aromatic compound is not rich in electrons, the effectiveness of the acid-catalyzed reaction is significantly reduced, and since deuteration is limited depending on the position of the aromatic hydrogen, vigorous reaction conditions or microwaves are sometimes used to achieve a high exchange rate. Metal catalysts include PtO 2 , Pt (black), Pt-C, Pd(OH) 2 , PdCl 2 , Pd-C, Raney-Ni, etc. Heterogeneous metal catalysts can be easily removed by filtration after completion of the reaction and have the advantage of not requiring an additional work-up process. However, when using a heterogeneous catalyst, it is reported that some hydrogenated by-products or hydrolysis products are generated because hydrogen is treated to activate the catalyst. To solve this problem, Publication Patent No. 10-2021-0071543 proposed adding a dehydrogenation reaction after deuteration.
It is most desirable to use heavy water (D 2 O) as a solvent that acts as a deuterium source in terms of cost, but the light-emitting material of organic light-emitting devices has a structure containing two or more aromatic rings and has low solubility in heavy water, so heavy water is used. When using only, the reaction time takes a long time and the deuterium substitution rate is low. Therefore, the solubility of the aromatic compound is increased by mixing it with another solvent that can dissolve the aromatic compound, or by using supercritical heavy water. Organic solvents used as deuterium sources other than heavy water include benzene-d 6 , acetone-d 6 , and DMSO-d 6 . Most of these are manufactured from heavy water, and considering the actual number of deuterium atoms transferred to the substrate, Because it is very expensive, its actual usage should be reduced as much as possible for economic efficiency. Supercritical heavy water has a critical temperature of 374°C and a critical pressure of 221 bar. In order to maintain the supercritical state, the reaction must be carried out at a high temperature/high pressure above the critical temperature and critical pressure, so it cannot be applied to compounds that are unstable at high temperatures and contains excess deuterium. Because it uses , there is a problem of high process cost.
Supercritical carbon dioxide has a critical temperature of 31℃ and a critical pressure of 7.5 MPa. It reaches the supercritical state at a lower temperature and pressure than heavy water, but it is chemically stable and insoluble, so it can be used safely. Therefore, problems when using supercritical heavy water can be avoided. Due to the nature of supercritical fluid, even if a separate organic solvent is not used or only a small amount is used, the hydrogen-deuterium exchange reaction by heavy water is possible because the solubility of aromatic compounds is high. It is expected that this will proceed efficiently.
In order to solve the problems of the prior art, the present invention While minimizing the practical use of deuterium sources, it is possible to prevent a decrease in deuteration reaction yield due to the solubility of aromatic compounds, and it does not require high temperature conditions, suppressing the production of by-products and producing deuterated water through a hydrogen-deuterium exchange reaction with high efficiency. The purpose is to provide a method for producing digested aromatic compounds.
Another object of the present invention is to provide a method for producing deuterated aromatic compounds that can separate/purify deuterated aromatic compounds without undergoing complicated processes after completion of the reaction, and that allow recovery and reuse of the main raw materials.
Even if the technical problem to be solved by the present invention is not mentioned above, it will be clearly understood by those skilled in the art. Additionally, when describing the invention, if it is determined that a detailed description of known technology related to the invention may unnecessarily obscure the gist of the invention, the detailed description will be omitted.
The present invention for achieving the above-mentioned object relates to a method for deuterating aromatic compounds, characterized in that an aromatic compound, a deuterium source, and a deuteration reaction catalyst are reacted with carbon dioxide in a supercritical state.
Because supercritical carbon dioxide has solvent characteristics similar to hydrocarbons, it has high solubility in aromatic compounds. The solubility can be easily adjusted by adjusting temperature and pressure, and it is non-flammable and inert, so it can be used safely. The mole fraction of water in a fluid containing supercritical carbon dioxide and water in a uniform state measured at 100°C and 10 MPa, which is supercritical conditions, is about 2.9%, and below 150°C, the mole fraction increases as temperature and pressure increase. When the reaction proceeds in a separated phase, the solubility of heavy water in the separated phase (i.e., the fraction of heavy water) has a significant effect on the reaction rate. Naturally, the larger the fraction of heavy water in the phase in which the aromatic compound is dissolved, the faster the reaction rate because the contact between the aromatic compound and heavy water increases outside the interface. Considering that the solubility of water in benzene, an organic solvent mainly used for deuteration, is only 0.00362M, supercritical carbon dioxide is highly miscible with water, so the deuteration reaction can proceed more effectively. The critical conditions of the entire reaction system may vary depending on the composition of the reaction solution, but the critical conditions of the mixed system have not yet been clearly explained. However, regardless of the type of aromatic compound, catalyst, or heavy water source used in the reaction, the temperature and pressure of the reaction mixture in a closed container can be maintained uniformly, so that it exceeds the critical temperature of carbon dioxide of 31°C and the critical pressure of 7.5 MPa. The conditions stipulate that it constitutes supercritical carbon dioxide. Therefore, in the present invention, carbon dioxide in a supercritical state means a temperature of 31°C or higher and a pressure of 7.5 MPa or higher. As the reaction temperature and reaction pressure increase, the reaction speed becomes faster, so production efficiency can increase. However, as the temperature and pressure increase, more costs are incurred for equipment and operation, so the deuteration reaction temperature of the present invention is 40 to 200 ° C. , it is more preferable that it is 7.5 to 15 MPa.
The aromatic compound may be a compound containing one or more aromatic rings, a heteroaromatic compound, or an aromatic amine compound. When the aromatic compound contains two or more (hetero) aromatic rings, the two or more (hetero) aromatic rings may be directly connected without a linker, and/or the two or more (hetero) aromatic rings may be fused to each other. The expression "(hetero) aromatic" means "hetero aromatic or aromatic". For example, two or more (hetero) aromatic rings are fused to each other, which means that an aromatic ring and an aromatic ring are fused, or a hetero aromatic ring It indicates that a heteroaromatic ring is fused, or a heteroaromatic ring and an aromatic ring are fused.
The present invention is characterized by using supercritical carbon dioxide as a solvent during the deuteration reaction of conventional aromatic compounds, and the deuterium source or deuteration reaction catalyst used in the conventional deuteration reaction is not used in the conventional hydrogen-deuterium exchange reaction. A deuterium source or a deuteration reaction catalyst can be used.
For example, the deuterium source may be one or more selected from the group consisting of D 2 O, D 2 , benzene-d 6 , toluene- d 8 , xylene-d 10 , acetone-d 6 , DMSO-d 6 and CDCl 3 You can. Aromatic compounds, which are usually used as light-emitting materials in organic light-emitting devices, have very low solubility in water, so they are used by mixing heavy water with a deuterated organic solvent that can dissolve aromatic compounds, such as benzene- d6 . shall. However, in the present invention, since aromatic compounds can be dissolved using supercritical carbon dioxide that is inert and does not contain hydrogen, only heavy water, which is the most economical of deuterium sources, is used or a hydrogen-deuterium exchange reaction is performed using a minimal amount of deuterated organic solvent. You can proceed. Alternatively, if necessary, only a deuterated organic solvent other than heavy water, such as benzene-d 6 , toluene-d 8 , xylene-d 10 or CDCl 3 , may be used as the deuterium source. In this case, in the deuterated organic solvent recovered after completion of the reaction, some of the deuterium has been replaced with hydrogen. After recovering this, the deuterated organic solvent can be easily regenerated by mixing heavy water and a deuteration reaction catalyst and reacting with carbon dioxide in a supercritical state. Additionally, D 2 can also be used as a deuterium source.
The equivalent ratio of the deuterium source to the aromatic compound is 1 or more. As the equivalent weight of the deuterium source increases, the reaction rate and deuteration rate can increase. However, an increase in reaction capacity results in a decrease in productivity, so it is difficult to set an appropriate equivalent weight depending on the aromatic compound. It will be easy for those skilled in the art. Generally, the preferred equivalence ratio is 3 to 30 equivalents, but it is natural that the optimal equivalent ratio may vary regardless of the above range depending on the reaction scale, type of aromatic compound, desired hydrogen-deuterium substitution rate, etc.
In this specification, 'deuteration reaction catalyst' refers to a material that can increase the hydrogen-deuterium exchange reaction rate, and an acid catalyst or a metal catalyst used in the hydrogen-deuterium exchange reaction in the prior art can be used. For example, acid catalysts include sulfuric acid (H 2 SO 4 ), trifluoroacetic acid (CF 3 CO 2 H), methanesulfonic acid (CH 3 SO 3 H), and benzenesulfonic acid (C 6 H 5 SO 3 H). ), fluorosulfonic acid (FSO 3 H), hydrochloric acid (HCl), acetic acid (AcOH), phosphoric acid (H 3 PO 4 )/BF 3 , trifluoromethanesulfonic acid (CF 3 SO 3 H) and their deuterium. One or more selected from the group consisting of cargo may be used. Normal Since the acid catalyst is used in a small amount and the deuterium source is used in an excessive amount, even if the acid is in a non-deuterated form, it does not significantly affect the deuteration rate of the aromatic compound. However, in cases where the use of a large amount of acid affects the deuteration rate of aromatic compounds, or when the supply of deuterated acid is easy and low in price, it may be desirable to use the deuterated form of the acid. is natural. When heavy water is used as a deuterium source, organic compounds that are hydrolyzed by heavy water to form the acid can also have the same effect on the acid catalyst. For example, trifluoroacetic anhydride is hydrolyzed by heavy water to produce deuterium-trifluoroacetic acid, respectively, so trifluoroacetic acid is considered an acid catalyst.
The metal catalyst may be one or more selected from the group consisting of platinum group metals such as platinum, palladium, rhodium, and ruthenium, nickel, cobalt, oxides thereof, and complexes thereof. The metal catalyst may be supported on a carbon support or a metal oxide support. It goes without saying that the metal catalyst may undergo an activation step before use, if necessary.
One of the characteristics of the method for producing deuterated aromatic compounds of the present invention is that the post-reaction process is simple because the solubility of supercritical carbon dioxide varies greatly depending on temperature and pressure. The post-reaction process is that ⓐ aromatic compounds are dissolved in supercritical carbon dioxide, ⓑ supercritical carbon dioxide is immiscible with water (heavy water), and ⓒ supercritical carbon dioxide can be easily removed by vaporizing it by controlling the temperature and pressure. Use it.
For example, when a water-soluble catalyst is used and only heavy water is used as a heavy water source, when the reaction is completed, the temperature and pressure are adjusted as in the Example to simply vaporize and remove carbon dioxide, thereby obtaining a deuterated aromatic compound as a precipitate. can do. Alternatively, in the case of a mass production system, heavy water containing a catalyst is removed from supercritical carbon dioxide containing deuterated aromatic compounds by layer separation, and the carbon dioxide is vaporized and removed by controlling the temperature and pressure. Digested aromatic compounds can be obtained. When using a heterogeneous catalyst such as Pt/C, in a mass production system, the heterogeneous catalyst is removed by passing it through a filter, and then the supercritical carbon dioxide and deuterium source are sequentially removed to obtain a deuterated aromatic compound. If the deuterium source is heavy water, a deuterated aromatic compound can be obtained by filtration, and if the deuterium source is a deuterated organic solvent other than heavy water, the deuterated aromatic compound can be obtained by concentrating the deuterium source.
In the present invention, the deuteration rate of the aromatic compound may be 70% or more, preferably 80% or more, more preferably 90%, 95% or more, 97% or more, 98% or more, or 99% or more. If the deuteration rate obtained by the reaction is low compared to the required deuteration rate, increase the ratio of the deuterium source used, increase the reaction time, or repeat the deuteration reaction using the deuteration product as the starting material. It is natural that the deuterium digestion rate can be increased. The deuteration rate refers to the degree to which H molecules contained in an aromatic compound are replaced by D molecules, and can be measured by methods known in the art. Examples of methods for measuring the deuteration rate include NMR analysis or analysis using mass spectrophotometry, such as atmospheric solid analysis probe mass spectrophotometry (ASAP-MS).
In the following examples, supercritical carbon dioxide was not recovered and used because the reaction was performed using an autoclave for laboratory-level reaction, but it is natural that it can be recycled and reused.
In addition to supercritical carbon dioxide, when organic solvents other than heavy water are used as a deuterium source, it can be recycled and reused. That is, in the deuterium source recovered after the reaction is completed, some of the deuterium has been replaced with hydrogen as a result of the hydrogen-deuterium exchange reaction. Therefore, before reusing it, it can be regenerated by reacting it with carbon dioxide in a supercritical state in the presence of heavy water and a catalyst and then reused.
Figure 1 is a schematic diagram showing the above-described regenerative cycle manufacturing process, and relates to an example of a process for deuterating carbazole (CBZ) using heavy water and benzene-d 6 as deuterium sources and acid as a catalyst. In the first reactor, the catalyst, heavy water, benzene- d6 (Bz-d6), and carbazole are reacted with supercritical carbon dioxide (ScCO2) to produce deuterated carbazole (CBZ-d8). When the reaction is completed, the heavy water layer containing the acid catalyst is removed by layer separation, and ScCO2, Bz-d6 and CBZ-d8 are transferred to the second reactor. By controlling the pressure and temperature in the second reactor, ScCO2 and Bz-d6 are removed to the third reactor to obtain the reaction product CBZ-d8. By adjusting the pressure of the third reactor, carbon dioxide is vaporized and recovered into the carbon dioxide storage, and Bz-d6 is transferred to the fourth reactor. A regeneration reaction is performed by adding heavy water, an acid catalyst, and ScCO2 to the fourth reactor containing Bz-d6. Once the reaction is completed, the heavy water containing the catalyst is discarded by layer separation and transferred to the fifth reactor containing ScCO2 and Bz-d6. Separate. By adjusting the pressure of the fifth reactor, carbon dioxide is vaporized and recovered, and the regenerated Bz-d6 is used again for deuteration of aromatic compounds. In the above description, the first to fifth reactors are described separately, but the same reactor may be used for different purposes depending on the situation. For example, if carbon dioxide is first separated into a gaseous state in the second reactor and Bz-d6 is sequentially concentrated and removed, the second reactor can also serve as a third reactor. Additionally, when only heavy water is used as the deuterium source, the process for enriching and regenerating Bz-d6 can be omitted. A more detailed reactor configuration can be designed appropriately depending on the type of deuterium source or catalyst used.
As described above, according to the present invention using supercritical carbon dioxide, supercritical carbon dioxide acts as a solvent for aromatic compounds, so even if only heavy water is used or the amount of expensive deuterium sources other than heavy water is minimized, hydrogen- It can solve the problems of slow speed and low yield of deuterium exchange reaction.
In addition, the method for producing a deuterated aromatic compound of the present invention does not require a complicated multi-step process in the workup process for obtaining the deuterated aromatic compound after completion of the reaction, and does not require recovery of carbon dioxide and metal catalyst, raw materials used as deuterium sources, and Because it is easy to regenerate, deuterated aromatic compounds can be produced economically.
